Duck hepatitis B virus infection, aflatoxin B1 and liver cancer in domestic Chinese ducks.

The oncogenicity of Duck hepatitis B virus (DHBV) is unclear since hepatocellular carcinomas (HCCs) have been reported only in domestic ducks in Qidong, an area of China where hepatitis B virus (HBV) and aflatoxin B1 (AFB1) are risk factors for liver cancer in man. In order to better define the association between DHBV infection, AFB1 and HCC we analysed a series of 16 duck liver samples collected from local farms in Qidong. HCC was found in eight and cirrhosis in one of these samples. Furthermore bile duct proliferation, characteristic of AFB1 exposure in ducks and other animal species, was f

Influence of aflatoxin B1 intoxication on duck livers with duck hepatitis B virus infection.

In an attempt to determine the effect of aflatoxin B1 (AFB) intoxication on livers with duck hepatitis B virus (DHBV) infection, domestic ducks were given 0.1 mg of AFB/kg body weight twice a week for a maximum period of 54 weeks employing various experimental designs. The ducks were infected with DHBV by i.v. inoculation of DHBV-positive sera within 24 h posthatch. The livers were examined histologically, immunohistochemically, and ultrastructurally, and the livers and sera were examined by molecular hybridization for DHBV DNA. AFB administration induced hepatocellular necrosis and marked bil

Transcriptional insights into aflatoxin B1 induced hepatotoxicity and comparative effects of medicinal herbs in pigs.

BACKGROUND: Aflatoxin B1 (AFB1) contamination in animal feed poses a serious risk to livestock health due to its hepatotoxic effects. Many medicinal herbs which may be used as feed additives exhibit antioxidant and anti-inflammatory properties with potential hepatoprotective outcomes. We investigated effects of AFB1 in three concentrations (30 µg/kg BW, 60 µg/kg BW, 120 µg/kg BW) as well as three medicinal herbs, i.e., kalmegh (Andrographis paniculata), milk thistle (Silybum marianum), and turmeric (Curcuma longa) in pigs. Hepatic expression of genes involved in biotransformation, detoxificati

Mitigation of aflatoxin B1-induced toxicopathology and immunosuppression in broilers using a multimodal antioxidant formulation.

Aflatoxin B1 (AFB1) contamination of poultry feed impairs growth, immunity, and organ function, causing substantial economic losses. This study evaluated the protective efficacy of a multimodal, water-soluble antioxidant formulation (HI-RESIST®) against AFB1-induced toxicosis in broiler chickens. A total of 240 one-day-old Indian River broilers were randomized into six groups (n = 40): control, AFB1 (150 µg/kg feed), HI-RESIST® 1 (1 mL/L water), HI-RESIST® 2 (2 mL/L), AFB1 + HI-RESIST® 1, and AFB1 + HI-RESIST® 2. Treatments continued for 35 days. Outcome measures included body weight and feed

Unravelling the antifungal mechanism of phytic acid on Aspergillus flavus development and aflatoxin B1 biosynthesis.

Aspergillus flavus (A. flavus) is a plant pathogenic fungus that produces toxic and carcinogenic aflatoxins, posing a threat to food safety and human health. As a kind of food additive, phytic acid (PA) exhibits excellent antimicrobial activity. However, the antifungal effects of PA against A. flavus are not well understood. Here, transcriptomic and biochemical analyses were carried out to investigate the inhibitory effects of PA against A. flavus and potential mechanisms. Our findings demonstrated that PA effectively inhibited the vegetative growth, conidiation and aflatoxin B1 (AFB1) product

Chronic exposure to aflatoxin B1 increases hippocampal microglial pyroptosis and vulnerability to stress in mice.

