Cell-specific behavior of P2X7 receptors in mouse parotid acinar and duct cells.

P2X7 receptors (P2X7Rs) affect many epithelial cell functions including transcellular ion transport, secretion, and cell death. Here we used parotid acinar and duct cells to reveal the unique cell-specific assembly and gating of the P2X7R channels. Immunolocalization indicated expression of P2X7Rs in the luminal membrane of both cell types. Stimulation with 5 mm ATP raised [Ca2+]i levels in a cell-specific manner and activated multiple currents. The current mediated by P2X7R was isolated by infusing the cells with high [EGTA]. The initial activation of acinar cell P2X7Rs by ATP was slow requir

Cell surface galactosyltransferase acts as a general modulator of rat acinar cell proliferation.

Several physiological parameters were examined for inducing acinar cell proliferation and corresponding increased expression of beta 1-4 galactosyltransferase. In this study, dietary changes causing acinar cell proliferation included the following: the introduction of animals to a liquid diet (causing gland atrophy) followed by reintroduction of solid chow, gustatory stimulation provided by the introduction of 0.5% citric acid to animal drinking water, and removal of the submandibular gland with subsequent reliance on the parotid gland for salivary protein. Alterations in growth factor levels

GZMK+CD8+ T cells target a specific acinar cell type in Sjögren's disease.

OBJECTIVES: Sjögren's disease (SjD) is a systemic autoimmune disorder characterized by dysfunction of exocrine glands, particularly the salivary and lacrimal glands, with no clear etiology or effective therapy. This study explores the complex interplay of varied cell types in the salivary glands and their role in the pathology of Sjögren's disease. METHODS: Utilizing single-cell and spatial transcriptomics alongside spatial immunophenotyping to analyze human minor salivary glands, we developed a comprehensive understanding of the cellular landscape of non-SjD salivary glands and how that land

RBPJ Deficiency Sensitizes Pancreatic Acinar Cells to KRAS-Mediated Pancreatic Intraepithelial Neoplasia Initiation.

BACKGROUND AND AIMS: Development of pancreatic ductal adenocarcinoma (PDAC) is a multistep process intensively studied; however, precocious diagnosis and effective therapy still remain unsatisfactory. The role for Notch signaling in PDAC has been discussed controversially, as both cancer-promoting and cancer-antagonizing functions have been described. Thus, an improved understanding of the underlying molecular mechanisms is necessary. Here, we focused on RBPJ, the receiving transcription factor in the Notch pathway, examined its expression pattern in PDAC, and characterized its function in mou

The actin-specific reagent jasplakinolide induces apoptosis in primary rat parotid acinar cells.

Jasplakinolide is a reagent that stabilizes and polymerizes actin filaments and is a commonly used tool in cell biology. In primary rat parotid acinar cells, jasplakinolide partially inhibited the release of amylase induced by β-adrenergic receptor activation, as previously reported. However, in confocal microscopic observation with fluorescence conjugated anti-actin antibody, the jasplakinolide-treated cells not only showed decreased fluorescence intensity and aggregation of cortical F-actin but also revealed events characteristic of apoptosis such as cell shrinkage, membrane blebbing and apo

Cell surface markers CD44 and CD166 localized specific populations of salivary acinar cells.

OBJECTIVE: Experimental approaches tested to date for functional restoration of salivary glands (SGs) are tissue engineering, gene transfer, and cell therapy. To further develop these therapies, identifying specific cell surface markers for the isolation of salivary acinar cells is needed. To test a panel of cell surface markers [used in the isolation of mesenchymal stem cells, (MSCs)] for the localization of salivary acinar cells. MATERIALS: Human submandibular and parotid glands were immunostained with a panel of MSC markers and co-localized with salivary acinar cell differentiation markers

HCO(3) (-)-dependent transient acidification induced by ionomycin in rat submandibular acinar cells.

