Associations of postpartum lying time with culling, milk yield, cyclicity, and reproductive performance of lactating dairy cows.

The objectives were to evaluate the associations of lying time (LT) during the first 14 d in milk (DIM) with milk yield, cyclicity (CYC), culling within 60 DIM (CULL), and reproductive performance of lactating dairy cows. A total of 1,052 Holstein cattle (401 nulliparous heifers and 651 parous cows) from 3 commercial dairy farms had electronic data loggers (IceQube, IceRobotics, Edinburgh, UK) placed on a hind leg 14 ± 3 d before the expected parturition date and removed at 14 ± 3 DIM to assess their LT. Serum concentrations of β-hydroxybutyrate were determined at 7 ± 3 and 14 ± 3 DIM. Cases o

Herd health monitoring in dairy farms - discover metabolic diseases. An overview.

The transition period, 3 weeks before and 3 weeks after calving, is inevitably accompanied by a negative energy balance, which sometimes causes metabolic disturbances, such as ketosis. Subclinical ketosis (SCK) is defined as an increase in the β-hydroxybutyrate (BHB) concentration to ≥ 1.2 mmol/l in the blood. According to a recent study, a value of ≥ 0.7 mmol/l of non-esterified fatty acids (NEFA) in the blood indicates the potential development of the poor metabolic adaption syndrome (PMAS). With a herd prevalence of 21 %, and an incidence of approximately 40 % within the first 2 weeks after

Short communication: Effects of body fat mobilization on macrophage infiltration in adipose tissue of early lactation dairy cows.

Intense lipolysis triggers an inflammatory response within adipose tissue characterized by adipose tissue macrophage (ATM) infiltration; however, the mechanisms triggering this process are poorly characterized in transition dairy cows. The aim of this study was to determine the association between ATM infiltration and body fat mobilization in the transition period, markers of excessive lipolysis, and adipose tissue expression of genes related to chemotactic and inflammatory responses. Subcutaneous adipose tissue samples were taken from the tailhead of 9 multiparous Holstein cows, 27 ± 2.2 d (f

Hysteroscopic and laparoscopic management of caesarean scar (niche) defects in symptomatic patients.

INTRODUCTION: Worldwide, rates of caesarean section are rising. The term 'niche' describes the presence of an hypoechoic area within the myometrium of the lower uterine segment, reflecting a discontinuation of the myometrium at the site of a previous caesarean section. A defect in caesarean section scar is associated with symptoms like abnormal uterine bleeding, infertility and complications in subsequent pregnancy including: risk of rupture and morbidly adherent placenta. It can also increase rates of complications during gynaecological procedures: IUCD insertion, evacuation of retained produ

Prevalence and risk factors for transition period diseases in grazing dairy cows in Brazil.

The aim of this cross-sectional study was to describe the incidence risk of dystocia, retained placenta (RP), pathological recumbence (down cow), the prevalence of metritis and subclinical ketosis (SCK), and the risk factors for SCK, metritis, and RP in grazing dairy herds in Southern Brazil. Fifty-three herds were visited 2-6 times from February to October of 2015. Body condition score (BCS), breed, days in milk (DIM), parity and disease status were recorded for each cow that was between 3 and 21 DIM at the time of the visits. Management practices were determined using a survey and environmen

Randomized clinical trial of a calcium supplement for improvement of health in dairy cows in early lactation.

Prophylactic Ca supplementation immediately after calving is a common strategy to prevent clinical and subclinical hypocalcemia in parturient dairy cows. The objective of this study was to evaluate the effect of prophylactic administration of an injected Ca supplement on blood Ca concentration at 24 and 48h after treatment, incidence risk of clinical disease and culling, milk production in early lactation, and probability of pregnancy at first insemination. Cows without signs of visible milk fever (n=984) from 7 farms were blocked by parity and randomly assigned to receive either Ca gluconate

Lying behavior and postpartum health status in grazing dairy cows.

Many cows have difficulty making the transition from pregnancy to lactation, as evidenced by the high incidence of disease that occurs in the weeks after calving. Changes in lying behavior can be used as an indicator of illness, yet no work to date has evaluated this relationship in dairy cows on pasture. The objectives of this study were to describe the lying behavior of grazing dairy cows during the first 3 wk after calving and determine the relationships between transition diseases and lying behavior. Our convenience sample included 227 multiparous and 47 primiparous Holstein cows from 6 co

Multi-Omics Identification of Key Immune Molecules in Gestational Diabetes Mellitus: FKBP5 and HLA-DQA1 as Candidate Biomarkers.

BACKGROUND: Gestational diabetes mellitus (GDM) is a major metabolic complication of pregnancy in which immune dysregulation has been implicated. The Systemic Immune-Inflammation Index (SII) has been significantly associated with GDM risk, highlighting the importance of the placental immune microenvironment in GDM pathogenesis. Yet comprehensive cross-modality integration of immune molecular data remains limited. This study aimed to systematically identify and validate key placental immune molecules in GDM. PATIENTS AND METHODS: Bulk transcriptome data (training set: 10 GDM/10 controls; valid

Lay attitudes and misconceptions and their implications for the control of brucellosis in an agro-pastoral community in Kilombero district, Tanzania.

