Absence of learning and memory deficits in the vasopressin-deficient rat (Brattleboro strain).

Active and passive avoidance behavior was compared between vasopressin (ADH)-deficient rats of the Brattleboro strain and normal Long-Evans rats. In retention of a passive avoidance task across three levels of footshock (0.5, 1.0 and 1.5 mA) the ADH-deficient rats exhibited a slight but overall superior passive avoidance performance. In a one-way avoidance task, diabetes insipidus (D.I.) and normal rats had closely comparable performance; whereas, in a shuttle-box avoidance task the D.I. rats made significantly more avoidance response than the Long-Evans rats. Flinch-jump testing indicated tha

Amphetamine-induced memory impairment in a discriminative avoidance task is state-dependent in mice.

In both humans and laboratory animals, the reports of cognitive effects following acute amphetamine (Amph) administration are mixed and depend, for example, on the timing of administration (e.g. before or after task acquisition) and/or on the memory model used. Besides its cognitive effects, Amph produces other important behavioural effects, including alterations in anxiety and general activity, which could modify the subject's internal state, thereby facilitating state-dependent learning. Importantly, state-dependency has been linked to drug dependence in humans. This study evaluates the role

Ether-induced retrograde amnesia for one-trial conditioning in mice.

Mice were tested in a situation permitting them to step readily from a small, restrictive platform to a larger one. Conditioned avoidance was established by a single training trial when animals received a shock by stepping from one platform to another. At various intervals after the single punishing shock, subjects were anesthetized with diethyl ether. Interference with retention, tested 1 day later, was shown for intervals up to 24 minutes, at which time ether no longer appeared to influence the subsequent response.

Restricted lesions to ventral prefrontal subareas block reversal learning but not visual discrimination learning in rats.

Previous studies have shown that extensive damage to the medial prefrontal cortex (mPFC) of rats causes reversal learning deficits. The mPFC of rats, however, consists of several subareas that are different from each other in both cytoarchitecture and neural connectivity, suggesting a functional dissociation among the mPFC subareas. In the present study, selective lesions of the mPFC of rats were made with a specially designed microknife whose intracranial placement could be controlled stereotaxically. Restricted lesions were made to each of the 3 parts of the mPFC: the anterior cingulate area

Beyond Fear Learning in Isolation: Integration of Social Dimensions into Aversive Learning.

Aversive learning paradigms in laboratory settings have led to valuable decades of research on the acquisition and expression of memories that produce fear and avoidance. However, the majority of these studies have focused on situations in which animals undergo aversive learning while they are in isolation. Although these studies have provided critical insights into the behavioral and neural processes guiding aversive learning, as well as translational insights into neuropsychiatric disorders, how social dimensions that are embedded in naturalistic aversive experiences influence these processe

Avoidance behaviours while circumventing to the left or right of someone with different shoulder widths and facing directions: How do side, width, or orientation matter?

BACKGROUND: Collision avoidance during locomotion is influenced by a variety of situational factors. When circumventing around an inanimate object, the amount of clearance is dependent on the side of avoidance. When avoiding other pedestrians, individuals most often choose to walk behind a moving pedestrian, and avoid people differently depending on their body size. However, side of avoidance has not been evaluated with human obstacles, nor facing direction of a stationary pedestrian, nor the size of a single pedestrian. Therefore, the aim of this study is to evaluate these knowledge gaps conc

I C U but I don't care: Callous-unemotional traits and approach-avoidance behaviour in conduct disorder.

BACKGROUND AND OBJECTIVES: Conduct disorder (CD) is associated with deficits in social-emotional behaviour, such as increased levels of aggression. Callous-unemotional (CU-) traits foster those deficits and contribute to severe rates of instrumental aggression in CD. Previous studies of that increase in aggression have mainly focused on intentional aspects of behaviour. Unintentional behaviour, such as automatic approach and avoidance, has not been taken into account despite being highly relevant for behaviour. Therefore, the relevance of CU-traits for automatic actions and the feasibility of

Light avoidance by a non-ocular photosensing system in the terrestrial slug Limax valentianus.

Although the eye is the best-studied photoreceptive organ in animals, the presence of non-ocular photosensing systems has been reported in numerous animal species. However, most of the roles that non-ocular photosensory systems play remain elusive. We found that the terrestrial slug Limax valentianus avoids light and escapes into dark areas even if it is blinded by the removal of the bilateral superior tentacle. The escape behaviour was more evident for short-wavelength light. Illumination to the head with blue but not red light elicited avoidance behaviour in the blinded slugs. Illumination t

Drug discrimination learning: Interoceptive stimulus control of behavior and its implications for regulated and dysregulated drug intake.