BACKGROUND: Chronic aflatoxin B1 (AFB1) exposure may increase the risk of multiple neuropsychiatric disorders. Stress is considered one of the main contributors to major depressive disorder. Whether and how chronic AFB1 exposure affects vulnerability to stress is unclear. METHODS: Mice were exposed for three weeks to AFB1 (100 µg/kg/d) and/or chronic mild stress (CMS). The vulnerability behaviors in response to stress were assessed in the forced swimming test (FST), sucrose preference test (SPT), and tail suspension test (TST). Microglial pyroptosis was investigated using immunofluorescence,

Aflatoxin B1-activated heterophil extracellular traps result in the immunotoxicity to liver and kidney in chickens.

Aflatoxin B1 (AFB1) is a mycotoxin with strong toxicity and play a large proportion in aspergillosis. Heterophil extracellular traps (HETs) was considered as an innate immune response of chickens to resist pathogens. AFB1 has been reported to trigger macrophages extracellular traps (METs) in THP-1 cells and RAW264.7 cells, but whether AFB1 could also activate HETs release, and the mechanism underlying AFB1-activated HETs in chicken remains unclear. In this study, we confirmed that AFB1could induce HETs release, which was a network of DNA-based structures consist of citrullinated histone 3 (cit

Biotransformation of aflatoxin B1 by Lactobacillus helviticus FAM22155 in wheat bran by solid-state fermentation.

Lactobacillus helveticus FAM22155 was the most efficient among five lactic acid bacteria at removing aflatoxin B1 (AFB1) during solid-state fermentation on wheat bran substrate. The mechanism of removal was explored by comparing different fermentation modes. Liquid fermentation had little effect on the breakdown of AFB1. However, a protein extract from the fermented bran was equally effective at degrading aflatoxin B1 as living cell digestion. After treatment with heat and protease K, the degrading capacity of the protein extract was significantly reduced. Taken together, the observed biotrans

Electrochemical Aflatoxin B1 immunosensor based on the use of graphene quantum dots and gold nanoparticles.

Electrochemical immunosensor for aflatoxin B1 (AFB1) is described that uses a composite prepared from graphene quantum dots (GQDs) and gold nanoparticles (Au NPs). The GQD-AuNP conjugate was obtained by using 2-aminothiophenol (ATP) as a linker where the carboxy groups of GQD bind to the amino groups of crosslinker via conjugation of thiol binding to the AuNP. To evaluate the conjugation of the GQD-AuNP composite, Fourier transform infrared spectroscopy (FT-IR), and transmission electron microscopy (TEM) was applied. The composite was placed on an indium tin oxide (ITO) electrode and then modi

Pathological effects of concurrent administration of aflatoxin B1 and fowl adenovirus-4 in broiler chicks.

The current study was designed to investigate pathological effects of fowl adenovirus in broilers exposed to aflatoxin B1. Fowl Adenovirus-4 (FAdV-4) infection is remerging in all types of poultry birds in Pakistan. Poultry feed contamination with mycotoxin (aflatoxin) is another important global issue. A total of 125-day old broiler birds were divided into six equal groups. Group A served as control. B and C groups were administered with aflatoxin B1 (AFB1) 100 and 200 μg/kg feed. Group D was infected with FAdV-4, while groups E and F administered with both AFB1 (100 & 200 μg/kg) along with F

Mutational spectra of aflatoxin B1 in vivo establish biomarkers of exposure for human hepatocellular carcinoma.

Aflatoxin B1 (AFB1) and/or hepatitis B and C viruses are risk factors for human hepatocellular carcinoma (HCC). Available evidence supports the interpretation that formation of AFB1-DNA adducts in hepatocytes seeds a population of mutations, mainly G:C→T:A, and viral processes synergize to accelerate tumorigenesis, perhaps via inflammation. Responding to a need for early-onset evidence predicting disease development, highly accurate duplex sequencing was used to monitor acquisition of high-resolution mutational spectra (HRMS) during the process of hepatocarcinogenesis. Four-day-old male mice w

Natural occurrence of aflatoxin B1 in marketed foods and risk estimates of dietary exposure in Koreans.

