Decreasing Pasteurization Treatment Efficiency against Amoeba-Grown Legionella pneumophila-Recognized Public Health Risk Factor.

Legionellae are gram-negative bacteria most commonly found in freshwater ecosystems and purpose-built water systems. In humans, the bacterium causes Legionnaires' disease (LD) or a Pontiac fever. In this study, the different waters (drinking water, pool water, cooling towers) in which Legionella pneumophila has been isolated were studied to assess the possible risk of bacterial spreading in the population. The influence of physical and chemical parameters, and interactions with Acanthamoeba castellanii on L. pneumophila, were analyzed by Heterotrophic Plate Count, the Colony-forming units (CFU

Amoeba Predation of Cryptococcus neoformans Results in Pleiotropic Changes to Traits Associated with Virulence.

Amoeboid predators, such as amoebae, are proposed to select for survival traits in soil microbes such as Cryptococcus neoformans; these traits can also function in animal virulence by defeating phagocytic immune cells, such as macrophages. Consistent with this notion, incubation of various fungal species with amoebae enhanced their virulence, but the mechanisms involved are unknown. In this study, we exposed three strains of C. neoformans (1 clinical and 2 environmental) to predation by Acanthamoeba castellanii for prolonged times and then analyzed surviving colonies phenotypically and genetic

Adaptation to an amoeba host drives selection of virulence-associated traits in Vibrio cholerae.

Predation by heterotrophic protists drives the emergence of adaptive traits in bacteria, and often these traits lead to altered interactions with hosts and persistence in the environment. Here we studied adaptation of the cholera pathogen, Vibrio cholerae during long-term co-incubation with the protist host, Acanthamoeba castellanii. We determined phenotypic and genotypic changes associated with long-term intra-amoebal host adaptation and how this impacts pathogen survival and fitness. We showed that adaptation to the amoeba host leads to temporal changes in multiple phenotypic traits in V. ch

Amoeba-inspired Tug-of-War algorithms for exploration-exploitation dilemma in extended Bandit Problem.

The true slime mold Physarum polycephalum, a single-celled amoeboid organism, is capable of efficiently allocating a constant amount of intracellular resource to its pseudopod-like branches that best fit the environment where dynamic light stimuli are applied. Inspired by the resource allocation process, the authors formulated a concurrent search algorithm, called the Tug-of-War (TOW) model, for maximizing the profit in the multi-armed Bandit Problem (BP). A player (gambler) of the BP should decide as quickly and accurately as possible which slot machine to invest in out of the N machines and

Encephalitis due to a free-living amoeba (Balamuthia mandrillaris): case report with literature review.

BACKGROUND: Amebic infections can spread to the central nervous system with a lengthy but usually fatal course. A typical case is presented to raise awareness of this increasingly reported infectious process that may have a more favorable outcome if diagnosed in its early stages. CASE DESCRIPTION: A 38-year-old male presented with an ulcerating 10 x 8 cm mass on his thigh and smaller skin nodules. In less than 6 months seizures developed due to granulomatous lesions of the brain. Biopsies/excisions of the thigh lesion, a subcutaneous nodule, and a brain lesion were performed. He failed to res

Displacement of cell-surface associated calcium inhibits phagocytosis and Ca-ATPase activity in amoeba.

Displacement of calcium from the cell surface region was observed in cells treated with either chlorpromazine or reserpine with chlorotetracycline being used as a calcium-fluorescent probe. The drugs also significantly inhibit the intensity of phagocytosis and Ca-ATPase activity. The possible role of Ca associated with the cell surface region in regulation of both phagocytosis and Ca-ATPase was discussed.

Integrating free-living amoebae into climate-microbiome and One Health frameworks.

Climate change is altering microbial ecosystems at a global scale, yet the role of free-living amoebae within these shifting microbiomes remains incompletely understood. Free-living amoebae occupy soil, water, air, and built environments, where they contribute to microbial diversity, regulate nutrient cycling, and host diverse bacterial, fungal, and viral microorganisms. As temperature, moisture, and land-use patterns change, amoebal behaviour and their associated microbiomes may shift in ways that may affect soil fertility, water quality, microbial network stability, and the persistence of op

Molecular evolution and adaptations of Legionella pneumophila from amoebae hosts to macrophages.

Legionella pneumophila is an environmental bacterium that emerged from a prolonged co-evolution and adaptation to free-living amoebae as the natural hosts. Within these protozoan hosts, L. pneumophila evolved to evade amoebae predation and remodel their vacuoles into endoplasmic reticulum (ER)-derived vacuoles that evade phagosomal-lysosomal fusion. The L. pneumophila-amoebae co-evolution fortuitously has facilitated infection of human alveolar macrophages, resulting in pneumonia known as Legionnaires' Disease. Intracellular replication and host manipulation are orchestrated by the Dot/Icm Typ

Naegleria amoebae seek confinement and crawl persistently through narrow spaces.

