A detailed study of the periodate oxidation of sialic acids in glycoproteins.

Periodate oxidation of terminal N-acetyl- and N-glycoloylneuraminic acid residues in the mucins from edible bird nest substance and pig submandibular gland, respectively, can be carried out under conditions which exclusively give rise to the formation of the C-7 analogues of these sialic acids. In contrast, the C-8 compounds can be obtained in a maximum yield of about 40%. Under identical conditions, N-glycoloylneuraminic acid is oxidized about 1.5 times faster than the N-acetylated derivative. After release of the sialic acids by acid hydrolysis, the characterization of the oxidation products

Sequential 1H and 13C resonance assignments for an octa- and decasaccharide of the N-acetyllactosamine type by multiple-step relayed correlation and heteronuclear correlation nuclear magnetic resonance.

400 MHz 1H-NMR and 100 MHz 13C-NMR spectra of a neutral octasaccharide and of a disialyldecasaccharide of the N-acetyllactosamine type were studied. The resonance assignments were made by combining multiple-relayed coherence-transfer chemical-shift-correlated spectroscopy (multiple-RELAY-COSY) and 1H/13C-shift correlated 2D experiments. The complete analysis of the 1H and 13C spectra was performed.

Chondroitin sulfate inhibits secretion of TNF and CXCL8 from human mast cells stimulated by IL-33.

Glycosaminoglycans (GAGs) are linear, highly negatively charged carbohydrate chains present in connective tissues. Chondroitin sulfate (CS) and heparin (Hep) are also found in the numerous secretory granules of mast cells (MC), tissue immune cells involved in allergic and inflammatory reactions. CS and Hep may inhibit secretion of histamine from rat connective tissue MC, but their effect on human MC remains unknown. Human LAD2 MC were pre-incubated with CS, Hep, or dermatan sulfate (DS) before being stimulated by either the peptide substance P (SP, 2 μM) or the cytokine IL-33 (10 ng/mL). Prein

Histochemical detection of expression of binding sites for labelled hyaluronic acid and carrier-immobilized synthetic (histo-blood group trisaccharides) or biochemically purified (ganglioside GM1) glycoligands in nasal polyps and other human lesions including neoplasms.

This study is intended to demonstrate the versatility and feasibility of custom-made oligosaccharide-exposing neoglycoconjugates including histo-blood group epitopes in various human lesions, including nasal polyps. The binding of the biotinylated probes was determined on formalin-fixed paraffin-embedded sections from archive materials. The general aspects of our results may be interpreted as follows: the neoglycoconjugates used here can readily detect differences in the ability of cells to bind glycan residues in tissue sections, thereby enabling the extent of the binding capacity of various

Total plasma N-glycomic patterns of COVID-19 disease.

Total plasma N-glycans alter in disease states, with few studies focused on COVID-19. A discovery cohort of 310 COVID-19 patients, replicated with 97 COVID-19 patients and tested with 100 COVID-19 patients, was used to unearth the N-glycans capable of distinguishing infection, as well as prognosis of intensive care unit (ICU) admission and mortality. All significant bisected glycans were decreased in patients compared to controls, whilst fucosylated tri-antennary glycans and sialylated tetra-antennary glycans were increased in patients. For both those admitted to the ICU and those who died, Pe

N-glycan signatures of immunoglobulins-depleted serum proteins in colorectal cancer.

Protein glycosylation has been implicated in the pathogenesis of cancer, with colorectal cancer (CRC) strongly associated with serum N-glycosylation and IgG N-glycosylation. However, the N-glycosylation profile of non-immunoglobulin serum proteins (IgS-D) in CRC progression remains largely unexplored. Serum IgS-D was isolated using affinity chromatography, followed by the release of N-glycans, methylamidation, and MALDI-MS analysis. The potential of the IgS-D N-glycome profile to reflect CRC progression was assessed through Mann-Whitney U tests and multivariate analysis. A total of 43 distinct

Enhanced anti-proliferative activity of Jacalin, a plant lectin in combination with taxol in MDA-MB-468 triple-negative breast cancer cells.

