BACKGROUND: Despite widespread implementation of malaria control measures, including insecticide-treated nets (ITNs), indoor residual spraying (IRS), and seasonal malaria chemoprevention (SMC), malaria remains a major public health concern in Burkina Faso. The persistence of transmission is often attributed to increasing insecticide resistance in Anopheles gambiae sensu lato and drug resistance in Plasmodium spp. However, additional factors, such as climatic variability, ecological change, and shifts in vector species composition, may also play a role. This study investigated the geographic di

A system for biocontrol of malaria and filarial mosquito vectors has been developed using herbal extracts of three Spilanthes species, S. acmella L.var oleraceae Clarke, S. calva L. and S. paniculata Wall ex DC. Cent percent mortalities was achieved against the late third/early fourth instar larvae of A. stephensi Liston, A. culicifacies species C and C. quinquefasciatus Say using crude hexane extract obtained from flower heads of Spilanthes spp. Of the three plant species, S. acmella extract proved to be the most effective in inducing complete lethality at minimum doses, the respective LC50 a

BACKGROUND: Chromosomal inversion polymorphisms have been associated with adaptive behavioral, physiological, morphological and life history traits in the two main Afrotropical malaria vectors, Anopheles coluzzii and Anopheles gambiae. The understanding of the adaptive value of chromosomal inversion systems is constrained by the feasibility of cytological karyotyping. In recent years in silico and molecular approaches have been developed for the genotyping of most widespread inversions (2La, 2Rb and 2Rc). The 2Ru inversion, spanning roughly 8% of chromosome 2R, is commonly polymorphic in West

MOTIVATION: Insect odorant receptors (iORs) essential for olfaction function as 7-transmembrane-domain (7TM) ion channels. iORs form tetrameric complexes that include a highly conserved odorant receptor co-receptor (Orco). Functional Orco homotetramers have also known to form ex vivo. Blocking Orco ex vivo and in vivo disrupts olfactory signalling, making it a promising target for functional intervention. Understanding its gating behaviour is critical for developing strategies to disrupt odour-guided behaviours in disease-vector insects. Thus, insights into the dynamic gating of the Anopheles

Malaria remains one of the main public health problems in Mauritania, and it is essential to identify the factors that determine the distribution and productivity of Anopheles breeding sites in order to develop more effective control strategies. A longitudinal survey with repeated measurements was conducted in Nouakchott between May 2023 and April 2024, in order to examine the factors influencing the distribution and productivity of Anopheles larval habitats. The larvae were collected by immersion in 60 water points, once a month during the dry season and twice a month during the rainy season,

Insecticide-based interventions such as IRS and LLINs have significantly reduced malaria transmission globally. However, their sustainability is increasingly threatened by insecticide resistance. While insecticide and pesticide use are known resistance drivers, the role of environmental contaminants remains underexplored. This study investigated the impact of environmental contaminants on insecticide resistance by sampling Anopheles larvae from six sites in Ghana, including petroleum spill sites, mining areas and industrial zones. WHO bioassays revealed resistance to clothianidin (Mortality ra

BACKGROUND: Mass endectocide or ectocide treatment of humans or livestock has been suggested as a possible malaria vector control tool. This work provides guidance for in vitro endectocide and ectocide experiments and raises biological points for further evaluation.
METHODS: Three experiments with ivermectin were performed with Anopheles dirus and Anopheles minimus. The first experiment assessed the impact of a sugar diet (10% sucrose, "Sucrose"; multivitamin syrup, "Multivitamin"; or multivitamin syrup followed by 10% sucrose, "Mix") on mosquito mortality following ingestion of a range of iv

Anopheles gambiae is a highly anthropophilic (human-preferring) malaria vector that prefers to blood feed frequently and selectively on humans. This mosquito species exhibits a strong innate olfactory preference to seek out human scent over other animals, and certain humans over others-key behavioral traits with the potential to drive heterogeneity in biting risk and malaria transmission. Here, we describe the application of a large-scale, semi-field system in Zambia for the quantification of An. gambiae olfactory preferences toward whole body odor sourced from individual humans. We detail ste

Despite efforts to minimize the impacts of malaria and reduce the number of primary vectors, malaria has yet to be eliminated in Zambia. Understudied vector species may perpetuate malaria transmission in pre-elimination settings. Anopheles squamosus is one of the most abundantly caught mosquito species in southern Zambia and has previously been found with Plasmodium falciparum sporozoites, a causal agent of human malaria. This species may be a critical vector of malaria transmission, however, there is a lack of genetic information available for An. squamosus. We report the first genome data an

