Cigarette smoke is a major risk factor for chronic respiratory diseases, yet the molecular mechanisms underlying its effects on pulmonary defense remain incompletely understood. Although transporters such as P-glycoprotein and CFTR have been implicated in smoking-related dysfunction, little is known about the impact of cigarette smoke on peptide transporter 2 (PEPT2), which plays a dual role in xenobiotic disposition and host defense by mediating the uptake of di- and tripeptides, bacterial fragments, and peptide-like drugs. Here, we investigated the effects of cigarette smoke extract (CSE) on

Particulate matter (PM2.5) contains a complex mixture of compounds with diverse physicochemical and toxicological properties. Although, solvent-extractable organic compounds of PM2.5 are known to induce oxidative and metabolic alterations, their impact on cellular structural integrity remains insufficiently characterized. In this study, we evaluated the impact of the PM2.5 extract obtained with dichloromethane (SEOM-PM2.5) on human alveolar epithelial cells (A549). Cells were exposed to non-cytotoxic concentrations of SEOM-PM2.5 and assessed for cytoskeletal organization, nuclear morphology, c

Tuberculosis is one of the leading causes of death due to a single infectious agent. Upon infection, Mycobacterium tuberculosis (M.tb) is deposited in the alveoli and encounters the lung mucosa or alveolar lining fluid (ALF). We previously showed that, as we age, ALF presents a higher degree of oxidation and inflammatory mediators, which favors M.tb replication in human macrophages and alveolar epithelial cells (ATs). Here, we define the transcriptional profile of M.tb when exposed to healthy ALF from adult (A-ALF) or elderly (E-ALF) humans before and during infection of ATs. Prior to infectio

BACKGROUND: Exposure to particulate matter (PM) with an aerodynamic diameter less than 2.5 μm (PM2.5) is a risk factor for developing pulmonary diseases and the worsening of ongoing disease. Mitochondrial fission and fusion are essential processes underlying mitochondrial homeostasis in health and disease. We examined the role of mitochondrial fission and fusion in PM2.5-induced alveolar epithelial cell damage and lung injury. Key genes in these processes include dystrophin-related protein 1 (DRP1) and optic atrophy 1 (OPA1) respectively.
METHODS: Alveolar epithelial (A549) cells were treated

Dyskeratosis congenita (DC) is a rare genetic disorder characterized by deficiencies in telomere maintenance leading to very short telomeres and the premature onset of certain age-related diseases, including pulmonary fibrosis (PF). PF is thought to derive from epithelial failure, particularly that of type II alveolar epithelial (AT2) cells, which are highly dependent on Wnt signaling during development and adult regeneration. We use human induced pluripotent stem cell-derived AT2 (iAT2) cells to model how short telomeres affect AT2 cells. Cultured DC mutant iAT2 cells accumulate shortened, un

BACKGROUND AIMS: Bronchopulmonary dysplasia (BPD), a chronic lung disease characterized by disrupted lung growth, is the most common complication in extreme premature infants. BPD leads to persistent pulmonary disease later in life. Alveolar epithelial type 2 cells (AEC2s), a subset of which represent distal lung progenitor cells (LPCs), promote normal lung growth and repair. AEC2 depletion may contribute to persistent lung injury in BPD. We hypothesized that induced pluripotent stem cell (iPSC)-derived AECs prevent lung damage in experimental oxygen-induced BPD.
METHODS: Mouse AECs (mAECs),

A disintegrin and metalloproteinase (ADAM) 17, constitutively expressed in alveolar epithelium, is the pivotal shedding enzyme mediating acute lung inflammation. On the other hand, angiotensin (Ang)-(1-7)/Mas signaling has been shown to improve acute respiratory distress syndrome and protect alveolar epithelial cells from apoptosis. In this study, we explored the effect of Ang-(1-7)/Mas signaling on the expression and activity of ADAM17 and assessed its impact on apoptosis in lipopolysaccharide (LPS)-treated human alveolar epithelial cells. LPS markedly induced the shedding activity of ADAM17

Affinity of different organs for verapamil is highly variable and organ-specific. For example, the drug exhibits high levels of accumulation in lung tissues. A transporter recognising verapamil as a substrate has previously been identified in human retinal pigment epithelial (RPE) and in rat retinal capillary endothelial (TR-iBRB2) cells. This transporter is distinct from any of the cloned organic cation transporters. Therefore, we hypothesised that the verapamil transporter is also functionally expressed in the human respiratory mucosa. Moreover, we tested the hypothesis that this transporter

BACKGROUND: Edaravone (3-methyl-1-penyl-2-pyrazolin-5-one) is a potent free-radical scavenger and has the antioxidant ability to inhibit lipid peroxidation. The study aimed to examine the effect of edaravone on protecting the acute injury of human type II alveolar epithelial cells (A549 cells) induced by paraquat (PQ) and the change of production of reactive oxygen species (ROS), malondialdehyde (MDA), superoxide dismutase (SOD).
METHODS: A549 cells were cultured and divided into PQ group (group P), edaravone-treated group (group E) and normal control group (group C). The cells in group P wer

BACKGROUND: Fatal human respiratory disease associated with influenza A subtype H5N1 has been documented in Hong Kong, and more recently in Vietnam, Thailand and Cambodia. We previously demonstrated that patients with H5N1 disease had unusually high serum levels of IP-10 (interferon-gamma-inducible protein-10). Furthermore, when compared with human influenza virus subtype H1N1, the H5N1 viruses in 1997 (A/Hong Kong/483/97) (H5N1/97) were more potent inducers of pro-inflammatory cytokines (e.g. tumor necrosis factor-a) and chemokines (e.g. IP-10) from primary human macrophages in vitro, which s

