We report the identification, cloning, heterologous expression and functional characterization of a novel antifungal peptide named lucimycin from the common green bottle fly Lucilia sericata. The lucimycin cDNA was isolated from a library of genes induced during the innate immune response in L. sericata larvae, which are used as therapeutic maggots. The peptide comprises 77 amino acid residues with a molecular mass of 8.2 kDa and a pI of 6.6. It is predicted to contain a zinc-binding motif and to form a random coil, lacking β-sheets or other secondary structures. Lucimycin was active against f

Olive branch dieback, which is caused mainly by fungal species in Neofusicoccum genus (Botryosphaeriaceae), leads to branch death and irreversible damage. This re-emerging disease in rainfed olive orchards in Spain is linked to crop intensification, the prohibition of the burning of pruning residues and drought. Lepra disease, caused by Phlyctema vagabunda (Desm.) Höhn, is another re-emerging disease associated with shoot dieback in high-density olive orchards. In this study, the behavior of Neofusicoccum mediterraneum Crous, M.J. Wingf. & A.J.L. Phillips, Neofusicoccum parvum (Van Niekerk & C

UNLABELLED: Rock-inhabiting fungi thrive in subaerial oligotrophic environments, such as desert rocks, solar panels, and marble monuments, where organic carbon and nitrogen are scarce. We tested whether the rock-inhabiting fungus Knufia petricola showed a preference regarding nitrogen ([Formula: see text] or [Formula: see text]) and carbon (glucose or sucrose) sources, and whether it was sensitive toward carbon and nitrogen limitation. As this fungus produces the carbon-rich, nitrogen-free 1,8-dihydroxynaphthalene (DHN) melanin, we tested whether a melanin-deficient mutant would be less sensit

Forests are an indispensable resource for human existence in the different regions of the world. Forests are often concentrated in a few parts and vary in size. The most important and extensive green reserve on Earth is the Amazon rainforest. Unfortunately, the forests are continually suffering massive losses due to abiotic stresses and deforestation to obtain other arable lands and microbial diseases, particularly those caused by pathogenic fungi. Pathogenic fungi play an important role in inducing oak decline, which is one of the most relevant forest diseases globally. The pathogens also aff

LysM proteins are widely distributed in fungi of diverse lifestyles, including pathogens of humans and animals. Using bioinformatic methods, LysM proteins have been identified in representative yeasts of medical importance. However, LysM proteins have been found in only four ascomycete yeasts (Candida subhashii, C. tropicalis, Debaryomyces fabryi, and D. hansenii). In contrast, almost all basidiomycete yeasts contain at least one LysM effector but lack subgroup C chitinases. Notably, a unique LysM effector is highly conserved among all the lipid-dependent Malassezia species. The repertoire of

Most of the currently known fungal laccases show their maximum activity under acidic environmental conditions. It is known that a decrease in the activity of a typical laccase at neutral or alkaline pH values is the result of an increase in the binding of the hydroxide anion to the T2/T3 copper center, which prevents the transfer of an electron from the T1 Cu to the trinuclear copper center. However, evolutionary pressure has resolved the existing limitations in the catalytic mechanism of laccase, allowing such enzymes to be functionally active under neutral/alkaline pH conditions, thereby giv

Powdery mildew disease caused by Podosphaerapannosa is the most widespread disease in global cut-rose production, as well as a major disease in garden and pot roses. In this study, the powdery mildew resistance of different wild rose varieties was evaluated. Rose varieties with high resistance and high sensitivity were used for cytological observation and transcriptome and expression profile analyses to study changes at the morphological and molecular levels during the interaction between Rosa multiflora and P. pannosa. There were significant differences in powdery mildew resistance among thre

