Evidence of impaired afferent vagal function in patients with diabetes gastroparesis.

Two patients, a 28-year-old male and a 70-year-old female, with chronic insulin dependent diabetes mellitus and evidence of autonomic neuropathy were studied using cortical evoked responses following esophageal balloon and electrical stimulation. Both patients had symptomatic gastroparesis, poor gastric emptying, and reduced gastroduodenal motility including abnormal results of scintigraphy and manometry. There was slowing of afferent vagal conduction but good evoked potential responses were recorded even though one patient could not feel electrical stimulation of either the proximal or distal

Signal processing in the vagus nerve: Hypotheses based on new genetic and anatomical evidence.

Each organism must regulate its internal state in a metabolically efficient way as it interacts in space and time with an ever-changing and only partly predictable world. Success in this endeavor is largely determined by the ongoing communication between brain and body, and the vagus nerve is a crucial structure in that dialogue. In this review, we introduce the novel hypothesis that the afferent vagus nerve is engaged in signal processing rather than just signal relay. New genetic and structural evidence of vagal afferent fiber anatomy motivates two hypotheses: (1) that sensory signals inform

Convergence onto interneurons subserving primary afferent depolarization of group I afferents.

The aim of the study was to investigate whether common or independent neuronal pathways are used to evoke primary afferent depolarization (PAD) from selectively activated group Ia and Ib afferents of different muscles. To this end, the spatial facilitation of effects of various afferents, indicating convergence on the same interneurons, was used as a test. Its occurrence was assessed on dorsal root potentials (DRPs) evoked in unspecified fibers or using intra-axonal recording from identified group Ia muscle spindle afferents or group Ib tendon organ afferents. Spatial facilitation has been fou

A single GABA neuron receives contacts from myelinated primary afferents of two adjacent peripheral nerves. A possible role in neuropathic pain.

GAD67-EGFP mice were used in a series of experiments to provide anatomical evidence for the role of the reduction in myelinated primary afferent input to GABA spinal neurons in the production of neuropathic pain following peripheral L5 nerve injury. First, we confirmed that L5 injury in these mice produced mechanical and thermal hyperalgesia in the ipsilateral foot. Second, we injected a mixture of cholera toxin subunit-B (CTb) and isolectin B4 (IB4) in the sciatic nerve to selectively label its myelinated and unmyelinated primary afferents. Results showed that primary afferents of sciatic ner

Changes in afferent activity after spinal cord injury.

AIMS: To summarize the changes that occur in the properties of bladder afferent neurons following spinal cord injury. METHODS: Literature review of anatomical, immunohistochemical, and pharmacologic studies of normal and dysfunctional bladder afferent pathways. RESULTS: Studies in animals indicate that the micturition reflex is mediated by a spinobulbospinal pathway passing through coordination centers (periaqueductal gray and pontine micturition center) located in the rostral brain stem. This reflex pathway, which is activated by small myelinated (Adelta) bladder afferent nerves, is in turn

Neural mechanisms of respiratory interoception.

Respiratory interoception is one of the internal bodily systems that is comprised of different types of somatic and visceral sensations elicited by different patterns of afferent input and respiratory motor drive mediating multiple respiratory modalities. Respiratory interoception is a complex system, having multiple afferents grouped into afferent clusters and projecting into both discriminative and affective centers that are directly related to the behavioral assessment of breathing. The multi-afferent system provides a spectrum of input that result in the ability to interpret the different

Mini Review: Central Organization of Airway Afferent Nerve Circuits.

Airway afferents monitor the local chemical and physical micro-environments in the airway wall and lungs and send this information centrally to regulate neural circuits involved in setting autonomic tone, evoking reflex and volitional respiratory motor outflows, encoding perceivable sensations and contributing to higher order cognitive processing. In this mini-review we present a current overview of the central wiring of airway afferent circuits in the brainstem and brain, highlighting recent discoveries that augment our understanding of airway sensory processing. We additionally explore how a

The sensory trigeminal system in birds: input, organization and effects of peripheral damage. A review.

The primary sensory trigeminal system in birds comprises the mesencephalic trigeminal nucleus and the trigeminal ganglion with projections to the principal sensory nucleus (PrV) and the descending tract with its subnuclei. Other cranial nerves can contribute to PrV and the descending system that together form the somatosensory system of the head. There is also a proprioceptive component. The somatosensory system comprises a component serving tactile sense and a nociceptive component. The former processes information from many mechanoreceptors in beak and tongue; both PrV and subnuclei of the d

The pattern of trunk lateral line afferents and efferents in the rainbow trout (Salmo gairdneri).

The primary projections of the mechanosensory posterior lateral line nerve of the rainbow trout Salmo gairdneri, a teleost without lateral line specializations, were studied by applying horseradish peroxidase (HRP). The afferents project nearly exclusively to the nucleus medialis and a small nucleus caudalis in the hindbrain, and to the eminentia granularis. In addition, there is a sparse projection to the cerebellum, but a projection to the magnocellular nucleus is lacking. The afferent projection to the lateral part of the eminentia granularis is more dense than the projection to the medial

Blockade of nociceptive inhibition of plasma extravasation by opioid stimulation of the periaqueductal gray and its interaction with vagus-induced inhibition in the rat.

