Identification of hormone-producing cells of the endocrine pancreas of the sea bass, Dicentrarchus labrax, by ultrastructural immunocytochemistry.

The hormone-producing cells of the endocrine pancreas of the sea bass Dicentrarchus labrax have been identified by ultrastructural immunocytochemistry. The glucagon cells have "clear" cytoplasm and contain characteristic electron-dense polygonal granules surrounded by a "halo" of electron-lucent material. The insulin cells have numerous, tightly packed, electron-dense granules that are almost twice as large as the peripherally located granules of the somatostatin cells. The pancreatic polypeptide cells have granules with variable electron density. When specific antisera are applied in the pero

Oral immunization of largemouth bass with recombinant Bacillus methylotrophicus expressing LMBV MCP induces protective immune responses.

Viral diseases have emerged as a critical bottleneck in the sustainable aquaculture of largemouth bass (Micropterus salmoides). Among these, largemouth bass virus (LMBV) has caused recurrent epizootics characterized by acute mortality and substantial economic losses, yet no licensed vaccine or effective antiviral treatment is currently available. To address this challenge, a probiotic Bacillus methylotrophicus strain with antagonistic activity against Aeromonas hydrophila was engineered as an oral vaccine carrier to express the major capsid protein (MCP) of LMBV. The recombinant B. methylotrop

Pathologic and immunohistochemical findings in sea bass (Dicentrarchus labrax) naturally infected with Aeromonas veronii biovar veronii.

Tissue samples from European sea bass (Dicentrarchus labrax) naturally infected with Aeromonas veronii biovar veronii were subjected to bacteriological, molecular, pathological, and immunohistochemical analyses. The fish were sourced from aquaculture farms in the Aegean region of Turkiye. Clinical and postmortem examinations revealed granuloma formation as the most prominent lesion, primarily affecting the skin, spleen, kidneys, and liver. Sequencing of the 16S rRNA gene from all isolates showed 96-98% identity with A. veronii biovar veronii in the BLASTN 2.13.0+ database. Histopathological an

Aloe-emodin provides dual protection against largemouth bass virus via antiviral activity and host immune enhancement.

Largemouth bass virus (LMBV) is a common pathogen in aquaculture, and with no approved therapies to curb its transmission or mortality, it has caused substantial economic losses to the largemouth bass farming industry. Here, we demonstrate that aloe-emodin (AE), a natural anthraquinone, exhibits potent antiviral activity against LMBV in vitro and in vivo. In vitro, AE (10 mg/L) suppressed viral replication by 91.8 % (p < 0.01), significantly reducing viral titers from 105.92 (96 hpi.) to 104.17 (96 hpi.) TCID50/0.1 mL. AE alleviated the cytopathic effect (CPE) of LMBV infection, maintaining cy

Co-modulation of Liver Genes and Intestinal Microbiome of Largemouth Bass Larvae (Micropterus salmoides) During Weaning.

In recent years, largemouth bass have become one of the most commonly aquacultured species in China, however, its low survival rate during larval weaning has always been a bottleneck that has restricted industrial development. Understanding the changes in liver metabolism and intestinal microflora during the weaning of largemouth bass larvae can help to design better weaning strategies and improve survival. In this study, liver mRNA and intestinal microflora 16S rRNA genes were analyzed using high-throughput sequencing at the pre, mid, and post weaning stages [15, 30, 45 days post hatching; to

Role of axial muscles in powering mouth expansion during suction feeding in largemouth bass (Micropterus salmoides).

Suction-feeding fishes capture food by fast and forceful expansion of the mouth cavity, and axial muscles probably provide substantial power for this feeding behavior. Dorsal expansion of the mouth cavity can only be powered by the epaxial muscles, but both the sternohyoid, shortening against an immobile pectoral girdle to retract the hyoid, and the hypaxial muscles, shortening to retract both the pectoral girdle and hyoid, could contribute ventral expansion power. To determine whether hypaxial muscles generate power for ventral expansion, and the rostrocaudal extent of axial muscle shortening

Differential expression of largemouth bass (Micropterus salmoides) estrogen receptor isotypes alpha, beta, and gamma by estradiol.

The expression levels of three estrogen receptor (ER) isotypes alpha, beta, and gamma were quantified in female largemouth bass (Micropterus salmoides) (LMB) liver, ovary, brain, and pituitary tissues. ER alpha and beta expression predominated in the liver, while ERs beta and gamma predominated in the other tissues. Temporally in females, ER alpha was highly up-regulated, ER gamma was slightly up-regulated, and ER beta levels remained unchanged in the liver when plasma 17-beta estradiol (E2) and vitellogenin (Vtg) levels were elevated in the spring. In ovarian tissue from these same fish, all

The class B scavenger receptor 2 act as a proviral factor in largemouth bass ranavirus infection.

Largemouth bass ranavirus (LMBRaV) is a serious epidemic pathogen of Largemouth bass (Micropterus salmoides) and cause severe economic losses to aquaculture. As a key receptor in innate immune responses, the function of scavenger receptor class B member 2 (SRB2) during virus infection has not been reported in fish to date. This study found that M. salmoides SRB2 (MsSRB2) is a relatively conserved sequence with two transmembrane domains and shares 65.41%-98% sequence identities with SRB2 from other vertebrates. Quantitative real-time PCR (qRT-PCR) results showed that MsSRB2 is predominantly exp

The activation of NLRP3 inflammasome and its function in anti-infection immunity of largemouth bass (Micropterus salmoides).