Ionomycin (IM, 5 microM), which exchanges 1 Ca2+ for 1 H+, changed intracellular pH (pHi) with Ca2+ entry into rat submandibular acinar cells. IM-induced changes in pHi consisted of two components: the first is an HCO3--dependent transient pHi decrease, and the second is an HCO3--independent gradual pHi increase. IM (1 microM), which activates store-operated Ca2+ channels, induced an HCO3--dependent and transient pHi decrease without any HCO3--independent pHi increase. Thus, a gradual pHi increase was induced by the Ca2+/H+ exchange. The HCO3--dependent and transient pHi decrease induced by IM

Redox-sensitive modulation of CD45 expression in pancreatic acinar cells during acute pancreatitis.

CD45, a transmembrane protein tyrosine phosphatase required for signal transduction in leukocytes, has recently been found in pancreatic acinar cells. We have investigated the relationship between kinetic expression of CD45 on acinar cells during acute pancreatitis (AP) and the ability of these cells to produce tumour necrosis factor-alpha (TNF-alpha) through mechanisms sensitive to the cellular redox state. Flow cytometric analysis showed a significant decrease in the constitutive expression of CD45 in acinar cells from six hours onwards after inducing AP by bile-pancreatic duct obstruction (

Effects of lipopolysaccharides on calcium homeostasis in isolated pancreatic acinar cells of rat.

AIM: To investigate the effects of lipopolysaccharides (LPS, endotoxin) on the calcium content in pancreatic acinar cells and the origin of Ca2+ during calcium overload induced by LPS, further to explore the mechanism of LPS in inducing calcium overload and pancreatic acinar cell injury. METHODS: Male rat pancreatic acinar cells were isolated by collagenase digestion and loaded with Fluo-3/AM, then exposed to varying doses of LPS (from 1 mg/L to 20 mg/L). The dynamic change of [Ca2+]i in single pancreatic acinar cell in the absence and presence of Ca2+ in extracellular fluid was determined by

CCK independently activates intracellular trypsinogen and NF-kappaB in rat pancreatic acinar cells.

In the cholecystokinin (CCK) hyperstimulation model of acute pancreatitis, two early intracellular events, activation of trypsinogen and activation of nuclear factor-kappaB (NF-kappaB), are thought to be important in the development of the disease. In this study, the relationship between these two events was investigated. NF-kappaB activity was monitored by using a DNA binding assay and mob-1 chemokine gene expression. Intracellular trypsin activity was measured by using a fluorogenic substrate. Protease inhibitors including FUT-175, Pefabloc, and E-64d prevented CCK stimulation of intracellul

Activation of phospholipase D by ADP-ribosylation factor in rat submandibular acinar cells.

The hydrolysis of membrane phosphatidylcholine by the enzyme phospholipase D is a key initial step in the intracellular release of the signalling molecules phosphatidic acid, diacylglycerol and arachidonic acid. Guanine nucleotide-dependent pathway leading to PLD activation were investigated in enzymatically dispersed rat submandibular acinar cells. Guanosine 5'-O-[gamma-thio]triphosphate (GTP gamma S) caused the time- and concentration-dependent stimulation of PLD in permeabilized cells. This effect was lost in prepermeabilized cells, from which cytosolic components had been allowed to leak,

Confocal and electron microscopy to characterize sialoglycoconjugates in mouse sublingual gland acinar cells.

Double lectin labeling for confocal microscopy and lectin-protein A-gold binding for electron microscopy were applied to the mouse sublingual gland in order to study surface and cytoplasmic sialoglycoconjugates. For this purpose, serially cut sections were submitted to sialidase followed by incubation with lectins recognizing usually acceptor sugars for terminal sialic acids. At the electron microscope level, the residues subtended to sialic acid were individually identified on adjacent sections by an indirect technique of labeling, whereas with confocal microscopy the above sugars were simult

cAMP modulation of a Ca2+-dependent K+ conductance in rat submandibular acinar cells.

