Cilia are microtubule-based cellular protrusions critical for cell signaling and motility. The ciliary transition zone connects axonemal microtubules to the ciliary membrane and functions as a gatekeeper by regulating selective targeting and sorting of proteins. However, the molecular mechanisms underlying its assembly remain incompletely understood. Herein, we identify an important role for cilia and flagella associated protein 100 (CFAP100) in the transition zone assembly. Cfap100 knockout mice exhibit ciliary defects in multiple organs, including the brain, trachea, and kidney. Super-resolu

Protein-RNA phase separation is at the center of membraneless biomolecular condensates governing cell physiology and pathology. Using an archetypical viral protein-RNA condensation model, we determined the sequence of events that starts with sub-second formation of a protomer with two RNAs per protein dimer. Association of additional RNA molecules to weaker secondary binding sites in this protomer kickstarts crystallization-like assembly of a molecular condensate. Primary nucleation is faster than the sum of secondary nucleation and growth, which is a multistep process. Protein-RNA nuclei grow

The genome stores and processes approximately 1.5 gigabytes of encoded information. In this article, we propose that the eukaryotic genome and its adaptable three-dimensional packing in the form of chromatin offer a valuable template for the system architecture of DNA-based digital computers. We examine embryonic and stem cells, which exhibit distinct chromatin-associated condensates enriched in transcription machinery. These dynamic biomolecular condensates facilitate the spatial association of genes, genomic control elements, and molecular machinery responsible for reading the genomic code.

Accumulating evidence indicates that metabolic enzymes can directly couple metabolic signals to transcriptional adaptation and cell differentiation. Glycogen synthase 1 (GYS1), the key metabolic enzyme for glycogenesis, is a nucleocytoplasmic shuttling protein compartmentalized in the cytosol and nucleus. However, the spatiotemporal regulation and biological function of nuclear GYS1 (nGYS1) microcompartments remain unclear. Here, we show that GYS1 dynamically reorganizes into nuclear condensates under conditions of glycogen depletion or transcription inhibition. nGYS1 complexes with the transc

In biological systems, liquid and solid-like biomolecular condensates may contain the same molecules but their behaviour, including movement, elasticity, and viscosity, is different on account of distinct physicochemical properties. As such, it is known that phase transitions affect the function of biological condensates and that material properties can be tuned by several factors including temperature, concentration, and valency. It is, however, unclear if some factors are more efficient than others at regulating their behaviour. Viral infections are good systems to address this question as t

The aggregation and misfolding of the neuronal microtubule-associated protein tau is closely linked to the pathology of Alzheimer's disease and several other neurodegenerative diseases. Recent evidence suggest that tau undergoes liquid-liquid phase separation in vitro and forms or associates with membrane-less organelles in cells. Biomolecular condensation driven by phase separation can influence the biological activities of tau including its ability to polymerize tubulin into microtubules. In addition, the high concentrations that tau can reach in biomolecular condensates provide a mechanism

Biomolecular phase separation (BPS), encompassing liquid-liquid, liquid-gel, and liquid-solid transitions, has emerged as a fundamental principle governing cellular organization and function. Dysregulation of these phase states acts as a pathogenic driver of diverse intractable diseases, including neurodegeneration, cancer, and autoimmune disorders. The intrinsic properties of biomolecular condensates, self-assembly, dynamic reversibility, and environmental responsiveness, also offer a robust blueprint for next-generation biomaterial design. Despite rapid progress across the field, a cohesive

Proper protein production is an essential biological process for all known living organisms. The cell depends on a functional protein homeostasis, or proteostasis, network to facilitate proper gene expression, transcription, translation, polypeptide folding, and degradation. Several condensates are involved in the "birth" and maturation of newly synthesized proteins. In conditions of cellular stress, gene expression and protein production are altered, and protein degradation often increases. Under various types of cellular stress, new condensates often form. This review will highlight the cond

Condensate-modifying therapeutics (c-mods), comprising low-molecular-weight compounds (~1.5 kDa) and related agents such as RNAs and metabolites like ATP, are emerging as promising drug candidates for neurodegenerative disorders, cancer, and viral infections. These compounds modulate liquid-liquid phase separation (LLPS), a biophysical process driven by intrinsically disordered regions, low-complexity domains, and multivalent non-covalent interactions (hydrogen bonds, van der Waals forces, hydrophobic, and electrostatic interactions). LLPS governs the formation of biomolecular condensates esse

Liquid-liquid phase separation (LLPS) is a physical process that drives the formation of biomolecular condensates, which coordinate cellular biochemical reactions in space and time. Nevertheless, these metastable condensates are susceptible to various disturbances, which trigger the occurrence of abnormal phase transitions. In neurodegeneration, this process promotes the development of misfolded proteins into toxic fibrils. In cancer, LLPS affects multiple stages of its occurrence and progression, including oncogenic signaling, gene expression, angiogenesis, metabolic reprogramming, and invasi

