The prevalence of encephalomyocarditis virus neutralizing antibodies among various human populations.

The prevalence of encephalomyocarditis virus (EMC) antibodies among selected human populations in various regions of the world was determined by the plaque reduction neutralization method. Antibody rates among children ranged from 1.0 to 33.9%, while those among adults varied from 3.2 to 50.6%. No differences between sexes were found in the frequency of EMC infection. The pattern of age-specific antibody rates observed among the study populations suggests that EMC infection occurs primarily during childhood. There appeared to be no association between the presence of EMC antibodies and potenti

Transplacental transfer of anti-SARS-CoV-2 neutralizing antibodies in comparison to other pathogens total antibodies.

BACKGROUNDS: Due to immaturity of their immune system, passive maternal immunization is essential for newborns during their first months of life. Therefore, in the current context of intense circulation of SARS-CoV-2, identifying factors influencing the transfer ratio (TR) of neutralizing antibodies against SARS-CoV-2 (NAb) appears important. METHODS: Our study nested in the COVIPREG cohort (NCT04355234), included mothers who had a SARS-CoV-2 PCR positive during their pregnancy and their newborns. Maternal and neonatal NAb levels were measured with the automated iFlash system. RESULTS: For t

Quantification of Neutralizing Antibodies of Tick-Borne Viruses Using Focus Neutralization Assays.

Tick-borne flaviviruses, such as Powassan virus (POWV) or Deer tick virus (DTV), can elicit robust humoral responses in animal models of disease (Hermanceet al., PLoS Negl Trop Dis 14(6):e0008359, 2020; Stone et al., Cell Rep 38(7):110388, 2022; Reynolds et al., Comp Med 67:232-241, 2017). Several aspects of humoral immunity to tick-borne flaviviruses are unknown; however, convalescent serum can offer partial protection against a POWV challenge naïve immune-competent animals (Stone et al., Cell Rep 38(7):110388, 2022). Accurately quantifying the neutralizing titers of protective antibodies all

Development of a rapid neutralization assay for the detection of neutralizing antibodies against coxsackievirus B1.

Coxsackievirus B1 (CVB1) is a major pathogen that causes viral myocarditis and aseptic meningitis and is implicated as a cause of type 1 diabetes mellitus. The rapid detection of neutralizing antibodies can help in the prevention and diagnosis of viral infection. The traditional cytopathic effect (CPE)-based neutralization assay (Nt-CPE) is time-consuming and labor-intensive. In this study, an efficient neutralization test based on an enzyme-linked immunospot assay and a monoclonal antibody 2E6 against CVB1 (Nt-Elispot) was developed. In this optimal Nt-Elispot, a multiplicity of infection (MO

Advances in Detection Methods for Human Respiratory Syncytial Virus Neutralizing Antibodies.

Human respiratory syncytial virus (HRSV) is a major cause of severe lower respiratory tract infections in infants, young children, and older adults worldwide. With the approval of nirsevimab and HRSV vaccines, accurate measurement of neutralizing antibody levels has become essential for vaccine evaluation, immunization strategy design, and seroepidemiology. The plaque reduction neutralization test (PRNT) remains the gold standard, but it is slow, low-throughput, and requires high biosafety. In recent years, newer methods including focus reduction neutralization testing (FRNT), pseudovirus neut

Broadly Sarbecovirus-Neutralizing Antibodies Induced by Ancestral SARS-CoV-2 Infection.

The COVID-19 pandemic, driven by SARS-CoV-2, continues to challenge global health due to emerging variants and the potential risk posed by related sarbecoviruses. Neutralizing antibodies targeting the spike (S) glycoprotein, particularly the receptor-binding domain (RBD), play a crucial role in viral neutralization and vaccine design. Although broadly neutralizing anti-RBD antibodies have been identified, the nature of cross-reactive humoral responses induced by natural infection with ancestral SARS-CoV-2 strains remains incompletely understood. Here, we isolated 105 S-specific monoclonal anti

A Fulminant Case of Refractory Sarcoidosis Triggered by Infliximab-Neutralizing Antibodies.