Plant lectins derived from a variety of plant sources can inhibit the proliferation of certain types of human cancer cells and have been studied extensively. In the present study, the effects of Jacalin, a Thomsen–Friedenreich disaccharide-binding lectin isolated from jackfruit seeds, on the triple-negative breast cancer (TNBC) cell line MDA-MB-468 was investigated in combination with Taxol. It was observed that Jacalin inhibited the proliferation of these cancer cells. The antiproliferative effect of Jacalin on MDA-MB-468 was found to be reversible. However, when Jacalin was removed from the

Macrophage modulation accounts for the anti-inflammatory effect of Hypnea cervicornis agglutinin in rat arthritis induced by zymosan.

Previous study demonstrated that Hypnea cervicornis lectin (HCA) reduces inflammation and nociception via interaction with the lectin carbohydrate-binding site, modulating gene expression of the interleukins IL-1β, TNF-α and of iNOS. This study evaluated the effect of HCA in the rat model of arthritis induced by zymosan in the tibiotarsal joint and the involvement of macrophage-derived mediators. In vitro, macrophages were stimulated with zymosan before being incubated with HCA and the supernatant was injected into the joint. In vivo, HCA was administered by intravenous route after intra-artic

Structural dynamics and stability analysis of a novel ARSK variant confirms its role in MPS type X pathogenesis.

Mucopolysaccharidoses (MPS) are heterogeneous group of disorders caused by the deficiency of lysosomal enzymatic activities. Deficiencies in these enzymes are required for proper degradation of glycosaminoglycans (GAGs), thus referred as inborn errors of lysosomal GAG metabolism. Arylsulfatase K (ARSK) has been recently characterized as lysosomal hydrolase involved in GAG degradation removing the 2-O-sulfate group from 2-sulfoglucuronate and has been associated with MPS X. A 12-year-old female presented with developmental delay, progressive musculoskeletal abnormalities, and skeletal dysplasia

Review on macrophage polarization during visceral leishmaniasis and impact of glycoprotein.

Visceral Leishmaniasis is one of the neglected vector-borne tropical disease escalating resistance against available drugs which have toxicity beyond therapeutic level leading to the requirement of alternative therapy. Macrophages, the primary cells of the immune system, being the host cells for the Leishmanial parasite undergo functional phenotypic changes based on its infected or uninfected states leading to release of specific cytokines. The M2 functional phenotype is suitable for parasite survival, while, M1 phenotype boosts the immune system leading to release of superoxide responsible fo

Evaluation of acylated/deacylated ghrelin, CCL20/CCR6 concentration, and IgG N-Glycan alterations in different dysplasia grades of colon adenoma.

Colorectal adenomas represent precursor lesions in the adenoma-carcinoma sequence and are characterized by chronic low-grade inflammation. Chemokine CCL20/CCR6 signaling and metabolic hormone ghrelin have been implicated in colorectal tumorigenesis, while IgG glycosylation reflects systemic immune status. However, their interrelationship across different stages of adenoma dysplasia remains unclear. A total of 70 patients with histologically confirmed colorectal adenomas were stratified into low-grade (n = 36) and high-grade dysplasia (n = 34). Serum CCL20, CCR6, acylated and deacylated ghrelin

GM1 oligosaccharide-mediated rescue in GBA-linked Parkinson's disease via modulation of lysosomal and mitochondrial dysfunctions.

Mutations in the glucocerebrosidase GBA gene, encoding the lysosomal enzyme β-glucocerebrosidase, represent the most frequent genetic risk factor for Parkinson's disease, leading to lysosomal dysfunction, α-synuclein aggregation, and mitochondrial impairment. In this study, we investigated the therapeutic potential of GM1 ganglioside and its oligosaccharide portion (OligoGM1) in a cellular model of GBA-associated Parkinson's disease, using SH-SY5Y neuroblastoma cells carrying the L444P GBA mutation. We observed that both GM1 and OligoGM1 reduced α-synuclein accumulation and improved cell viabi

High molecular weight fucosylated glycosaminoglycan promotes the binding of antithrombin and thrombin: a template mechanism and structure-activity relationship study.

Native fucosylated glycosaminoglycan (FG), a structurally unique polysaccharide with chondroitin sulfate like backbone and sulfated fucose side chains, has multiple anticoagulant mechanisms. Analyzing the interaction between FG and coagulation proteins will help to further understand its pharmacological mechanisms. Previously, we have reported that high molecular weight depolymerized FG (dFG) and unfractionated heparin (UFH) exhibit different binding models for antithrombin (AT), which is a primary natural anticoagulant in plasma by inhibiting coagulation proteases including thrombin (FIIa). Spatial single-cell maps reveal ST6GAL1 promoting ovarian cancer metastasis.

