The function of the gallbladder as a lesser biliary circulation.

The function of the gallbladder is questioned in a hypothesis postulating that all bile which enters this reservoir is totally absorbed to the liver and none of it flows back to the cystic duct and duodenum. Cholecystokinesis is believed to increase and maintain a gradient of intravesical pressure which increases the filtration pressure necessary for the absorption of bile to the liver. The absorption is through the tributaries of the cystic veins, the ducts of Luschka and Rokitansky-Aschoff sinuses. Although the hypothesis lacks experimental support, it is based on a number of anatomical, his

Anatomy, physiology and function of the nasal cavities in health and disease.

In studying drug absorption from the nasal mucous membrane, it is essential to have a clear understanding of anatomy and physiology of the nose, and how it relates to the characteristics of the delivery system used. The human nose is characterized by an individually varying shape and caliber, which might interfere with standard recommendations of intranasal medication. It is also of significance that there is a tendency for reflex- induced and profuse watery hypersecretion from glands, and for quick and considerable changes of mucosal thickness due to the presence of large venous sinusoids. Molecular aspects of intestinal calcium absorption.

Intestinal Ca(2+) absorption is a crucial physiological process for maintaining bone mineralization and Ca(2+) homeostasis. It occurs through the transcellular and paracellular pathways. The first route comprises 3 steps: the entrance of Ca(2+) across the brush border membranes (BBM) of enterocytes through epithelial Ca(2+) channels TRPV6, TRPV5, and Cav1.3; Ca(2+) movement from the BBM to the basolateral membranes by binding proteins with high Ca(2+) affinity (such as CB9k); and Ca(2+) extrusion into the blood. Plasma membrane Ca(2+) ATPase (PMCA1b) and sodium calcium exchanger (NCX1) are mai

Enteroendocrine Regulation of Nutrient Absorption.

Enteroendocrine cells (EECs) in the intestine regulate many aspects of whole-body physiology and metabolism. EECs sense luminal and circulating nutrients and respond by secreting hormones that act on multiple organs and organ systems, such as the brain, gallbladder, and pancreas, to control satiety, digestion, and glucose homeostasis. In addition, EECs act locally, on enteric neurons, endothelial cells, and the gastrointestinal epithelium, to facilitate digestion and absorption of nutrients. Many recent reports raise the possibility that EECs and the enteric nervous system may coordinate to re

The effect of esomeprazole on the upper GI tract release and systemic absorption of mesalazine from colon targeted formulations.

The aim of the present study was to investigate the effect of coadministration of the proton pump inhibitor (PPI) esomeprazole on the upper GI tract behavior and systemic exposure of mesalazine from two mechanistically different colon targeted delivery systems: Claversal (pH-dependent release) and Pentasa (prolonged release). To this end, gastric, jejunal and systemic concentrations of mesalazine and its metabolite N-acetyl mesalazine were monitored in 5 healthy volunteers following oral intake of Pentasa or Claversal with or without PPI pre-treatment (cross-over study). Our exploratory study

Biliary epithelium: a new chapter in cell biology.

The intrahepatic biliary epithelium lines the biliary tree from the duct of Hering to the extrahepatic bile ducts. Until the early '90's, the biliary tree was considered to play only a mechanic role in transporting bile into the duodenum. In the last few years, a number of experimental models have generated a bulk of new knowledge showing how the biliary epithelium displays extraordinary properties in term of secretion, absorption, proliferation and signalling toward the other liver parenchymal and mesenchymal cells. It is now clearly emerging that the functions of biliary epithelium are tight

The influence of playing surface on external demands and physiological responses during a soccer match simulation.

We investigated the effects of playing surfaces with different impact absorption characteristics on external demand and physiological responses. Fifteen participants completed a soccer match simulation on natural grass, synthetic turf and concrete surfaces. Accelerometry-derived PlayerLoadTM per minute (PL·min-1) and average net force (AvFNet) were used to quantify external demands at the centre of mass (CoM), upper-back, mid-back and hip. Heart rate, oxygen uptake, energy expenditure and RPE quantified physiological responses. The concrete surface exhibited the least impact absorption, with p

Zinc Absorption from Micronutrient Powders Is Low in Bangladeshi Toddlers at Risk of Environmental Enteric Dysfunction and May Increase Dietary Zinc Requirements.

BACKGROUND: Environmental enteric dysfunction (EED), a chronic inflammatory disorder of the small bowel, is suspected to impair absorption of micronutrients, including zinc. OBJECTIVE: The objective of this study was to compare zinc absorption from micronutrient powder (MNP) over a range of zinc doses in young children screened for EED with use of the lactulose:mannitol ratio (L:M). METHODS: Bangladeshi children aged 18-24 mo, grouped according to high and low L:M (≥0.09 and <0.09, respectively), were randomly assigned to MNP with 0, 5, 10, or 15 mg Zn/sachet (10 subjects per dose per L:M gr

Model-Based Analysis of Biopharmaceutic Experiments To Improve Mechanistic Oral Absorption Modeling: An Integrated in Vitro in Vivo Extrapolation Perspective Using Ketoconazole as a Model Drug.