Drug discrimination research has generated rich evidence for the capacity of interoceptive drug stimuli to control behavior by serving as discriminative cues. Owing to its neuropharmacological specificity, drug discrimination learning has been widely used to characterize the stimulus effects and neuropharmacological underpinning of drugs. Apart from such utility, discriminative drug stimuli may help regulate drug use by disambiguating conditioned associations and post-intake outcomes. First, this review summarizes the evidence supporting interoceptive regulation of drug intake from the literat

Aversive social learning with a dead conspecific is achieved by Pavlovian conditioning in crickets.

Social learning, learning from other individuals, has been demonstrated in many animals, including insects, but its detailed neural mechanisms remain virtually unknown. We showed that crickets (Gryllus bimaculatus) exhibit aversive social learning with a dead conspecific. When a learner cricket was trained to observe a dead cricket on a drinking apparatus, the learner avoided the odor of that apparatus thereafter. Here we investigated the hypothesis that this social learning is achieved by first-order Pavlovian conditioning of an odor (conditioned stimulus) and a dead conspecific (unconditione

Halothane anesthesia causes state-dependent retrieval failure in mice.

Effects of exposure to halothane on memory processing was studied using single-trial inhibitory avoidance learning to measure retention. Mice were anesthetized with halothane either before training, immediately after training, or both before training and before testing. Results showed that memory was not impaired by posttraining halothane exposure, indicating that the anesthetic does not cause retrograde amnesia. Mice trained after recovery from halothane showed a robust memory loss 24 h later. This deficit could be alleviated by reexposure to the anesthetic before the retention test. Mice giv

Effects of chronic ingestion and withdrawal of sodium barbitone on learning in rats.

Rats were submitted to three different manipulations: chronic ingestion of sodium barbitone, which was added to the drinking water; chronic administration of barbitone and subsequent withdrawal of the drug; and drinking water only. Both groups of experimental animals showed deficient acquisition in both shuttle-box avoidance and passive-avoidance response when compared to the control animals. Nevertheless, no impairment was observed in passive avoidance when the period of withdrawal was 15 days. Neither was impairment observed when the animals were tested in a T-maze or in another appetitive t

The effects of receptor blockers on brain natriuretic peptide-32-induced action on passive avoidance behavior in rats.

The effects of several doses of porcine brain natriuretic peptide-32 (pBNP-32) administered into the lateral brain ventricle were tested as regards the consolidation of passive avoidance learning in rats. The peptide was found to increase the passive avoidance latency in a dose-dependent manner. In order to clarify which transmitter systems might be involved in the action of pBNP-32, the experimental animals were pretreated with different receptor blockers in selected doses which did not influence the behavioral paradigm. Four of the receptor blockers (haloperidol, atropine, phenoxybenzamine a

Effects of propentofylline on disorder of learning and memory in rodents.

Effects of a newly synthesized xanthine derivative propentofylline (3,7-dihydro-3-methyl-1-(5-oxohexyl)-7-propyl-1H-purine-2,6-dione) on learning and memory of rodents were examined in the two different paradigms. In a shuttle box active avoidance paradigm, propentofylline (25 mg/kg/day, p.o.) improved the decreased learning ability of 12-month-old spontaneously hypertensive rats. Normotensive Wistar-Kyoto rats at a comparable age showed rapid acquisition of avoidance learning, which was not influenced by propentofylline. Step-down passive avoidance task was carried out as the other paradigm.

Avoidance in Anorexia Nervosa: Towards a research agenda.

Anorexia Nervosa is a severe and disabling mental disorder and a huge challenge to treat. Intense fears of e.g., food, eating, weight gain and social evaluation are core features of anorexia nervosa and obstacles during treatment. The perceived threats trigger avoidance and safety behaviors like highly restrictive eating, strict eating rules, vomiting and body checking, to minimize feared outcomes. The role of avoidance in anorexia nervosa is however hardly studied experimentally. In the present article, the focus is on a new transdiagnostic research agenda featuring both basic and clinical ex

Effects of nimodipine, felodipine and amlodipine on electroconvulsive shock-induced amnesia in the rat.

The effects of various doses (0.03, 0.1, 0.3 or 1.0 mg/kg) of the Ca2+ channel blockers nimodipine, felodipine and amlodipine on the learning ability of rats exposed to electroconvulsive shock were examined. The electroconvulsive shock was given immediately after the learning trial response had been acquired. A passive avoidance retention test was performed 24 h later. It was found that electroconvulsive shock strongly impaired the retention of the passive avoidan

Chronic developmental exposure to phenytoin has long-term behavioral consequences.

Anti-epileptic compounds have been linked to several developmental disorders. Specifically, fetal exposure to phenytoin is linked to fetal hydantoin syndrome in humans. We have developed a rat model of fetal hydantoin syndrome in an effort to explore the relationship between drug exposure, development, and learning and memory. Previous studies of this animal model have used various embryological periods of exposure; however, the human syndrome is reported in the offspring of mothers that maintain drug regimens throughout gestation and nursing. To that end, the present study investigated associ
