Cascade Coupling of Dinitrogen, Carbon Monoxide, Carbon Dioxide, and Alkynes in a Dititanium Framework.

Dinitrogen (N2), carbon monoxide (CO), and carbon dioxide (CO2) are all abundant and readily available chemical feedstocks. The functionalization of N2 with CO and CO2 is of considerable interest and importance, but remains a formidable challenge, owing to their high thermodynamic and kinetic stability. Here, we report an unprecedented cascade coupling of N2, CO, CO2, and alkynes within a dititanium framework. Sequential reactions of N2, CO, alkynes (R1C≡CR2), and CO2 with a dititanium tetrahydride complex at room temperature selectively afforded the corresponding four-component coupling produ

Enantioselective Ni-Al Bimetal-Catalyzed Cycloaddition of Cyclopropyl Ketones with Alkynes under Mild Conditions.

Transition metal-catalyzed enantioselective intermolecular cycloaddition of cyclopropyl ketones and π-unsaturated compounds represents a long-standing challenge in the asymmetric C-C bond activation of cyclopropanes. Developed strategies rely on substrate modifications to mitigate racemization or to stabilize radical intermediates. In contrast, an efficient approach governed by transition-metal catalysts for general cyclopropyl ketones remains an elusive challenge. Herein, we report a highly active chiral diamine-phosphine oxide-ligated Ni-Al bimetallic catalyst that promotes enantioselective

1,1-Oxycarbonation of Terminal Alkynes via Sequential Borylation, 1,2-Migration, and Oxidation with Oxone.

Alkynes are readily available and multifunctional synthetic intermediates, but their 1,1-oxofunctionalization remains challenging. Herein, we report a 1,1-oxycarbonation of terminal alkynes to construct ketones through sequential borylation, 1,2-carbon migration, and oxidation with Oxone as the proton source and oxidant. The synthetic potential of this transformation is showcased by the broad functional groups, scale-up synthesis, and diverse product transformations.

Visible-light induced metal-free intramolecular reductive cyclisations of ketones with alkynes and allenes.

A visible-light-induced, intramolecular, reductive cyclisation of ketones with an unsaturated hydrocarbon moiety was developed. In contrast to conventional protocols requiring resource precious or hazardous metal sources, this method enables facile access to ketyl radicals under metal-free and mild reaction conditions. By polarity-reversed, ketyl radical hydroalkoxylation of alkynes and allenes, a variety of five-membered (hetero-)cyclic products were generated in good yields with good to excellent stereoselectivities. The embedded homoallylic tertiary alcohol could be transformed into other u

Synthesis of α-Boryl Ketones via Hydration or Oxidation of B(MIDA)-Decorated Alkynes.

α-Boryl ketones are traditionally challenging targets in organic synthesis. Reported herein is a mild and metal-free synthesis of α-boryl ketones via the hydration or oxidation of N-methyliminodiacetyl boronate (B(MIDA))-decorated alkynes. A new hydration system comprised of AcCl and H2O in HFIP allows the hydration of arylethynyl B(MIDA)s at room temperature with decent functional group tolerance. An oxidative carbon deletion process of propargylic B(MIDA)s is also developed for the synthesis of aliphatic α-boryl ketones. An intriguing β-boron effect was observed to account for the unique sit

Cobalt nanoparticles-catalyzed aerobic oxygenation and esterification of alkynes via C≡C bonds cleavage.

An unprecedented efficient protocol is developed for the oxidative cleavage of C≡C bonds in alkynes to produce structure-diverse esters using heterogeneous cobalt nanoparticles as catalyst with molecular oxygen as the oxidant. A diverse set of mono- and multisubstituted aromatic and aliphatic alkynes can be effectively cleaved and converted into the corresponding esters. Characterization analysis and control experiments indicate high surface area and pore volume, as well as nanostructured nitrogen-doped graphene-layer coated cobalt nanoparticles are possibly responsible for excellent catalytic

Synthesis of primary propargylic alcohols from terminal alkynes using rongalite as the C1 unit.

Here, an efficient leaving group-activated methylene alcohol strategy for the preparation of primary propargyl alcohols from terminal alkynes by employing the bulk industrial product rongalite as the C1 unit has been described. The reaction avoids the low-temperature reaction conditions and inconvenient lithium reagents required for the classical method of preparing primary propargylic alcohols. Preliminary mechanistic studies showed that the reaction may not proceed via formaldehyde intermediates, but through the direct nucleophilic attack of the terminal alkyne on the carbon atom of rongalit

Synthesis of α,β-unsaturated ketones through nickel-catalysed aldehyde-free hydroacylation of alkynes.