Mechanistic modeling of in vitro data generated from metabolic enzyme systems (viz., liver microsomes, hepatocytes, rCYP enzymes, etc.) facilitates in vitro-in vivo extrapolation (IVIV_E) of metabolic clearance which plays a key role in the successful prediction of clearance in vivo within physiologically-based pharmacokinetic (PBPK) modeling. A similar concept can be applied to solubility and dissolution experiments whereby mechanistic modeling can be used to estimate intrinsic parameters required for mechanistic oral absorption simulation in vivo. However, this approach has not widely been a

An objective skin-type classification based on non-invasive biophysical parameters.

BACKGROUND: Despite the invention of various non-invasive bioengineering tools, skin-type analysis has largely been based on subjective assessments. However, advancements in the functional cosmetic industry and artificial intelligence-assisted dermatology are creating a greater demand for an objective skin-type classification system. OBJECTIVES: To propose an objective skin-type classification system solely based on non-invasive, bioengineering devices; provide reference values applicable to the Korean population; and compare our reference values with those of published studies. Topical carvedilol delivery prevents UV-induced skin cancer with negligible systemic absorption.

The β-blocker carvedilol prevents ultraviolet (UV)-induced skin cancer, but systemic drug administration may cause unwanted cadiovascular effects. To overcome this limitation, a topical delivery system based on transfersome (T-CAR) was characterized ex vivo and in vivo. T-CAR was visualized by Transmission Electron Microscopy as nanoparticles of spherical and unilamellar structure. T-CAR incorporated into carbopol gel and in suspension showed similar drug permeation and deposition profiles in Franz diffusion cells loaded with porcine ear skin. In mice exposed to a single dose UV, topical T-CAR

An overview of epidermal lamellar bodies: Novel roles in biological adaptations and secondary barriers.

The epidermal lamellar bodies (LBs) are specialized organelles that contain pro-barrier lipids imparting a fully lamellar internal structure, but also other cargoes such as enzymes (lipid metabolizing and proteolytic), enzyme inhibitors, and antimicrobial peptides. Thus, the LB secretory system, by virtue of delivering these cargoes to the stratum corneum (SC) interstices, is essential for forming the various skin barriers located in the SC. Ultrastructural studies have suggested that the morphologic features of LBs reflect the functional status of the SC. Several ichthyotic skin diseases as w

Vitamin supplementation and physical exercise performance.

Vitamins, just as minerals and trace elements, meet with great interest in the world of sports because of their supposed role in enhancing physical performance. Of the 13 compounds now considered as vitamins, most water-soluble vitamins and vitamin E are involved in mitochondrial energy metabolism. The influence of vitamin supplementation on mitochondrial metabolism is largely unknown. The principal argument for vitamin supplementation is the assumed increased vitamin requirement of athletes. Theoretically, an increased requirement can be caused by decreased absorption by the gastrointestinal

A novel nucleoside rescue metabolic pathway may be responsible for therapeutic effect of orally administered cordycepin.

Although adenosine and its analogues have been assessed in the past as potential drug candidates due to the important role of adenosine in physiology, only little is known about their absorption following oral administration. In this work, we have studied the oral absorption and disposition pathways of cordycepin, an adenosine analogue. In vitro biopharmaceutical properties and in vivo oral absorption and disposition of cordycepin were assessed in rats. Despite the fact that numerous studies showed efficacy following oral dosing of cordycepin, we found that intact cordycepin was not absorbed f

Importance of zinc in the elderly: the ZENITH study.

The elderly are at nutritional risk as a result of multiple physiological, social, psychological, and economic factors. Physiological functions naturally decline with age, which may influence absorption and metabolism. Social and economic conditions can adversely affect dietary choices and eating patterns. However, at the same time, the nutrient needs of the elderly for certain nutrient (such as vitamins, minerals, proteins) is higher than for younger adults. This article reviews the importance of zinc (Zn) in elderly people, particularly for behavioural and mental function, micronutrient stat

A Physiological-Based Model for Simulating the Bioavailability and Kinetics of Sulforaphane from Broccoli Products.

There are no known physiological-based digestion models that depict glucoraphanin (GR) to sulforaphane (SR) conversion and subsequent absorption. The aim of this research was to make a physiological-based digestion model that includes SR formation, both by endogenous myrosinase and gut bacterial enzymes, and to simulate the SR bioavailability. An 18-compartment model (mouth, two stomach, seven small intestine, seven large intestine, and blood compartments) describing transit, reactions and absorption was made. The model, consisting of differential equations, was fit to data from a human interv

Semimechanistic model describing gastric emptying and glucose absorption in healthy subjects and patients with type 2 diabetes.

The integrated glucose-insulin (IGI) model is a previously published semimechanistic model that describes plasma glucose and insulin concentrations after glucose challenges. The aim of this work was to use knowledge of physiology to improve the IGI model's description of glucose absorption and gastric emptying after tests with varying glucose doses. The developed model's performance was compared to empirical models. To develop our model, data from oral and intravenous glucose challenges in patients with type 2 diabetes and healthy control subjects were used together with present knowledge of s
