CtChi19: a valuable gene for improving resistance to Botrytis cinerea and Alternaria alternata in Carthamus tinctorius.

Safflower (Carthamus tinctorius L.) is an economically important crop with both medicinal and oil-producing values, However, its production is severely threatened by fungal diseases, including those caused by Alternaria alternata and other pathogenic fungi. To date, molecular investigations into the disease resistance mechanisms of safflower remain largely limited. In this study, proteomic analysis was performed on safflower following infection by A. alternata, identifying 490 and 921 differentially expressed proteins (DEPs) at 48 hours post-infection (hpi) and 72 hpi, respectively, Among thes

Discovery of the Alternaria mycotoxins alterperylenol and altertoxin I as novel immunosuppressive and antiestrogenic compounds in vitro.

Alternaria mycotoxins may pose significant challenges to food safety and public health due to the wide spectrum of reported adverse effects. Despite this, critical information on the immunomodulatory and antiestrogenic properties of most of these contaminants is still lacking. The present study aimed to identify the mycotoxins responsible for the immunosuppressive and antiestrogenic effects of a complex extract of Alternaria mycotoxins (CE) obtained by growing an Alternaria alternata strain on rice. Through a toxicity-guided fractionation procedure involving the production of CE-fractions by s

Toxoplasma gondii and Alternaria sp.: An Original Association in an Immunosuppressed Dog with Persistent Skin Lesions.

Dogs and cats may suffer from a variety of diseases, mainly immune mediated, that require the administration of immunosuppressive drugs. Such therapies can cause adverse effects either by the toxicity of the drugs or as a consequence of immune suppression and associated opportunistic infections. Here we present an, yet unknown, association of Toxoplasma gondii and Alternaria fungus, within cutaneous lesions in a dog under long-term immunosuppressive therapy. The diagnosis of such infections is laborious and not obvious at first glance, since the clinical signs of cutaneous toxoplasmosis, neosp

The methionine biosynthesis regulator AaMetR contributes to oxidative stress tolerance and virulence in Alternaria alternata.

The tangerine pathotype of A. alternata, which produces a unique host-selective ACT toxin causes brown spots on citrus leaves and fruits. In this study, we report a methionine biosynthesis regulator (MetR), which belong to bZIP transcription factor, is required for methionine metabolism, oxidative stress tolerance and pathogenicity. We generated two ΔAaMetR mutants in the tangerine pathotype of Alternaria alternata and investigated the resulting mutant phenotypes. The ΔAaMetR disruption mutant grew poorly in the absence of methionine and unable to produce conidia. Furthermore, pathogenicity te

Successful Posaconazole Therapy of Disseminated Alternariosis due to Alternaria infectoria in a Heart Transplant Recipient.

We report a case of phaeohyphomycosis caused by Alternaria infectoria in a 61-year-old heart transplant recipient with multiple skin lesions and pulmonary infiltrates. The infection spread via the haematogenous route from the primary cutaneous lesions into the lungs. The diagnosis was based on the histopathological examination, direct microscopy, skin lesion cultures and detection of Alternaria DNA in the bronchoalveolar lavage fluid using molecular methods. The treatment consisted of a combination of surgical excision and systemic antifungal therapy. Voriconazole was the first agent used but

The Bdtf1 gene in Alternaria brassicicola is important in detoxifying brassinin and maintaining virulence on Brassica species.

Brassinin is an antifungal compound induced in Brassica plants after microbial infection. Molecular evidence is incomplete, however, in supporting the importance of brassinin in plant resistance to pathogens. To test the importance of brassinin in plant defense, we studied the functions of the gene Bdtf1 in the necrotrophic fungus Alternaria brassicicola. Several strains of mutants of this gene were weakly virulent on Brassica species, causing lesions 70% smaller in diameter than the wild type on three Brassica species. These mutants, however, were as virulent as the wild type on Arabidopsis t

Chitin deacetylases AaCDAs enable Alternaria alternata to evade host immune responses via cell wall modification.

Chitin deacetylases (CDAs) are essential for phytopathogenic fungi to evade plant immune defenses by deacetylating chitin oligomers into chitosan. However, the function of CDAs in Alternaria alternata, the causal agent of pear black spot, remains unclear. In this study, two CDA genes, AaCDA1 and AaCDA2, were cloned and identified. Gene knockout and complementation analyses revealed that ΔAaCDA1 and ΔAaCDA2 mutants showed no significant changes in colony morphology, biomass, or conidiation. However, the ΔAaCDA1 mutant exhibited increased extracellular melanin, linked to the upregulation of the

Ursolic acid nanoemulsion system for inhibition of goji berry black mold caused by Alternaria alternata.

Ursolic acid (UA) possesses antifungal activity against A. alternata, however, its poor solubility poses a challenge for practical application. We prepared UA nanoemulsions consisting of ethyl oleate, Cremophor EL, and Transcutol HP. The particle size of the UA nanoemulsions ranged from 20.53 to 27.88 nm. Electron microscopy revealed a nearly spherical structure. Fourier-transform infrared analysis proved that UA was successfully encapsulated, and the encapsulation efficiency ranged from 82 to 84%. UA nanoemulsions showed stable characteristics for 15 days of storage. The minimum inhibitory co

Three new shikimate-conjugated meroterpenoids from Alternaria alternata isolated from an entomopathogenic fungus Apis mellifera.