Aflatoxin B1 (AFB1) is an unavoidable food contaminant. To evaluate the potential health risk of AFB1 to Koreans posed by food consumption, we determined the natural occurrence of AFB1 in food and estimated the excess risk for liver cancer through dietary exposure to AFB1. A total of 694 food samples collected from six different regions of South Korea were analyzed for their AFB, content. One hundred four of the 694 samples were found to give positive enzyme-linked immunosorbent assay (ELISA) readings for AFB1 and were further investigated with high-performance liquid chromatography. The role of duck hepatitis B virus and aflatoxin B1 in the induction of oxidative stress in the liver.

The aim of our study was to use the Pekin duck model to investigate the interactions between hepadnaviral infection and aflatoxin B1 (AFB1) exposure including the role of both factors in the induction of oxidative stress in the liver. AFB1 exposure of duck hepatitis B virus (DHBV) infected Pekin ducks induced a significant increase in viral replication associated with an intense biliary ductular cells proliferation. Interestingly, extremely high levels of AFB1-DNA adducts (40-120 pmol AFB1-Fapy/mg DNA) and AFB1-albumin adducts (1,500-3,000 pg AFB1-lys Eq/mg albumin) were detected in duck liver

Low frequency of p53 gene mutation in tumors induced by aflatoxin B1 in nonhuman primates.

Aflatoxin B1 has been suggested as a causative agent for a G to T mutation at codon 249 in the p53 gene in human hepatocellular carcinomas from southern Africa and Qidong in China. To test this hypothesis, nine tumors induced by aflatoxin B1 in nonhuman primates were analyzed for mutations in the p53 gene. These included four hepatocellular carcinomas, two cholangiocarcinomas, a spindle cell carcinoma of the bile duct, a hemangioendothelial sarcoma of the liver, and an osteogenic sarcoma of the tibia. None of the tumors showed changes at the third position of codon 249 by cleavage analysis of

Effect of protein calorie malnutrition and cell replication on aflatoxin B1-induced hepatocarcinogenesis in rats.

Malnourished and well-fed neonatal Holtzman rats 10 days of age were exposed to 3 doses of aflatoxin B1 [(AFB1) CAS: 1162-65-8] at intervals of 96 hours to study the combined effect of malnutrition and cell replication in AFB1-induced hepato-carcinogenesis. The neonatal model made use of the fact that cell replication persists in the liver for 3 weeks of postnatal life. Malnutrition during suckling was induced by adopting the techniques of Widdowson and McCance of increasing the litter size to 16. Following AFB1 administration, the malnourished animals were rehabilitated on a high-protein pell

Induction of mutations in DNA-repair deficient bacteria by a liver microsomal metabolite of aflatoxin B1.

Certain strains of Salmonella typhimurium and Escherichia coli, particularly those which are very sensitive to u.v. light, are killed when incubated with rat liver mixed function oxidases and aflatoxin B(1). UvrA or recA strains of E. coli are more susceptible than the wild-type strain, while the double mutant uvrA recA is the most sensitive strain yet tested. The aflatoxin B(1) metabolite is also able to induce reverse mutations in 2 histidine auxotrophic strains of S. typhimurium, one strain of which is reverted specifically by frame shift mutagens and the other by agents inducing base pair

A light-based DNA hydrogel valve biosensor for naked-eye sensitive detection of aflatoxin B1.

DNA hydrogels have garnered significant attention in biosensing due to their high programmability and excellent biocompatibility. However, the synthesis of their key building block, DNA-polymer, is typically cumbersome, time-consuming, and involves hazardous reagents, which significantly hinders their broader application. Herein, we report a rapid (<1 min) and green synthesis strategy for DNA-polymer via photo-initiated (405 nm) covalent grafting of DNA strands onto gelatin methacryloyl (GelMA). Based on this DNA-polymer, we constructed a light-based DNA hydrogel integrated with a paper substr
