An analytical study of the physiology and pathology of the propagation of cardiac action potentials.

Many cardiac diseases are caused by the abnormal propagation of electrical waves. Previous experimental and modelling work is reviewed, then a detailed study of the mathematics of cardiac propagation is presented. Pathologies are examined in the context of the models by varying parameters in the models to mimic different pathological states. Ionic models of cells are simplified to form analytically tractable models of the propagation of electrical cardiac waves. The roles that sodium channel activation and inactivation play in determining the conduction velocity are studied in detail, and the

Electrical rhythmicity and spread of action potentials in longitudinal muscle of guinea pig distal colon.

Using simultaneous intracellular recordings, we have characterized 1) electrical activity in the longitudinal muscle (LM) of isolated segments of guinea pig distal colon free to contract spontaneously and 2) extent of propagation of spontaneous action potentials around the circumference of the colon. In all animals, rhythmical spontaneous depolarizations (SDs) were recorded that are usually associated with the generation of action potentials. Recordings from pairs of LM cells, separated by 100 microm in the circumferential axis, revealed that each action potential was phase locked at the two e

Calcium transients in subcompartments of the leech Retzius neuron as induced by single action potentials.

Regional Ca(2+) influx into neurons plays an essential role for fast signal processing, yet it is little understood. We have investigated intracellular Ca(2+) transients induced by a single action potential (AP) in Retzius neurons in situ of isolated ganglia of the leech Hirudo medicinalis using confocal laser scanning microscopy in the cell body, in different axonal branches, and in dendrites. In the cell body, a single AP induced a Ca(2+) transient in submembrane regions, while in central regions no fluorescence change was detected. Burst activity evoked a much larger Ca(2+) influx, which el

The membrane actions of estrogens can potentiate their lordosis behavior-facilitating genomic actions.

The membrane actions of estrogens can facilitate their genomic actions. To determine whether this facilitation bears on CNS mechanisms for estrogen-dependent behaviors, ovariectomized rats were subjected to a two-pulse treatment of estrogen directly in the hypothalamic ventromedial nucleus. Two days later, each rat was given progesterone and then tested for lordosis behavior, the induction of which requires the genomic actions of estrogen. When estrogen was given in both pulses (15 min to 2 h duration, and 5 h apart) lordosis was induced. Based on results from studies on neuroblastoma cells, w

An overview of actionable and potentially actionable TSC1 and TSC2 germline variants in an online Database.

Tuberous Sclerosis Complex (TSC) is caused by loss of function germline variants in the TSC1 or TSC2 tumor suppressor genes. Genetic testing for the detection of pathogenic variants in either TSC1 or TSC2 was implemented as a diagnostic criterion for TSC. However, TSC molecular diagnosis can be challenging due to the absence of variant hotspots and the high number of variants described. This review aimed to perform an overview of TSC1/2 variants submitted in the ClinVar database. Variants of uncertain significance (VUS), missense and single nucleotide variants were the most frequent in clinica

Summating-potential/action-potential ratio in normal ears: effects of dehydration.

INTRODUCTION: Studies using electrocochleography (ECoG) in patients with Meniere's disease and in patients with perilymph fistula have demonstrated abnormally large summating-potential (SP) components compared with the amplitude of the action-potential (AP). The possible influence of normal physiological variation, methodological differences, and test-retest variations are often difficult to interpret. The purpose of this study is to specify what constitutes a clinically important change in the SP/AP ratio in normal hearing subjects under the condition of dehydration induced by urea. MATERIAL

Antiviral properties of phages: potential mechanisms of actions.

Bacteriophages (phages) have long been known to treat bacterial infections, although some early studies showed that phages also have potential antiviral mechanisms. This review provides a descriptive summary of ideas on how phages might have a significant role in inhibiting viral infections. Phages are known to directly modulate the host immunity that subsequently influences the immune responses against viral infections. It is also reasonable to explore the phage potential to directly inhibit viral infection in humans, including herpes simplex virus (HSV) and severe acute respiratory syndrome

Computational modeling of the human compound action potential.

The auditory nerve (AN) compound action potential (CAP) is an important tool for assessing auditory disorders and monitoring the health of the auditory periphery during surgical procedures. The CAP has been mathematically conceptualized as the convolution of a unit response (UR) waveform with the firing rate of a population of AN fibers. Here, an approach for predicting experimentally recorded CAPs in humans is proposed, which involves the use of human-based computational models to simulate AN activity. CAPs elicited by clicks, chirps, and amplitude-modulated carriers were simulated and compar

The potential therapeutic actions of melatonin in colorectal cancer.

Colorectal cancer (CRC) is the third most common cancer and lethal disease worldwide. Melatonin, an indoleamine produced in pineal gland, shows anticancer effects on a variety of cancers, especially CRC. After clarifying the pathophysiology of CRC, the association of circadian rhythm with CRC, and the relationship between shift work and the incidence of CRC is reviewed. Next, we review the role of melatonin receptors in CRC and the relationship between inflammation and CRC. Also included is a discussion of the mechanism of gene regulation, control of cell proliferation, apoptosis, autophagy, a

Parsing the Potential Neuroendocrine Actions of FGF21 in Primates.

Fibroblast growth factor (FGF) 21, a unique, largely liver-derived endocrine member of the FGF superfamily, is often thought of as a fasting factor owing to its induction in rodents during starvation. However, FGF21 is not increased by fasting for periods of <7 days in humans; instead, it rises sharply after acute alcohol and sugar intake and also after several days of overfeeding, suggesting another role in states of positive energy balance. Recent studies suggest that in the postingestive state, FGF21 may regulate energy intake and discourage consumption of alcohol and sugars, most likely th

CCN proteins as potential actionable targets in scleroderma.

