Epididymal St3gal2-mediated α2,3-sialylation regulates murine sperm motility and correlates with N-glycan alterations in human asthenozoospermia.

Sialic acid residues on the sperm surface play crucial roles in sperm maturation, capacitation, and sperm-oocyte interactions. Here, we investigated the dynamic regulation of α2,3-sialylation on the sperm membrane during epididymal maturation in mice, with a focus on the sialyltransferase St3gal2. St3gal2 is highly expressed in the caput epididymis and mediates the progressive accumulation of α2,3-linked sialic acid on the murine sperm surface during epididymal transit. Enzymatic removal of surface α2,3-sialic acid significantly reduced mouse sperm progressive motility, whereas co-incubation o

Reduced eccDNA content in idiopathic asthenozoospermia sperm is associated with compromised DNA repair capacity and elevated DNA damage.

BACKGROUND: Despite representing approximately 30% of male infertility cases, idiopathic asthenozoospermia (iAZS) remains etiologically uncharacterized. Extrachromosomal circular DNA (eccDNA) is mobile and circular DNA outside of linear chromosomes. Although eccDNA has been identified in human sperm, its biogenesis and potential role in iAZS pathogenesis requires further study. METHODS: We enrolled 31 patients with idiopathic asthenozoospermia (iAZS) and 31 healthy controls (normozoospermia, NZS), collecting sperm samples with progressive motility (PR) ranging from 0.3% to 90.9%. Sperm eccDNAs

Proteomic analysis of seminal plasma from asthenozoospermia patients reveals proteins that affect oxidative stress responses and semen quality.

Asthenozoospermia (AS) is a common cause of human male infertility. In one study, more than 80% of the samples from infertile men had reduced sperm motility. Seminal plasma is a mixture of secretions from the testis, epididymis and several male accessory glands, including the prostate, seminal vesicles and Cowper's gland. Studies have shown that seminal plasma contains proteins that are important for sperm motility. To further explore the pathophysiological character of AS, we separated the seminal plasma proteins from AS patients and healthy donors using sodium dodecyl sulfate polyacrylamide

4D-quantitative proteomics signature of asthenozoospermia and identification of extracellular matrix protein 1 as a novel biomarker for sperm motility.

Asthenozoospermia (AZS), diagnosed by reduced sperm motility, is one of the major causes of male infertility. However, AZS has no effective therapeutic treatment and the underlying molecular mechanism remains largely unclear. In this study, state-of-the-art 4D-quantitative proteomics analysis was used to compare the protein profiling between 7 normozoospermic and 11 asthenozoospermic sperm samples. Overall, 4718 proteins were identified and 1430 differential abundant proteins were found in the two groups. The differentially expressed proteins were analyzed by GO and KEGG. The core deregulated

Predictive value of the sperm DNA fragmentation index for low or failed IVF fertilization in men with mild-to-moderate asthenozoospermia.

INTRODUCTION: It has been observed that there is an increased incidence of total fertilization failure (TFF) and a low fertilization rate (LFR, <25 %) during conventional in vitro fertilization (IVF) treatments involving men with poor sperm motility. These men also exhibit a high sperm DNA fragmentation index (DFI), which has adverse effects on various IVF outcomes. However, the relationship between a high DFI and an increased TFF or LFR during IVF cycles has not been elucidated. Here, we aimed to investigate the association between the sperm DFI and TFF or LFR in IVF cycles involving men with

Further studies on knockout mice lacking a functional dynein heavy chain (MDHC7). 2. A developmental explanation for the asthenozoospermia.

Male mice had been previously generated in which the inner dynein arm heavy chain 7 gene (MDHC7) was disrupted. MDHC7-/- animals show asthenozoospermia and are sterile. Very few of their spermatozoa can achieve forward progression, but for those that can, we add here the information (1) that the three-dimensional aspects of their movement are normal; (2) that their maximum velocity is less than that of wild-type controls; and (3) that they are entirely unable to penetrate media of raised viscosity (25-4,000 cP). However, the large majority of the spermatozoa can achieve only a low amplitude vi

FFAR4 Mediates High-Altitude Hypoxia-Induced Asthenozoospermia through Oxidative Stress and Mitochondrial Dysfunction: An Integrative Bioinformatics and Experimental Study.

High-altitude hypoxia is a significant environmental factor contributing to male infertility, particularly asthenozoospermia, yet the underlying molecular mechanisms remain elusive. To address this, we performed an integrated bioinformatics analysis of transcriptome datasets (SRP418387 and SRP418442) to identify candidate genes, followed by the establishment of a high-altitude hypoxia mouse model (simulated 5000 m, 12.5% O₂) for validation. Functional mechanisms were elucidated using AAV-mediated FFAR4 knockdown and pharmacological modulation, alongside comprehensive assessments of sperm motil

Seminal Plasma Exosomal miRNA Profiling Reveals hsa-miR-7-5p as a Key Regulator of Sperm Motility in Asthenozoospermia.

BACKGROUND: Asthenozoospermia is a significant contributing factor to male infertility. Accumulating evidence indicates that impaired sperm motility is closely linked to dysregulated microRNA expression during spermatogenesis. Seminal plasma exosomes are enriched with diverse microRNAs, which play pivotal roles in modulating sperm motility, the acrosome reaction, capacitation, and fertilization. OBJECTIVE: To investigate the association between exosomal microRNA profiles and sperm motility by comparing microRNA expression patterns in seminal plasma exosomes from asthenozoospermic patients and

Identification of a Novel Splice-Site variant in TACR3 (c.888 + 1G > A) Associated with Asthenozoospermia and Hypogonadotropic Hypogonadism in an Iranian Family.

BACKGROUND: TACR3 encodes the receptor for neurokinin B, a key regulator of the hypothalamic-pituitary-gonadal axis. Disruption of this pathway can impair gonadotropin release and male reproductive function. Given the genetic heterogeneity of male infertility, this study aimed to identify novel variants in TACR3 that may underlie asthenozoospermia and related hormonal abnormalities. METHODS: Fifteen infertile men with confirmed asthenozoospermia were enrolled. Whole-exome sequencing (WES) was performed on genomic DNA from peripheral blood, and the candidate variant was validated by Sanger seq

Protective Effect of Ursolic Acid in Prunella vulgaris L. on LPS-Induced Asthenozoospermia via Bcl-2/Bax Apoptosis Signaling Pathway.

BACKGROUND: Asthenozoospermia, also known as lack of sperm motility, accounts for about 27.8% of male infertility as a separate factor, and is often associated with abnormal quantity and morphology of spermatozoa. Therefore, oligozoospermia has become one of the most important factors affecting male infertility. METHODS: Ursolic Acid (UA), also known as wusu acid, is the main active component isolated from Prunella vulgaris L. and has a variety of pharmacological effects. However, the protective effect of UA on asthenozoospermia disease has not been reported. In the current study, the purpose

Blood plasma trimethylamine N-oxide and related metabolites and asthenozoospermia odds: a hospital-based matched case-control study in China.

STUDY QUESTION: Are blood plasma trimethylamine N-oxide (TMAO) and related metabolites linked to the odds of asthenozoospermia? SUMMARY ANSWER: Increased blood plasma TMAO levels were positively associated with the odds of asthenozoospermia, while elevated levels of choline and L-carnitine were related to reduced asthenozoospermia odds, implying that TMAO and its related metabolites might play an important role in the development of asthenozoospermia. WHAT IS KNOWN ALREADY: Sperm motility and concentration are profoundly impaired by excessive reactive oxygen species (ROS). A positive correla
