Monitoring of hearing during treatment of leukaemia with special reference to the use of amikacin.

Thirty-nine patients with leukaemia were followed audiometrically during treatment with broad-spectrum antibiotics. Amikacin was given during neutropenic febrile episodes. Five patients reported a deterioration of the hearing function after termination of amikacin treatment. Significant hearing threshold loss occurred in 20 patients (51%). The hearing threshold changes were small in general, except for two patients who exhibited bilateral hearing threshold changes in the frequency range 0.5-8 kHz. Using multiple linear regression analysis 22% of the changes in hearing thresholds was estimated

Mycobacterium fortuitum keratitis. A comparison of topical ciprofloxacin and amikacin in an animal model.

The effect of topical amikacin or topical ciprofloxacin on Mycobacterium fortuitum keratitis was studied in a rabbit model. Two strains of M fortuitum were used: ATCC-6841 [for which mean inhibitory concentrations (MICs) indicated in vitro sensitivity to both drugs] and a cutaneous isolate from a human infection (for which MICs indicated relative in vitro resistance to amikacin but in vitro sensitivity to ciprofloxacin). Both drugs reduced the number of organisms in eyes infected with either strain (all p values < or = 0.01), but in no cases were organisms eliminated from eyes after 4 days of

Therapy of experimental cerebral nocardiosis with imipenem, amikacin, trimethoprim-sulfamethoxazole, and minocycline.

A mouse model of cerebral nocardiosis was used to determine relative antibiotic efficacy by reducing bacterial colony counts per gram of brain tissue. The antimicrobial agents employed were demonstrated in vitro to be inhibitory to most strains of Nocardia asteroides at very low concentrations. The agents used in this study were imipenem-cilastatin, amikacin, trimethoprim-sulfamethoxazole, and minocycline. Antibiotics were administered every 4 h for 72 h before animal sacrifice. Bacterial colony counts were assayed at various time points before the completion of therapy. Imipenem-cilastatin an

Evidence for a chromosomal site specifying amikacin resistance in multiresistant Serratia marcescens.

We studied 21 strains of amikacin-resistant Serratia marcescens from three different U.S. cities, Twenty of the 21 strains contained conjugative R plasmids mediating gentamicin and tobramycin resistance. Amikacin-resistant S. marcescens from two cities predominated in protracted outbreaks. Conversely, the amikacin-resistant Charleston strain (serotype 02/03:nonmotile) was isolated from only four patients during an outbreak of gentamicin- and tobramycin resistant, amikacin-susceptible S. marcescens (serotype O19:O17). Five different representative amikacin-resistant S. marcescens, each containi

N-Acetylcysteine Attenuates Amikacin-Induced Biochemical, Inflammatory, and Histopathological Alterations in Mice.

Background: Amikacin is an effective aminoglycoside antibiotic, but its clinical use is limited by nephrotoxicity, hepatotoxicity, and neurotoxicity. N-acetylcysteine (NAC), a glutathione precursor and antioxidant, may help reduce these side effects. This study evaluated whether NAC attenuated selected amikacin-induced biochemical, inflammatory, and histopathological alterations in mice. Methods: Male Swiss albino mice were divided into four groups: control, amikacin, NAC, and amikacin-NAC, and the behavioral performance was assessed using the light-dark box test. Serum samples were collected

Amikacin Liposomal Inhalation Suspension: Advances in Application Against Refractory Mycobacterium avium Complex.

Mycobacterium avium complex (MAC) is the most prevalent pathogen among slow-growing Nontuberculous mycobacteria (NTM), and the global incidence of MAC pulmonary disease (MAC-PD) continues to rise steadily. Its intrinsic resistance to conventional anti-mycobacterial agents poses a major therapeutic obstacle. Currently, macrolide-based multidrug regimens remain the standard of care; however, treatment options for macrolide-resistant MAC-PD are critically limited, leading to poor outcomes. Refractory MAC-PD is conventionally defined as persistent sputum culture positivity after at least six month

Real-world effectiveness and lung abnormalities associated with amikacin liposome inhalation suspension.

BACKGROUND: The guideline recommends the addition of amikacin liposome inhalation suspension (ALIS) for patients with refractory Mycobacterium avium complex pulmonary disease (MAC-PD). However, factors influencing culture conversion and managing ALIS-related adverse events, particularly ALIS-related lung abnormalities (ALIS-RLAs) remain insufficient. METHODS: This was a two-center retrospective cohort study involving 87 patients with refractory MAC-PD who received ALIS from August 2021 to September 2023. Refractory MAC-PD was defined as failure to achieve sputum culture conversion after six m

Amikacin sulphate loaded chitosan-diopside nanoparticles composite scaffold for infectious wound healing.

A wound dressing material should inhibit infections that may occur at the wound site, and at the same time, it should enhance the healing process. In this study, we developed an amikacin sulphate (AK) incorporated chitosan (Ch) and Diopside nanoparticles composite dressing (Ch-nDE-AK) for controlling wound infection and healing. The diopside nanoparticles (nDE) were prepared using sol-gel synthesis and characterized using XRD, FT-IR, and FESEM. nDE shows a size range of 142 ± 31 nm through FESEM analysis. Later, the developed composite dressing was characterized using SEM, EDS, and FT-IR analy

M. tuberculosis ferritin (Rv3841): Potential involvement in Amikacin (AK) & Kanamycin (KM) resistance.

Tuberculosis is an infectious disease, caused by one of the most successful human pathogen, Mycobacterium tuberculosis. Aminoglycosides, Amikacin (AK) & Kanamycin (KM) are commonly used to treat drug resistant tuberculosis. They target the protein synthesis machinery by interacting with several steps of translation. Several explanations have been proposed to explain the mechanism of aminoglycoside resistance but still our information is inadequate. Iron storing/interacting proteins were found to be overexpressed in aminoglycosides resistant isolates. Iron assimilation and utilization in M. tub

Mutant APH(2'')-IIa enzymes with increased activity against amikacin and isepamicin.