Dosage compensation (DC), a process countering chromosomal imbalance in individuals with heteromorphic sex chromosomes, has been molecularly characterized only in mammals, Caenorhabditis elegans, and fruit flies.1 In Drosophila melanogaster males, it is achieved by an approximately 2-fold hypertranscription of the monosomic X chromosome mediated by the MSL complex.2,3 The complex is not assembled on female X chromosomes because production of its key protein MSL-2 is prevented due to intron retention and inhibition of translation by Sex-lethal, a female-specific protein operating at the top of

In mammals, the serine protease plasmin degrades extracellular proteins during blood clot removal, tissue remodeling, and cell migration. The zymogen plasminogen is activated into plasmin by two serine proteases: tissue-type plasminogen activator (tPA) and urokinase-type plasminogen activator (uPA), a process regulated by plasminogen activator inhibitor 1 (PAI-1), a serine protease inhibitor that specifically inhibits tPA and uPA. Plasmodium gametes and sporozoites use tPA and uPA to activate plasminogen and parasite-bound plasmin degrades extracellular matrices, facilitating parasite motility

Key behaviours, physiologies and gene expressions in Anopheles mosquitoes impact the transmission of Plasmodium. Such mosquito factors are rhythmic to closely follow diel rhythms. Here, we set to explore the impact of the mosquito circadian rhythm on the tripartite interaction between the vector, the parasite and the midgut microbiota, and investigate how this may affect the parasite infection outcomes. We assess Plasmodium falciparum infection prevalence and intensity, as a proxy for gametocyte infectivity, in Anopheles gambiae mosquitoes that received a gametocyte-containing bloodfeed and me

Anopheles stephensi mosquitoes share urban breeding sites with Aedes aegypti and Culex quinquefasciatus mosquitoes in the Republic of Djibouti. We present evidence that A. stephensi mosquitoes might be responsible for an increase in malaria incidence in this country. We also document resistance of Plasmodium falciparum to dihydroartemisinin/piperaquine.

A possible malaria control approach involves the dissemination in mosquitoes of inherited symbiotic microbes to block Plasmodium transmission. However, in the Anopheles gambiae complex, the primary African vectors of malaria, there are limited reports of inherited symbionts that impair transmission. We show that a vertically transmitted microsporidian symbiont (Microsporidia MB) in the An. gambiae complex can impair Plasmodium transmission. Microsporidia MB is present at moderate prevalence in geographically dispersed populations of An. arabiensis in Kenya, localized to the mosquito midgut and

BACKGROUND: Understanding the contribution of outdoor-resting Anopheles mosquitoes to residual malaria transmission is important in terms of scaling up vector control towards malaria elimination in South Africa. The aim of this project was to assess the potential role of Anopheles parensis and other Anopheles species in residual malaria transmission, using sentinel surveillance sites in the uMkhanyakude District of northern KwaZulu-Natal Province.
METHODS: Monthly vector surveillance was conducted at the sentinel sites from January 2017 to May 2018. Outdoor-placed clay pot resting traps were

Plasmodium sporozoites (SPZs) must traverse the mosquito salivary glands (SGs) to reach a new vertebrate host and continue the malaria disease cycle. Although SGs can harbor thousands of sporozoites, only 10 to 100 are deposited into a host during probing. To determine how the SGs might function as a bottleneck in SPZ transmission, we have characterized Anopheles stephensi SGs infected with the rodent malaria parasite Plasmodium berghei using immunofluorescence confocal microscopy. Our analyses corroborate findings from previous electron microscopy studies and provide new insights into the inv

Small laboratory cage trials of non-drive and gene-drive strains of the Asian malaria vector mosquito, Anopheles stephensi, were used to investigate release ratios and other strain properties for their impact on transgene spread during simulated population modification. We evaluated the effects of transgenes on survival, male contributions to next-generation populations, female reproductive success and the impact of accumulation of gene drive-resistant genomic target sites resulting from nonhomologous end-joining (NHEJ) mutagenesis during Cas9, guide RNA-mediated cleavage. The Anopheles hyrcanus group is widely distributed in the Palearctic and Oriental regions. Among the reported members, 25 species have definitive scientific names. Some are determined as vectors of malaria, lymphatic filariasis, and Japanese encephalitis. However, it is extremely difficult to morphologically discriminate the group members. With the development of techniques like crossing experiment, karyotype analysis, and molecular phylogeny, the classification of the Hyrcanus Group has been changed. In this review we update the list of the Hyrcanus Group, including addition of newly discover

BACKGROUND: Mosquito-borne viral diseases cause significant burden in much of the developing world. Although host-virus interactions have been studied extensively in the vertebrate host, little is known about mosquito responses to viral infection. In contrast to mosquitoes of the Aedes and Culex genera, Anopheles gambiae, the principal vector of human malaria, naturally transmits very few arboviruses, the most important of which is O'nyong-nyong virus (ONNV). Here we have investigated the A. gambiae immune response to systemic ONNV infection using forward and reverse genetic approaches.