Rat submandibular acinar cells exhibit a tetraethylammonium (TEA)-insensitive, Ca2+-activated K+ conductance, which may play an important role in Ca2+-dependent fluid secretion by this gland (T. Ishikawa, M. Murakami, and Y. Seo. Pfluegers Arch. 428: 516-525, 1994; T. Ishikawa and M. Murakami. Pfluegers Arch. 429: 748-750, 1995). We have now investigated whether this conductance would be modified during the potentiation of Ca2+-activated fluid secretion by an adenosine 3',5'-cyclic monophosphate (cAMP)-mediated mechanism in the rat submandibular gland. In isolated, vascularly perfused rat subm

Localization of salivary proteins in granules of human parotid and submandibular acinar cells.

Electron microscopic immunocytochemistry was used to investigate the distribution of four secretory proteins within the granules of human parotid and submandibular gland acinar cells. The results demonstrate that the distribution of the individual proteins differs significantly. Amylase is concentrated within the electron-dense regions of the granules, whereas agglutinin is present mainly in the regions of lower density. In contrast, the proline-rich proteins and histatins are distributed throughout the content of the granules and do not appear to be concentrated in either the high- or low-den

Pancreatic acinar cells: ionic dependence of the membrane potential and acetycholine-induced depolarization.

1. Intracellular recordings of membrane potentials have been made in vitro from the exocrine acinar cells of the mouse pancreas using glass micro-electrodes.2. The mean membrane potential of the acinar cells during superfusion with Krebs-Henseleit solution was -39.2 mV. Increasing [K](o) tenfold decreased the membrane potential by 28 mV when [K](o) was above 10 mM. This depolarization was not affected by atropine (1.4 x 10(-6)M). Strophanthin-G (10(-3)M) slowly depolarized the cells at about 10 mV hr(-1).3. Brief exposure to acetylcholine (ACh), 5.5 x 10(-5)M, or pancreozymin resulted in a sho

Precision oncology promise in the management of pancreatic acinar cell carcinoma.

Pancreatic acinar cell carcinoma (PACC) is a rare malignancy, accounting for less than 2 % of all pancreatic cancers that exhibits distinct clinical and molecular features compared to the more common pancreatic ductal adenocarcinoma (PDAC). Historically, treatment approaches for PACC have been adapted from PDAC due to limited available prospective and/or randomized data to guide management. However, the emergence of next-generation sequencing (NGS) has revealed a unique genomic landscape in PACC, creating new opportunities for precision oncology. Unlike PDAC, PACC typically lacks KRAS, TP53, a

A case of breast acinar cell carcinoma combined with metaplastic carcinoma.

BACKGROUND: Breast acinar cell carcinoma (AcCC) is a rare subtype of triple-negative breast cancer. Its histological origin remains controversial, and it exhibits poor sensitivity to conventional chemotherapy. High-grade transformation and co-existence with matrix-producing metaplastic carcinoma are uncommon, and their clinical and molecular genetic features are poorly understood. CASE PRESENTATION: A 42-year-old woman incidentally discovered a breast mass for 10 days. Ultrasound revealed an irregular hypoechoic lesion in the upper inner quadrant of the left breast, classified as BI-RADS 4b, l

S100A11 Promotes Acute Pancreatitis by Upregulating Acinar Cell Ferroptosis.

Acute pancreatitis (AP) is a common gastrointestinal disease that can cause systemic inflammation and lead to dysfunction of multiple organs. In pancreatitis, ferroptosis promotes disease progression and organ damage by regulating oxidative stress and inflammatory response. Here, ferroptosis was significantly elevated in the AP rat model and participated in regulating disease progression. Meanwhile, the expression of S100A11 was significantly upregulated in the pancreatic tissue of rats with AP, as determined by tandem mass spectrometry (TMT) proteomics. This phenomenon was also confirmed in p
