A stable transformation platform in pomegranate uncovers PgMYB10 as a key regulator of anthocyanin biosynthesis.

An efficient genetic transformation platform enables functional validation of PgMYB10, identifying it as a master regulator governing anthocyanin biosynthesis in pomegranate. Limited availability of stable genetic transformation systems restricts functional genomics research in pomegranate. Here, we established efficient in vitro regeneration and Agrobacterium tumefaciens-mediated transformation systems for 'Taishanhong' pomegranate using stem segment explants. Optimized medium combinations produced high-frequency regeneration: a 93.3% shoot-induction rate on MS medium with 1.5 mg/L 6-benzylam

Anthocyanin-Rich Berries and Their Bioactive Compounds in Ocular Health: Mechanisms and Therapeutic Potential.

As a global public health challenge, ocular diseases are driven by interconnected mechanisms, including oxidative stress, vascular abnormalities, and neurodegeneration. This review analyzes the multitarget mechanisms of berry-derived bioactive components (anthocyanins, proanthocyanidins, vitamin C, lutein, zeaxanthin, quercetin, and resveratrol) in regulating ocular homeostasis. Beyond antioxidant and anti-inflammatory effects, these compounds delay the progression of age-related macular degeneration, diabetic retinopathy, and glaucoma through mechanisms such as improving microcirculation, pro

Fabrication and characterization of anthocyanin-loaded double Pickering emulsions stabilized by β-cyclodextrin.

Anthocyanins, one of the important water-soluble pigments, are sensitive to environmental factors, which limits the application of anthocyanins in food field. In order to overcome this limitation, double Pickering emulsions stabilized by β-cyclodextrin were developed. The optimum preparation conditions of the emulsions were determined firstly and the performance and structure of emulsions were investigated. Results showed that the optimum preparation conditions of emulsions were the ratio of (W1/O): W2 = 6:4 and 4 % β-cyclodextrin concentration. Optical microscope and confocal laser scanning m

Transcription factors BZR1 and PAP1 cooperate to promote anthocyanin biosynthesis in Arabidopsis shoots.

Anthocyanins play critical roles in protecting plant tissues against diverse stresses. The complicated regulatory networks induced by various environmental factors modulate the homeostatic level of anthocyanins. Here, we show that anthocyanin accumulation is induced by brassinosteroids (BRs) in Arabidopsis (Arabidopsis thaliana) shoots and shed light on the underlying regulatory mechanism. We observed that anthocyanin levels are altered considerably in BR-related mutants, and BRs induce anthocyanin accumulation by upregulating the expression of anthocyanin biosynthetic genes. Our genetic analy

Extraction and characterization of anthocyanin pigments from Iris flowers and metal complex formation.

Exploring the chemical processes and factors influencing the stability of the blue color derived from anthocyanins is a crucial objective in agricultural and food chemistry research. The ability of these compounds to bind with metals could potentially stabilize anthocyanins extracted from plant-based foods or enable modifying their hues for application as natural food colorants. This study had two core objectives - first, to extract and identify the major anthocyanin pigments responsible for iris flower coloration. Second, to selectively complex purified iris anthocyanins with aluminum (Al3+)

Identification and functional analysis of three new anthocyanin R2R3-MYB genes in Petunia.

We identified three novel members of the R2R3-MYB clade of anthocyanin regulators in the genome of the purple flowering Petunia inflata S6 wild accession, and we called them ANTHOCYANIN SYNTHESIS REGULATOR (ASR). Two of these genes, ASR1 and ASR2, are inactivated by two different single base mutations in their coding sequence. All three of these genes are absent in the white flowering species P. axillaris N and P. parodii, in the red flowering P. exserta, and in several Petunia hybrida lines, including R27 and W115. P. violacea and other P. hybrida lines (M1, V30, and W59) instead harbor funct

The Role of AOPP in Age-Related Bone Loss and the Potential Benefits of Berry Anthocyanins.

Age-related bone loss is a major factor in osteoporosis and osteoporotic fractures among the elderly. Because bone homeostasis involves a balance between bone formation and resorption, multiple mechanisms may induce age-dependent changes in bone. Oxidative stress is one such factor that contributes to the pathology of aging-associated osteoporosis (AAO). Advanced oxidation protein products (AOPP) are a biomarker of oxidant-mediated protein damage, and can also act to increase the production of reactive oxygen species (ROS), thereby perpetuating oxidative damage. AOPP is a relatively novel mark

Regulatory mechanisms and multidimensional applications of blueberry anthocyanins: a review of current gaps and future directions.