Directed evolution by random PCR mutagenesis of the gene for the aminoglycoside 2''-IIa phosphotransferase generated R92H/D268N and N196D/D268N mutant enzymes, resulting in elevated levels of resistance to amikacin and isepamicin but not to other aminoglycoside antibiotics. Increases in the activities of the mutant phosphotransferases for isepamicin are the result of decreases in K(m) values, while improved catalytic efficiency for amikacin is the result of both a decrease in K(m) values and an increase in turnover of the antibiotic. Enzymes with R92H, D268N, and D268N single amino acid substi

Postantibiotic effect of amikacin, rifampin, sparfloxacin, clofazimine and clarithromycin against Mycobacterium avium.

Antimycobacterial drugs acting efficiently against Mycobacterium avium complex have in common low MICs and MBC/MIC ratios. The recently reported clinical efficacy of some of the newer drugs is also clearly linked to their pharmacokinetic properties such as higher serum level and/or intracellular concentrations and half-life. In the present investigation, comparative postantibiotic effects (PAEs) of amikacin, rifampin, sparfloxacin, clofazimine and clarithromycin were investigated. Bacteria were exposed to MIC, MIC x 4 and MIC x 8 concentrations of each drug for 2 h, the drug was removed by cen

Chromosomal origin of acetyltransferase AAC(6') specifying amikacin resistance in Serratia marcescens.

Two clinical isolates of Serratia marcescens resistant to aminoglycoside-aminocyclitols and other antibiotics have been examined for aminoglycoside-modifying enzymes. Both strains were amikacin-resistant, and this resistance was mediated by an acetyltransferase AAC(6'). S. marcescens 737 contains a single conjugative plasmid, pUZ 737, of 135 kilobases, which confers resistance to gentamicin and tobramycin by a nucleotidyltransferase, ANT(2"), and to kanamycin, neomycin, butirosin and lividomycin by a phosphotransferase, APH(3'). S. marcescens 1830 does not contain extrachromosomal DNA, and it

Empirical antimicrobial therapy with Timentin plus amikacin in febrile granulocytopenic cancer patients.

Timentin (ticarcillin plus clavulanic acid) plus amikacin was administered as an empirical regimen to 52 febrile granulocytopenic patients with cancer and appeared as effective as the other commonly recommended combinations of antimicrobial agents. A favourable response was observed in 61% of episodes with bacteraemia and in 83% of the episodes without bacteraemia. However, the efficacy in Gram-positive bacteraemia was suboptimal and the emergence of superinfections caused by Gram-positive cocci may represent a clinical challenge.

Population Pharmacokinetics of Amikacin in Non-Neonatal Pediatric Patients: A Scoping Review.

BACKGROUND: Amikacin remains an important agent in managing severe Gram-negative infections in children. However, its clinical use is limited by a narrow therapeutic index and pronounced pharmacokinetic (PK) variability. Population pharmacokinetic (PopPK) modeling offers a framework for individualized dosing, yet evidence in non-neonatal pediatric populations remains fragmented. METHODS: A scoping review was conducted across PubMed, Scopus, and Web of Science (last updated July 2025) to identify PopPK studies evaluating intravenous amikacin in children aged 1 month to 18 years. Amikacin Protection Assay for Quantification and Visualization of Escherichia coli Cell Invasion.

Epithelial cell invasion is a key phenotype for many enteric pathogens, including Salmonella, Shigella, and Vibrio. An emerging invasive pathogen is adherent-invasive Escherichia coli (AIEC), which is implicated in the pathogenesis of inflammatory bowel disease (IBD). AIEC strains lack virulence factors commonly associated with other invasive pathogens, making genetic identification challenging. As a result, AIEC strains are classified phenotypically based on their ability to invade epithelial cells using a field-standard gentamicin protection assay. However, since AIEC strains are often isola

Time-dependent pharmacodynamics of amikacin on Mycobacterium abscessus growth and resistance emergence.

Mycobacterium abscessus pulmonary disease is increasing in prevalence globally, particularly for individuals with cystic fibrosis. These infections are challenging to treat due to a high rate of resistance. Amikacin is critical to treatment, but the development of toxicity, amikacin resistance, and treatment failure are significant challenges. Amikacin has been characterized previously as peak-dependent and extended-interval dosing is commonly used. In our hollow fiber infection model of M. abscessus, amikacin exhibited time-dependent rather than the expected peak-dependent pharmacodynamics. H

Elimination and penetration of amikacin into urine in patients with decreased glomerular filtration rate.

BACKGROUND: Amikacin monotherapy is recommended for urinary tract infection (UTI) treatment with multi-resistant pathogens. Even though amikacin efficacy in the treatment of UTIs is dependent on its urinary concentration, there are no robust data proving that sufficiently high urinary concentration is reached in patients with reduced glomerular filtration rate (GFR). METHODS: A prospective study to monitor amikacin penetration into urine of 70 patients [40 males, median (interquartile range) age 70 (65-79) years] with different levels of glomerular filtration decline, including patients treat
