The effects of atropine on the tolerance and the convulsions seen after withdrawal from forced barbital drinking in the rat.

Male rats were forced to drink a barbital solution as their only drinking fluid for 33 weeks. During the last part of the treatment the average dose of barbital was around 200 mg/kg/day. In the abstinence period after barbital treatment, tolerance was recorded with a hexobarbital anaesthesia threshold utilizing an EEG criterium. Convulsions were recorded in jiggle cages. On days 3 and 28 of the abstinence period, i.p. pretreatment with atropine (2, 4, and 8 mg/kg) was given 1.5 h before threshold determinations. The barbital treatment induced a clear tolerance to the hexobarbital, which still

pH-dependency in diffusion of some weakly acidic drugs, warfarin, sulfaethidole, and barbital, into organic phases.

The diffusion of three weakly acidic drugs (warfarin, sulfaethidole, and barbital) in cyclohexane-buffer and n-octanol-buffer systems was studied by two different methods; shaking and model cell type of experimentation. At low pH values the drugs moved generally more readily into the organic phases than at higher pH. Moreover, when the shaking method was used the drugs moved readily to the n-octanol phase even at higher pH levels. Lecithin improved the diffusion at low more than at high pH levels. Thus, the results varied according to the organic phase selected and the method used. Poor correl

Learning deficits in aged rats pretreated chronically with barbital and tested late in abstinence: alleviation by tetrahydroaminoacridine.

Physostigmine and tetrahydroaminoacridine (THA) have been reported to improve cognitive function in patients with Alzheimer's disease. Two experiments were conducted to examine the effects of these anticholinesterase agents on learning in aged rats pretreated chronically with barbital. In the first experiment animals received barbital in their drinking water for 46 weeks. Controls were given only water. On days 100-104 of abstinence, when the animals were 20 months old, acquisition of the Morris maze task was initiated after treatment with physostigmine. It was found that physostigmine improve

Factors involved in the differential response to ethanol, barbital and pentobarbital in rats selectively bred for ethanol sensitivity.

Two lines of rats, 'least affected' (LA) and 'most affected' (MA), had been selectively bred for their differential sensitivity to ethanol. Both males and females of the LA strain were observed to be less sensitive than their MA counterparts to the acute hypnotic and motor-impairing effects of ethanol. However, a lower ethanol metabolic rate of the MA males suggests that both CNS and metabolic factors contribute to their enhanced sensitivity to ethanol. By contrast, no differences were observed between the LA and MA males with respect to the hypnotic and subhypnotic effects of pentobarbital or

Studies on acute tolerance to ethanol in the abstinence period after long-term barbital treatment in the rat.

Male rats were treated with sodium barbital in the drinking water for 18 weeks. The average weekly dosage was around 215 mg/kg in groups 1, 3 and 5. Groups 2, 4 and 6 were controls. Every week in the abstinence period after the barbital treatment the sensitivity to hexobarbital was tested with a threshold method after three different pretreatments. These pretreatments consisted of saline (groups 1 and 2) and ethanol (2.0 g/kg intraperitoneally) given either 0.25 hr (groups 3 and 4) or 2.5--3 hrs (group 5 and 6) prior to the hexobarbital test. Tolerance seen as a difference in the sensitivity t

Long-term effects of barbital on spontaneous activity of rats trained to use the drug as a discriminative stimulus.

This investigation sought to determine if the discriminative stimulus properties of barbital are reflected in the pattern of spontaneous motor activity induced by the drug. Rats were trained in a Skinner box to discriminate the effects of sodium barbital (80 mg/kg), injected 60 min prior to training, from those of saline. Half of the animals (Group I) were taught the drug discrimination by rewarding them for bar pressing only when they were in the drug condition. The other half of the animals (Group II) were rewarded only in the absence of the effects of barbital. Spontaneous motor activity wa

Subepithelial capillary blood flow estimated from blood-to-lumen flux of barbital in ileum of rats.

The outflux of barbital from the blood into the small intestine perfused with an isosmotic buffer of pH 9.5 in anesthetized rats was measured to determine the subepithelial capillary blood flow in the gut. It was shown that barbital clearance is blood flow limited when total blood flow to the small intestine varied between 0.3 and 1.3 ml/min per g wet gut. The barbital clearance amounted to an average of 50.3% of the total blood flow. The total mucosal blood flow determined by the use of the distribution of microspheres in the layers of gut wall was 62.76% of the total blood flow. It is conclu

Effects of withdrawal from long-term barbital treatment on open-field behaviour and seizure susceptibility of rats.

Rats withdrawn from long-term barbital treatment were tested both for their activity in an open-field and for their sensitivity to convulsant stimuli. Barbital withdrawal induced a significant increase on locomotion and rearing frequency. Drug removal also made the rats more sensitive to convulsions elicited by sound and maximal electroshock. A positive correlation was found between locomotion plus rearing frequency and audiogenic-induced seizures 3, 4 and 5 days after barbital withdrawal. The results show that exploratory behaviour evaluated through the open-field method can be used as a reli

Age-dependence of succinate dehydrogenase (SDH) in rat liver and its inducibility after barbital and phenylbutazone pretreatment.

SDH activity in liver homogenates increases threetimes from one day before to one day after birth. The plateau till the 13th day of life is followed by a minimum on the 15th day of life, thereafter the increase leads to a maximum on the 120th day. The influence of phenylbutazone and barbital on body and liver weight as well as on hepatic SDH activity was investigated in different age groups. Both drugs were administered in three single daily doses, phenylbutazone additionally 31 days long. Hepatic SDH activity based on liver wet weight or liver protein did not change in any of the experimental

Effect of long-term forced oral barbital administration on endogenous acetylcholine in different regions of rat brain.

Rats received a solution of sodium barbital as their only drinking fluid for 25 and 30 weeks. Four groups were studied: (1) control; (2) barbital until sacrifice; (3) barbital withheld (abstinent) for 3 days; (4) abstinent for 12 days. Abstinence convulsions in groups 3 and 4 were recorded with jiggle cages. The rats were killed by decapitation and the concentration of acetylcholine (ACh) was measured in 3 parts of the brain: striatum, hippocampus + cerebral cortex, cerebellum + medulla oblongata + midbrain. In animals receiving barbital until sacrifice, no significant change in ACh content wa

Initiation by nickel acetate and promotion by sodium barbital of renal cortical epithelial tumors in male F344 rats.

Soluble nickel(II) ion, given to male F344/NCr rats as a single i.p. injection of nickel(II) acetate tetrahydrate at a dose of 90 mumols/kg body weight at 5 weeks of age, proved an effective initiator of renal cortical epithelial tumors. The tumors were revealed by subsequent dosing with the known renal tumor promoter, sodium barbital (5,5-diethylbarbituric acid, sodium salt) dissolved in drinking water at a concentration of 500 p.p.m. Only one rat given the nickel injection without subsequent promotion developed a single renal cortical adenoma, while multiple tumors were common in nickel(II)

Increase of low-Km NADPH-dependent aldehyde reductase activity in the brain by chronic treatment of rats with barbital.

Kinetics studies showed two apparent Km values for p-nitrobenzaldehyde. The high-Km aldehyde reductase (ALR) had a lower Ki value for barbital than the low-Km ALR. The value for maximal velocity (Vmax) for the high-Km ALR was not changed by chronic barbital treatment. However, the Vmax value for the low-Km ALR was significantly increased by chronic barbital treatment. These results suggest that during barbital treatment low-Km ALR which is less sensitive to barbital is induced in compensation for the decreased aldehyde metabolizing capacity of brain.
