Journal of Pediatric Surgery-Sponsored Fred McLoed Lecture. Undescended testis: the underlying mechanisms and the effects on germ cells that cause infertility and cancer.

Testicular descent is a complex morphological process that occurs in at least 2 stages, with different hormonal control. Insl3 controls the first step of gubernacular enlargement, although the abnormality long gubernacular cord in persistent Műllerian duct syndrome remains unexplained. Androgens control inguinoscrotal migration, which may be triggered by local signalling from the mammary line, and which requires the genitofemoral nerve. However, there is still much to learn about this phase, which when abnormal frequently leads to cryptorchidism. Orchidopexy is being recommended in the first y

Low paternal dietary folate alters the mouse sperm epigenome and is associated with negative pregnancy outcomes.

Epidemiological studies suggest that a father's diet can influence offspring health. A proposed mechanism for paternal transmission of environmental information is via the sperm epigenome. The epigenome includes heritable information such as DNA methylation. We hypothesize that the dietary supply of methyl donors will alter epigenetic reprogramming in sperm. Here we feed male mice either a folate-deficient or folate-sufficient diet throughout life. Paternal folate deficiency is associated with increased birth defects in the offspring, which include craniofacial and musculoskeletal malformation

CIB1 is essential for mouse spermatogenesis.

CIB1 is a 22-kDa calcium binding, regulatory protein with approximately 50% homology to calmodulin and calcineurin B. CIB1 is widely expressed and binds to a number of effectors, such as integrin alphaIIb, PAK1, and polo-like kinases, in different tissues. However, the in vivo functions of CIB1 are not well understood. To elucidate the function of CIB1 in whole animals, we used homologous recombination in embryonic stem cells to generate Cib1(-/-) mice. Although Cib1(-/-) mice grow normally, the males are sterile due to disruption of the haploid phase of spermatogenesis. This is associated wit

Comparison of antioxidant capacity and apoptosis-related molecular changes in testicular tissue of Tibetan sheep at different altitudes.

The high-altitude hypoxic environment threatens the reproductive function of male animals. Although Tibetan sheep are well-adapted to the plateau, the molecular mechanisms underlying testicular functional maintenance remain unclear. Using testicular tissues from Tibetan sheep at three altitudes (2300, 3300, and 4000 m), this study systematically compared histomorphology, antioxidant status, apoptosis-related molecular changes, and related gene expression through histological, biochemical, molecular, and cellular assays. The aim was to multi-dimensionally analyze altitudinal differences in test

A simplified two-dimensional culture system supports meiotic progression during mouse spermatogenesis in vitro.

In vitro spermatogenesis (IVS) remains a major challenge due to its complexity and extended duration. Although organ culture of neonatal mouse testes can support complete spermatogenesis, its three-dimensional structure limits precise observation and control of culture conditions, resulting in heterogeneous oxygen and nutrient gradients. Two-dimensional (2-D) systems provide greater accessibility but often induce accelerated or abnormal meiosis. In this study, we present a simplified, serum free 2-D culture system that enables meiotic progression of mouse germ cells to the mid pachytene stage.

Dissection of Obesity-Induced Spermatogenic Dysfunction at Single-Cell Resolution.

BACKGROUND: Obesity is a major risk factor for male infertility; however, how different obesity etiologies reshape testicular cellular states and transcriptional programs remains poorly understood. OBJECTIVES: This study aimed to characterize cell-type-specific transcriptional alterations associated with testicular dysfunction in dietary and genetic obesity models using single-cell RNA sequencing. METHODS: Normal diet (ND) mice served as controls, whereas high-fat diet (HFD) and leptin-deficient obese (OB) mice represented diet-induced and genetic obesity models, respectively. Testicular phe

Ssrp governs germline development independent of the FACT complex in Drosophila spermatogenesis.

BACKGROUND: The heterodimeric FACT complex (SSRP1/Ssrp and SUPT16H/Dre4) is primarily recognized as a chromatin remodeler. While mutations in this complex are linked to human intellectual disability and it maintains neural stem cell fate in flies, single-cell RNA sequencing reveals robust FACT complex expression in human testicular germ cells. Nevertheless, its specific functions during spermatogenesis remain unexplored. METHODS: This study utilized Drosophila melanogaster as a model to investigate the roles of FACT complex during spermatogenesis. Germline-specific and somatic-specific knockd

Dose-Dependent Effects of Intratesticular Adipose-Derived Mesenchymal Stem Cell Injection on Heat-Induced Spermatogenesis Disorder in Wistar Rats: Focus on Apoptosis and Oxidative Stress.

BACKGROUND & OBJECTIVE: Spermatogenesis is a temperature-dependent process, and testicular heat stress can cause spermatogenic failure by inducing cell apoptosis and oxidative stress, ultimately leading to male infertility. Adipose-derived mesenchymal stem cells (AMSCs) have been considered an effective therapy for various tissue degenerations, demonstrating the ability to stimulate testicular regeneration and restore spermatogenesis. The current study focuses on the therapeutic potential of AMSCs on semen quality, testicular morphological changes, and oxidative stress parameters in rats expos

Structure and function of vasa gene in gonadal gametogenesis of Pacific abalone.

