Diverse progenitor cells preserve salivary gland ductal architecture after radiation-induced damage.

The ductal system of the salivary gland has long been postulated to be resistant to radiation-induced damage, a common side effect incurred by head and neck cancer patients receiving radiotherapy. Yet, whether the ducts are capable of regenerating after genotoxic injury, or whether damage to ductal cells induces lineage plasticity, as has been reported in other organ systems, remains unknown. Here, using the murine salivary gland, we show that two ductal progenitor populations, marked exclusively by KRT14 and KIT, maintain non-overlapping ductal compartments after radiation exposure but do so

The E3 ubiquitin ligase APC/C-Cdh1 coordinates neurogenesis and cortical size during development.

The morphology of the adult brain is the result of a delicate balance between the symmetric divisions to maintain the progenitor cell pool, and the asymmetric divisions to generate a newly differentiated neuron. Neurogenesis is a complex process that relies on an as yet unknown molecular switch that tightly coordinates the cell cycle exit with the start of the differentiation process. The cell cycle length is a key factor that determines the balance between the maintenance of progenitor cells and neuronal differentiation. In fact, neurogenesis in the cerebral cortex is stimulated by lengthenin

PD-1 blockade increases the self-renewal of stem-like CD8 T cells to compensate for their accelerated differentiation into effectors.

PD-1+TCF-1+ stem-like CD8 T cells act as critical resource cells for maintaining T cell immunity in chronic viral infections and cancer. In addition, they provide the proliferative burst of effector CD8 T cells after programmed death protein 1 (PD-1)-directed immunotherapy. However, it is not known whether checkpoint blockade diminishes the number of these stem-like progenitor cells as effector cell differentiation increases. To investigate this, we used the mouse model of chronic lymphocytic choriomeningitis virus (LCMV) infection. Treatment of chronically infected mice with either αPD-1 or α

[Mitochondrial metabolism and the NADPH-cholesterol axis in hematopoietic stem cell fate determination].

Mitochondrial metabolism actively and precisely contributes to hematopoietic stem cell (HSC) fate determination by ensuring proper segregation of mitochondria and by modulating fatty acid oxidation (FAO) activity and mitochondrial NADPH during asymmetric (or symmetric) cell division. Elevated NADPH levels in HSCs promote anabolic processes, notably cholesterol biosynthesis, redirecting acetyl-CoA away from the TCA cycle toward cholesterol production, which in turn supports multiple downstream biosynthetic pathways essential for HSC maintenance. The NADPH-cholesterol axis facilitates the biogen

Targeting proteins to the cell wall of sporulating Bacillus anthracis.

Dormant spores of Bacillus anthracis germinate during host infection and their vegetative growth and dissemination precipitate anthrax disease. Upon host death, bacilli engage a developmental programme to generate infectious spores within carcasses. Hallmark of sporulation in Bacillus spp. is the formation of an asymmetric division septum between mother cell and forespore compartments. We show here that sortase C (SrtC) cleaves the LPNTA sorting signal of BasH and BasI, thereby targeting both polypeptides to the cell wall of sporulating bacilli. Sortase substrates are initially produced in dif

lin-17/Frizzled and lin-18 regulate POP-1/TCF-1 localization and cell type specification during C. elegans vulval development.

The Caenorhabditis elegans vulva is comprised of highly similar anterior and posterior halves that are arranged in a mirror symmetric pattern. The cell lineages that form each half of the vulva are identical, except that they occur in opposite orientations with respect to the anterior/posterior axis. We show that most vulval cell divisions produce sister cells that have asymmetric levels of POP-1 and that the asymmetry has opposite orientations in the two halves of the vulva. We demonstrate that lin-17 (Frizzled type Wnt receptor) and lin-18 (Ryk) regulate the pattern of POP-1 localization and

FtsZ-dependent localization of GroEL protein at possible division sites.