Three new shikimate-conjugated meroterpenes, tricycloalternarenes Z1-Z3 (1-3), were isolated from Alternaria alternate, an endophytic fungus residing in Apis mellifera. The structures of new compounds were characterized by analyses of their NMR and HRESIMS data. The absolute configurations were assigned through electronic circular dichroism (ECD) calculations and CD spectra. Bioassay results showed that tricycloalternarene Z1 (1) exhibited moderate anti-inflammatory activity against the LPS-induced production of NO in RAW 264.7 cells, with an IC50 value of 0.93 µM.

Alternaria alternata-induced airway epithelial signaling and inflammatory responses via protease-activated receptor-2 expression.

Inhalation of the fungus Alternaria alternata is associated with an increased risk of allergic asthma development and exacerbations. Recent work in acute exposure animal models suggests that A. alternata-induced asthma symptoms, which include inflammation, mucus overproduction and airway hyperresponsiveness, are due to A. alternata proteases that act via protease-activated receptor-2 (PAR2). However, because other active components present in A. alternata may be contributing to asthma pathophysiology through alternative signaling, the specific role PAR2 plays in asthma initiation and maintenan

Orbital apex syndrome caused by Alternaria species: A novel invasive fungus and new treatment paradigm.

Orbital apex syndrome as a result of invasive fungal sinusitis is a disease entity most commonly found in immunocompromised patients. Infectious invasion affecting the orbital apex can have devastating visual and life-threatening consequences. Mucormycosis and Aspergillus species are the most common causes of such infections. Alternaria fungal sinusitis is a known entity, but its ability to cause an orbital apex syndrome has not yet been reported. Here, we present a case of orbital apex syndrome in an immunocompromised patient with invasive fungal sinusitis caused by Alternaria species. The pa

Alternaria Induces Production of Thymic Stromal Lymphopoietin in Nasal Fibroblasts Through Toll-like Receptor 2.

PURPOSE: Chronic rhinosinusitis with nasal polyps is a chronic inflammatory disease with markedly increased eosinophils, Th2-type lymphocytes, fibroblasts, and goblet cells. Fungi are commonly associated with airway inflammatory diseases, and thymic stromal lymphopoietin (TSLP) is important in the development of Th2 inflammatory responses. The aim of this study was to investigate the interaction between airborne fungi and nasal fibroblasts in TSLP mRNA and protein expression. METHODS: Inferior turbinate and nasal polyp fibroblasts were stimulated with Alternaria and Aspergillus, respectively,

Folic acid induces salicylic acid-dependent immunity in Arabidopsis and enhances susceptibility to Alternaria brassicicola.

Folates are essential for one-carbon transfer reactions in all organisms and contribute, for example, to de novo DNA synthesis. Here, we detected the folate precursors 7,8-dihydropteroate (DHP) and 4-amino-4-deoxychorismate (ADC) in extracts from Arabidopsis thaliana plants by Fourier transform ion cyclotron resonance-mass spectrometry. The accumulation of DHP, but not ADC, was induced after infection of plants with Pseudomonas syringae delivering the effector protein AvrRpm1. Application of folic acid or the DHP precursor 7,8-dihydroneopterin (DHN) enhanced resistance in Arabidopsis to P. syr

Multiple phytohormone signalling pathways modulate susceptibility of tomato plants to Alternaria alternata f. sp. lycopersici.

Three phytohormone molecules - ethylene (ET), jasmonic acid (JA) and salicylic acid (SA) - play key roles in mediating disease response to necrotrophic fungal pathogens. This study investigated the roles of the ET, JA, and SA pathways as well as their crosstalk during the interaction between tomato (Solanum lycopersicum) plants and a necrotrophic fungal pathogen Alternaria alternata f. sp. lycopersici (AAL). Both the ET and JASMONIC ACID INSENSITIVE1 (JAI1) receptor-dependent JA signalling pathways are necessary for susceptibility, while SA response promotes resistance to AAL infection. In add

Bacillus sp. Z26 suppresses Alternaria alternata and its mycotoxins by targeting AaPex13-mediated peroxisome biogenesis.

Alternaria alternata is a destructive postharvest pathogen that causes fruit decay and produces multiple mycotoxins, including alternariol (AOH), alternariol monomethyl ether (AME), and tenuazonic acid (TeA), posing a serious threat to crop quality and food safety. Biocontrol stands out as a promising strategy to mitigate both fungal infection and mycotoxin production. This study investigated that Bacillus sp. Z26 effectively inhibits the mycelial growth and mycotoxin accumulation of A. alternata. Comparative transcriptomic analysis revealed that strain Z26 did not directly repress canonical m

Autophagy-related protein 4 contributes to siderophore biosynthesis, toxin production, and virulence in Alternaria alternata.

The tangerine pathotype of Alternaria alternata infects multiple citrus cultivars, causing brown spot disease. The role of autophagy in toxin production and siderophore biosynthesis remains to be further confirmed. This study identifies autophagy-related protein 4 (AaAtg4) as a critical regulator of fungal growth, development, stress resistance, iron homeostasis, and virulence, as determined by genetic and biochemical analyses. Targeted deletion of the AaAtg4 gene using split-marker recombination generated two ΔAaAtg4 mutants, which displayed reduced growth on minimal medium, impaired conidiat