The mammalian epidermis forms a critical barrier against environmental insults and water loss. The formation of its outermost layer, the stratum corneum, involves a rapid terminal differentiation process that has traditionally been explained by the "bricks and mortar" model. Recent advances reveal a more dynamic mechanism governed by intracellular liquid-liquid phase separation (LLPS). This review proposes that the lifecycle of the granular layer is orchestrated by LLPS. Evidence is synthesized showing that keratohyalin granules (KGs) are biomolecular condensates formed by the phase separation

Intrinsically disordered proteins (IDPs) often associate into biomolecular condensates. While micron-sized condensates are observed above a well-defined saturation concentration, the formation of nanoscale clusters also occurs at subsaturated concentrations. The diversity and physical nature of such nanoscale clusters remain underexplored with respect to the well-established condensate state. Here we show that the Parkinson's disease-associated protein α-synuclein forms highly monodisperse assemblies at low ionic strength, distinct from the larger polydisperse assemblies previously observed at

The onset and development of Alzheimer's disease is linked to the accumulation of pathological aggregates formed from the normally monomeric amyloid-β peptide within the central nervous system. These Aβ aggregates are increasingly successfully targeted with clinical therapies at later stages of the disease, but the fundamental molecular steps in early stage disease that trigger the initial nucleation event leading to the conversion of monomeric Aβ peptide into pathological aggregates remain unknown. Here, we show that the Aβ peptide can form biomolecular condensates on lipid bilayers both in m

The biomolecular aggregates generated and maintained through phase separation contribute to the establishment of functionally oriented and ordered compartments in biological systems, which is crucial for intracellular activities. However, the mechanisms underlying the aggregation and dynamic transformation of biomacromolecules are not fully understood. In this paper, the phase separation and self-assembly of DNA and ferrous ions are reported to achieve a two-step structural change. DNA-Fe2+ condensates are initially formed by phase separation, followed by a phase transition from primary cluste

Thousands of genetic variants in protein-coding genes have been linked to disease. However, the functional impact of most variants is unknown as they occur within intrinsically disordered protein regions that have poorly defined functions1-3. Intrinsically disordered regions can mediate phase separation and the formation of biomolecular condensates, such as the nucleolus4,5. This suggests that mutations in disordered proteins may alter condensate properties and function6-8. Here we show that a subset of disease-associated variants in disordered regions alter phase separation, cause mispartitio

Biomolecular condensates are dynamic membraneless compartments that regulate a myriad of cellular functions. A particular type of physiological condensate called stress granules (SGs) has gained increasing interest due to its role in the cellular stress response and various diseases. SGs, composed of several hundred RNA-binding proteins, form transiently in response to stress to protect mRNAs from translation and disassemble when the stress subsides. Interestingly, SGs contain several aggregation-prone proteins, such as TDP-43, FUS, hnRNPA1, and others, which are typically found in pathologica

The crowdedness of the cell calls for adequate intracellular organization. Biomolecular condensates, formed by liquid-liquid phase separation of intrinsically disordered proteins and nucleic acids, are important organizers of cellular fluids. To underpin the molecular mechanisms of protein condensation, cell-free studies are often used where the role of crowding is not investigated in detail. Here, we investigate the effects of macromolecular crowding on the formation and material properties of a model heterotypic biomolecular condensate, consisting of nucleophosmin (NPM1) and ribosomal RNA (r

A significant proportion of global photosynthetic carbon fixation relies on the pyrenoid, a biomolecular condensate found in the chloroplast of most unicellular algae, where the CO2-fixing enzyme Rubisco is exposed to saturating concentrations of the gas. In this review, we highlight recent advances in our understanding of the molecular basis of diverse pyrenoids. Phase separation of phylogenetically distant Rubiscos is mediated by convergently evolved linker proteins, with an emerging theme of pyrenoid condensation being organized via Rubisco-binding motifs. To minimize CO2 leakage out of the

The transactive response DNA binding protein 43 (TDP-43) is an RNA/DNA-binding protein that is involved in a number of cellular functions, including RNA processing and alternative splicing, RNA transport and translation, and stress granule assembly. It has attracted significant attention for being the primary component of cytoplasmic inclusions in patients with amyotrophic lateral sclerosis or frontotemporal dementia. Mounting evidence suggests that both cytoplasmic aggregation of TDP-43 and loss of nuclear TDP-43 function contribute to TDP-43 pathology. Furthermore, recent studies have demons

Mast cell (MC) degranulation is commonly viewed as the rapid release of preformed inflammatory mediators into the extracellular environment. In a recent study published in the Journal of Experimental Medicine, Salm et al. described a more spatially controlled form of MC communication following sterile peritoneal injury. Sensory neurons activated MCs through substance P signaling, promoting the release of extracellular granules that persisted as membraneless biomolecular condensates. These condensates, enriched in more than 200 proteins, remained associated with the surface of large peritoneal