When Escherichia coli is treated with penicillin, the envelopes bulge at the centre of the cells and the cells then lyse. The bulges expand into vesicle-like structures termed penicillin-induced vesicles. We have developed a method to isolate these structures and have shown that they contain mainly membrane proteins plus a high concentration of a 60 kDa protein. The N-terminal amino acid sequence of the protein is identical to that of GroEL protein. Western blotting analysis using anti-GroEL antibody showed that GroEL is indeed concentrated in the vesicles. Indirect immuno-fluorescence microsc

Dynamic centrosomes: functional diversity and remodeling across cell types.

The centrosome is an evolutionarily conserved, microtubule-based organelle that has pronounced cell type-specific functions. In somatic cells, it serves as the primary microtubule-organizing center (MTOC), regulating chromosome segregation and contributing to asymmetric cell division and ciliogenesis. Although traditionally considered structurally stable, increasing evidence indicates that centrosomes can undergo extensive remodeling or even complete elimination in specialized cell types. Recent studies have begun to elucidate the molecular basis of these processes. For example, during oogenes

Cell polarity: cell type-specific regulators, common pathways, and polarized vesicle transport.

Cell polarity, the asymmetric organization of cellular components, is evolutionarily conserved from unicellular and multicellular organisms and is crucial for many biological processes. Polarity is required to maintain cell and tissue integrity by regulating cell division, migration, orientation, cell-cell interactions, and morphogenesis. Impaired polarity leads to dysregulation of cellular functions and is associated with disease. Understanding how polarity is established, maintained, and regulated is thus critical to improving our knowledge of pathologies and devising novel therapies. Here,

Contact area-dependent cell communication and the morphological invariance of ascidian embryogenesis.

Marine invertebrate ascidians display embryonic reproducibility: Their early embryonic cell lineages are considered invariant and are conserved between distantly related species, despite rapid genomic divergence. Here, we address the drivers of this reproducibility. We used light-sheet imaging and automated cell segmentation and tracking procedures to systematically quantify the behavior of individual cells every 2 minutes during Phallusia mammillata embryogenesis. Interindividual reproducibility was observed down to the area of individual cell contacts. We found tight links between the reprod

Dark Side of Escherichia coli Biogenic Inner Membrane: Overabundance of Three Main Phospholipids on Cytoplasmic Leaflet.

Transmembrane (TM) asymmetry of glycerophospholipids in the plasma membrane is well established, yet still a complex and evolving area of study regarding the exact mechanisms, magnitude, and composition. The emerging controversy challenges "Fluid Mosaic Sandwich" model simplicity by combining compositional (different types of lipids on each side) and numerical (different total amounts of phospholipids) asymmetries. While Gram-negative Escherichia coli phosphatidylethanolamine (PE) distribution is asymmetric (mostly cytoplasmic), the TM distribution of phosphatidylglycerol (PG) and cardiolipin

Toxoplasma gondii phosphatidylserine flippase complex ATP2B-CDC50.4 critically participates in microneme exocytosis.

Regulated microneme secretion governs motility, host cell invasion and egress in the obligate intracellular apicomplexans. Intracellular calcium oscillations and phospholipid dynamics critically regulate microneme exocytosis. Despite its importance for the lytic cycle of these parasites, molecular mechanistic details about exocytosis are still missing. Some members of the P4-ATPases act as flippases, changing the phospholipid distribution by translocation from the outer to the inner leaflet of the membrane. Here, the localization and function of the repertoire of P4-ATPases was investigated ac

Genetically Induced Mouse Model for Colon-specific Epithelial Cell Tumorigenesis Driven by Loss of K8 and Apc.

BACKGROUND & AIMS: Loss of keratin 8 (K8) has been shown to increase susceptibility towards colonocyte hyperproliferation and tumorigenesis. However, most colorectal cancer (CRC) mouse models require carcinogen, develop small intestinal tumors, or have a long latency period. The aim was to establish a genetic, colon-specific, and more human-like CRC model driven by loss of K8 and adenomatous polyposis coli (Apc). METHODS: Colon-specific targeting using CDX2P-CreERT2 mice was used to generate K8flox/flox; CDX2P-CreERT2 and K8flox/flox; CDX2P-CreERT2; Apcflox/+ mice. Disease activity was monito

GPSM2 mutations cause the brain malformations and hearing loss in Chudley-McCullough syndrome.

