Endophytic fungi from two isolated Bethencourtia palmensis populations: metabolomic and bioactivity fingerprinting.

BACKGROUND: Bethencourtia palmensis (Nees) Choisy is an endemic plant of the Canary Islands, with different populations on La Palma and Tenerife, that contains biopesticidal compounds and whose endophytic fungi have also shown potential for biopesticide production. This study explores the metabolic and biopesticide potential of endophytic culturable fungi isolated from two island populations of Bethencourtia palmensis [Tenerife (T) and La Palma (P)]. RESULTS: The two plant populations showed distinct chemical profiles: 11β-acetoxy-5α-(angeloyloxy)silphinen-3-one and tetratriacontane were majo

Ecological patterns of the gut mycobiome and microbiome in ulcerative colitis across life stages.

INTRODUCTION: Age-related variations in the gut microbial communities may influence immune regulation and inflammatory processes in inflammatory bowel diseases (IBD). However, distinguishing age effects from differences in clinical characteristics remains challenging. METHODS: We investigated life-stage-associated patterns of the gut microbiome and mycobiome while accounting for clinical heterogeneity between paediatric and adult ulcerative colitis (UC) populations. We analysed 73 targeted metagenomes of bacteria and 69 targeted metagenomes of fungi from 26 paediatric and 47 adult patients wi

Characterization of the gut mycobiome in patients with non-alcoholic fatty liver disease and correlations with serum metabolome.

BACKGROUND: Emerging evidence suggests that the gut microbiome plays a key role in metabolic diseases such as non-alcoholic fatty liver disease, yet the contribution of the gut mycobiome remains largely overlooked. METHODS: We performed a comprehensive analysis of publicly available fecal metagenomic sequencing data and matched serum metabolomic profiles from 90 non-alcoholic fatty liver disease patients and 90 healthy controls. A curated fungal genome database was constructed for taxonomic profiling. We integrated fungal, bacterial, and metabolomic data to assess taxon-specific associations,

Systematic Review of Tinea Nigra: A Clinical Approach.

Tinea nigra (TN) is a superficial fungal infection caused by the melanized fungus Hortaea werneckii, characterized by irregular dark patches, typically on the palms. This systematic review aims to evaluate the epidemiology, demographic characteristics, lesion distribution, diagnostic approaches, causative agents, and treatment outcomes of TN. The PubMed and MEDLINE databases were systematically searched using relevant keywords from January 1990 to January 2025, yielding a total of 102 cases across 42 studies. TN is more prevalent in tropical regions, with a higher incidence in the Americas (64

Multi-omics analysis and the remedial effects of Swertiamarin on hepatic injuries caused by CCl4.

Hepatic diseases pose a significant threat to community health, impacting the quality of life and longevity of millions worldwide. Despite revolutionary advancements in treatment, liver diseases remain a pressing issue, necessitating the development of more effective therapeutic approaches. Here, we conducted a comprehensive multi-omics analysis to investigate the underlying mechanism of Swertiamarin in alleviating hepatic injuries induced by CCl4 in mice. We divided 100 Kunming mice into five groups: RC (control), RM (CCl4), RD (15 mg/Kg Swertiamarin), RZ (30 mg/Kg Swertiamarin), and RG (60 m

Novel symbiotic protoplasts formed by endophytic fungi explain their hidden existence, lifestyle switching, and diversity within the plant kingdom.

Diverse fungi live all or part of their life cycle inside plants as asymptomatic endophytes. While endophytic fungi are increasingly recognized as significant components of plant fitness, it is unclear how they interact with plant cells; why they occur throughout the fungal kingdom; and why they are associated with most fungal lifestyles. Here we evaluate the diversity of endophytic fungi that are able to form novel protoplasts called mycosomes. We found that mycosomes cultured from plants and phylogenetically diverse endophytic fungi have common morphological characteristics, express similar

Contribution of DNA Metabarcoding to the Environmental Fungal Assessments in Hospitals.

Hospitals are particularly sensitive environments where immunosuppressed patients might acquire invasive fungal infections. Therefore, it is necessary to carry out periodical environmental microbiological assessments that evaluate the fungal bioburden in air and surfaces from different hospital zones. Current microbiological monitoring protocols at healthcare settings are mostly based on cultivation, while environmental DNA (eDNA) assessments are still scarce and should be further evaluated. To fill this gap, this study combines a large sampling scheme, comprising > 200 samples (air, surface,

Plant growth-promoting properties of the phosphate-solubilizing red yeast Rhodosporidium paludigenum.

Phosphorus (P) is one of the essential elements that are necessary for plant development and growth. However, the availability of soluble forms of P for plants in the soils is limited, because a large proportion of it is bound to soil constituents. Thus, the concentration of P available to plants at any time is very low and, moreover, its availability depends on the soil pH. As a solution, phosphate-solubilizing microorganisms (PSMs) are employed that render inorganic P available to plants in soluble form. Thus far, research into PSMs has been insufficient, and only few such organisms have bee

Metagenomic analysis of developmental-category-associated fungal communities in the gut contents of Achatina fulica, an intermediate host of Angiostrongylus cantonensis.