α,β-Unsaturated ketones are common feedstocks for the synthesis of fine chemicals, pharmaceuticals, and natural products. Transition metal-catalysed hydroacylation reactions of alkynes using aldehydes have been recognised as an atom-economical route to access α,β-unsaturated ketones through chemoselective aldehydic C-H activation. However, the previously reported hydroacylation reactions using rhodium, cobalt, or ruthenium catalysts require chelating moiety-bearing aldehydes to prevent decarbonylation of acyl-metal-hydride complexes. Herein, we report a nickel-catalysed anti-Markovnikov select

Recent advances in visible-light-mediated functionalization of olefins and alkynes using copper catalysts.

Over the past decade, visible-light photoredox catalysis has blossomed as a powerful strategy and offers a discrete activation mode complementary to thermal controlled reactions. Visible-light-mediated photoredox catalysis also offers exciting opportunities to achieve challenging carbon-carbon and carbon-heteroatom bond formations with excellent chemo-, regio-, or stereoselectivity under mild and ecologically benign conditions. The greater part of photoredox reactions depends on heavy metals like ruthenium(II) or iridium(III) based complexes as photoredox catalysts owing to their strong absorp

Light-enabled, AlCl3-catalyzed regioselective intramolecular nucleophilic addition of non-nucleophilic alkyls to alkynes.

Light-enabled, AlCl3-catalyzed regioselective intramolecular nucleophilic cyclization of alkynes using non-nucleophilic alkyls as the nucleophile is reported. Upon photoexcitation, o-alkylphenyl alkynyl ketones can be transferred into (E)-photoenols. Thus, a nucleophilic methylene is formed from the non-nucleophilic alkyl. An AlCl3 catalyst can stabilize the (E)-photoenol intermediate and facilitate further intramolecular nucleophilic cyclization. DFT calculations indicated that the AlCl3-catalyzed cyclization is the regioselectivity determining step.

Photoinduced, Copper-Catalyzed Radical Cross-Coupling of Cycloketone Oxime Esters, Alkenes, and Terminal Alkynes.

A photoinduced, copper-catalyzed three-component radical cross-coupling of cycloketone oxime esters, alkenes, and terminal alkynes is described for the first time. Key to the success of this process was the integration of photoinduced iminyl radical-mediated C-C bond cleavage with the conceptual simplicity of copper-catalyzed radical cross-coupling. This protocol provides access to cyanoalkyl-containing propargylic compounds in good yields.

[2+2+2] cyclotrimerization of alkynes and isocyanates/isothiocyanates catalyzed by ruthenium-alkylidene complexes.

Ruthenium carbene catalysts are able to catalyze crossed [2+2+2] cyclotrimerizations of α,ω-diynes with isocyanates, isothiocyanates, and carbon disulfide. Both aliphatic and aromatic isocyanates can be used to produce fused 2-pyridones, although aliphatic isocyanates were more reactive. Aromatic isocyanates give better results when they bear electron-donating substituents. The reaction of unsymmetrical α,ω-diynes gave a product only with the substituent adjacent to the 2-pyridone nitrogen. Isothiocyanates gave thiopyranimines upon reaction with the C═S bond, whereas CS2 reacted efficiently to

Cobalt-catalyzed asymmetric three-component reductive addition of alkynes with alkyl halides and imines.

The catalytic dicarbofunctionalization of alkynes is a highly efficient strategy for the rapid assembly of complex, multi-substituted acyclic alkenes from simple feedstocks. Despite significant progress having been made in various racemic transformations, the direct enantioselective construction of multi-substituted acyclic alkenes bearing sterically congested tetrasubstituted stereocenters via multicomponent radical pathways remains a formidable and unsolved challenge in organic chemistry. Herein, we report a cobalt-catalyzed asymmetric three-component radical addition of terminal alkynes, ke

Unexpected Reactivity of 1,3,4-Thiadiazole-N-oxides with Strained Alkynes Enables Enhanced Antibody Loading.

Protein bioconjugates are at the forefront of precision medicine, enabling the design of highly selective drugs for diagnostic and therapeutic applications. While increasing the number of payloads attached to a protein is a proven strategy to enhance the potency of a bioconjugate, the introduction of additional conjugation sites often risks compromising the protein's targeting properties. In this study, we investigate the potential of 1,3,4-thiadiazole-N-oxides (TNO), a heterocyclic scaffold exhibiting excellent stability in biological media. We demonstrate that this reactive partner undergoes

Catalyst-Free Direct Hydrocarbonation of Terminal Alkynes Toward E-Alkene Substituted Stabilized Sulfoxonium Ylides.

Amide and alkene moieties are frequently found in natural products and are privileged structures in pharmaceuticals and agrochemicals. Moreover, vinyl sulfoxonium ylide can be converted into a broad range of high-value compounds, thus they have been widely employed in organic synthesis. However, the synthesis of alkene-substituted amide-sulfoxonium ylides via intermolecular hydrocarbonation of alkynes remains underexplored. This study describes the development of a high-throughput approach to provide diverse functionalized E-alkene substituted (hetero)amide-sulfoxonium ylides. The reaction occ
