Revitalization of plant growth-promoting plant allied microbes: an eco-friendly approach for agricultural and environmental sustainability.

Plants are increasingly recognized as metaorganisms because they harbor diverse and dynamic microbial communities that establish intricate symbiotic associations and collectively regulate plant growth, development, and adaptation to changing environmental conditions. Plant-associated microorganisms colonize distinct ecological niches, including the rhizosphere, phyllosphere, endosphere, and surrounding soil, where they contribute to nutrient acquisition, phytohormone production, disease suppression, and tolerance to biotic and abiotic stresses. These multifunctional interactions have positione

Chitosan-crosslinked microwave-engineered biochar alleviates lead stress in Brassica napus by improving soil functioning and plant performance in Pb-contaminated soil.

Lead (Pb) contamination in agricultural soils reduces crop productivity and threatens food safety, highlighting the need for in situ amendments that lower Pb bioavailability while supporting soil functioning and plant performance. This study upgraded poultry manure-derived biochar through microwave activation and genipin-crosslinked chitosan modification (MPBCH), then evaluated its performance in a greenhouse pot experiment with Brassica napus grown in field-collected Pb-contaminated soil. Six treatments were compared: untreated Pb-stressed soil (CK), chitosan (CH), poultry manure biochar (PBC

Spatio-Temporal Plasticity of Root Exudation in Three Temperate Tree Species: Effects of Season, Site, and Soil Characteristics.

Root exudation provides a constant carbon input to the rhizosphere and is therefore a very important factor in shaping this hotspot of biological activity. Nonetheless, data on root exudation and its spatio-temporal plasticity are scarce. This study provides insights into compound-specific root exudation in three temperate tree species in two seasons (late spring and late summer) and two soil compartments (forest floor and the top mineral soil), including the effect of soil chemistry. At four sites with differing mean annual temperatures and soil phosphorus levels, root exudates were sampled u

Integrative spatial transcriptomic analysis pinpoints the role of the ferroxidase, TaMCO3, in wheat root tip iron mobilization.

Roots play a critical role in the sensing and absorption of essential minerals from the rhizosphere. Iron (Fe) deficiency, for example, triggers a well-known series of physiological and molecular responses within roots that facilitate uptake, which differs between monocots and dicots. In monocots, little is known about the molecular responses that occur within specific root development zones in response to iron deprivation, and how these differences result in overall nutrient uptake. Here, we conducted a transcriptome analysis of wheat root tips under Fe deficiency (-Fe) and performed a compar

Production of kojic acid by Aspergillus flavus OL314748 using box-Behnken statistical design and its antibacterial and anticancer applications using molecular docking technique.

Kojic acid is a wonderful fungal secondary metabolite that has several applications in the food, medical, and agriculture sectors. Many human diseases become resistant to normal antibiotics and normal treatments. We need to search for alternative treatment sources and understand their mode of action. Aspergillus flavus ASU45 (OL314748) was isolated from the caraway rhizosphere as a non-aflatoxin producer and identified genetically using 18S rRNA gene sequencing. After applying the Box-Behnken statistical design to maximize KA production, the production raised from 39.96 to 81.59 g/l utilizing

Bacillus velezensis WB induces systemic resistance in watermelon against Fusarium wilt.

BACKGROUND: Our previous findings indicated that Bacillus velezensis WB could control Fusarium wilt by changing the structure of the microbial community in the watermelon rhizosphere. However, there are few studies on its mechanism in the pathogen resistance of watermelon. Therefore, in this study, we determined the mechanism of B. velezensis WB-induced systemic resistance in watermelon against Fusarium wilt through glasshouse pot experiments. RESULTS: The results showed that B. velezensis WB significantly reduced the incidence and disease index of Fusarium wilt in watermelon. B. velezensis W

In silico analysis of comparative affinity of phytosiderophore and bacillibactin for iron uptake by YSL15 and YSL18 receptors of Oryza sativa.

