Fungicidal activity of a phospholipase-A2-derived synthetic peptide variant against Candida albicans.

pEM-2, a 13-mer synthetic peptide variant derived from myotoxin II, a phospholipase A(2) homologue present in Bothrops asper snake venom, has shown potent bactericidal activity in previous studies due to the combination of cationic and hydrophobic amino acids, including three tryptophan-substituted residues in its sequence. This study reports that pEM-2 also exerts potent fungicidal activity against a variety of clinically relevant Candida species, killing 100% of yeasts at concentrations near 10 mg/l (5 microM), as indicated by plate counting assays. Thus, this peptide displays a broad-spectr

Physicochemical and immunological effects of adjuvant formulations with snake venom antigens for immunization of horses for antivenom production.

Enhancement of antivenom immune responses in horses through adjuvant technology improves antivenom production efficiency, but substantial local reactogenicity associated with some traditional veterinary adjuvants limits their usability. To explore modern adjuvant systems suitable for generating antivenom responses in horses, we first assessed their physicochemical compatibility with Bothrops asper snake venom. Liposome and nanoparticle aluminum adjuvants exhibited changes in particle size and phospholipid content after mixing with venom, whereas squalene emulsion-based adjuvants remained stabl

Structural characterization and phylogenetic relationships of myotoxin II from Atropoides (Bothrops) nummifer snake venom, a Lys49 phospholipase A(2) homologue.

In order to analyze its structure-function relationships, the complete amino acid sequence of myotoxin II from Atropoides (Bothrops) nummifer from Costa Rica was determined. This toxin is a Lys49-type phospholipase A(2) (PLA(2)) homologue, devoid of catalytic activity, structurally belonging to class IIA. In addition to the Asp49 --> Lys change in the (inactive) catalytic center, substitutions in the calcium-binding loop suggest that its lack of enzymatic activity is due to the loss of ability to bind Ca(2+). The toxin occurs as a homodimer of basic subunits of 121 residues. Its sequence has h

Homogenates of skeletal muscle injected with snake venom inhibit myogenic differentiation in cell culture.

INTRODUCTION: Viperid snakebite envenomings are characterized by muscle necrosis and a deficient regenerative response. METHODS: Homogenates from gastrocnemius muscles of mice injected with the venom of the snake Bothrops asper or with 2 tissue-damaging toxins were added to cultures of C2C12 myogenic cells. Myoblasts proliferation and fusion were assessed. Venom was detected by immunoassay in mouse muscle during the first week after injection. RESULTS: Homogenates from venom-injected muscle induced a drop in the number of proliferating myoblasts and a complete elimination of myotube formatio

Membrane independent activation of fibroblast proMMP-2 by snake venom: novel roles for venom proteinases.

ProMMP-2 activation by Bothrops asper venom was investigated in mouse gastrocnemius muscle, mammalian cell culture and a cell-free system. Zymography revealed an increment of latent and activated forms of MMP-2 in muscle homogenates 1-3 days after venom injection. To clarify if venom can induce expression and activation of MMP-2, independently of the inflammatory response, venom was added to cultured human fibroblasts, endothelial and skeletal muscle cells, which expressed proMMP-2 constitutively. Venom activated proMMP-2 without promoting its expression. Venom also activated and degraded proM

Venoms and toxins that induce anticoagulation in vitro do not cause major alterations in coagulation parameters in vivo in a mouse model.

Snake venoms exert profound effects on blood coagulation, mostly associated with consumption coagulopathy due to the procoagulant activity of venom proteinases, which result in drastic alterations of laboratory clotting tests. In addition, snake venoms contain enzymes [mostly phospholipases A2 (PLA2s) and metalloproteinases (SVMPs)] that have strong anticoagulant activity in vitro. However, their actual effect on coagulation parameters in vivo has not been studied in detail. In the present work, we evaluated the alterations in standard clotting tests (prothrombin time, activated partial thromb

Coagulopathy induced by viperid snake venoms in a murine model: Comparison of standard coagulation tests and rotational thromboelastometry.

