The effects of neuron morphology and spatial distribution on the selectivity of dorsal root ganglion stimulation.

Objective. For prosthesis users, sensory feedback that appears to come from the missing limb can improve function, confidence, and phantom limb pain. Numerous pre-clinical studies have considered stimulation via penetrating microelectrodes at the dorsal root ganglion (DRG) as a potential approach for somatosensory neuroprostheses. However, to develop clinically translatable neuroprosthetic devices, a less invasive approach, such as stimulation via epineural macroelectrodes, would be preferable. This work explores the feasibility of using such electrodes to deliver focal sensory feedback by exam

Morphological relations between the receptor neurons, sustentacular cells and Schwann cells in the olfactory mucosa of the salamander.

The morphological characteristics of olfactory receptor neurons and their relations to the sustentacular cells and to the sheath cells of Schwann in the olfactory mucosa of the salamander (Ambystoma maculatum et tigrinum) were studied using a modification of the rapid Golgi technique. The bipolar receptor neurons had a fusiform-shaped cell body whose apical pole gave rise to a surface-reaching dendrite and a basal pole which gave rise to an axon. The length and width of the dendrite, although variable, were positively correlated with the relative depth at which the cell body was located in the

Cerebellar basket cell filtering of Purkinje cell responses elicited by low frequency parallel fibre transmission.

Basket cells (BC) are inhibitory interneurons of the cerebellar molecular layer (ML) forming peri-somatic synapses on Purkinje cells (PC). BC physiological and computational properties remained poorly understood and not clearly differentiated from those of stellate cells (SC). We identified BCs in acute mouse cerebellar slices and measured their intrinsic excitability and synaptic responsiveness. BCs and SCs were similar in some respects, although BCs showed stronger and faster synaptic excitation in response to parallel fibre (pf) bursts. The analysis of BC inhibition of PCs was extended over

Mapping spatial organization of in vitro neuronal networks using high-content imaging.

Neuronal network formation is an intricate process by which individual neurons connect into a functional circuitry. At the subcellular level, neuronal connectivity is characterized by the number, size and strength of synapses. At the cellular level, in vitro network characterization remains a challenge due to the large number of neurons involved, spreading widely across a culture dish. Here, we demonstrate a pipeline using high-content confocal microscopy and automated image analysis to study spatial organization of individual neurons in an in vitro cellular network. With this approach, we ena

Cost of auditory sharpness: Model-Based estimate of energy use by auditory brainstem "octopus" neurons.

Octopus cells (OCs) of the mammalian auditory brainstem precisely encode timing of fast transient sounds and tone onsets. Sharp temporal fidelity of OCs relies on low resting membrane resistance, which suggests high energy expenditure on maintaining ion gradients across plasma membrane. We provide a model-based estimate of energy consumption in resting and spiking OCs. Our results predict that a resting OC consumes up to 2.6 × 109 ATP molecules (ATPs) per second which remarkably exceeds energy consumption of other CNS neurons. Glucose usage by all OCs in the rat is nevertheless low due to thei

Increases in collateral axonal growth rostral to a thoracic hemisection in neonatal and weanling rat.

The spinal cords of newborn and weanling rats were hemisected at the mid-thoracic level. Control studies revealed that Fink-Heimer positive debris was absent in the gray matter at three months postoperative. The remaining animals were given a second lesion, a high cervical spinal hemisection, at five to seven months after the original thoracic hemisection. The pattern of degeneration rostal to the thoracic lesion was compared with similar regions of the spinal cord from animals receiving only a cervical hemisection at the adult stage. In neither experimental group of doubly hemisected rats was

Neuropathic Pain Creates an Enduring Prefrontal Cortex Dysfunction Corrected by the Type II Diabetic Drug Metformin But Not by Gabapentin.