BACKGROUND: The invasive giant African snail (Achatina fulica) poses serious agricultural and ecological threats and serves as an important intermediate host of the zoonotic parasite Angiostrongylus cantonensis, which causes human eosinophilic meningitis. While its gut bacterial microbiome has been extensively characterized, the gut mycobiome remains largely uncharacterized. In particular, variations in gut fungal communities across developmental stages remain unknown, limiting our understanding of the A. fulica gut holobiont. METHODS: In November 2025, giant African snails were collected fro

Unveiling the diversity, ecology, and biotechnological potential of culturable marine yeasts in Western Mediterranean coastal ecosystems.

Coastal ecosystems are dynamic environments characterized by strong seasonal variability in physicochemical parameters and biological communities. In the western Mediterranean, the Region of Murcia (southeastern Spain) is characterized by notable biodiversity and ecological heterogeneity, encompassing diverse habitats, including rocky shores, sandy beaches, seagrass meadows, and the Mar Menor, one of Europe's largest hypersaline lagoons. Within these systems, marine yeasts play critical ecological roles in organic matter recycling and produce metabolites of biotechnological and medical interes

Fungal morphotype detection and quantification in microscopic images with TU_MyCo-vision: a user-friendly deep learning object detection tool.

Morphological switching in response to environmental stimuli is a well-known phenomenon in fungi, leading to diverse morphotypes. Microscopic observation remains a widely used approach to study these phenotypes, but variation in sample preparation and operator skill can limit the scale of sample processing or introduce operator bias. Although several image-based cell detection tools have been developed, most are tailored to specific applications or limited to a particular taxon. To address the need for a tool applicable to the polymorphic, yeast-like fungus Aureobasidium pullulans, and with po

Efficacy of N-163 beta-glucan in beneficially improving biomarkers of relevance to muscle function in patients with muscular dystrophies in a pilot clinical study.

BACKGROUND: Muscular dystrophies other than Duchenne muscular dystrophy (DMD) are genetic diseases characterized by increasing muscle weakness, loss of ambulation, and ultimately cardiac and respiratory failure. There are currently no effective therapeutics available. Having demonstrated the efficacy of a N-163 strain of Aureobasidium Pullulans (Neu-REFIX) produced B-1, 3-1,6-Glucan in pre-clinical and clinical studies of Duchenne muscular dystrophy (DMD) earlier, we assessed the effectiveness of this novel Beta glucan in the other muscular dystrophies in the present study. METHODS: In this 6

Microbial quality and nutritional aspects of Norwegian brand waters.

The microbiological quality of the five leading brands of Norwegian bottled still waters was investigated. All brands were free for the enteric indicator organisms and named pathogens whose absence is demanded in current quality directives. The relatively nutrient-poor agar R₂A revealed large heterogeneous bacterial populations which grew slowly, or not at all, on clinical media specified for use in substrate-utilization approaches to identification. The main approach used for identification was cultivation of microbes on R₂A, followed by amplification and partial sequencing of 16S rDNA genes.

Fungal diversity of Norway spruce litter: effects of site conditions and premature leaf fall caused by bark beetle outbreak.

Fungi play an important role in leaf litter decomposition due to their ability to break down the lignocellulose matrix, which other organisms are unable to digest. However, little is known regarding the factors affecting components of fungal diversity. Here, we quantified richness of internal fungi in relation to litter nutrient and phenolic concentrations, sampling season (spring or fall), and premature leaf shedding due to low precipitation and infestation of bark beetles (mainly Ips typographus and Ips duplicatus). The study was conducted in 37-year-old Norway spruce [Picea abies (L.) Karst

Seasonal Dynamics of Culturable Yeasts in Ornithogenically Influenced Soils in a Temperate Forest and Evaluation of Extracellular Enzyme Secretion in Tausonia pullulans at Different Temperatures.

The culturable yeast communities in temperate forest soils under the ornithogenic influence were studied in a seasonal dynamic. To investigate the intense ornithogenic influence, conventional and "live" feeders were used, which were attached to trees in the forest and constantly replenished throughout the year. It was found that the yeast abundance in the soil under strong ornithogenic influence reached the highest values in winter compared to the other seasons and amounted to 4.8 lg (cfu/g). This was almost an order of magnitude higher than the minimum value of yeast abundance in ornithogenic

Analysis of gut microbiota characteristics in osteoarthritis patients.

OBJECTIVE: Osteoarthritis (OA) is a chronic and debilitating disease that represents a leading cause of disability among middle-aged and elderly individuals. With the aging population, the incidence of OA and its associated socioeconomic burden is increasing; however, the underlying pathogenesis remains unclear. Based on the "gut-bone axis" theory, this study systematically characterized the gut microbiota of patients with OA to identify potential biological targets for clinical diagnosis and treatment. METHODS: Fecal samples from 32 OA patients and 26 healthy controls were analyzed for gut b
