Basal subtype cancers are deadly malignancies but the molecular events driving tumor lethality are not completely understood. Ataxia-telangiectasia group D complementing gene (ATDC, also known as TRIM29), is highly expressed and drives tumor formation and invasion in human bladder cancers but the factor(s) regulating its expression in bladder cancer are unknown. Molecular subtyping of bladder cancer has identified an aggressive basal subtype, which shares molecular features of basal/squamous tumors arising in other organs and is defined by activation of a TP63-driven gene program. Here, we dem

Cancers of the breast and prostate are one of the leading causes of cancer deaths in women and men, respectively. Although several treatment options have been developed to transform these cancers into manageable chronic diseases, they still contribute to over 70 000 deaths each year in the United States. Though majority of these cancers belong to slow growing differentiated subtypes, the cancers evolve over time due to treatment-related pressure into aggressive treatment-resistant types. A mechanism attributed to the transformation of hormonal and other cancers into aggressive treatment-refrac

The authors have described three cases of voyeurism. They could state the following with regard to the psychopathological connections, the effect of the consumption of alcohol and the question of sanity: 1. In two cases, the sexual drive anomaly remained permanently hidden in the conduct of life. One case was an episodical depression associated with voyeurism and exhibitionism. 2. The effect of the alcohol has not only promoted the occurrence of the abnormal sexual drive but also caused an aggressiveness and dangerous incautiousness. 3. The soundness of mind was in no case restricted.

We investigated the relationship between work stress arising from effort-reward imbalance at work (ERI) and driving anger in a community sample of workers in Nagoya, a mid-sized city in Japan. We hypothesised that ERI would exert a positive effect on driving anger via its influence on trait anger. The study also pioneered the use of the Driving Anger Scale (DAS) in a non-western country and explored cultural differences in the experience of anger on the road. A random sample of 215 (N = 138, 64% females; N = 77, 36% males) full-time Japanese workers was obtained through random selection of one

How environmental and physiological signals interact to influence neural circuits underlying developmentally programmed social interactions such as male territorial aggression is poorly understood. We have tested the influence of sensory cues, social context, and sex hormones on progesterone receptor (PR)-expressing neurons in the ventromedial hypothalamus (VMH) that are critical for male territorial aggression. We find that these neurons can drive aggressive displays in solitary males independent of pheromonal input, gonadal hormones, opponents, or social context. By contrast, these neurons c

BACKGROUND: Risky behaviors are common in Huntington's disease (HD) and can lead to significant adverse consequences. However, the prevalence and scope of these symptoms have not been studied systematically, and no empirically validated measures are available to screen for them.
OBJECTIVE: To test a novel screening tool designed to assess risk-taking behaviors in HD.
METHODS: We administered the Risk Behavior Questionnaire (RBQ-HD) to HD patients and caregivers at Vanderbilt University Medical Center between 2018-2019. Patients completed the questionnaire based on self-report; caregivers pro

Driving behavior and speed enforcement are both important to road safety and affect vehicle exhaust emissions. Relationships between driving characteristics and safety or emissions have been assessed in multiple studies. However, there is scant information on whether safe driving also reduces emissions and how this relationship changes across urban areas. This study makes use of two similar GPS datasets collected in the metropolitan areas of Toronto and Beijing to conduct a comparative analysis of driving characteristics, speed limit violations, and emissions. Emissions for all trips were comp

BACKGROUND: The majority of crashes cause "minor" injuries (i.e., treated and released from the emergency department [ED]). Minor injury crashes are poorly studied.
OBJECTIVES: This study aims to determine the prevalence of driver-related risk factors and subsequent outcome in drivers involved in minor crashes.
METHODS: We interviewed a convenience sample of injured drivers, aged over 17 years, who were treated and released from the ED. Follow-up interviews were conducted 6 months after the crash.
RESULTS: We approached 123 injured drivers; baseline interviews were completed in 69 and follo

BACKGROUND: Aggressive driving is the leading behavior resulting in fatal or nonfatal disabling injuries. Therefore, programs with the main focus on driving behavior could reduce the risk of traffic injuries remarkably. We aimed to investigate the role of non-punitive performance feedback on drivers' behavior and evaluate the persistence of the altered behavior by using in-vehicle telematics.
METHODS: This study was done as a randomized controlled trial by repeated-measures design to evaluate the non-punitive behavioral intervention's efficacy on the behavioral transition to safer driving for

Melanoma pathogenesis from normal neural crest-derived melanocytes is often fatal due to aggressive cell invasion throughout the body. The identification of signals that reprogram de-differentiated, metastatic melanoma cells to a less aggressive and stable phenotype would provide a novel strategy to limit disease progression. In this study, we identify and test the function of developmental signals within the chick embryonic neural crest microenvironment to reprogram and sustain the transition of human metastatic melanoma to a neural crest cell-like phenotype. Results reveal that co-culture of

