BACKGROUND: Blood-based biomarkers hold significant promise for the early detection and diagnosis of Alzheimer's disease (AD) and other dementias. Age-related changes in blood levels of AD biomarkers are well-documented but poorly understood. Epigenetic clocks are mathematical models based on DNA methylation patterns that reflect various aspects of the multidimensional aging process. We investigated the associations of epigenetic aging with five blood-based AD biomarkers in 2656 Hispanic/Latino adults (mean age 62.5 years; 65% females) from the Hispanic Community Health Study/Study of Latinos.

Biological aging clocks across organ systems and tissues have advanced understanding of human aging and disease. In this study, we expand this framework to develop seven magnetic resonance imaging-based multi-organ biological age gaps (MRIBAGs), including the brain, heart, liver, adipose tissue, spleen, kidney and pancreas. Using data from 313,645 individuals curated by the MULTI Consortium, we link the seven MRIBAGs to 2,923 plasma proteins, 327 metabolites and 6,477,810 common genetic variants. Genome-wide associations identify 53 MRIBAG-locus pairs (P < 5 × 10-8). Genetic correlation and Me

This study aimed to determine the nature of the relationship between the internalized stigma of chronic pain (ISCP), the pace of biological aging, and racial disparities in nonspecific chronic low back pain (CLBP). We used Dunedin Pace of Aging from the Epigenome (DunedinPACE), Horvath's, Hannum's, and PhenoAge clocks to determine the pace of biological aging in adults, ages 18 to 82 years: 74 no pain, 56 low-impact pain, and 76 high-impact pain. Individuals with high-impact pain reported higher levels of ISCP and DunedinPACE compared to those with low-impact or no pain (p < 0.001). There was

BACKGROUND: Frailty reflects diminished physiological reserve and increased vulnerability to adverse health outcomes. It has been linked to biological aging, including epigenetic age acceleration (EAA), a DNA methylation-based marker of aging, but the extent to which EAA accounts for the frailty-mortality association remains unclear.
METHODS: We analyzed three U.S. cohorts-NHANES (1999-2002), HRS (2016), and HANDLS (2004-2009)-with mortality follow-up through 2019-2022. Frailty was defined using harmonized adaptations of the Fried phenotype and FRAIL scale. EAA was derived from five epigeneti

Schizophrenia spectrum disorders (SSD) are debilitating psychiatric illnesses that require extensive pharmacologic, cognitive, and functional management. SSD patients are often prescribed different medications, most commonly antipsychotics, which bear numerous side effects. Recently, accumulating evidence has shown epigenetic aging changes in SSD. However, the effects of antipsychotic medications on this phenomenon remain unexplored. We investigated whether antipsychotic medications are associated with epigenetic age acceleration (EAA) in 153 SSD patients. EAA was estimated using six different

The SARS-CoV-2 pandemic has affected millions worldwide, with aging being a key risk factor for severe disease outcomes. This study examines the rate of epigenetic aging, as measured by DNA methylation-based aging markers, in COVID-19 patients versus healthy individuals. We found that PCGrimAge, a next-generation epigenetic clock associated with immune dysregulation and inflammation, showed the strongest correlation with the chronological age of the European COVID-19 patients. Several other next-generation epigenetic clocks, including PCGrimAge, DunedinPACE, and ZhangY2017, also exhibited acce

This paper reviews studies that addressed the influence of diet on circadian rhythmicity in mice and, in turn, circadian clock chronodisruption and its role in the development of metabolic disorders. Studies from the past 14 years were selected via a systematic search conducted using the PubMed electronic database. After applying the inclusion and exclusion criteria, 291 studies were selected, of which 13 were chosen using the following inclusion criteria: use of a high-fat diet for mice, evaluation of clock gene expression, and the association between chronodisruption and lipid metabolism dis

BACKGROUND: Repetitive head impacts in former contact sport athletes are associated with cognitive impairment, accelerated cerebral atrophy and risk of neurodegenerative disease. Epigenetic clocks derived from age-associated DNA methylation (DNAm) profiles may capture accelerated biological ageing in neurodegenerative conditions; however, their application in former athletes remains unexplored. Here we aim to explore the application of epigenetic clocks as a measure of accelerated biological ageing in a cohort of former athletes.
METHODS: In 126 former athletes (96% male; mean age: 54.5±14.4

Epigenetic clocks estimate biological age, yet evidence linking decelerated epigenetic aging to healthy cognitive performance in community-dwelling older adults is limited. Cross-sectional study within the population-based ALEXANDROS cohort (Santiago, Chile). Healthy Cognitive Agers (HCA) were participants aged ≥80 years who consistently maintained a MMSE score =30 across all cohort assessments. Controls were cognitively normal peers from the same cohort (MMSE 28.5 ± 2.9). Epigenetic age was estimated using the Horvath, GrimAge, and PhenoAge clocks. Epigenetic age acceleration (EAA) was calcul

The increase in life expectancy over the past century has been accompanied by the recognition of age as the primary risk factor for a wide range of pathologies, including cardiovascular and neurodegenerative diseases, as well as cancer. Thus, the development of interventions that slow the underlying biological processes of aging could have beneficial effects on the prevention or progression of these diseases. To evaluate such geroprotective interventions, quantifying biological damage via aging clocks-particularly those based on transcriptomic biomarkers-has become highly relevant, as they pro

