Manta ray-inspired soft swimmers actuated by bistable actuators.

Manta rays exhibit highly agile and complex swimming patterns through undulatory pectoral fin flapping, inspiring the development of efficient and controllable swimming robots. However, integrating high propulsion efficiency, maneuverability, and untethered operation into a single robotic platform remains a grand challenge. Here, we propose a bioinspired bistable flapping soft swimming robot that mimics manta ray motion to address this issue. The flapping wings employ pre-stretched elastomeric matrix composites to achieve bistable shapes, enabling reconfigurable mono/bistable transitions and m

Loss of Cathepsin Z enhances pro-inflammatory macrophage responses and promotes tissue regeneration.

The plasticity of macrophages is well documented, with fundamental roles in modulating inflammation and promoting tissue repair, notably aiming to maintain homeostasis in multicellular organisms. However, the precise factors that regulate their polarization remain poorly understood. Cathepsin Z (CTSZ) encodes an enzyme highly expressed in macrophages and involved in various processes, such as migration, maturation and signal transduction, but its roles in regeneration are not described. Therefore, we used zebrafish models to investigate the roles of ctsz in macrophage polarization and regenera

Transcriptome analysis of the pectoral fin degeneration in half-smooth tongue sole (Cynoglossus semilaevis).

Appendage degeneration is a notable morphological feature of some teleosts with specialized benthic lifestyles. The half-smooth tongue sole (Cynoglossus semilaevis) undergoes severe pectoral fin regression during metamorphosis. However, the molecular basis underlying rapid pectoral fin degeneration remains unclear. Here, we performed time-series transcriptome sequencing on pectoral fins at pre-metamorphosis, metamorphosis peak and post-metamorphosis to characterize the molecular changes associated with pectoral fin degeneration. Transcriptional dynamics and functional enrichment showed that no

A new species of sexually dimorphic and rheophilic ghost knifefish (Apteronotidae: Gymnotiformes) from the Amazon basin.

A new species of ghost knifefish, Apteronotus, is described from high-energy environments in the Rios Mapuera and Trombetas (at Cachoeira Porteira waterfalls), Brazil. X-ray microcomputed tomography (μCT scan) was used to access the internal anatomy of the type series. The new species is distinguished from all congeners by the anteriormost position of the anus, with its posterior margin extending less than one eye diameter beyond the vertical through the caudal limit of the posterior nostril, the low number of anal-fin rays (117-125) and the reduced number of branchiostegal rays (three). A ser

Multiplex real-time PCR assay to detect illegal trade of CITES-listed shark species.

The Convention on International Trade in Endangered Species of Wild Fauna and Flora (CITES) is a multilateral environmental agreement to ensure that the international trade of threatened species is either prohibited (Appendix I listed species) or being conducted legally, sustainably, and transparently (Appendix II listed species). Twelve threatened shark species exploited for their fins, meat, and other products have been listed under CITES Appendix II. Sharks are often traded in high volumes, some of their products are visually indistinguishable, and most importing/exporting nations have limi

Using the whole body as a sucker: combining respiration and feeding with an attached lifestyle in hill stream loaches (Balitoridae, Cypriniformes).

Small fishes living in fast-flowing rivers face a harsh environment as they can easily be swept away by the rapid currents. To survive such circumstances, teleosts evolved a wide variety of attachment mechanisms, based on friction, negative pressure or both. Balitorinae (Balitoridae, Cypriniformes) are exceptional in using their whole body as an adhesive apparatus. We investigated the morphological adaptations of Balitorinae by studying the osteology and myology of four species (Beaufortia leveretti, Sewellia lineolata, Pseudogastromyzon myersi, and Gastromyzon punctulatus) using clearing and

Molas have mighty mitochondria: intensified purifying selection on mtDNA despite presumed "low-energy" lifestyles in ocean sunfishes (family Molidae).

Ocean sunfish (family Molidae) are often perceived as "low-energy" drifters composed largely of connective tissue with noticeably missing fins and even cited as "mistakes" of natural selection. Because locomotory capacity has been identified as a driver of evolution in animal mitochondrial genomes (mtDNA), ocean sunfish are expected to show signatures of relaxed selection on mtDNA compared to closely related, more energetic species. Contrary to expectations, ocean sunfish showed lower dN/dS ratios (the ratio of non-synonymous to synonymous substitution rates) in all mitochondrial-encoded genes

Epithelial sodium channel (ENaC) family: Phylogeny, structure-function, tissue distribution, and associated inherited diseases.

The epithelial sodium channel (ENaC) is composed of three homologous subunits and allows the flow of Na(+) ions across high resistance epithelia, maintaining body salt and water homeostasis. ENaC dependent reabsorption of Na(+) in the kidney tubules regulates extracellular fluid (ECF) volume and blood pressure by modulating osmolarity. In multi-ciliated cells, ENaC is located in cilia and plays an essential role in the regulation of epithelial surface liquid volume necessary for cilial transport of mucus and gametes in the respiratory and reproductive tracts respectively. The subunits that for

reg6 is required for branching morphogenesis during blood vessel regeneration in zebrafish caudal fins.