The "brain-eating amoeba" Naegleria fowleri dwells in ponds where it normally feeds on bacteria, but if it enters the brain it can cause a deadly infection. To establish infection, N. fowleri must migrate through different environments-along olfactory axons, through openings in the cribriform plate, and within brain tissue-yet how it does so remains unknown. As a model for N. fowleri migration within these environments, we examine how its nonpathogenic relative, Naegleria gruberi, navigates environments of distinct geometries. We show that Naegleria uses both actin-rich protrusions and membran

Novel Antiamoebic Tyrocidine-Derived Peptide against Brain-Eating Amoebae.

Acanthamoeba castellanii (A. castellanii) can cause Acanthamoeba keratitis, a sight-threatening infection, as well as a fatal brain infection termed granulomatous amoebic encephalitis, mostly in immunocompromised individuals. In contrast, Naegleria fowleri (N. fowleri) causes a deadly infection involving the central nervous system, recognized as primary amoebic encephalitis, mainly in individuals partaking in recreational water activities or those with nasal exposure to contaminated water. Worryingly, mortality rates due to these infections are more than 90%, suggesting the need to find altern

Neutrophils vs. amoebas: Immunity against the protozoan parasite Entamoeba histolytica.

Entamoeba histolytica is a protozoan parasite with high prevalence in developing countries, and causes amoebiasis. This disease affects the intestine and the liver, and is the third leading cause of human deaths among parasite infections. E. histolytica infection of the intestine or liver is associated with a strong inflammation characterized by a large number of infiltrating neutrophils. Consequently, several reports suggest that neutrophils play a protective role in amoebiasis. However, other reports indicate that amoebas making direct contact with neutrophils provoke lysis of these leukocyt

A Legionella pneumophila amylase is essential for intracellular replication in human macrophages and amoebae.

Legionella pneumophila invades protozoa with an "accidental" ability to cause pneumonia upon transmission to humans. To support its nutrition during intracellular residence, L. pneumophila relies on host amino acids as the main source of carbon and energy to feed the TCA cycle. Despite the apparent lack of a requirement for glucose for L. pneumophila growth in vitro and intracellularly, the organism contains multiple amylases, which hydrolyze polysaccharides into glucose monomers. Here we describe one predicted putative amylase, LamB, which is uniquely present only in L. pneumophila and L. ste

Prevalence of Acanthamoeba and other naked amoebae in South Florida domestic water.

The purpose was to identify the prevalence of naked amoebae in tap water in south Florida to ascertain the risk of amoebal infections of the cornea in contact lens wearers. Over the course of a 2-year period, water samples were collected from sites throughout Broward, Palm Beach, and Dade counties, Florida. The presence of amoebae in samples was based on an enrichment cultivation method appropriate for Acanthamoeba. Amoebae were identified using diagnostic features discernable by light microscopy. A total of 283 water samples were processed and amoebae were noted in 80 of these. Acanthamoeba w

Evidence that an amoeba acquired a chloroplast by retaining part of an engulfed eukaryotic alga.

Chlorarachniophytes are amoeboid algae with unusual chloroplasts. Instead of the usual two membranes that surround the chloroplasts of plants, green algae, and red algae, the chloroplasts of chlorarachniophytes have four bounding membranes. The extra membranes may reflect an unusual origin of chlorarachniophyte chloroplasts. Rather than inheriting the organelle directly from their ancestors, chlorarachniophytes may have adopted the chloroplast of an algal cell ingested as prey. Parts of the algal cell are postulated to remain within the amoeba as a reduced eukaryotic endosymbiont [Hibberd, D.

[Penetration of amoebae into ganglion cells of the cerebral cortex in primary amoebic meningoencephalitis].

The intracellular penetrating activity of amoebas of Limax-type (vs. Naegleria fowleri) was observed in a previously published case of the PAME with fulminant lethal course in man. This invasion concerned some ganglion cells of the cerebral cortex and - exceptionally - also the Purkinje-cells in the cerebellum. As to the formal genesis, adjacent and penetrating forms were identified in relation to the ganglion cells, and finally invaded neurons containing solitary or - exceptionally - two parasites were noted. This phenomenon of the intraneuronal lesion due to amoebae in PAME is said to be in

Electrophoretic isoenzyme patterns of the pathogenic and non-pathogenic intestinal amoebae of man.

Cultured stocks of Entamoeba hartmanni, Endolimax nana, Iodamoeba buetschlli and Dientamoeba fragilis were compared with the four Entamoeba histolytical groups already described (SARGEAUNT et al., 1978), by the electrophoretic patterns of three enzymes: glucose phosphate isomerase (GPI), phosphoglucomutase (PGM) and L-malate: NADP+ oxidoreductase (oxalacetate-decarboxylating) (ME). All the species were easily distinguished by their characteristic patterns.