Chronic pain patients suffer from pain-related cognitive deficits, even when taking commonly prescribed analgesics. These deficits are likely related to pain-related maladaptive plasticity in the frontal cortex. We sought to model cognitive deficits in mice with neuropathic pain to examine maladaptive morphological plasticity in the mPFC and to assess the effects of several therapeutics. We used an attentional set-shifting task in mice with spared nerve injury (SNI) who received either a single intrathecal injection of an analgesic dose of clonidine, 7 d of 100 mg/kg gabapentin, or 7 d of 200

Sensory neurons selectively upregulate synthesis and transport of the beta III-tubulin protein during axonal regeneration.

The effects of peripheral nerve injury on the content, synthesis, and axonal transport of the class III beta-tubulin protein in adult rat dorsal root ganglion (DRG) neurons were examined. Recent reports of selective increases in the steady-state levels of the beta III-tubulin mRNA during axonal regeneration (Moskowitz et al., 1993) led to the hypothesis that upregulated levels of expression of the beta III-tubulin isotype that alter the composition of neuronal microtubules is important for effective axonal regrowth. If this is the case, the increases in mRNA levels must be translated into incr

Mechanical property changes of glial LC and RGC axons in response to high intraocular pressure.

INTRODUCTION: Pathological high intraocular pressure (IOP) is an important risk factor for glaucoma. The lamina cribrosa (LC) area in the optic nerve head is the initial site of optic nerve injury for glaucoma. LC deformation caused by elevated IOP will compress the retinal ganglion cells (RGC) axons passing through it, thereby leading to the damage of the RGC axons. The deformation of LC is highly correlated with its mechanical properties. Therefore, changes in mechanical properties of LC with the duration of high IOP is of great significance. METHODS: To investigate the impact of chronic hi

Combined pharmacological targeting of NAD+ metabolism preserves central and peripheral axons in an ex vivo model of Wallerian degeneration.

Wallerian degeneration (WD) is a conserved axonal self-destruction program implicated in a spectrum of neurological conditions, including peripheral neuropathies, traumatic brain injury, and possibly neurodegenerative diseases. Wallerian degeneration is triggered by depletion of the NAD+-synthesizing enzyme NMNAT2, subsequent NMN accumulation, and activation of the NAD+-hydrolyzing SARM1, ultimately driving axonal fragmentation. While pharmacological inhibition of SARM1 is an emerging therapeutic strategy, current challenges due to paradoxical toxicity with orthosteric base-exchange SARM1 inhi

Modulation of sciatic nerve expression of class 3 semaphorins by nerve injury.

Beginning with the unexpected finding by cDNA array analysis that neuropilin-2 is induced in sciatic nerve distal to a transection, we document, for the first time, up-regulation in the axotomized adult peripheral nervous system of class 3 semaphorins and their receptors, which are known to play prominent roles in axonal guidance during neural development. Previously, we described the use of cDNA arrays to screen for novel peripheral nervous system axotomy-induced candidate neurotrophic proteins. A novel finding of that prior study was substantial induction of neuropilin 2 (NP2) mRNA in the ax

Peripheral neuropathy after concomitant dimethyl sulfoxide use and sulindac therapy.

The case is presented of a 63-year-old man with a long history of degenerative arthritis who took sulindac (Clinoril) 200 mg BID for 6 months with no untoward effects. Then, without physician knowledge, he began using 90% dimethyl sulfoxide (DMSO) topically to his upper and lower extremities. Shortly thereafter, he developed a profound mixed sensorimotor peripheral neuropathy. Serial electromyographic and nerve conducion studies performed at intervals of several months for 1 year suggested both segmental demyelination and axonal neuropathy. The patient experienced initial deterioration followe

Central demyelination of the Vth nerve root in trigeminal neuralgia associated with vascular compression.

We have examined the ultrastructure of the trigeminal sensory nerve root in three patients with medically intractable trigeminal neuralgia. In one patient, the nerve root was sandwiched between a large vein and a small pontine artery, in the others compression was due to marked dolichoectasia of a verterbal artery. Because these were not amenable to microvascular decompression, a caudal rhizotomy was performed, by excising a short inferior segment of nerve root in the region of indentation. In all cases, examination revealed a zone of chronic demyelination in the proximal (centrally myelinated

The Equine Movement Disorder "Shivers" Is Associated With Selective Cerebellar Purkinje Cell Axonal Degeneration.

