Subversion of the Endocytic and Secretory Pathways by Bacterial Effector Proteins.

Intracellular bacteria have developed numerous strategies to hijack host vesicular trafficking pathways to form their unique replicative niches. To promote intracellular replication, the bacteria must interact with host organelles and modulate host signaling pathways to acquire nutrients and membrane for the growing parasitophorous vacuole all while suppressing activation of the immune response. To facilitate host cell subversion, bacterial pathogens use specialized secretion systems to deliver bacterial virulence factors, termed effectors, into the host cell that mimic, agonize, and/or antago

Genome-Guided Dissection of ser and shlB Uncovers Multifactorial Virulence Mechanisms of Serratia marcescens PXG6 in Plutella xylostella.

The rapid evolution of pesticide resistance in Plutella xylostella has reduced the effectiveness of conventional pest control methods in cruciferous crops. This study investigated the virulence of Serratia marcescens PXG6 against P. xylostella and identified two putative virulence factors, ser (serralysin) and shlB (hemolysin transporter). Whole-genome sequencing revealed a GC content of 59.65%, 4650 predicted genes, multiple secretion systems (types I, V, and VI), and 715 putative virulence-related genes. Bioinformatic analyses predicted type IV pili, serralysin, hemolysin, and flagella as pa

Arginine GlcNAcylation of Rab small GTPases by the pathogen Salmonella Typhimurium.

Salmonella enterica serovar Typhimurium, an intracellular Gram-negative bacterial pathogen, employs two type III secretion systems to deliver virulence effector proteins to host cells. One such effector, SseK3, is a Golgi-targeting arginine GlcNAc transferase. Here, we show that SseK3 colocalizes with cis-Golgi via lipid binding. Arg-GlcNAc-omics profiling reveals that SseK3 modifies Rab1 and some phylogenetically related Rab GTPases. These modifications are dependent on C-termini of Rabs but independent of the GTP- or GDP-bound forms. Arginine GlcNAcylation occurs in the switch II region and

The Shigella T3SS needle transmits a signal for MxiC release, which controls secretion of effectors.

Type III secretion systems (T3SSs) are key determinants of virulence in many Gram-negative bacteria, including animal and plant pathogens. They inject 'effector' proteins through a 'needle' protruding from the bacterial surface directly into eukaryotic cells after assembly of a 'translocator' pore in the host plasma membrane. Secretion is a tightly regulated process, which is blocked until physical contact with a host cell takes place. Host cell sensing occurs through a distal needle 'tip complex' and translocators are secreted before effectors. MxiC, a Shigella T3SS substrate, prevents premat

Engineering of Surface Display Systems in E. coli for Recombinant Protein Production.

Isolation of various groups of protein preparations is a crucial task in many areas of biochemical research. Creating an effective, simple and economically feasible system for recombinant protein production is highly valuable for both basic research and applied biotechnology. Most laboratory studies focus on bacterial systems for heterologous gene expression. However, such systems can encounter major difficulties with target proteins that have specific properties. Some proteins can aggregate, form insoluble inclusion bodies or undergo proteolytic cleavage during overproduction. Moreover, they

The Fem cell-surface signaling system is regulated by ExsA in Pseudomonas aeruginosa and affects pathogenicity.

Bacterial interspecies interactions shape microbial communities and influence the progression of polymicrobial infections. FemI-FemR-FemA, a cell-surface signaling system, in Pseudomonas aeruginosa, is involved in the uptake of iron-chelating mycobactin produced by Mycobacterium spp. In this report, we present the data that indicates the femA-PA1909 operon is positively regulated by ExsA, a master regulator for the type three secretion system (T3SS), connecting the Fem system with T3SS. Intriguingly, the Fem system also influenced virulence factors in P. aeruginosa, including the quorum sensin

Brucella pathogenesis, genes identified from random large-scale screens.

Pathogenicity islands, specialized secretion systems, virulence plasmids, fimbriae, pili, adhesins, and toxins are all classical bacterial virulence factors. However, many of these factors, though widespread among bacterial pathogens, are not necessarily found among bacteria that colonize eukaryotic cells in a pathogenic/symbiotic relationship. Bacteria that form these relationships have developed other strategies to infect and grow in their hosts. This is particularly true for Brucella and other members of the class Proteobacteria. Thus far the identification of virulence factors for Brucella

Expression of a gene encoding a scorpion insectotoxin peptide in yeast, bacteria and plants.

The nucleotide sequence encoding the scorpion insectotoxin I5A was chemically synthesized and expressed in yeast, bacteria and tobacco. The I5A peptides produced in these organisms were purified using an immunoaffinity chromatography procedure. I5A produced using the bacterial secretion system was efficiently secreted and released into the culture medium. In contrast, only a trace amount of I5A was detected in bacterial cytosols when expressed from a direct expression vector, suggesting that I5A was unstable in bacterial cells. I5A secreted from yeast using an alpha-factor signal sequence was

Cytolysin A is an intracellularly induced and secreted cytotoxin of typhoidal Salmonella.

Typhoidal Salmonella enterica serovars, such as Typhi and Paratyphi A, cause severe systemic infections, thereby posing a significant threat as human-adapted pathogens. This study focuses on cytolysin A (ClyA), a virulence factor essential for bacterial dissemination within the human body. We show that ClyA is exclusively expressed by intracellular S. Paratyphi A within the Salmonella-containing vacuole (SCV), regulated by the PhoP/Q system and SlyA. ClyA localizes in the bacterial periplasm, suggesting potential secretion. Iron availability and oxygen tension regulate the Yersinia Ysc type III secretion system to enable disseminated infection.