Autosomal-recessive inheritance, severe to profound sensorineural hearing loss, and partial agenesis of the corpus callosum are hallmarks of the clinically well-established Chudley-McCullough syndrome (CMS). Although not always reported in the literature, frontal polymicrogyria and gray matter heterotopia are uniformly present, whereas cerebellar dysplasia, ventriculomegaly, and arachnoid cysts are nearly invariant. Despite these striking brain malformations, individuals with CMS generally do not present with significant neurodevelopmental abnormalities, except for hearing loss. Homozygosity m

Integration of Metabolic Pathways and Micronutrient Signals Influences Neurodevelopment by Modulating Neural Stem Cell Dynamics.

Central nervous system (CNS) development commences in third week of gestation with neural stem cells (NSCs), which, through symmetric division, expand the pool of stem cells and generate diverse types of neuronal and glia cells of CNS via asymmetric division. During neurodevelopment, spatiotemporal coordination is fundamental for appropriate morphogenesis and producing neuronal connections. Besides gene regulatory networks, external and internal factors guide NSCs during self-renewal, fate determination and differentiation. Recent studies indicate metabolism as one of the common converging int

Can Unicellular Organisms Sequester a Germline? The Yeast-Germline Hypothesis.

Germline mutations can affect future generations, while somatic mutations cannot. This germline-soma distinction does not seem to make sense for unicellular organisms. We challenge this view, arguing that baker's yeast (Saccharomyces cerevisiae) has a germline. Under aerobic conditions yeast cells use mainly fermentation of glucose to produce ethanol. Only when glucose is exhausted, cells switch to full respiration of the produced ethanol. We hypothesize that only a subset of the cells continue dividing and switch to respiration. A change from exponential to linear growth is consistent with as

A missense mutation in human INSC causes peripheral neuropathy.

PAR3/INSC/LGN form an evolutionarily conserved complex required for asymmetric cell division in the developing brain, but its post-developmental function and disease relevance in the peripheral nervous system (PNS) remains unknown. We mapped a new locus for axonal Charcot-Marie-Tooth disease (CMT2) and identified a missense mutation c.209 T > G (p.Met70Arg) in the INSC gene. Modeling the INSCM70R variant in Drosophila, we showed that it caused proprioceptive defects in adult flies, leading to gait defects resembling those in CMT2 patients. Cellularly, PAR3/INSC/LGN dysfunction caused tubulin a

Engineering E. coli for Magnetic Control and the Spatial Localization of Functions.

The fast-developing field of synthetic biology enables broad applications of programmed microorganisms including the development of whole-cell biosensors, delivery vehicles for therapeutics, or diagnostic agents. However, the lack of spatial control required for localizing microbial functions could limit their use and induce their dilution leading to ineffective action or dissemination. To overcome this limitation, the integration of magnetic properties into living systems enables a contact-less and orthogonal method for spatiotemporal control. Here, we generated a magnetic-sensing Escherichia

NuMA-related LIN-5, ASPM-1, calmodulin and dynein promote meiotic spindle rotation independently of cortical LIN-5/GPR/Galpha.

The spindle apparatus dictates the plane of cell cleavage, which is critical in the choice between symmetric or asymmetric division. Spindle positioning is controlled by an evolutionarily conserved pathway, which involves LIN-5/GPR-1/2/Galpha in Caenorhabditis elegans, Mud/Pins/Galpha in Drosophila and NuMA/LGN/Galpha in humans. GPR-1/2 and Galpha localize LIN-5 to the cell cortex, which engages dynein and controls the cleavage plane during early mitotic divisions in C. elegans. Here we identify ASPM-1 (abnormal spindle-like, microcephaly-associated) as a novel LIN-5 binding partner. ASPM-1, t