We have previously shown that stimulation of cutaneous or visceral nociceptors suppresses inflammation measured as bradykinin-induced synovial plasma extravasation in the knee joint of the rat. This suppression occurs through the activation of a spinal as well as a supraspinal reflex pathway leading to activation of the adrenal medullae and probably the release of epinephrine. These nociceptive-neuroendocrine reflex pathways are tonically inhibited by activity in abdominal vagal afferents acting through an inhibitory descending pathway projecting through the dorsolateral funiculus (DLF) ipsila

Organization of the Auditory Brainstem in a Lizard, Gekko gecko. II. Afferent and Efferent Projections of Nuclei of the Lateral Lemniscus and the Torus Semicircularis.

In reptiles, two taxa, lepidosaurs and archosaurs, have ears with thin tympanic membranes that permit sensitive hearing of high-frequency sounds in air. The organization of their central auditory systems may reflect this increased sensitivity to sound. To understand auditory processing in lepidosaurs further, we used tract tracing techniques to examine the organization and connections of the lemniscal and midbrain auditory nuclei in the Tokay gecko. The nuclei of the lateral lemniscus (LLD) consist of anterior (LLDa), medial (LLDm), and posterior (LLDp) subdivisions and receive ascending proje

From Anatomy to Function: Critical Questions on the Dynamic Properties of Segregated Visceral Afferent Circuits.

The recent study by Wang et al. (2025) provides a seminal description of the distinct brainstem projections of splanchnic and pelvic spinal afferents from the mouse colorectum, offering a crucial anatomical framework for visceral sensation. In this letter, we critically appraise these findings and delineate essential frontiers for future research. While the anatomical segregation is compelling, we argue that the molecular logic governing synaptic specificity and signal transmission within these circuits remains unexplored. Furthermore, the translational relevance of this map must be tested in

Gaps in our understanding of how vagal afferents to the small intestinal mucosa detect luminal stimuli.

Vagal afferents to the gastrointestinal tract are crucial for the regulation of food intake, signaling negative feedback that contributes to satiation and positive feedback that produces appetition and reward. Vagal afferents to the small intestinal mucosa contribute to this regulation by sensing luminal stimuli and reporting this information to the brain. These afferents respond to mechanical, chemical, thermal, pH, and osmolar stimuli, as well as to bacterial products and immunogens. Surprisingly, little is known about how these stimuli are transduced by vagal mucosal afferents or how their

Telencephalic afferents to the caudolateral neostriatum of the pigeon.

The pigeon caudolateral neostriatum (NCL) shares a dopaminergic innervation with mammalian frontal cortical areas and is implicated in the regulation of avian cognitive behavior. Retrograde tracing methods were used to identify forebrain projections to NCL and to suggest a possible role of this area in mediating spatial behavior. NCL receives telencephalic projections from the hyperstriatum accessorium, cells along the border of hyperstriatum dorsale and hyperstriatum ventrale, anterolateral hyperstriatum adjacent to the vallecula, confined cell groups within the anterior neostriatum, and subd

Alteration of human cutaneous afferent discharges as the result of long-lasting vibration.

The unitary activities of slowly (15 SAI) and fast-adapting type I (12 FAI) skin mechanoreceptive afferent units innervating the anterior part of the human leg and foot were recorded by using the microneurographic method. The recordings were performed both at rest and on application of cutaneous stimuli of various intensities before and after exposure of the corresponding receptive fields to vibration (0.5 mm peak to peak, 100 pulses/s, 10 min). The results show that 11% of the units tested, which were previously silent, developed a bursting pattern of postvibratory activity, which lasted 12 m

Characterization of mouse lumbar splanchnic and pelvic nerve urinary bladder mechanosensory afferents.

Sensory information from the urinary bladder is conveyed via lumbar splanchnic (LSN) and sacral pelvic (PN) nerves to the spinal cord. In the present report we compared the mechanosensitive properties of single afferent fibers in these two pathways using an in vitro mouse bladder preparation. Mechanosensitive primary afferents were recorded from the LSN or PN and distinguished based on their response to receptive field stimulation with different mechanical stimuli: probing (160 mg to 2 g), stretch (1-25 g), and stroking of the urothelium (10-1,000 mg). Four different classes of afferent were r

Role of C1 neurons in anti-inflammatory reflex: Mediation between afferents and efferents.

Neuroimmune communication, the connection between the autonomic regulatory pathway and immune cells, has been implicated in the regulation of immune function and inflammation. The role of afferents (vagal afferent and somatic sensory nerves) and efferents (autonomic nervous and hypothalamic-pituitary-adrenal systems) in the inflammatory reflex has been well studied; however, the central pathway remains unknown. C1 neurons include both catecholaminergic and glutamatergic neurons, which are located in the rostral ventrolateral medulla. C1 neurons project to the spinal cord, dorsal motor nucleus

The renal afferent nerves in the pathogenesis of hypertension.

The renal nerves play a role in the pathogenesis of hypertension in a number of experimental models. In the deoxycorticosterone acetate - salt (DOCA-NaCl) hypertensive rat and the spontaneously hypertensive rat (SHR) of the Okamoto strain, total peripheral renal denervation delays the development and blunts the severity of hypertension and causes an increase in urinary sodium excretion, suggesting a renal efferent mechanism. Further, selective lesioning of the renal afferent nerves by dorsal rhizotomy reduces hypothalamic norepinephrine stores without altering the development of hypertension i