Iron is an important micronutrient for plant growth and development. In the case of Oryza sativa, iron is made available primarily with the help of iron chelators called phytosiderophores i.e. variants of deoxymugineic acid (DMA). They bind with ferric ions and get internalized through Yellow Stripe Like transporters viz. YSL15 and YSL18. However, due to low amount of secretion of phytosiderophores, rice suffers from iron deficiency. Alternatively, siderophores of plant growth promoting rhizobacteria may support iron uptake and make it available to plants via transporting ferric ions possibly

Isolation, screening, characterization, and optimization of bacteria isolated from calcareous soils for siderophore production.

The most effective agricultural practice to prevent iron deficiency in calcareous soils is fertilizing with synthetic chelates. These compounds are non-biodegradable, and persistent in the environment; hence, there is a risk of leaching metals into the soil horizon. To tackle iron deficiency-induced chlorosis (IDC) in crops grown on calcareous soils, environmentally friendly solutions are needed rather than chemical application as it affects the soil health further. Hence, the present work focused on isolating and screening calcareous soil-specific bacteria capable of producing iron-chelating

The Integrated Effect of Microbial Inoculants and Biochar Types on Soil Biological Properties, and Plant Growth of Lettuce (Lactuca sativa L.).

Numerous reports confirm the positive effect of biochar application on soil properties and plant development. However, the interaction between root-associated beneficial microbes and different types of biochar is not well understood. The objective of this study was to evaluate the plant growth of lettuce after the application of three types of biochar in loamy, sandy soil individually and in combination with plant-beneficial microbes. Furthermore, total microbial activity in rhizosphere soil of lettuce was measured by means of fluorescein diacetate (FDA) hydrolase and enzyme activities linked

Effective biofertilizer Trichoderma spp. isolates with enzymatic activity and metabolites enhancing plant growth.

Trichoderma species have been widely recognized as biofertilizer fungi for their ability to produce phytohormones and enhance plant growth. In our current study, fifteen strains of Trichoderma spp. (T1-T15) were screened for their capacity to produce phytohormones and metabolites eliciting plant growth. The stains were previously isolated from olive rhizosphere soil in northern Algeria. Plant growth promoting (PGP) potential of Trichoderma spp. was evaluated in vitro through the production of phosphatases, siderophores, hydrogen cyanide (HCN), and ammonia (NH3). Besides, plant growth phytohorm

Root exudates from citrus plants subjected to abiotic stress conditions have a positive effect on rhizobacteria.

Plants are constantly releasing root exudates to the rhizosphere. These compounds are responsible for different (positive or negative) interactions with other organisms, including plants, fungi or bacteria. In this work, the effect of root exudates obtained from in vitro cultured citrus plants on two rhizobacteria (Pseudomonas putida KT2440 and Novosphingobium sp. HR1a) was evaluated. Root exudates were obtained from two citrus genotypes differing in their sensitivity to salt and heat stress and differentially affected the growth of both rhizobacteria. Root exudates from salt-stressed plants o

Label-Free Proteomics of a Defined, Binary Co-culture Reveals Diversity of Competitive Responses Between Members of a Model Soil Microbial System.

Interactions among members of microbial consortia drive the complex dynamics in soil, gut, and biotechnology microbiomes. Proteomic analysis of defined co-cultures of well-characterized species provides valuable information about microbial interactions. We used a label-free approach to quantify the responses to co-culture of two model bacterial species relevant to soil and rhizosphere ecology, Bacillus atrophaeus and Pseudomonas putida. Experiments determined the ratio of species in co-culture that would result in the greatest number of high-confidence protein identifications for both species.

Climatic Drivers for Multi-Decadal Shifts in Solute Transport and Methane Production Zones within a Large Peat Basin.

Northern peatlands are an important source for greenhouse gases but their capacity to produce methane remains uncertain under changing climatic conditions. We therefore analyzed a 43-year time series of pore-water chemistry to determine if long-term shifts in precipitation altered the vertical transport of solutes within a large peat basin in northern Minnesota. These data suggest that rates of methane production can be finely tuned to multi-decadal shifts in precipitation that drive the vertical penetration of labile carbon substrates within the Glacial Lake Agassiz Peatlands.
