In vivo activity of orally administered antibiotics and chemotherapeutics against acute septicaemic pasteurellosis in rabbits.

Different antibiotics and chemotherapeutics were tested for therapeutic efficacy in rabbits, in an experimental model using a Pasteurella multocida strain which causes hyperacute septicaemia in this animal species. Only enrofloxacin, administered in the drinking water at a concentration of 50-100 mg/l cured the rabbits, provided that a daily intake of 5 mg/kg body weight was achieved. The other drugs tested (tetracycline, spiramycin, erythromycin and a combination of sulfamerazine with trimethoprim), at doses recommended for rabbits, showed little or no activity at all, with the exception of c

Interaction of quinolones with the theophylline metabolism in man: investigations with lomefloxacin and pipemidic acid.

The single-dose pharmacokinetics of theophylline alone and at the end of a five-day treatment period with a new difluorinated quinolone antibacterial, lomefloxacin (400 mg/d) and pipemidic acid (400 mg b.i.d.) were investigated in 12 healthy male volunteers. Concentration of theophylline and its metabolites 1-methylxanthine, 1-methyluric acid, 1.3-dimethyluric acid and 3-methylxanthine were determined in plasma, and (except 1-methyluric acid) in urine by HPLC separation. The elimination half-life of theophylline before quinolone treatment was 5.7 +/- 2.0 h and did not change significantly unde

Acquired genetic mechanisms of a multiresistant bacterium isolated from a treatment plant receiving wastewater from antibiotic production.

The external environment, particularly wastewater treatment plants (WWTPs), where environmental bacteria meet human commensals and pathogens in large numbers, has been highlighted as a potential breeding ground for antibiotic resistance. We have isolated the extensively drug-resistant Ochrobactrum intermedium CCUG 57381 from an Indian WWTP receiving industrial wastewater from pharmaceutical production contaminated with high levels of quinolones. Antibiotic susceptibility testing against 47 antibiotics showed that the strain was 4 to >500 times more resistant to sulfonamides, quinolones, tetrac

Dog/cat-origin quinolone-resistant Streptococcus agalactiae isolates with point mutations in quinolone resistance-determining regions: Relatedness with clonal complex 10.

OBJECTIVE: We aimed to investigate dog/cat-origin quinolone-resistant Streptococcus agalactiae isolates with point mutations in quinolone resistance-determining regions (QRDRs) and to define the relatedness between quinolone-resistant isolates and their microbiological features of capsular genotype, sequence type (ST)/clonal complex (CC), and antimicrobial resistance (AMR) gene. METHODS: With dog/cat-origin 22 isolates, type strain, and human-origin 6 isolates, we performed antimicrobial susceptibility testing by agar plate dilution method using levofloxacin, ciprofloxacin, and moxifloxacin.

In vitro activity of plazomicin against quinolone-resistant gram-negative bacteria isolated from catheter-associated urinary tract infections.

Quinolone resistance among uropathogens is an increasing concern. Plazomicin is a new aminoglycoside that shows promising results against resistant bacteria. However, no study has yet tested its effect specifically on quinolone-resistant organisms. This study aimed to evaluate the in vitro activity of plazomicin and comparator drugs against quinolone-resistant Gram-negative isolates of catheter-associated urinary tract infections (CAUTI). Plazomicin demonstrated high inhibiting activity against Enterobacteriaceae isolates (95.9% at MIC≤ 2 mg/L), with MIC50/90 was 1/2 mg/L. High MICs values wer

Suppression of the SOS response modifies spatiotemporal evolution, post-antibiotic effect, bacterial fitness and biofilm formation in quinolone-resistant Escherichia coli.

BACKGROUND: Suppression of the SOS response has been proposed as a therapeutic strategy for potentiating quinolones against susceptible, low-level quinolone-resistant (LLQR) and resistant Enterobacteriaceae. OBJECTIVES: To monitor the functionality of the SOS response in the evolution towards clinical quinolone resistance and study its impact on the evolution of spatiotemporal resistance. METHODS: An isogenic collection of Escherichia coli (derived from the strain ATCC 25922) carrying combinations of chromosomally and plasmid-mediated quinolone resistance mechanisms (including susceptible, L

Quinolones versus macrolides in the treatment of legionellosis: a systematic review and meta-analysis.

BACKGROUND: Legionellosis is a life-threatening disease. The clinical superiority of quinolones or macrolides for treating patients with legionellosis has not been established. METHODS: We performed a systematic review and meta-analysis of studies reporting data that allowed the comparison of quinolones versus macrolides in the treatment of proven legionellosis published from 1 January 1985 to 31 January 2013. We collected baseline aggregate patient characteristics. Studied outcomes included mortality, clinical cure, time to apyrexia, length of hospital stay and occurrence of complications in

Temafloxacin in acute purulent exacerbations of chronic bronchitis.

Temafloxacin hydrochloride, a new fluoroquinolone, was given orally in doses of 300 or 600 mg twice daily for ten days to 36 patients, all hospitalized because of severe acute purulent exacerbations of chronic bronchitis. Sputum cultures before, during and after treatment showed that the infection was eliminated in 12/18 evaluable patients given 300 mg and in 13/16 receiving the 600 mg doses. Haemophilus influenzae, Branhamella catarrhalis and Streptococcus pneumoniae were effectively eliminated, but only half the Pseudomonas aeruginosa infections were eradicated. MICs for most pathogens were

Spatial distribution and structural discrimination of quorum sensing metabolites in Pseudomonas aeruginosa by MALDI-MSI.