Blueberries are among the richest fruit sources of anthocyanins, which contribute significantly to fruit quality, flavor, and plant physiology. Anthocyanins also exhibit various health-promoting effects, making them valuable in the food, pharmaceutical, and cosmetic industries. Although the structures of anthocyanins and the general framework of their biosynthesis have been largely elucidated, the precise regulatory mechanisms governing anthocyanin production in blueberries remain incompletely understood. Here, we analyze recent advances in understanding blueberry anthocyanin regulation at the

Anthocyanins Delay D-Galactose-Induced Mouse Liver Aging by Regulating the NF-κB/IKK Signaling Pathway.

Aging is an intricate pathophysiological phenotype. It is the result of the combined action of various inflammatory factors and cytokines. Aging is closely related to inflammation, apoptosis, tumors, and other diseases. Anthocyanins are a kind of natural flavonoid, mainly contained in plant fruits such as bilberry, grape, purple sweet potato, and so on. These flavonoids have antioxidation, antiaging, and anti-inflammatory properties. It has been found that anthocyanins can effectively delay liver, ovary, and other organ aging. However, the biological mechanism by which anthocyanins alleviate a

Preparation and texture assessment of purple red rice bran anthocyanins-rice starch based dysphagia food masses.

The decline in physiological functions caused by aging increases the prevalence of dysphagia. Anthocyanins play a dual role in enhancing the nutrition of the food and influencing its swallowing properties. The objective of this study was to investigate the impact of anthocyanins from purple red rice bran on the viscosity, rheological and textural properties, and IDDSI classification of rice starch-based dysphagia food masses. The results showed that anthocyanins increased the peak viscosity (PV) and contributed to amylose leaching from the food masses. In contrast, the addition of 1% and 2% ant

Lingonberry Anthocyanins Inhibit Hepatic Stellate Cell Activation and Liver Fibrosis via TGFβ/Smad/ERK Signaling Pathway.

Phytochemicals from lingonberry have rich pharmacological value and may play an essential role in treating liver diseases. We investigated the regulatory role of lingonberry anthocyanins (LA) on HSC activation in vitro and liver fibrogenesis in vivo. The viability of HSCs treated with LA was significantly reduced in a dose-dependent manner at the concentration of 25-100 μg/mL, in which the monomers of LA also reduced the proliferation of HSCs via IC50 assay. The inducer transforming growth factor β1 (TGFβ1) and the effector α-smooth muscle actin (α-SMA) of HSC activation were all decreased bot

Phenolic Compounds and Anthocyanins in Legumes and Their Impact on Inflammation, Oxidative Stress, and Metabolism: Comprehensive Review.

Inflammation, oxidative stress, and metabolic diseases are intricately linked in a complex, self-reinforcing relationship. Inflammation can induce oxidative stress, while oxidative stress can trigger inflammatory responses, creating a cycle that contributes to the development and progression of metabolic disorders; in addition, these effects can be observed at systemic and local scales. Both processes lead to cellular damage, mitochondrial dysfunction, and insulin resistance, particularly affecting adipose tissue, the liver, muscles, and the gastrointestinal tract. This results in impaired met

Dual targeting of FSP1 and xCT: Potential mechanism of anthocyanins in alleviating neuronal ferroptosis in vascular dementia.

BACKGROUND: VaD, the second most prevalent type of dementia in the elderly following Alzheimer's disease, is marked by significant cognitive and motor deficits, with few effective treatment options currently available. Ferroptosis, a type of regulated cell death driven by iron-mediated lipid peroxidation, has recently emerged as a key pathological mechanism in the development of VaD. Ferroptosis drives neuronal damage in VaD, making it a promising therapeutic target to reduce neuronal death and preserve cognitive function. Health Risks Associated with a Long-Term Gluten-Free Diet and the Potential Protective Role of Anthocyanins.

Gluten-free diets (GFDs) are gaining popularity worldwide. Originally, GFDs were designed for individuals with gluten intolerance, including conditions such as celiac disease, non-celiac gluten sensitivity, gluten ataxia, or dermatitis herpetiformis. However, many individuals without gluten sensitivity have subsequently adopted this diet, leading to a growing demand for gluten-free food products. Meanwhile, the health risks associated with a long-term gluten-free diet (LTGFD) and the inherent related issues, such as nutrient imbalances, remain uncertain. Scientific studies have indicated that

Starch based hydrogel NPs loaded by anthocyanins might treat glycogen storage at cardiomyopathy in animal fibrotic model.

Many reports have been published recently confirmed the limitation of cargo molecules delivered into the heart. This failure is mostly associated with lymphatic or vascular channels washing or to the immune system recognition. Delivery of anthocyanins by encapsulation may augment it retention in the heart at early time points as the capsules are too large to wash out by lymphatic or venous channels and the physical structure of the capsule may shield the anthocyanins from immunoglobulins and cellular components of the immune system. In the current study, the cardiac dysfunction was induced by