As an intracellular sensor, NLRP3 can recognize a variety of pathogen-associated molecular patterns (PAMPs) and damage-associated molecular patterns (DAMPs). Upon activation, it assembles with the adaptor protein ASC and the effector molecule pro-Caspase-1 to form inflammasome complex, which is closely associated with various infectious and inflammatory diseases. However, the NLRP3 inflammasome has only been identified in a few teleost species, and its activation mechanism remains unclear. In the present study, four NLRP3 genes (MsNLRP3) were cloned and identified from the largemouth bass (Mic

Transcriptomic analysis of immune and metabolic responses in largemouth bass (Micropterus salmoides) challenged with Citrobacter freundii.

Citrobacter freundii, a common zoonotic pathogen affecting humans, livestock and fish, is recognized for its substantial impact on largemouth bass (Micropterus salmoides) mortality. However, the mechanisms of C. freundii infection in largemouth bass remain poorly understood. Based on the results of extracellular enzyme activity detection, it was speculated that the production of extracellular enzymes may be related to bacterial pathogenicity. To elucidate the immune response mechanism, transcriptomic analysis was conducted on spleen tissues from largemouth bass infected with C. freundii. Follo

Study on the potential impact of sustained high temperatures during non-breeding season on largemouth bass.

With the growing scale of largemouth bass breeding, the demand for seedlings is increasing. As global temperatures rise, it is crucial to study the effects of high temperature their regulatory mechanisms in largemouth bass. In this study, we simulated a high water temperature (28 °C) in the non-breeding season in aquaculture ponds for 28 days to examine the growth, reproduction, metabolism, apoptosis, and methylation markers in largemouth bass; transcriptome analysis was also performed. The results showed no significant difference in body weight between male and female largemouth bass. However

Proteomic analysis of hepatic fibrosis induced by a high starch diet in largemouth bass (Micropterus salmoides).

Largemouth bass is sensitive to the dietary starch level and excess starch can induce metabolic liver diseases (MLD). Hepatic fibrosis is a typical pathological phenotype of MLD in largemouth bass, but the molecular basis underlying is largely unclear. This study fed fish with a low or high starch diet for 4 weeks. Liver tissues with or without fibrotic symptoms were recognized through histopathological and molecular markers analysis of hepatic fibrosis, following TMT Quantitative proteomics and conducted Parallel Reaction Monitoring (PRM) analyses. 2455 differentially expressed proteins with

Identification and functional analysis of circRNAs in the skeletal muscle of juvenile and adult largemouth bass (Micropterus salmoides).

Circular RNA (circRNA) is a novel emerging type of endogenous regulatory non-coding RNA molecules with a covalent closed-loop configuration, which exerts important functions in multiple biological processes. CircRNAs are known to regulate gene expression as functional regulators interacting with miRNAs by sponge, which have been reported to regulate skeletal muscle development. Nevertheless, the information of circRNAs involved in regulating muscle growth and development in fish is largely unknown. Here, we first identified 312 and 511 circRNAs in skeletal muscle of juvenile and adult largemou

Gonadal Development in Smallmouth Bass (Micropterus Dolomieu) Reared in the Absence and Presence of 17-α-Ethinylestradiol.

Testicular oocytes in wild adult bass (Micropterus spp.) are considered a potential indication of exposure to estrogenic compounds in municipal, agricultural, or industrial wastewater. However, our ability to interpret links between testicular oocyte occurrence in wild fish species and environmental pollutants is limited by our understanding of normal and abnormal gonadal development. We previously reported low-to-moderate testicular oocyte prevalence (7%-38%) among adult male bass collected from Minnesota waters with no known sources of estrogenic compounds. In the present study, two experime

The influence of 17β-oestradiol on lymphopoiesis and immune system ontogenesis in juvenile sea bass, Dicentrarchus labrax.

The female sex steroid 17β-oestradiol (E2) is involved in the regulation of numerous physiological functions, including the immune system development and performance. The role of oestrogens during ontogenesis is, however, not well studied. In rodents and fish, thymus maturation appears to be oestrogen-dependent. Nevertheless, little is known about the function of oestrogen in immune system development. To further the understanding of the role of oestrogens in fish immune system ontogenesis, fingerlings of European sea bass (Dicentrarchus labrax) were exposed for 30 days to 20 ng E2·L-1, at two

Overwinter Changes in the Lipid Profile of Young-of-the-Year Striped Bass (Morone saxatilis) in Freshwater Ponds.

Young-of-the-year (YOY) striped bass (Morone saxatilis) suffer significant mortality during their first winter. While causes of this mortality are unclear, lipids may play role in adapting to winter stresses, including thermal change and food scarcity. To address this, YOY striped bass were placed in mesh cages in freshwater ponds in the fall (November) and were held until the end of winter, in March. Liver and white muscle tissue were sampled at the beginning and end of the study to compare concentrations of specific lipid classes and fatty acid composition. Muscle-tissue total lipid and tria