Fungal phytopathogens require different skills to infect plants and complete their lifecycle. Some proteins in fungi are essential for pathogenesis and their expression is regulated by epigenetic mechanisms via chromatin-remodeling. Macrophomina phaseolina is an important phytopathogenic fungus that leads to considerable losses of different crops, especially during drought conditions. Some biological features of the fungus have been described. However, the epigenetics mechanisms involved in the development and virulence of M. phaseolina have not been fully studied. In this work, chemical inhib

Tasteful morels (Morchella sp.) are one of the most popular mushrooms, both economically and scientifically. Due to their beneficial constituents, they are classified as functional foods. This study focuses on the chemical composition and biological activities of a wild edible mushroom, Morchella steppicola Zerova. The metal composition of this species reveals high levels of biogenic elements. However, according to the levels of iron and cobalt, and calculated health risk indices, this mushroom can be recommended for occasional consumption. Also, M. steppicola is found to be rich in gallic aci

Dematiaceous Fonsecaea monophora is one of the major pathogens of chromoblastomycosis. It has been well established that melanization is catalyzed by the type I polyketide synthase (PKS) in F. monophora. Multidomain protein Type I PKS is encoded by six genes, in which the last enzyme thioesterase (TE) catalyzes the cyclization and releases polyketide. Two PKS genes AYO21_03016 (pks1) and AYO21_10638 have been found in F. monophora and both PKS loci have the same gene arrangement but the TE domain in AYO21_10638 is truncated at 3'- end. TE may be the key enzyme to maintain the function of pks1.

Separate identification keys for females, and for males where known, of the 40 genera of Mymaridae (Hymenoptera: Chalcidoidea) in the Afrotropical region are given. The subgenera of four genera are also included in the key to females. The genera are illustrated with over 300 photographs. The 122 named, valid species reported from the region are catalogued. Reliable host records are reported for 6 genera and 11 species from rearings undertaken in the region. An appendix lists the 27 species from northern Africa and offshore islands (Canary Islands, Madeira) but not recorded in the Afrotropical

Sclerotinia sclerotiorum is a devastating plant pathogen that causes substantial losses in various agricultural crops. Although plants have developed some well-known defense mechanisms against invasive fungi, much remains to be learned about plant responses to fungal pathogens. In this study, we investigated how S. sclerotiorum infection affects plant primary and secondary metabolism in the model plant Arabidopsis thaliana. Our results showed that soluble sugar and amino acid content changed significantly in A. thaliana leaves upon fungal colonization, with a decrease in sucrose and an increas

OBJECTIVE: The mango tree Mangifera indica is known as one of the botanical sources of propolis in Tropical regions. There are two different materials which bees can collect from a mango tree to produce propolis: the resin of the tree bark, and the latex found on the fruits. We performed the study of the chemical profile of mango resin in comparison with propolis in order to clarify its importance as propolis source.
RESULTS: We compared the chemical profiles (by GC-MS analysis of ethanol extracts after silylation) of the resin and samples of propolis: of stingless bees (3 Vietnames, 2 Indone

The detection of muriform cells in direct mycological or anatomopathological examination is considered pathognomonic for chromoblastomycosis. The morphological aspect that these fungal structures acquire were called "Borelli spiders", when associated with hyphae. Reports of this association have been described for decades, initially related to more pathogenic agents of this mycosis. More recent studies have shown aspects related to the host's immunity that participate in this process, as well as an association with a worse disease prognosis. The present study discloses the findings of compleme

BACKGROUND: Sclerotinia stem rot (SSR) caused by Sclerotinia sclerotiorum threatens oilseed rape cultivation, and the emergence of fungicide-resistant strains has led to control failures worldwide. Identifying novel chemical alternatives with different modes of action and high antifungal activities is thus crucial. Herein we evaluated the antifungal effects of 3-(2-pyridyl)methyl-2-(4-chlorphenyl)imino- thiazolidine (PMAS) on S. sclerotiorum to determine its efficacy for SSR management.
RESULTS: PMAS had an inhibitory effect on mycelial growth; the EC50 values were 17.83 and 21.15 μg mL-1 for