Bacterial biofilms are complex bacterial communities embedded within an extracellular polymeric substance (EPS) that protects the cells and increases survival. Once established on an orthopaedic implant, a biofilm can cause difficult-to-treat and severe infections. Pseudomonas aeruginosa is of particular concern due to its strong biofilm-forming capability. Within biofilm communities, quorum sensing (QS) networks regulate biofilm formation, virulence, and intercellular communication. This is achieved through the production of signalling molecules, QS metabolites. In P. aeruginosa, quinolones s

Pharmacokinetics of gyrase inhibitors, Part 1: Basic chemistry and gastrointestinal disposition.

This review describes the physicochemistry and pharmacokinetic principles of gyrase inhibitors in relation to their distribution in the gastrointestinal tract. Their basicity and lipophilicity at physiologic pH, which affect their disposition in the gastrointestinal tract, are weakened or strengthened by substitutions on the quinoline nucleus. Heterocyclic substitution at position 7 significantly increases basicity and affects lipophilicity considerably. At physiologic pH, the 3-carboxyl group adds significant negative charge. The balance between the negatively charged carboxyl group and the p

[Comparison of the adverse effect profile of different substances such as penicillins, tetracyclines, sulfonamides and quinolones].

The penicillins, the tetracyclines and the sulfonamides have often been used in the last few decades in spite of their well-known side effects. Hypersensitivity reactions to penicillins are among the most important adverse reactions in these antibiotics; in every case a careful medical history has to be taken before a new course of penicillin treatment. The use of tetracyclines in women during the last six months of pregnancy or in children under the age of eight years is contraindicated. Patients with severe blood, kidney or liver disease should not be treated with sulfonamides. Toxic reactio

Quinolones in the treatment of Salmonella carriers.

Infections caused by Salmonella typhi are commonly followed by a chronic carrier state despite positive clinical and initial bacteriologic responses. The use of primary antibiotics like chloramphenicol, ampicillin, and trimethoprim-sulfamethoxazole has several major drawbacks, including in some instances the failure to prevent the carrier state. The appearance worldwide of strains with multiple resistance to the most commonly used regimens has prompted the search for new forms of therapy. Among the agents studied have been third-generation cephalosporins and quinolones, which are active in vit

Quinolones in urology.

The new quinolones have broad antimicrobial spectra covering all aerobic gram-negative and gram-positive bacteria encountered in urinary tract infections. All are administered orally, some also parenterally, low degree of resistance, few side effects and bacteriological and clinical cure rates similar to or higher than traditional antimicrobials make them especially suitable for treatment of complicated urinary tract infections including bacterial prostatitis. Non-critical use of quinolones in simple infections where standard drugs may be equally effective and safe should be discouraged.

Current experience with quinolones in the treatment of urinary tract infection.

Urinary infection in some clinical situations remains difficult to treat with standard oral therapy. Particular problems occur in individuals with underlying genitourinary abnormalities, with intolerance to multiple oral agents, with infections due to resistant organisms, and in men with prostatitis. The fluoroquinolone antimicrobials have an extended spectrum of activity compared to standard oral therapy, achieve high concentrations in the urine following the usual therapeutic doses, and are relatively free of major adverse effects. Thus, they may be effective therapy for some of the clinical

Use of quinolones for the treatment of osteomyelitis and septic arthritis.

The low minimal inhibitory concentrations and minimal bactericidal concentrations of the quinolones for most pathogenic gram-negative and many gram-positive organisms, the ease of their administration, and their good oral absorption make them good candidates for the treatment of chronic bone infections. Data presently available suggest that the quinolones are effective in the treatment of experimental osteomyelitis due to Pseudomonas aeruginosa, osteomyelitis due to other gram-negative organisms, and (when combined with rifampin) in the treatment of gram-positive osteomyelitides. Quinolones ha

Results of treatment with quinolones of sexually transmitted diseases in the Far East: a brief summary.

The emergence of penicillinase-producing Neisseria gonorrhoeae (PPNG) strains in 1976 and the increasing frequency of chromosomally mediated resistance to penicillin has resulted in a decline in the effectiveness of penicillin for the treatment of gonorrhea. In Hong Kong, the emergence of bacterial resistance to kanamycin or tetracycline has also been observed. Quinolones were first introduced in Hong Kong in 1985, and since then the percentage of PPNG-related cases of gonorrhea has dropped dramatically. The use of quinolones for the treatment of nongonococcal urethritis has not been as succes

Antibiotics in the management of shigellosis in children: what role for the quinolones?

In the last 20 years, epidemic bacillary dysentery due to Shigella dysenteriae type 1 has become an important cause of serious morbidity and death in children less than 5 years of age in a number of developing countries; in many instances the infecting strains have been resistant to many or all of the usually recommended antibiotics. Moreover, an increasing proportion of Shigella strains of other serotypes isolated in all parts of the world are now resistant to the most commonly used antibiotics. The new fluoroquinolones have been shown to be active against Shigella in vitro and effective for

Quinolone antibacterial-agent-induced cutaneous phototoxicity: ear swelling reactions in Balb/c mice.

The phototoxic potentials of quinolone derivatives and the possibility of free radical contribution to their phototoxicity were investigated by measuring increments in ear thickness. Balb/c mice, fasted overnight, were orally administered ofloxacin (OFLX), lomefloxacin (LMFX), enoxacin (ENX), ciprofloxacin (CPFX) and DR-3355 (the s-isomer of OFLX), and immediately exposed to ultraviolet-A light (UVA: 320-400 nm) for 4 h (21.6 joules/cm2). Measurement of ear thickness was carried out 0, 24 and 48 h after the end of irradiation. The time-course profiles of ear thickness varied with both the dose