Sarcoidosis is a multisystem granulomatous disorder, commonly involving the lungs, heart, and skin. Despite recent dramatic advancements in the field, the exact cause remains an enigma; yet, we believe the pathogenesis represents an interplay between environmental, infectious, and genetic factors. An abnormal T regulatory response, in parallel with an enhanced Th1 response, results in the release of cytokines, including IL-2, IL-12, IL-18, tumor necrosis factor-alpha (TNF-α), and interferon-gamma (IFN-γ), which initiates granuloma formation. Meanwhile, enhanced Th17 production stimulates the J

Screening of Neutralizing Antibodies against FaeG Protein of Enterotoxigenic Escherichia coli.

The misuse of antibiotics in veterinary medicine presents significant challenges, highlighting the need for alternative therapeutic approaches such as antibody drugs. Therefore, it is necessary to explore the application of antibody drugs in veterinary settings to reduce economic losses and health risks. This study focused on targeting the F4ac subtype of the FaeG protein, a key adhesion factor in enterotoxigenic Escherichia coli (ETEC) infections in piglets. By utilizing formaldehyde-inactivated ETEC and a soluble recombinant FaeG (rFaeG) protein, an antibody library against the FaeG protein

HIResist: a database of HIV-1 resistance to broadly neutralizing antibodies.

MOTIVATION: Changing the course of the human immunodeficiency virus type I (HIV-1) pandemic is a high public health priority with approximately 39 million people currently living with HIV-1 (PLWH) and about 1.5 million new infections annually worldwide. Broadly neutralizing antibodies (bnAbs) typically target highly conserved sites on the HIV-1 envelope glycoproteins (Envs), which mediate viral entry, and block the infection of diverse HIV-1 strains. But different mechanisms of HIV-1 resistance to bnAbs prevent robust application of bnAbs for therapeutic and preventive interventions. RESULTS:

Potent neutralizing human monoclonal antibodies protect from Rift Valley fever encephalitis.

Rift Valley fever (RVF) is an emerging arboviral disease affecting both humans and livestock. In humans, RVF displays a spectrum of clinical manifestations, including encephalitis. To date, there are no FDA-approved vaccines or therapeutics for human use, although several are in preclinical development. Few small-animal models of RVF encephalitis exist, further complicating countermeasure assessment. Human mAbs RVFV-140, RVFV-268, and RVFV-379 are recombinant potently neutralizing antibodies that prevent infection by binding the RVFV surface glycoproteins. Previous studies showed that both RVF

Intratumoral co-injection of NK cells and NKG2A-neutralizing monoclonal antibodies.

NK-cell reactivity against cancer is conceivably suppressed in the tumor microenvironment by the interaction of the inhibitory receptor NKG2A with the non-classical MHC-I molecules HLA-E in humans or Qa-1b in mice. We found that intratumoral delivery of NK cells attains significant therapeutic effects only if co-injected with anti-NKG2A and anti-Qa-1b blocking monoclonal antibodies against solid mouse tumor models. Such therapeutic activity was contingent on endogenous CD8 T cells and type-1 conventional dendritic cells (cDC1). Moreover, the anti-tumor effects were enhanced upon combination wi

Complement-dependent mpox-virus-neutralizing antibodies in infected and vaccinated individuals.

Mpox virus (MPXV) caused a multi-country outbreak in non-endemic areas in 2022. Following historic success of smallpox vaccination with vaccinia virus (VACV)-based vaccines, the third generation modified vaccinia Ankara (MVA)-based vaccine was used as prophylaxis for MPXV, but its effectiveness remains poorly characterized. Here, we applied two assays to quantify neutralizing antibodies (NAbs) in sera from control, MPXV-infected, or MVA-vaccinated individuals. Various levels of MVA NAbs were detected after infection, historic smallpox, or recent MVA vaccination. MPXV was minimally sensitive to

Two Akabane virus glycoprotein Gc domains induce neutralizing antibodies in mice.