Pacific abalone (Haliotis discus hannai) is a marine gastropod mollusc with significant economic importance in both global fisheries and aquaculture. However, studies exploring the gonadal development and regulatory mechanisms of Haliotis discus hannai are limited. This study aimed to explore whether the vasa gene acted as a molecular marker for germ cells. Initially, the vasa gene was successfully cloned using the cDNA-end rapid amplification technique. The cloned gene had a 2478-bp-long open reading frame and encoded 825 amino acids. Then, a recombinant expression vector was constructed base

Involvement of Nrf2-PPAR-γ signaling in Coenzyme Q10 protecting effect against methotrexate-induced testicular oxidative damage.

Studies have identified Coenzyme Q10 (CoQ10) as a promising agent in improving idiopathic male infertility; however, its role in chemically or environmentally induced testicular dysfunction is not well-established. We investigated the potential of CoQ10 to attenuate methotrexate (MTX)-induced testicular damage and to identify molecular targets of CoQ10 effects. Wistar rats received a single intraperitoneal dose of 20 mg/kg MTX on the fifth day of the 10-day experimental protocol. 100 mg/kg CoQ10 was given orally daily for ten days, alone or combined with MTX. The testes of MTX-treated animals

Transcription factor PBX4 regulates limb development and haematopoiesis in mice.

The mammalian Pre-B cell leukaemia transcription factors 1-4 (PBX1-4) constitutes the PBC class of the homeodomain (HD)-containing proteins, which play important roles in diverse developmental processes. The functions and the underlying molecular mechanisms of PBX1-3 but not PBX4 have been extensively studied, and they have been reported to direct essential morphogenetic processes and organogenesis. In the present study, we generated knockin mice of FLAG-tagged PBX4 and the Pbx4 knockout (KO) mice and carried out in-depth characterisation of PBX4 expression and function. PBX4 was initially det

A Review of Animal Models Investigating the Reproductive Effects of Gender-Affirming Hormone Therapy.

Gender-affirming hormone therapy (GAHT) is an important component in the process of transitioning for many transgender and gender-diverse (TGD) individuals. Multiple medical organizations recommend fertility preservation counseling prior to initiation of GAHT; however, there remains little high-quality data regarding the impact of GAHT on fertility and reproductive function. A PubMed literature review was performed using Boolean search operators linking keywords or phrases such as "mouse", "rat", "primate", "animal model", "transgender", "gender", "estrogen", "testosterone", "fertility", and "

PACAP ameliorates the fertility of obese mice through PAC1/PKA/ERK/Nrf2 signal axis.

Obesity is strongly linked to male infertility. Apoptotic inflammatory response caused by oxidative stress in testicular spermatogenic cells is one of the important causes of obesity-related male infertility. As bioactive peptide is secreted by the pituitary gland, pituitary adenylate cyclase activating polypeptide (PACAP) has a powerful triple role of anti-oxidation, anti-apoptosis, and anti-inflammation, and it is involved in the male reproduction regulation, but the specific mechanism remains unknown. The purpose of this study is to explore the role of PACAP in obesity-related male infertil

Deciphering the autophagy regulatory network via single-cell transcriptome analysis reveals a requirement for autophagy homeostasis in spermatogenesis.

Background: Autophagy has been implicated as a crucial component in spermatogenesis, and autophagy dysfunction can lead to reproductive disorders in animal models, including yeast, C. elegans and mice. However, the sophisticated transcriptional networks of autophagic genes throughout human spermatogenesis and their biological significance remain largely uncharacterized. Methods: We profiled the transcriptional signatures of autophagy-related genes during human spermatogenesis by assessing specimens from nine fertile controls (including two normal persons and seven obstructive azoospermia (OA)

Variance in expression and localization of sex-related genes CgDsx, CgBHMG1 and CgFoxl2 during diploid and triploid Pacific oyster Crassostrea gigas gonad differentiation.

Several evolutionarily conserved classes of transcriptional regulators were involved in diverse sex determination and differentiation pathways across taxa, whereas their roles in most mollusks is still limited. The Pacific oyster Crassostrea gigas, a dioecious bivalve with sex reversal, could be an ideal model for this issue because of its complex sexuality and potential disruption of sex differentiation in triploid individuals. Here, two mRNA splicing isoforms of a DM domain gene CgDsx and two isoforms of a novel sex-related CgBHMG1 (ortholog of BHMG1 in mammals) were identified in C. gigas.

Transcriptome analysis of turkey (Meleagris gallopavo) reproductive tract revealed key pathways regulating spermatogenesis and post-testicular sperm maturation.

The application of transcriptomics to the study of the reproductive tract in male turkeys can significantly increase our current knowledge regarding the specifics of bird reproduction. To characterize the complex transcriptomic changes that occur in the testis, epididymis, and ductus deferens, deep sequencing of male turkey RNA samples (n = 6) was performed, using Illumina RNA-Seq. The obtained sequence reads were mapped to the turkey genome, and relative expression values were calculated to analyze differentially expressed genes (DEGs). Statistical analysis revealed 1,682; 2,150; and 340 DEGs