"Shivers" is a progressive equine movement disorder of unknown etiology. Clinically, horses with shivers show difficulty walking backward, assume hyperflexed limb postures, and have hind limb tremors during backward movement that resembles shivering. At least initially, forward movements are normal. Given that neither the neurophysiologic nor the pathologic mechanisms of the disease is known, nor has a neuroanatomic locus been identified, we undertook a detailed neuroanatomic and neuropathologic analysis of the complete sensorimotor system in horses with shivers and clinically normal control h

Demonstration of the potential for chronically injured neurons to regenerate axons into intraspinal peripheral nerve grafts.

Experiments were carried out to determine if neurons damaged by injury to the spinal cord retain the ability to regenerate their axonal process for a prolonged period of time after the initial response to injury and if peripheral nerve (PN) grafts could support the regrowth of these processes. True blue (TB) was injected into one side of the adult rat lumbar spinal cord to label neurons with axons coursing through this region. Seven days later spinal cord tissue surrounding the injection sites was removed by aspiration to create a hemisection cavity 3-4 mm in length. Four weeks later scar tiss

Cellular Mechanisms of Transcranial Magnetic Stimulation in Climbing Fibers and Purkinje Neurons in the Cerebellum.

Although transcranial magnetic stimulation (TMS) is widely used for brain stimulation, fundamental issues about its underlying mechanisms remain unresolved. We investigated some of these issues experimentally using an intact isolated turtle cerebellum in vitro, employing a novel chamber designed to deliver precisely calibrated induced electric fields along cortical depth. Our results show that single-pulse TMS can directly activate Purkinje cells and climbing fibers, and synaptically activate Purkinje cells via climbing fibers - all within the first 1.2 ms. Specifically, current source density

AP2A2 mutation and defective endocytosis in a Malian family with hereditary spastic paraplegia.

Hereditary spastic paraplegia (HSP) comprises a large group of neurogenetic disorders characterized by progressive lower extremity spasticity. Neurological evaluation and genetic testing were completed in a Malian family with early-onset HSP. Three children with unaffected consanguineous parents presented with symptoms consistent with childhood-onset complicated HSP. Neurological evaluation found lower limb weakness, spasticity, dysarthria, seizures, and intellectual disability. Brain MRI showed corpus callosum thinning with cortical and spinal cord atrophy, and an EEG detected slow background

Genetic disruption of ankyrin-G in adult mouse forebrain causes cortical synapse alteration and behavior reminiscent of bipolar disorder.

Genome-wide association studies have implicated the ANK3 locus in bipolar disorder, a major human psychotic illness. ANK3 encodes ankyrin-G, which organizes the neuronal axon initial segment (AIS). We generated a mouse model with conditional disruption of ANK3 in pyramidal neurons of the adult forebrain (Ank-G cKO). This resulted in the expected loss of pyramidal neuron AIS voltage-gated sodium and potassium channels. There was also dramatic loss of markers of afferent GABAergic cartridge synapses, resembling the cortical microcircuitry changes in brains from psychotic patients, and suggesting

Immediate anti-tumor necrosis factor-alpha (etanercept) therapy enhances axonal regeneration after sciatic nerve crush.

Peripheral nerve regeneration begins immediately after injury. Understanding the mechanisms by which early modulators of axonal degeneration regulate neurite outgrowth may affect the development of new strategies to promote nerve repair. Tumor necrosis factor-alpha (TNF-alpha) plays a crucial role in the initiation of degenerative cascades after peripheral nerve injury. Here we demonstrate using real-time Taqman quantitative RT-PCR that, during the time course (days 1-60) of sciatic nerve crush, TNF-alpha mRNA expression is induced at 1 day and returned to baseline at 5 days after injury in ne