Brucellosis is a priority zoonotic disease in Tanzania that causes ill-health in people and affects livestock productivity. Inadequate awareness and behavior risking transmission can impede control efforts. We conducted a cross-sectional survey of 333 livestock owners in three villages in the Kilombero district, Tanzania, to understand their awareness, knowledge and behavior associated with brucellosis. Six Focus Group Discussions (FGDs), two in each village, were conducted, as well as an additional FGD with male herders from one of the villages. Factors associated with knowledge on brucellosi

Maternal obesity and postpartum haemorrhage after vaginal and caesarean delivery among nulliparous women at term: a retrospective cohort study.

BACKGROUND: Increasing rates of postpartum haemorrhage in developed countries over the past two decades are not explained by corresponding changes in risk factors and conjecture has been raised that maternal obesity may be responsible. Few studies investigating risk factors for PPH have included BMI or investigated PPH risk among nulliparous women. The aim of this study was to determine in a cohort of nulliparous women delivering at term whether overweight and obesity are independent risk factors for major postpartum haemorrhage (PPH ≥1000ml) after vaginal and caesarean section delivery. Advancing Human Placental Modeling Through Stem-Cell-Derived Trophoblast Organoids and Reprogramming Innovations.

The human placenta is a temporary organ structured to optimize exchange between the maternal and fetal circulatory systems. Its fetal component consists of highly branched chorionic villi, which are anchored to the maternal uterine wall and project into the intervillous space. The outer surface of these villi is lined by a multinucleated, continuous layer called the syncytiotrophoblast, which is supported by an underlying layer of proliferative cytotrophoblast cells and the invasive extravillous trophoblast (EVT). This cellular bilayer forms a selective barrier that directly bathes in maternal

Vaccine Candidate Brucella melitensis 16MΔvjbR Is Safe in a Pregnant Sheep Model and Confers Protection.

As a natural host species for Brucella melitensis, pregnant sheep offer an ideal model to evaluate vaccine candidates for safety. B. melitensis strain Rev. 1 has been used almost exclusively to prevent brucellosis in small ruminants, but it causes abortions when given to pregnant animals. To evaluate the comparative safety of the candidate Brucella melitensis 16MΔvjbR, pregnant sheep (n = 6) were vaccinated subcutaneously with 1 × 1010 CFU/ml of 16MΔvjbR or 1 × 109 CFU/ml Rev. 1 at a highly susceptible stage of gestation (approximately 70 days). 16MΔvjbR resulted in only 1 abortion (1 of 6) co

Modification of endometrial arteries during invasion by cytotrophoblast cells in the pregnant macaque.

Fetal trophoblast cells invade endometrial blood vessels and gain access to maternal blood within two days after the onset of blastocyst implantation in macaques. Soon thereafter, cytotrophoblast cells migrate well into the lumina of arteries and subsequently invade arterial walls. Using electron microscopy and light microscopy we investigated the interactions between invasive cytotrophoblast cells and the cellular and extracellular components in the walls of endometrial arteries. The placentas and adjacent endometrium of 22 macaques (GD 17 to term) were examined. Spiral arteries containing mi

Placenta at single-cell resolution in early and late preeclampsia: insights and clinical implications.

Preeclampsia, one of the great obstetrical syndromes, manifests through diverse maternal and fetal complications and remains a leading contributor to adverse perinatal outcomes. In this review, we describe our work on single-cell and single-nuclei RNA sequencing to elucidate the molecular mechanisms that underlie early- and late-onset preeclampsia. Analysis of 46 cell types, encompassing approximately 90,000 cells from placental tissues collected after delivery, demonstrated cellular dysregulation in early-onset preeclampsia, whereas late-onset preeclampsia showed comparatively subtle changes.

Time profiles of energy balance in dairy cows in association with metabolic status, inflammatory status, and disease.

The early lactation period in dairy cows is characterized by complex interactions among energy balance (EB), disease, and alterations in metabolic and inflammatory status. The objective of this study was to cluster cows based on EB time profiles in early lactation and investigate the association between EB clusters and inflammatory status, metabolic status, oxidative stress, and disease. Holstein-Friesian dairy cows (n = 153) were selected and monitored for disease treatments during wk 1 to 6 in lactation. Weekly EB was calculated based on energy intake and energy requirements for maintenance

Prevalence and risk factors related to anovular phenotypes in dairy cows.

The objective of the current study was to evaluate the relationship of body condition score (BCS) at 35 d in milk (DIM), milk production, diseases, and duration of the dry period with prevalence of anovulation at 49 DIM and then, specifically, with the prevalence of each anovular phenotype. We hypothesized that anovular follicular phenotypes, classified based on maximal size of the anovular follicle, have different etiologies. A total of 942 lactating Holstein cows (357 primiparous and 585 multiparous) from 1 herd had ovaries evaluated by ultrasonography at 35 ± 3 and 49 ± 3 DIM to detect the
