Traditional systems of medicine have acted as a primary health care modality for resource-constrained regions of the world since time immemorial. This study investigates the anthelmintic activity of 17 ethnobotanically selected medicinal plants from the Swat region of Pakistan and characterises their phytochemical profiles. The anthelmintic activity of plant extracts prepared in five different solvents (hexane, acetone, ethanol, methanol, and water) was determined by using Caenorhabditis elegans as a model, followed by assessment of the phytochemistry as well as thin-layer chromatographic prof

African swine fever virus (ASFV) causes a viral disease in swine with a mortality rate of approximately 100%, threatening the global pig industry's economic development. However, vaccines are not yet commercially available, and other antiviral therapeutics, such as antiviral drugs, are urgently needed. In this study, berbamine hydrochloride, a natural bis-benzylisoquinoline alkaloid isolated from the traditional Chinese herb Berberis amurensis, showed significant antiviral activity against ASFV. The 50% cytotoxic concentration (CC50) of berbamine hydrochloride in porcine alveolar macrophages (

Berberine, a naturally occurring isoquinoline alkaloid found in several medicinal plants such as Berberis spp. and Coptis chinensis has long been used in traditional medicine. Recent evidence positions berberine as a pleiotropic modulator of multiple interconnected hallmarks of aging, making it a promising candidate for geroprotective interventions. This review critically examines the molecular mechanisms through which berberine influences energy metabolism, mitochondrial homeostasis, proteostasis, chronic inflammation, and cellular senescence. Berberine activates AMP-activated protein kinase

After cryopreservation, reactive oxygen species (ROS) can damage sperm. Antioxidants are the primary defense against oxidative damage. Berberine is a bioactive alkaloid found in Berberis vulgaris, Curcuma longa, and Ergon grape, and is a potent antioxidant. Due to the negative effects of free radicals in oxidative stress processes, antioxidant chemicals are required to protect sperm. However, berberine has low bioavailability, making it less effective. Loading techniques on nanoparticles and nanotechnology can help overcome this limitation. Selenium nanoparticles were synthesized with barberry

Berberine is a famous alkaloid extracted from Berberis plants and has been widely used as medications and functional food additives. Recent studies reveal that berberine exhibits neuroprotective activity in animal models of Parkinson's disease (PD), the second most prevalent neurodegenerative disorders all over the world. However, the actual site of anti-PD action of berberine remains largely unknown. To this end, we employed a fluorescently labeled berberine derivative BBRP to investigate the subcellular localization and blood brain barrier (BBB) permeability in a cellular model of PD and zeb

Cytochrome P450 reductase was purified to homogeneity from cell suspension cultures of the opium poppy Papaver somniferum, the enzyme was characterized (K(m) cytochrome c, 8.3 microM; K(m) NADPH, 4.2 microM; pH optimum, 8.0; M(r), 80 kDa), and the amino acid sequence of internal peptides was determined. Partial cDNA clones from P. somniferum and from Eschscholzia californica (California poppy) were then generated using the polymerase chain reaction and were used as hybridization probes to isolate full-length cDNAs. The Papaver and Eschscholzia cytochrome P450 reductases are 63% identical at th

UNLABELLED: Alzheimer's disease (AD) is one of the leading tauopathies in which several kinases phosphorylate Tau in response to Aβ oligomers, inflammation, calcium dysregulation, oxidative stress, mitochondrial dysfunction, or disruption of the key signaling pathways. Tau is phosphorylated by Ser/Thr kinases such as GSK3β, CDK5, MAPK14, and MARK2, as well as Tyr kinases like Fyn. Given the large crosstalk among these kinases, targeting one of these kinases is ineffective in treating tauopathies such as AD, highlighting the need for a multi-targeted approach. In this study, we targeted key kin

Uncapped and Glutathione capped Cupric oxide nanospheres were synthesized by the interaction of Berberis lycium (Bl) root extract with corresponding salt solution. CuO nanospheres were best optimized by mixing 2% Bl extract solution with 1 mM CuSO4·5H2O (pH 11, 90 °C) Reduced glutathione (0.25 mM) in solution form was added in respective emulsion after 24 h. Synthesis of nanospheres was ensured by distinct surface plasmonic resonance peaks shown by CuO (370-420 nm). Addition of glutathione resulted in sharp blue shift and lowered absorbance values in UV spectra suggesting the decrease in nanop

Cancer cells frequently evade therapies that depend on apoptosis, necessitating the exploration of alternative cell death mechanisms. Ferroptosis, an iron-dependent regulated cell death characterized by lethal lipid peroxidation, has emerged as a promising strategy for cancer treatment. Recent studies have identified berberine, an isoquinoline alkaloid derived from Coptis chinensis and Berberis species, as an inducer of ferroptosis in various malignancies through its multitarget effects. This review systematically elucidates the molecular pathways through which berberine induces ferroptosis. T