BACKGROUND: Alcohol consumption and drink/driving are positively correlated and many predictors of alcohol use also predict drink/driving. Past research has not fully distinguished the contributions of personal risk factors from the level of alcohol use in the prediction of drink/driving. As a result, the extent to which predictors are specific to drink/driving, versus due to a mutual association to alcohol use, is unclear.
METHODS: This study examined the unique and shared risk factors for drink/driving and alcohol use, and examined the attributable risk (AR) associated with predictors of dr

Triple-negative breast cancer (TNBC) is an aggressive and heterogeneous breast cancer subtype with poor patient outcomes. TNBC heterogeneity arises from multiple dysregulated pathways, including Notch and Hedgehog, which contribute to tumor initiation, progression, and drug resistance. Identifying common molecular regulators of TNBC aggressiveness is crucial for developing effective therapeutic strategies. Here, we demonstrate that the transcriptional coactivator MAML1 drives TNBC aggressiveness by regulating Notch1 and Gli1 stability through the E3 ubiquitin ligase Itch, functioning as an Itc

Road traffic collisions are a major issue for public health. Depression is characterized by mental, emotional and executive dysfunction, which may have an impact on driving behaviour. Patients with depression (N = 39) and healthy controls (N = 30) were asked to complete questionnaires and to drive on a driving simulator in different scenarios. Driving simulator data included speed, safety distance from the preceding vehicle and lateral position. Demographic and medical information, insomnia (Athens Insomnia Scale, AIS), sleepiness (Epworth Sleepiness Scale, ESS), fatigue (Fatigue Severity Scal

Transformed lymphoma is a complex syndrome that encompasses an array of different underlying low-grade lymphoproliferative conditions transforming into more aggressive disease as manifest by morphologic, clinical, and genetic features. Over the last decade, advances in chemoimmunotherapy have led to new options. Knowledge surrounding the genetic changes driving the process of transformation is leading to novel targeted therapies. This article focuses on the transformation of chronic lymphocytic leukemia and follicular lymphoma into diffuse large B-cell lymphoma.

OBJECTIVE: Taxis play an important role in public travel, yet limited concerns are devoted to taxi-involved safety compared with other travel modes. In particular, few studies have examined the heterogeneities of risky driving behaviors in taxi-involved crashes. This study quantifies the heterogeneity and characterizes the injury distribution of risky driving behaviors based on police-reported taxi crashes.
METHODS: Latent class clustering (LCC) is applied to identify the optimal number of clusters by maximizing heterogeneities across clusters. The number is determined by the Akaike informati

OBJECTIVES: A substantial body of literature documents the association between alcohol and drug use with impaired driving, increased risky driving behavior, and traffic crashes. However, most research has focused on offender drivers, with limited attention given to non-offenders. Thus, the applicability of these findings to non-offender drivers remains unclear. This study aimed to: (a) examine alcohol consumption patterns, drug problems, and non-prescription medication use among non-offender drivers; (b) investigate the relationship between driver characteristics (age, sex, education, and driv

Primary cutaneous CD8+ aggressive epidermotropic cytotoxic T-cell lymphoma (pcAECyTCL) is a rare provisionally categorized cutaneous lymphoma characterized by an aggressive course. Its pathogenesis and molecular mechanisms are still unknown, and only two individual cases have so far been molecularly characterized. The aim of this study was to define the pattern of numerical chromosomal alterations in tumor samples taken from 20 patients with pcAECyTCL at the time of diagnosis by means of array-comparative genomic hybridization (a-CGH). a-CGH detected numerous genomic aberrations in all the pat

Ovarian cancer is characterized by aggressive metastasis, chemoresistance, and poor survival outcomes. Gaps in understanding of factors that drive these phenotypes have hindered the development of actionable therapeutic targets. We demonstrate that Zinc Finger Protein 217 (ZNF217) is a key pro-metastatic factor in ovarian cancer cells. ZNF217 overexpression dramatically increases proliferation, metastasis, and chemoresistance while its depletion impairs these phenotypes. Consistently, ZNF217 overexpression is associated with poor prognosis in both mouse models and ovarian cancer patients. Inte

The systemic metabolic shifts that occur during aging and the local metabolic alterations of a tumor, its stroma and their communication cooperate to establish a unique tumor microenvironment (TME) fostering cancer progression. Here, we show that methylmalonic acid (MMA), an aging-increased oncometabolite also produced by aggressive cancer cells, activates fibroblasts in the TME, which reciprocally secrete IL-6 loaded extracellular vesicles (EVs) that drive cancer progression, drug resistance and metastasis. The cancer-associated fibroblast (CAF)-released EV cargo is modified as a result of re