The enteropathogen Yersinia pseudotuberculosis and the related plague agent Y. pestis require the Ysc type III secretion system (T3SS) to subvert phagocyte defense mechanisms and cause disease. Yet type III secretion (T3S) in Yersinia induces growth arrest and innate immune recognition, necessitating tight regulation of the T3SS. Here we show that Y. pseudotuberculosis T3SS expression is kept low under anaerobic, iron-rich conditions, such as those found in the intestinal lumen where the Yersinia T3SS is not required for growth. In contrast, the Yersinia T3SS is expressed under aerobic or anae

Small-molecule type III secretion system inhibitors block assembly of the Shigella type III secreton.

Type III secretion systems (T3SSs) are essential virulence devices for many gram-negative bacteria that are pathogenic for plants, animals, and humans. They serve to translocate virulence effector proteins directly into eukaryotic host cells. T3SSs are composed of a large cytoplasmic bulb and a transmembrane region into which a needle is embedded, protruding above the bacterial surface. The emerging antibiotic resistance of bacterial pathogens urges the development of novel strategies to fight bacterial infections. Therapeutics that rather than kill bacteria only attenuate their virulence may

Immunogenicity of trimeric autotransporter adhesins and their potential as vaccine targets.

The current problem of increasing antibiotic resistance and the resurgence of numerous infections indicate the need for novel vaccination strategies more than ever. In vaccine development, the search for and the selection of adequate vaccine antigens is the first important step. In recent years, bacterial outer membrane proteins have become of major interest, as they are the main proteins interacting with the extracellular environment. Trimeric autotransporter adhesins (TAAs) are important virulence factors in many Gram-negative bacteria, are localised on the bacterial surface, and mediate the

Right on Q: genetics begin to unravel Coxiella burnetii host cell interactions.

Invasion of macrophages and replication within an acidic and degradative phagolysosome-like vacuole are essential for disease pathogenesis by Coxiella burnetii, the bacterial agent of human Q fever. Previous experimental constraints imposed by the obligate intracellular nature of Coxiella limited knowledge of pathogen strategies that promote infection. Fortunately, new genetic tools facilitated by axenic culture now allow allelic exchange and transposon mutagenesis approaches for virulence gene discovery. Phenotypic screens have illuminated the critical importance of Coxiella's type 4B secreti

The type VI secretion system of the emerging pathogen Stenotrophomonas maltophilia complex has antibacterial properties.

Infections with the opportunistic pathogen Stenotrophomonas maltophilia complex can be fatal for immunocompromised patients. The mechanisms used by the bacterium to compete against other prokaryotes are not well understood. We found that the type VI secretion system (T6SS) allows S. maltophilia complex to eliminate other bacteria and contributes to the competitive fitness against a co-infecting isolate. The presence of T6SS genes in isolates across the globe highlights the importance of this apparatus as a weapon in the antibacterial arsenal of S. maltophilia complex. The T6SS may confer survi

Molecular mechanisms of Vibrio parahaemolyticus pathogenesis.

Vibrio parahaemolyticus is a Gram-negative halophilic bacterium that is mainly distributed in the seafood such as fish, shrimps and shellfish throughout the world. V. parahaemolyticus can cause diseases in marine aquaculture, leading to huge economic losses to the aquaculture industry. More importantly, it is also the leading cause of seafood-borne diarrheal disease in humans worldwide. With the development of animal model, next-generation sequencing as well as biochemical and cell biological technologies, deeper understanding of the virulence factors and pathogenic mechanisms of V. parahaemol

Beneath the Bark: Molecular Determinants of Adaptation and Infection in Bacterial Pathogens of Woody Hosts.

Bacterial pathogens of woody plants represent a pervasive yet underexplored threat to global agriculture and forestry, causing devastating diseases in fruit trees, shrubs, forest trees and ornamental species. Unlike pathogens of herbaceous plants, the molecular strategies enabling bacteria to colonise and damage woody tissues remain poorly understood, owing to the experimental challenges associated with woody host systems and the distinctive biology of these plants. Moreover, persistence within long-lived woody tissues requires prolonged modulation of host immune signalling, enabling pathogens

Transcriptomic profiling reveals RetS-mediated regulation of type VI secretion system and host cell responses in Pseudomonas aeruginosa infections.

Pseudomonas aeruginosa is a major opportunistic pathogen that causes chronic infections, particularly in patients with cystic fibrosis and chronic obstructive pulmonary disease (COPD). The type VI secretion system (T6SS) is a primary virulence factor of P. aeruginosa in chronic infections. The objective of this study was to elucidate the regulatory mechanisms and pathogenic effects of the T6SS during P. aeruginosa infection, utilizing transcriptome sequencing and functional assays. We found that T6SS expression is elevated in P. aeruginosa isolated from chronically infected patients. Deletion

Shigella IpaH Family Effectors as a Versatile Model for Studying Pathogenic Bacteria.

Shigella spp. are highly adapted human pathogens that cause bacillary dysentery (shigellosis). Via the type III secretion system (T3SS), Shigella deliver a subset of virulence proteins (effectors) that are responsible for pathogenesis, with functions including pyroptosis, invasion of the epithelial cells, intracellular survival, and evasion of host immune responses. Intriguingly, T3SS effector activity and strategies are not unique to Shigella, but are shared by many other bacterial pathogens, including Salmonella, Yersinia, and enteropathogenic Escherichia coli (EPEC). Therefore, studying Shi