Regeneration of alveolar epithelial cells is one of the important repair processes in many types of lung injury, including the adult respiratory distress syndrome (ARDS). We have examined the effects of growth factors and cytokines on alveolar epithelial cells in vitro, and also examined histopathology and surfactant protein gene expressions in alveolar epithelial cells in rats with endotoxin-induced lung injury. Keratinocyte growth factor only induced a marked increase in levels of mRNAs for SP-A and SP-B, accompanied by an increase in SP-A protein. Intratracheal administration of endotoxin i

Influenza A virus infection mediates the host's excessive immune response, wherein caspase-3-GSDME-mediated pyroptosis of lung alveolar epithelial cells can contribute to inducing cytokine storm, leading to acute lung injury (ALI) or acute respiratory distress syndrome (ARDS). Numerous studies have shown that mesenchymal stem cells (MSCs) possess potent immunomodulatory abilities and can mitigate virus-induced cytokine storm and lung injury. However, the role of MSCs in lung pyroptosis remains poorly understood. In this study, we established an ALI model using a mouse-adapted strain of avian i

Alveolar epithelial cell (AEC) senescence is a crucial driver of pulmonary fibrosis (PF). Epoxyeicosatrienoic acids (EETs) are synthesized from arachidonic acid through the cytochrome P450 cyclooxygenase (CYP) pathway. We have reported that EETs inhibit AEC senescence, alleviating bleomycin (BLM)-induced PF in mice. However, EETs have a short half-life in vivo and are rapidly degraded to low-active end-product diols by soluble epoxide hydrolase (sEH). We observed a significant increase in sEH expression in senescent AECs, but the underlying mechanism remains unclear. In this study, we found th

Persulfidation is a post-translational modification involving the specific conversion of thiols (-SH) to persulfide groups (-SSH) on cysteine residues in target proteins. This modification plays critical roles in numerous physiological and cellular processes. Previously, we demonstrated that host cell persulfidation levels influence the effectiveness of antifungal immunity against the fungal pathogen Aspergillus fumigatus. In this study, we sought to further elucidate the relationship between persulfidation and the immune response. To this end, we defined the persulfidated proteins (the persul

Mechanical ventilation is a key respiratory support measure for critically ill patients. During improper ventilation, continuous exposure of alveolar epithelial cells (AECs) to abnormal mechanical environment can lead to ventilator-induced lung injury (VILI). Heat shock transcription factor 1 (HSF1) is a stress-responsive transcriptional regulator that orchestrates cytoprotective heat shock protein (HSP) expression in response to diverse stresses, including thermal and oxidative stress, but its role in stretch-induced AEC injury remains unclear. In this study, A549 cells were subjected to biax

INTRODUCTION: Inhalation of drugs for the treatment of pulmonary diseases has been used since a long time. Due to lungs' larger absorptive surface area, delivery of drugs to the lungs is the method of choice for different disorders. Here we present the establishment of a comprehensive permeability model using Type II alveolar epithelial cells and Beclomethasone Dipropionate (BDP) as a model drug delivered by pressurized metered dose inhaler (pMDI).
METHODS: Using Type II alveolar epithelial cells, the method was standardized for parameters viz., cell density, viability, incubation period and

Alveolar epithelial type I (AE1) cells with their wide spatial expansion form approximately 95% of the outer surface area of the air-blood barrier inside the lung. Serial block-face scanning electron microscopy (SBF-SEM) investigations led to the hypothesis that AE1 cell mitochondria are preferentially distributed as aggregates in those parts of AE1 cells that are located above connective tissue pillars between capillaries, thus not increasing the thickness of the diffusion distance for oxygen and carbon dioxide. Furthermore, it was hypothesised that postnatal development requires adapting the

BACKGROUND/AIM: Despite evidence of an association between pulmonary diseases and periodontopathic bacteria, the molecular mechanisms remain unknown. Matrix metalloproteinase-9 (MMP9) plays important roles in pneumonia, chronic obstructive pulmonary disease, and asthma; therefore, we assessed the effects of Fusobacterium nucleatum on MMP9 expression in mouse lung and A549 human alveolar epithelial cells.
MATERIALS AND METHODS: Heat-killed F. nucleatum was administered to the trachea of mice or added to A549 cell cultures. MMP9 expression was determined using real-time PCR and western blotting

Sepsis-induced acute lung injury is a type of lung disease with a high fatality rate that is characterized by acute inflammation. In the present study, the underlying role and potential mechanism of the stomatin (STOM) protein were investigated in lipopolysaccharide (LPS)-induced oxidative stress and inflammation in a mouse lung epithelial cell line, MLE-12. The expression levels of STOM and CD36 were measured using reverse transcription-quantitative PCR and western blotting. Subsequently, the expression levels of STOM and CD36 in LPS-treated MLE-12 cells were knocked down or overexpressed, re

Alveolar type II (ATII) respiratory epithelial cells are essential to normal lung function. They may be also central to the pathogenesis of diseases such as acute lung injury, pulmonary fibrosis, and pulmonary adenocarcinoma. Hence, ATII cells are important therapeutic targets. However, effective ATII cell-specific drug delivery in vivo requires carriers of an appropriate size, which can cross the hydrophobic alveolar surfactant film and polar aqueous layer overlying ATII cells, and be taken up without inducing ATII cell dysfunction, pulmonary inflammation, lung damage, or excessive systemic s
