Prevalence of Aminoglycoside Resistance Genes in Acinetobacter baumannii Isolates.

BACKGROUND: Acinetobacter baumannii is one of the major causes of nosocomial infections and is resistant to most available antibiotics. Aminoglycosides remain as drugs of choice for treatment of Acinetobacter infections yet resistance to aminoglycosides has increased in the recent years. OBJECTIVES: The present study investigated the prevalence of genes encoding aminoglycoside-modifying enzymes in A. baumannii strains isolated from patients of Tabriz city, northwest of Iran. MATERIALS AND METHODS: A total of 103 Acinetobacter isolates were collected from Imam Reza Hospital of Tabriz Universi

Crystal structure of carbapenemase OXA-58 from Acinetobacter baumannii.

Class D β-lactamases capable of hydrolyzing last-resort carbapenem antibiotics represent a major challenge for treatment of bacterial infections. Wide dissemination of these enzymes in Acinetobacter baumannii elevated this pathogen to the category of most deadly and difficult to treat. We present here the structure of the OXA-58 β-lactamase, a major class D carbapenemase of A. baumannii, determined to 1.30-Å resolution. Unlike two other Acinetobacter carbapenemases, OXA23 and OXA-24, the OXA-58 enzyme lacks the characteristic hydrophobic bridge over the active site, despite conservation of the

A case of community-acquired Acinetobacter junii-johnsonii cellulitis.

Acinetobacter skin and soft tissue infection outside of the traumatic wound setting are rare occurrences. The majority of cases occur in the presence of significant comorbilities and by Acinetobacter baumanii. Herein a case is reported of community-onset, health-care-associated, non-traumatic cellulitis caused by Acinetobacter, species junii-johnsonii with bacteremia. This is the first reported case of Acinetobacter junii-johnsonii skin and soft tissue infection. Hemorrhagic bullae might be one of the clinical features of Acinetobacter cellulitis.

Two cases of necrotizing fasciitis due to Acinetobacter baumannii.

Necrotizing fasciitis has conventionally been associated with the streptococci, and when it is caused by other organisms, it is most often the result of a polymicrobial infection. We report on two cases of fatal monomicrobial necrotizing fasciitis due to Acinetobacter baumannii, an unusual finding that may be an indication of enhanced virulence of the organism.

[Bacteremia with Acinetobacter species--clinicopathological characteristics of 27 cases].

Acinetobacter spp. are non-fermented gram-negative rods that are widespread in the environment and colonize in the human skin. They are known to be a nosocomial pathogen causing, pneumonia, meningitis and bacteremia. Recently, they have been found increasingly in catheter-related infections (CRI). Thirty-seven cases of bacteremia were developed in our hospital during the past five years. Of these 27 cases were chosen out of the medical records for discussion in this paper. Twenty-three cases are blood positive for Acinetobacter anitratus and 4 cases for A. lwoffii. Most cases have an underlyin

Pathogenic Potential and Antimicrobial Resistance Trends in Acinetobacter Species.

Acinetobacter baumannii has emerged as one of the most potent multidrug-resistant Gram-negative bacteria causing healthcare-associated illnesses worldwide. Its exceptional ability to thrive in hospital settings, acquire antibiotic resistance determinants, build biofilms, and elude host immune responses has contributed to its growing therapeutic relevance. The fast rise of multidrug-resistant (MDR), extensively drug-resistant (XDR), and carbapenem-resistant A. baumannii has significantly reduced available therapeutic choices while increasing morbidity and mortality. This narrative review summar

New Insights into Acinetobacter baumannii Pathogenesis and Therapeutic Implications.

Acinetobacter baumannii is a leading cause of healthcare-associated infections and is classified among the highest-priority antimicrobial-resistant pathogens. Its clinical success reflects the convergence of antimicrobial resistance (AMR) and biological traits that promote environmental persistence and transmission. Acinetobacter baumannii has undergone a remarkable transformation over the past few decades, evolving from a relatively obscure environmental bacterium into a globally recognized multidrug-resistant pathogen. Its prevalence in healthcare settings, particularly intensive care units,

An aryloxyphenol potentiates polymyxin against multidrug-resistant Acinetobacter baumannii.

BACKGROUND: The increasing prevalence of antibiotic-resistant pathogens highlights the urgent need for new strategies to overcome antimicrobial resistance. Adjuvant compounds that potentiate antibiotic activity represent a promising alternative to the development of new antibiotics. METHODS: Through high-throughput screening of a chemical library, an aryloxyphenol compound, PA74, was identified as a polymyxin adjuvant candidate. Checkerboard analyses, serial passage assays, cytotoxicity assays, and functional analyses using FabI-overexpressing and point-mutant strains were performed. In vivo

A plasmid-associated immunoglobulin-binding protein in Acinetobacter baumannii.

BACKGROUND: Acinetobacter baumannii is a critical global health threat due to multidrug resistance and high mortality. Although antimicrobial resistance mechanisms are well characterised, the virulence determinants that drive severe infections remain poorly understood. METHODS: We screened 89 carbapenem-resistant clinical isolates of A. baumannii for virulence in animal infection models and combined comparative genomics with functional assays to identify virulence factors. An immunoglobulin-binding protein from A. baumannii (ImbA) encoded on the type D plasmid was selected and characterised.

Porphyrin and photodynamic therapy as tools against Acinetobacter baumannii.

Since the 1990s, Acinetobacter baumannii has become a major global concern owing to its high incidence in hospital-acquired infections. Although antimicrobial resistance is a natural process, it has been enhanced by the widespread use of antimicrobials in human and animal healthcare and is considered a global priority in the development of new drugs. Therefore, antibacterial photodynamic therapy has gained prominence for treating infections caused by multidrug-resistant microorganisms. This involves the use of photosensitizers, which, in the presence of light radiation at the appropriate wavel

The (un)successful global spread of Acinetobacter baumannii ST3.

Acinetobacter baumannii sequence type 3 (ST3) has been reported in multiple regions but has not achieved the global dominance of major clones such as ST1 and ST2. Here, we analysed 383 ST3 genomes, including 205 isolates from the Multidrug-Resistant Organism Repository and Surveillance Network (MRSN) and 178 publicly available genomes, to investigate the population structure, resistance gene repertoire and evolutionary dynamics of this lineage. Phylogenetic analysis revealed two major clades with multiple subclades, with 82% of genomes linked to the Middle East, highlighting this region as the

Investigation of colistin-resistant non-human Acinetobacter seifertii isolates.

The Acinetobacter calcoaceticus-baumannii (ACB) complex comprises multiple nosocomial infection-causing Acinetobacter species, the best-known being Acinetobacter baumannii. Once mainly considered opportunistic, Acinetobacter species are now being isolated from agricultural and food sources. We obtained and sequenced four Acinetobacter isolates from tank milk in Bogor, Indonesia. Using Average Nucleotide Identity and digital DNA:DNA hybridization, they were identified as Acinetobacter seifertii, a member of the ACB complex. Interestingly, all four isolates were resistant to colistin, a membrane
