Transfer of multiple antibiotic resistance between subspecies of Bacteroides fragilis.

Resistance to clindamycin, erythromycin, streptogramins, and tetracycline was shown to be transferable from a clinical isolate of Bacteroides fragilis subspecies distasonis to a sensitive strain of B. fragilis subspecies fragilis. Resistance to clindamycin, erythromycin, and streptogramins was transferable from a clinical isolate of B. fragilis subspecies fragilis to the sensitive strain of B. fragilis subspecies fragilis. Except for tetracycline, resistance to all of these antibiotics was spontaneously curable en bloc at a frequency of approximately 10(-2). These results suggest that resistan

Structural and functional characterisation of the dextran utilisome from Bacteroides thetaiotaomicron.

Bacteroides thetaiotaomicron (B. theta) is a model Bacteroidota of the healthy human gut microbiota and a specialist in glycan utilisation. Like other Bacteroides, B. theta has many highly regulated polysaccharide utilisation loci (PUL) that encode outer membrane (OM) TonB-dependent transporters (SusC), closely associated "lid" lipoproteins (SusD), and additional surface-exposed lipoproteins (SLPs) that bind and partially degrade specific glycans derived from host cells, diet, or other microbiota members. The canonical starch PUL products are thought to form a dynamic complex in the presence o

Synthesis and Docking Studies of Glycolipids Inspired by Bacteroides fragilis Lipid A.

Bacteroides fragilis, a prominent commensal of the human gut microbiota, plays a vital role in immune system regulation through its capsular polysaccharide A (PSA), which requires a glycolipid anchor structurally reminiscent of lipid A. While canonical Escherichia coli lipid A acts as a potent TLR4 agonist contributing to septic shock and inflammatory disorders, certain B. fragilis-derived glycolipids demonstrate antagonistic effects, offering potential as anti-inflammatory agents. In this study, we report the synthesis and preliminary computational evaluation of a library of glycolipids inspi

Comparative genomics reveals the evolution of antimicrobial resistance in Bacteroides nordii.

Bacteroides nordii, is an understudied member of the pathogenic B. fragilis group which comprises several multidrug-resistant (MDR) strains. Thus, it is of great interest to study the genome biology of Bacteroides nordii. However, no detailed study is available that characterized B. nordii at the genetic level and explored its role as a potential pathogen. We isolated an MDR strain viz., B. nordii PGMM4098 from the pus sample and subjected it to whole genome sequencing using Illumina technology. The draft genome was de-novo assembled and annotated, followed by comprehensive comparative genomic

Growth of succinate consumer Dialister hominis is supported by Bacteroides thetaiotaomicron.

This study revealed an interaction between the gut commensal bacterium Bacteroides thetaiotaomicron JCM 5827T and asaccharolytic bacterium Dialister hominis JCM 33369T, which uses succinate instead of carbohydrates for growth. D. hominis usually forms extremely small colonies on Brucella blood agar plates. However, when co-cultured with B. thetaiotaomicron, D. hominis grew noticeably and formed larger colonies than those in the single culture, especially near B. thetaiotaomicron colonies. Although D. hominis barely grew in Gifu anaerobic medium broth, adding 1% succinate improved its growth. I

An update on ampicillin resistance and β-lactamase genes of Bacteroides spp.

Introduction. There are several β-lactamase genes described for Bacteroides strains, of which cepA and cfiA are specific for Bacteroides fragilis and define two genetic divisions. The expression and phenotypic effects of these genes are usually regulated by insertional activation.Hypotheses/Gap Statement. Information is lacking about how cepA is regulated for most of the B. fragilis strains and whether there could be a genetic element for it.Aim. We aimed to investigate the molecular background of ampicillin (and other β-lactam) resistance among Bacteroides strains as mediated mainly by cepA a

A Case of Native Joint Septic Arthritis Caused by Bacteroides fragilis.

BACKGROUND Bacteroides fragilis (B. fragilis) is an uncommon cause of native joint septic arthritis (NJSA). It is an anaerobic gram-negative rod that is normally found in the oral cavity, gastrointestinal tract, genitourinary system, and skin, and thus is rarely isolated from the musculoskeletal system. Risk factors for B. fragilis NJSA include inflammatory arthritis, malignancy, sickle cell disease, and ischemic heart disease. CASE REPORT We discuss a case of a 65-year-old man with coronary artery disease, heart failure, chronic kidney disease, and gout, who presented with 5 days of right kne

The Mechanism of Bacteroides fragilis Toxin Contributes to Colon Cancer Formation.

The Bacteroides fragilis (B. fragilis) produce biofilm for colonisation in the intestinal tract can cause a series of inflammatory reactions due to B. fragilis toxin (BFT) which can lead to chronic intestinal inflammation and tissue injury and play a crucial role leading to colorectal cancer (CRC). The enterotoxigenic B. fragilis (ETBF) forms biofilm and produce toxin and play a role in CRC, whereas the non-toxigenic B. fragilis (NTBF) does not produce toxin. The ETBF triggers the expression of cyclooxygenase (COX)-2 that releases PGE2 for inducing inflammation and control cell proliferation.

Purulent Pericarditis Caused by Bacteroides fragilis: A Rare Complication of Cholangitis.