Epigenetic clocks based on DNA methylation (DNAm) accurately predict age, but their biological underpinnings remain unclear. One primary mechanism by which DNAm might influence gene regulation is by modulating transcription factor binding activity. This study investigates the regulatory potential of predictive CpGs in established epigenetic clocks. Our analysis reveals that generally most CpGs used by epigenetic clocks do not overlap known transcription factor binding sites (TFBS), indicating that clock accuracy is not primarily driven by changes in TF binding dynamics. However, analysis of Cp

To synchronize with the fluctuating environment, organisms have evolved an endogenous time tracking mechanism referred to as the biological clock(s). This clock machinery has been identified in almost all cells of vertebrates and categorized as central and peripheral clocks. In birds, three independent circadian clocks have been identified in the hypothalamus, the pineal and the retina which interact and generate circadian time at a functional level. However, there is a limited knowledge of molecular clockwork and integration between central and peripheral clocks in birds. Therefore, we studie

BACKGROUND: Biological ageing is one of the principal risk factors for neurodegenerative diseases. It is becoming increasingly clear that acceleration of DNA methylation age, as measured by the epigenetic clock, is closely associated with many age-related diseases.
METHODS: We searched the PubMed and Web of Science databases to identify eligible studies reporting epigenetic clocks in several neurodegenerative diseases, including Alzheimer's disease (AD), Parkinson's disease (PD), amyotrophic lateral sclerosis (ALS) and Huntington's disease (HD).
RESULTS: Twenty-three studies (12 for AD, 4 fo

In both mammals and fish, the circadian system is composed of oscillators that function at the cellular, tissue, and system levels and show the cyclic expression of clock genes. The organization and functioning of the biological clock in fish has not yet been characterized in detail, therefore, in the present study, an extensive analysis of the rhythmic expression of the main components of the biological clock in the central and peripheral organs of common carp was performed. The diurnal changes in clock gene expression were determined with respect to the subjective light cycle in fish exposed

In our consumer-oriented society, in which productivity requires around-the-clock activity and demanding shift work, the biologic system that regulates our internal rhythms is being compromised. Poor sleep patterns and hectic lifestyle are detrimental to harmonious physiological and metabolic body systems, with severe impact on public health. Over a trillion peripheral cellular clocks throughout the body, supervised by the master clock located in the hypothalamic suprachiasmatic nucleus, govern most aspects of physiology and behavior. To exemplify the importance of the biologic clock for healt

BACKGROUND: Biological clocks are promising tools for the evaluation of biological age deviations (i.e., positive/negative acceleration). Here, we explored the associations of biological age acceleration (BAA) assessed by Horvath's, Hannum's, PhenoAge, and GrimAge epigenetic clocks, as well as the iAge inflammation-based clock, with functional capacities across adulthood and tested if chronological age and sex moderate these associations.
METHODS: Cross-sectional analysis was conducted with baseline (2019-2021) data from 1014 participants (age range 20-104 years old, 62.82% female) drawn from

Worldwide increase in life expectancy has boosted research on aging. Overcoming the concept of chronological age, higher attention has been addressed to biological age, which reflects a person's real health state, and which may be the resulting combination of both intrinsic and environmental factors. As epigenetics may exert a pivotal role in the biological aging, epigenetic clocks were developed. They are based on mathematical models aimed at identifying DNA methylation patterns that can define the biological age and that can be adopted for different clinical scopes (i.e., estimation of the r

The circadian clock is a biological timekeeping system to govern temporal rhythms of the endocrine system and metabolism. The master pacemaker of biological rhythms is housed in the hypothalamic suprachiasmatic nucleus (SCN) where approximately 20,000 neurons exist and receive light stimulus as a predominant timed external cue (zeitgeber). The central SCN clock orchestrates molecular clock rhythms in peripheral tissues and coordinates circadian metabolic homeostasis at a systemic level. Accumulated evidence underscores an intertwined relationship between the circadian clock system and metaboli

Gene polymorphisms in the mammalian biological clock system influence individual rhythms. A single nucleotide polymorphism (SNP) in the 3' flanking region of CLOCK (3111 T/C; rs1801260) influenced diurnal preference in healthy humans and caused sleep phase delay and insomnia in patients affected by bipolar disorder. Genes of the biological clock are expressed in many brain structures other than in the 'master clock' suprachiasmatic nuclei. These areas, such as cingulate cortex, are involved in the control of many human behaviors. Clock genes could then bias 'nonclock' functions such as informa

Ageing biomarkers quantify molecular, physiological or functional features that vary between individuals as they age and may improve prediction of health outcomes beyond chronological age. Recently, plasma proteomics have been used to estimate the biological ages of 11 human organs. Although accelerated organ ageing is associated with higher mortality risk, systematic benchmarking against established ageing biomarkers is lacking. Here, we pursued two complementary aims. First, we benchmarked proteomic organ clocks against multimodal ageing biomarkers for associations with 16-year all-cause mor