Postnatal neovascularization is essential for wound healing, cancer progression, and many other physiological functions. However, its genetic mechanism is largely unknown. In this report, we study neovascularization in regenerating adult zebrafish fins using transgenic fish that express EGFP in blood vessel endothelial cells. We first describe the morphogenesis of regenerating vessels in wild-type animals and then the phenotypic analysis of a genetic mutation that disrupts blood vessel regeneration. In wild-type zebrafish caudal fins, amputated blood vessels heal their ends by 24 h postamputat

A novel cell line established from caudal fin tissue of Carassius auratus gibelio is susceptible to cyprinid herpesvirus 2 infection with the induction of apoptosis.

Herpesviral haematopoietic necrosis induced by cyprinid herpesvirus 2 (CyHV-2) in Carassius auratus gibelio has caused huge economic losses in aquaculture. To better characterize this pathogen at molecular and cellular levels, it is necessary to study its pathogenesis in a sustainable cell line. In this study, a new cell line (GiCF) from the caudal fin of Carassius auratus gibelio (C. a. gibelio) was established and characterised. Typical cytopathic effect represented by characteristic morphological changes was induced in the cells at the 7th day post infection with CyHV-2, which was isolated f

The pattern of c-fos activation in the CNS is related to behavior in the mudskipper, Periophthalmus cantonensis.

The effects of three types of behavior on c-fos activation in different brain regions of the mudskipper, Periophthalmus cantonensis, were studied by immunocytochemistry. Animals were divided into four groups: The control group did not undergo any specific treatment. The second group consisted of animals that were agitated for an hour with a glass rod at an irregular speed. The third group contained animals showing aggressive behavior during an hour of monitoring, i.e. documented territorial hostility by raising of dorsal fins and pursuit of intruders. In contrast to these three groups, which h

Modeling lacaziosis lesion progression in common bottlenose dolphins Tursiops truncatus using long-term photographic records.

Lacaziosis (lobomycosis) is a skin disease caused by Lacazia loboi, occurring naturally only in humans and dolphins. Attempts to culture the pathogen in vitro have been unsuccessful, and inoculation studies of lacaziosis development in mice have provided only limited, short-term data on the progression and propagation of L. loboi. The present study used photographic data from long-term photo-identification and health assessment projects to model and quantify the progression of lacaziosis lesions in 3 common bottlenose dolphins Tursiops truncatus from Sarasota Bay, Florida, USA. A conserved Hox9-10 code underlies the early establishment of hind appendage progenitors in zebrafish and the Iberian ribbed newt.

The acquisition of paired appendages was a key innovation in vertebrate evolution. Although HoxA/D paralog groups (PGs) 9-13 play central roles in patterning paired fins and limbs, the evolutionary origin of this paired appendage-patterning system remains unknown. Here, we show that HoxA/D PG9-10 genes are essential for the early establishment of pelvic fin progenitors in zebrafish, and that Hox9-10 function is similarly required for hindlimb progenitor formation in the Iberian ribbed newt. In zebrafish, combined disruption of five HoxA/D PG9-10 genes abolishes the detectable pelvic fin progen

Genetic Reprogramming of Positional Memory in a Regenerating Appendage.

Certain vertebrates such as salamanders and zebrafish are able to regenerate complex tissues (e.g., limbs and fins) with remarkable fidelity. However, how positional information of the missing structure is recalled by appendage stump cells has puzzled researchers for centuries. Here, we report that sizing information for adult zebrafish tailfins is encoded within proliferating blastema cells during a critical period of regeneration. Using a chemical mutagenesis screen, we identified a temperature-sensitive allele of the gene encoding DNA polymerase alpha subunit 2 (pola2) that disrupts fin reg

A new species of Peniculus (Copepoda: Siphonostomatoida) parasitizing mesopelagic myctophid fish: first discovery of colonization of the genus in deep water.

Peniculus hokutoae n. sp. is described on the basis of an ovigerous adult female parasitizing the caudal fin of the myctophid fish Symbolophorus evermanni (Gilbert, 1905), collected from Suruga Bay, Japan. This is the first record of parasitism by this genus on mesopelagic myctophid fish. The new species is easily distinguished from other congeners in: (1) the presence of a conical process anterior to the rostrum; (2) the secondary elongation of the first pedigerous somite; (3) the incorporation of the third and fourth pedigerous somites into the trunk; (4) the unilobate maxillule bearing two

Chronic Perfluorooctanoic Acid Exposure in Young Adult Zebrafish Affects Scale and Internal Skeleton Bone Metabolism, as Well as Offspring Bone Development.

Perfluorooctanoic acid (PFOA) is one of the most extensively investigated per- and polyfluoroalkyl substances (PFAS), a class of highly fluorinated organic compounds widely used in consumer products. While assessing the long-term health effects of chronic PFOA exposure in humans remains challenging, simple animal models like zebrafish serve as essential tools to identify potential toxicological risks. The aim of this study was to investigate the chronic effects of PFOA on bone metabolism in young adult zebrafish, as well as the potential transgenerational impacts on their offspring. Fish fed a

A Zebrafish Model of Human Fibrodysplasia Ossificans Progressiva.

Fibrodysplasia ossificans progressiva (FOP) is a rare, autosomal dominant genetic disorder in humans characterized by explosive inflammatory response to injury leading to gradual ossification within fibrous tissues, including skeletal muscle, tendons, and ligaments. A variety of animal models are needed to study and understand the etiology of human FOP. To address this need, here we present characterizations of the first adult zebrafish model for FOP. In humans, activating mutations in the Type I BMP/TGFβ family member receptor, ACVR1, are associated with FOP. Zebrafish acvr1l, previously know