Viperid snakebite envenoming is often characterized by a venom-induced consumption coagulopathy due to the procoagulant effect of venom components, resulting in the alteration of clotting laboratory tests. There is a growing trend to use rotational thromboelastometry in the assessment of clotting disturbances in a variety of pathologies, although its use in experimental models of envenoming has been limited. An in vivo murine model was implemented to assess the coagulopathy induced by three Central American viperid venoms which have different mechanisms of action on clotting factors, i.e., Bot

Site mutation of residues in a loop surrounding the active site of a PI snake venom metalloproteinase abrogates its hemorrhagic activity.

Abrogation of the hemorrhagic activity of BaP1, a PI Snake Venom Metalloproteinase (SVMP) from the venom of Bothrops asper, was achieved by the substitution of residues in the first part of the Ω loop surrounding the active site by the corresponding residues of a structurally-similar non-hemorrhagic PI SVMP from a related venom. Previous studies by molecular dynamic simulation showed higher flexibility in the first part of the loop in hemorrhagic SVMPs, as compared to non-hemorrhagic SVMPs. It has been suggested that the Ω loop is critical for protein-protein interface and may be involved in t

Discovery of small molecule inhibitors for the snake venom metalloprotease BaP1 using in silico and in vitro tests.

Snakebites represent an important public health problem, with a great number of victims with permanent sequelae or fatal outcomes, particularly in rural, agriculturally active areas. The snake venom metalloproteases (SVMPs) are the principal proteins responsible for some clinically-relevant effects, such as local and systemic hemorrhage, dermonecrosis, and myonecrosis. Because of the difficulties in neutralizing them rapidly and locally by antivenoms, the search and design of small molecules as inhibitors of SVMPs are proposed. The Bothrops asper metalloprotease P1 (BaP1) is hereby used as a t

Increased infectivity of Staphylococcus aureus in an experimental model of snake venom-induced tissue damage.

Soft-tissue infection is commonly found in patients bitten by Latin American Bothrops snakes. Staphylococcus aureus, which is not present in the mouth of the snake, is frequently isolated from these infections. The effects of B. asper venom on infection with S. aureus were analyzed in a model of infection in envenomated mouse gastrocnemius muscle. Inoculation of 50 colony-forming units (cfu) of S. aureus was enough to cause infection in envenomated muscle, compared with >5x104 cfu without venom. This effect was also achieved by injection of venom myotoxin III (an A(2) phospholipase). A sarA mu

Mechanical properties, microstructure, and strike biomechanics of tubular fangs in neotropical vipers.

Viper fangs represent one of the most mechanically sophisticated biological penetration devices found in nature. Despite extensive interest in their venom-delivery function, a comprehensive characterization of the structure, composition, and biomechanics of neotropical viper fangs remains lacking. This study presents the first integrated analysis of the tubular fangs of three Latin American viper species - Lachesis acrochorda, Crotalus durissus cumanensis, and Bothrops asper - combining optical microscopy, scanning electron microscopy, high-resolution X-ray computed tomography, attenuated tota

Isolation and characterization of myotoxin II from Atropoides (Bothrops) nummifer snake venom, a new Lys49 phospholipase A2 homologue.

Myotoxic phospholipases A2 of class II are commonly found in the venoms of crotalid snakes. As an approach to understanding their structure-activity relationship, diverse natural variants have been characterized biochemically and pharmacologically. This study describes a new myotoxic phospholipase A2 homologue, isolated from the venom of Atropoides (Bothrops) nummifer from Costa Rica. A. nummifer myotoxin 1 is a basic protein, with an apparent subunit molecular mass of 16 kDa, which migrates as a dimer in sodium dodecylsulfate-polyacrylamide gel electrophoresis under nonreducing conditions. Diversity and antibiotic resistance profiles of bacteria isolated from the oral cavity of captive snakes.

Venomous snake bites are a global public health issue, causing between 81000 and 138000 annual deaths and 400000 permanent disabilities. This study investigated the oral bacterial diversity and antibiotic resistance profiles in captive Viperidae snakes. Oral swabs from 48 specimens across four species (Porthidium lansbergii, Bothriechis nigroviridis, Cerrophidion sasai, and Bothrops asper) were analyzed using culture-dependent methods. Bacterial isolation and identification using the VITEK 2 automated system revealed 41 strains from 12 genera, predominantly Gram-negative bacteria, including Mo

Intramuscular delivery of mRNA-encoded single-chain variable fragments prevents myotoxin II-induced skeletal muscle damage in a preclinical model.