In this study, spatial and single-cell transcriptome techniques were used to investigate the role of beta-galactoside alpha-2,6-sialyltransferase 1 (ST6GAL1) in promoting peritoneal metastasis in ovarian cancer epithelial cells. We collected single-cell transcriptomic (GSE130000) and spatial transcriptomic datasets (GSE211956) from the Gene Expression Omnibus and RNA-sequencing data from The Cancer Genome Atlas. The Robust Cell Type Decomposition (RCTD) approach was implemented to integrate spatial and single-cell transcriptomic data. In addition, pseudo-time trajectory analysis, cell-cell com

In Silico and in vitro evaluation of Cucurbitaceae seed lectins as promising prophylactic and therapeutic agents against SARS-CoV-2.

Concern over COVID-19 has arisen from continuous mutations in SARS-CoV-2 genome leading to emergence of novel variants. A detailed study and understanding of N-linked glycans (NAGs) of SARS-CoV-2 spike protein are key for the development of vaccines and new therapeutic strategies. Potential of four Cucurbitaceae seed lectins (CSLs) to inhibit SARS-CoV-2 attachment to host cells via interaction with receptor binding domain (RBD), was determined by Molecular Docking using PRODIGY and LZerD web servers. Among them, Luffa cylindrica lectin (PDB ID: 1NIO) displayed the most significant binding (B.E

Mass spectrometric profiling reveals alterations in N-Glycans and O-Glycans in Tay-Sachs disease under Autophagy-Induced conditions.

Tay-Sachs disease is a rare neurodegenerative disorder caused by mutations in the HEXA gene. The HEXA gene encodes the α-subunit of the enzyme β-hexosaminidase A, which degrades GM2 ganglioside. Previously, we identified impaired autophagy in the brains of a mouse model of Tay-Sachs disease, which exhibited neuropathological and clinical abnormalities. Moreover, we demonstrated autophagosome clearance in Tay-Sachs cells under lithium-induced conditions. Here, we further aimed to evaluate N- and O-glycan changes in these cells and examine whether glycan alterations are linked to ER stress. The

Core-fucose-specific Pholiota squarrosa lectin decreased hepatic inflammatory macrophage infiltration in steatohepatitis mice.

Recent findings in glycobiology revealed direct evidence of the involvement of oligosaccharide changes in human diseases, including liver diseases. Fucosylation describes the attachment of a fucose residue to a glycan or glycolipid. We demonstrated that fucosylated proteins are useful serum biomarkers for nonalcoholic fatty liver disease. Among fucosyltransferases, expression of alpha-1, 6-fucosyltransferase (Fut8), which produces core fucose, is frequently elevated during the progression of human chronic liver diseases. Previously, we discovered core-fucose-specific Pholiota squarrosa lectin

Glycosylated SARs Cov 2 interaction with plant lectins.

Lectins are non-immune carbohydrate-binding proteins/glycoproteins that are found everywhere in nature, from bacteria to human cells. They have also been a valuable biological tool for the purification and subsequent characterisation of glycoproteins due to their carbohydrate binding recognition capacity. Antinociceptive, antiulcer, anti-inflammatory activities and immune modulatory properties have been discovered in several plant lectins, with these qualities varying depending on the lectin carbohydrate-binding site. The Coronavirus of 2019 (COVID-19) is a respiratory disease that has swept t

Increasing outer membrane complexity: the case of the lipopolysaccharide lipid A from marine Cellulophaga pacifica.

Gram-negative bacteria living in marine waters have evolved peculiar adaptation strategies to deal with the numerous stress conditions that characterize aquatic environments. Among the multiple mechanisms for efficient adaptation, these bacteria typically exhibit chemical modifications in the structure of the lipopolysaccharide (LPS), which is a fundamental component of their outer membrane. In particular, the glycolipid anchor to the membrane of marine bacteria LPSs, i.e. the lipid A, frequently shows unusual chemical structures, which are reflected in equally singular immunological propertie