Systemic sclerosis (SSc) is a complex autoimmune connective tissue disease combining inflammatory, vasculopathic and fibrotic manifestations. Skin features, which give their name to the disease and are considered as diagnostic as well as prognostic markers, have not been thoroughly investigated in terms of therapeutic targets. CCN proteins (CYR61/CCN1, CTGF/CCN2, NOV/CCN3 and WISP1-2-3 as CCN4-5-6) are a family of secreted matricellular proteins implicated in major cellular processes such as cell growth, migration, differentiation. They have already been implicated in key pathophysiological pr

Quantification of motor unit action potential energy.

OBJECTIVE: Motor unit action potentials (MUAPs) recorded by needle electrode reflect the functional state of the motor unit and its force-generating capacity, and are usually described morphologically (e.g. amplitude, duration). However, since the purpose of motor unit activation is force generation, MUAP energy seems a more physically meaningful measurement. METHODS: MUAPs were obtained by multi-MUAP decomposition of real interference patterns taken from human patients with neurological diseases. The energy content of each MUAP was measured from a time-frequency representation (TFR), specifi

Steroid inhibitors of androgen-potentiated actions on skin.

Antiandrogens, such as cyproterone acetate, and oestrogens both inhibit sebaceous secretion in rats and have a potentiality for the treatment of hirsutism and acne in the human female. However, they act at different points. In castrated rats treated with testosterone, 3 micrograms/day oestradiol produced a greater decrease in sebum secretion than a dose of cyproterone acetate over 1000 times larger; moreover the antiandrogen reduced the incidence of sebaceous mitoses whereas the oestradiol did not. In hirsute women, oral administration of 100 mg of cyproterone acetate daily caused a 40% reduct

Interactions of neurotoxins with the action potential NA + ionophore.

Four neurotoxins that activate the action potential Na+ionophore of electrically excitable neuroblastoma cells interact with two distinct classes of sites, one specific for the alkaloids veratridine, batrachotoxin, and aconitine, and the second specific for scorpion toxin. Positive heterotropic cooperativity is observed between toxins bound at these two classes of sites. Tetrodotoxin, a specific inhibitor of the action potential Na+ current, inhibits activation by each of these toxins in a noncompetitive manner (KI=4-8 nM). These results suggest the existence of three functionally separable co

Epidural spinal cord recordings (ESRs): sources of neural-appearing artifact in stimulation evoked compound action potentials.

Objective. Evoked compound action potentials (ECAPs) measured during epidural spinal cord stimulation (SCS) can help elucidate fundamental mechanisms for the treatment of pain and inform closed-loop control of SCS. Previous studies have used ECAPs to characterize neural responses to various neuromodulation therapies and have demonstrated that ECAPs are highly prone to multiple sources of artifact, including post-stimulus pulse capacitive artifact, electromyography (EMG) bleed-through, and motion artifact. However, a thorough characterization has yet to be performed for how these sources of art

Calmodulin D133H Disrupts Cav1.2 and Kv7.1 Regulation to Prolong Cardiac Action Potentials in Long QT Syndrome.

Calmodulin (CaM) plays a central role in cardiac excitation-contraction coupling by regulating ion channels, including the L-type calcium (Ca2+) channel Cav1.2 and the voltage-gated potassium (K+) channel Kv7.1. Mutations in CaM are linked to severe arrhythmogenic disorders such as Long QT syndrome (LQTS), yet the molecular mechanisms remain incompletely understood. Here, we investigate the structural and functional consequences of the arrhythmia-associated CaM variant D133H. Biophysical analysis revealed that D133H destabilises Ca2+ binding at the C-terminal lobe of CaM, altering its Ca2+-dep

Calibration of a biophysical model to replicate evoked compound action potentials in the swine vagus nerve.

Vagus nerve stimulation (VNS) is a promising application of bioelectronic medicine to treat many pathologies, ranging from epilepsy and depression to cardiovascular diseases. Conventional VNS is not optimized taking into account the topographic organization of the vagus nerve, resulting in suboptimal stimulation protocols, which can lead to severe adverse effects. The development of in vivo methods to determine topographic organization would allow more selective stimulation protocols and is thus pivotal in the development of future therapies. Here, we show that it may be possible to reverse-en

Mechanisms of altered skeletal muscle action potentials in the R6/2 mouse model of Huntington's disease.

Huntington's disease (HD) patients suffer from progressive and debilitating motor dysfunction for which only palliative treatment is currently available. Previously, we discovered reduced skeletal muscle Cl- channel (ClC-1) and inwardly rectifying K+ channel (Kir) currents in R6/2 HD transgenic mice. To further investigate the role of ClC-1 and Kir currents in HD skeletal muscle pathology, we measured the effect of reduced ClC-1 and Kir currents on action potential (AP) repetitive firing in R6/2 mice using a two-electrode current clamp. We found that R6/2 APs had a significantly lower peak amp

Late electrically-evoked compound action potentials as markers for acute micro-lesions of spiral ganglion neurons.

Cochlear implants (CIs) are the treatment of choice for profoundly hearing impaired people. It has been proposed that speech perception in CI users is influenced by the neural health (deafferentation, demyelination and degeneration) of the cochlea, which may be heterogeneous along an individual cochlea. Several options have been put forward to account for these local differences in neural health when fitting the speech processor settings, however with mixed results. The interpretation of the results is hampered by the fact that reliable markers of locally restricted changes in spiral ganglion