Antivenom is the only effective treatment for snakebites, but it often fails to tackle venom-induced local tissue/muscle damage, leading to permanent disabilities. We investigated whether monoclonal single-chain variable fragments (scFvs) targeting myotoxin II (M-II) from Bothrops asper venom, delivered via mRNA-lipid nanoparticles (LNPs), could neutralise M-II-induced adverse effects under diverse settings. Human cultured myotubes transfected with mRNA-LNPs expressed scFvs within 24 h and showed resistance to both M-II and venom. In a mouse model, a single intramuscular injection of mRNA-LNPs

Enzymatic labelling of snake venom phospholipase A2 toxins.

Almost all animal venoms contain secretory phospholipases A2 (PLA2s), 14 kDa disulfide-rich enzymes that hydrolyze membrane phospholipids at the sn-2 position, releasing lysophospholipids and fatty acids. These proteins, depending on their sequence, show a wide variety of biochemical, toxic and pharmacological effects and deserve to be studied for their numerous possible applications, and to improve antivenom drugs. The cellular localization and activity of a protein can be studied by conjugating it with a tag. In this work, we applied an enzymatic labelling method, using Streptomyces mobaraen

A synthetic biology approach for consistent production of plant-made recombinant polyclonal antibodies against snake venom toxins.

Antivenoms developed from the plasma of hyperimmunized animals are the only effective treatment available against snakebite envenomation but shortage of supply contributes to the high morbidity and mortality toll of this tropical disease. We describe a synthetic biology approach to affordable and cost-effective antivenom production based on plant-made recombinant polyclonal antibodies (termed pluribodies). The strategy takes advantage of virus superinfection exclusion to induce the formation of somatic expression mosaics in agroinfiltrated plants, which enables the expression of complex antibo

Edematic and coagulant effects caused by the venom of Bothrops rhombeatus neutralized by the ethanolic extract of Piper auritum.

ETHNOPHARMACOLOGICAL RELEVANCE: In Colombia, the only authorized treatment to cure snakebite envenomation is with the use of antivenom. The antivenom neutralizes the systemic effects properly, but is not very effective at neutralizing local effects, thus several cases have lead to complications. On the other hand, rural communities turn to the use of plants that are easily accessible and available for basic health care. One of these plants is named Piper auritum (PA), which is traditionally highlighted in some indigenous communities of Antioquia and Chocó. AIM OF THE STUDY: The main objective

[The development of the anti-phospholipase A2 antibody response in horses inoculated with venom for the production of polyvalent antisnake serum in Costa Rica].

The development of antibody response against phospholipase A2 activity of Bothrops asper venom was studied in a group of adult and healthy horses used in the production of the polyvalent antivenom at the Instituto Clodomiro Picado. Simultaneously, the general condition of the animals during the immunization schedule was also studied. There was a great individual variability in the immune response, although most of the horses studied reached the highest neutralizing titer after injection of doses of venom of 30 mg and 50 mg. On the other hand, in horses that had been previously immunized and we

Cytotoxicity of snake venom Lys49 PLA2-like myotoxin on rat cardiomyocytes ex vivo does not involve a direct action on the contractile apparatus.

Viperid snake venoms contain a unique family of cytotoxic proteins, the Lys49 PLA2 homologs, which are devoid of enzymatic activity but disrupt the integrity of cell membranes. They are known to induce skeletal muscle damage and are therefore named 'myotoxins'. Single intact and skinned (devoid of membranes and cytoplasm but with intact sarcomeric proteins) rat cardiomyocytes were used to analyze the cytotoxic action of a myotoxin, from the venom of Bothrops asper. The toxin induced rapid hypercontraction of intact cardiomyocytes, associated with an increase in the cytosolic concentration of c

Proteomic and functional analyses of Lachesis acrochorda snake venom from the Valle del Cauca Department of Colombia.

Lachesis acrochorda envenomation has a lethality rate of approximately 90%. Despite its high lethality, little is known about its local and systemic effects and its relationship with its protein content. Thus, to increase knowledge of L. acrochorda snake venom from the Southwestern ecoregion of Colombia, we developed a proteomic analysis using a "bottom-up shotgun proteomic profiling" approach. Besides, we evaluated toxinological properties and compared the effects with the Bothrops asper snake venom activities. The RP-HPLC profile showed similarities with the L. acrochorda snake venom from th