Bacterial infection is a rare cause of pericarditis especially in the post-antibiotic era. When compared to viral or idiopathic etiologies, purulent pericarditis carries a higher risk for complications. While most cases are due to Staphylococcus aureus, we present a rare case of pericarditis due to Bacteroides fragilis originating from a liver abscess and leading to pericardial effusion. Our case highlights the need to maintain a high clinical suspicion of bacterial infection when patients present with sepsis and have evidence of pericarditis.

Molecular insights into bacteroid development during Rhizobium-legume symbiosis.

Rhizobial soil bacteria can form a symbiosis with legumes in which the bacteria fix atmospheric nitrogen into ammonia that can be utilized by the host. The plant, in turn, supplies the rhizobia with a carbon source. After infecting the host cell, the bacteria differentiate into a distinct bacteroid form, which is able to fix nitrogen. The bacterial BacA protein is essential for bacteroid differentiation in legumes producing nodule-specific cysteine-rich peptides (NCRs), which induce the terminal differentiation of the bacteria into bacteroids. NCRs are antimicrobial peptides similar to mammali

A role for TLR1, TLR2 and NOD2 in cytokine induction by Bacteroides fragilis.

Bacteroides fragilis, an intestinal flora commensal microorganism, is frequently isolated from abscesses and soft tissue infections. This study aimed to identify pattern recognition receptors (PRRs) involved in B. fragilis recognition and to characterize the induced cytokine profile. Human PBMCs were stimulated with heat-killed B. fragilis and cytokine levels were determined by ELISA. Roles of individual PRRs were assessed using specific blockers of receptor signaling pathways and PBMCs carrying single nucleotide polymorphisms of PRR genes. Cell lines expressing human TLR2 or TLR4 were employe

Suppression of blastogenic transformation of lymphocytes by Bacteroides fragilis in vitro and in vivo.

Bacteroides species and Enterobacteriaceae are known to cause synergistic infections. However, the mechanisms behind this synergy are not completely understood. Several authors have shown that Bacteroides species may inhibit the phagocytosis of Enterobacteriaceae by polymorphonuclear leukocytes as well as by macrophages. With the present study we have addressed the question of whether Bacteroides fragilis (BF) is also capable of suppressing specific immune functions. When incubated together with murine lymphocytes, BF significantly inhibited the blastogenic transformation of these cells stimul

Respiration supported nitrogenase activity of isolated Rhizobium meliloti bacteroids.

Bacteroids having a high level of respiration-supported nitrogenase activity were isolated from nitrogen-fixing alfalfa root nodules. Gentle maceration under anaerobic conditions in the presence of sodium succinate and a fatty acid scavenging agent were employed in this method. A large proportion of isolated bacteroids retained a triple membrane structure as shown by transmission electron microscopy. Dicarboxylic acids of the TCA cycle (malate, fumarate, succinate), but not glutamate or aspartate, supported sufficient respiratory activity to supply the nitrogenase system with ATP and reducing

Nationwide study of the susceptibility of the Bacteroides fragilis group in the United States.

A nationwide susceptibility survey of the Bacteroides fragilis group was continued at New England Medical Center in 1983. A total of 555 strains were obtained from eight centers in the United States. In addition to the nine antimicrobial agents studied in the two previous years, three other agents were added to the evaluation: cefamandole, cefuroxime, and cefonicid. The results for the strains tested with the original nine drugs in 1983 were compared with those for 1,292 isolates tested in 1981 and 1982. The most active beta-lactam drug was piperacillin, which had an 8% resistance rate. Cefoxi

Uptake and incorporation of glucose and mannose by whole cells of Bacteroides thetaiotaomicron.

Glucose uptake by whole-cell suspensions of the obligate anaerobe Bacteroides thetaiotaomicron was two- to fourfold higher under aerobic conditions than during incubation under atmospheres of N(2) or H(2) gas. The O(2)-stimulated uptake activity was lost rapidly (>70% in 5 h) when cell suspensions were incubated aerobically, but this loss was prevented by the addition of crude catalase. Catalase had no apparent effect on cell viability during these incubations. Glucose uptake activity was strongly inhibited by a 10-fold excess of mannose or galactose but not by methyl-alpha-d-glucoside, fructo

In vivo protection of Fusobacterium necrophorum from penicillin by Bacteroides fragilis.

A mixed infection of Bacteroides fragilis and Fusobacterium necrophorum was resistant to treatment with penicillin even though a pure F. necrophorum infection could be successfully treated with penicillin. Since B. fragilis alone did not produce infection, these results indicate that B. fragilis can protect F. necrophorum from penicillin in vivo. The extent of protection afforded by a strain of B. fragilis was related to its level of resistance to penicillin. Only a few cells of B. fragilis were required in the initial bacterial injection. Moreover, protection was demonstrated when B. fragilis

Bacteroides thetaiotaomicron Improves Colorectal Cancer Prognosis by Remodeling the Immune Microenvironment.

BACKGROUND: Colorectal cancer (CRC) progression is closely associated with gut microbiota. Bacteroides thetaiotaomicron (B.theta), a key species in the human cecal and colonic microbiota, plays an incompletely defined role in CRC progression. METHODS: CRC animal models were established utilizing immunocompetent C57BL/6 mice and immunodeficient BALB/C nude mice. Histopathological changes were assessed by H&E staining. Immune cell infiltration was evaluated by IHC. CD8+ T cells and cancer cells were treated with B.theta culture supernatant. Cytokine secretion was measured by ELISA. Surface prot