Akabane virus (AKAV), belonging to the genus Orthobunyavirus and family Peribunyaviridae, causes reproductive and congenital abnormalities in ruminants. Its envelope glycoprotein Gc is a neutralizing antigen, on which at least five distinct antigenic regions have been identified. We attempted to identify the domains using truncated recombinant AKAV Gc proteins expressed in Escherichia coli and monoclonal antibodies (mAbs) with AKAV-neutralizing activity. Dot blot analysis revealed that amino acid positions 1-97 and 189-397 (Gc1-97 and Gc189-397) in the truncated recombinant proteins reacted wi

Multiscale affinity maturation simulations to elicit broadly neutralizing antibodies against HIV.

The design of vaccines against highly mutable pathogens, such as HIV and influenza, requires a detailed understanding of how the adaptive immune system responds to encountering multiple variant antigens (Ags). Here, we describe a multiscale model of B cell receptor (BCR) affinity maturation that employs actual BCR nucleotide sequences and treats BCR/Ag interactions in atomistic detail. We apply the model to simulate the maturation of a broadly neutralizing Ab (bnAb) against HIV. Starting from a germline precursor sequence of the VRC01 anti-HIV Ab, we simulate BCR evolution in response to diffe

Broadly neutralizing antibodies for treatment and prevention of HIV-1 infection.

PURPOSE OF REVIEW: Anti-HIV-1 broadly neutralizing antibodies (bNAbs) are promising agents in the fight against the AIDS epidemic. Multiple bNAbs have been already evaluated in clinical trials with encouraging results. This review discusses the use of bNAbs for the prevention and treatment of HIV-1 infection, focusing on manufactured products that have been evaluated in clinical settings. RECENT FINDINGS: More than 17 bNAbs have been evaluated for safety and pharmacokinetics in humans. The vast majority presented a well tolerated profile and were generally well tolerated. Serum half-life vari

Delineating the mechanism of anti-Lassa virus GPC-A neutralizing antibodies.

Lassa virus (LASV) is the etiologic agent of Lassa Fever, a hemorrhagic disease that is endemic to West Africa. During LASV infection, LASV glycoprotein (GP) engages with multiple host receptors for cell entry. Neutralizing antibodies against GP are rare and principally target quaternary epitopes displayed only on the metastable, pre-fusion conformation of GP. Currently, the structural features of the neutralizing GPC-A antibody competition group are understudied. Structures of two GPC-A antibodies presented here demonstrate that they bind the side of the pre-fusion GP trimer, bridging the GP1

Research progress on neutralizing epitopes and antibodies for the Rabies virus.

Rabies is a zoonotic infectious disease with a high fatality rate. It is caused by a virus in the genus Lyssavirus and is a global public health threat. The rabies virus invades and infects cells mainly via a glycoprotein, which may involve multiple receptors. Neutralizing antibodies against the rabies virus function by blocking the binding of the glycoprotein to a receptor or preventing the membrane fusion process. Vaccination combined with anti-rabies virus neutralizing antibodies is essential for postexposure prophylaxis for category III exposure to the rabies virus. In this review, we disc

Protective neutralizing antibodies from human survivors of Crimean-Congo hemorrhagic fever.

Crimean-Congo hemorrhagic fever virus (CCHFV) is a World Health Organization priority pathogen. CCHFV infections cause a highly lethal hemorrhagic fever for which specific treatments and vaccines are urgently needed. Here, we characterize the human immune response to natural CCHFV infection to identify potent neutralizing monoclonal antibodies (nAbs) targeting the viral glycoprotein. Competition experiments showed that these nAbs bind six distinct antigenic sites in the Gc subunit. These sites were further delineated through mutagenesis and mapped onto a prefusion model of Gc. Pairwise screeni
