Quantitative Susceptibility Mapping Suggests Altered Brain Iron in Premanifest Huntington Disease.

BACKGROUND AND PURPOSE: In patients with premanifest (nonsymptomatic) and advanced Huntington disease, changes in brain iron levels in the basal ganglia have been previously reported, especially in the striatum. Quantitative susceptibility mapping by using MR phase imaging allows in vivo measurements of tissue magnetic susceptibility, which has been shown to correlate well with iron levels in brain gray matter and is believed to be more specific than other imaging-based iron measures. The purpose of this study was to investigate the use of magnetic susceptibility as a biomarker of disease prog

Metabolic connectivity mapping reveals effective connectivity in the resting human brain.

Directionality of signaling among brain regions provides essential information about human cognition and disease states. Assessing such effective connectivity (EC) across brain states using functional magnetic resonance imaging (fMRI) alone has proven difficult, however. We propose a novel measure of EC, termed metabolic connectivity mapping (MCM), that integrates undirected functional connectivity (FC) with local energy metabolism from fMRI and positron emission tomography (PET) data acquired simultaneously. This method is based on the concept that most energy required for neuronal communicat

Microscopic R2* mapping of reduced brain iron in the Belgrade rat.

R2* mapping has recently been used to detect iron overload in patients with movement disorders. We demonstrate here that this technique can also be used to detect reduced brain iron, as in the case of a missense mutation in the iron-transporting protein divalent metal transporter 1. Surprisingly, we found that the same brain regions are affected (ie, the globus pallidus, substantia nigra, and cerebellar dentate nucleus); this suggests a much more extensive role for these structures in regulating overall brain iron homeostasis. Therefore, for the clinical monitoring of movement disorders for wh

Voxel-based mapping of brain hypometabolism in permanent amnesia with PET.

In this study, we used voxel-based mapping methods to compare the resting cerebral metabolic rate of glucose (CMRglc) measured with PET in five patients with permanent amnesia (three with chronic Wernicke-Korsakoff and two with postanoxia syndrome) to that of nine healthy age-matched subjects. We assessed (i) a group pattern of relative hypometabolism; and (ii) the consistency of this group pattern, if any, in individual subjects, according to etiology. The results from the group analysis documented that permanent amnesia is associated with hypometabolism in the thalamus, posterior cingulate c

Functional mapping of human brain in olfactory processing: a PET study.

This study describes the functional anatomy of olfactory and visual naming and matching in humans, using positron emission tomography (PET). One baseline control task without olfactory or visual stimulation, one control task with simple olfactory and visual stimulation without cognition, one set of olfactory and visual naming tasks, and one set of olfactory and visual matching tasks were administered to eight normal volunteers. In the olfactory naming task (ON), odors from familiar items, associated with some verbal label, were to be named. Hence, it required long-term olfactory memory retriev

[Neuropsychological findings of functional cortical mapping in patients with brain tumors].

In 3 patients with brain tumors involving language areas, we investigated functional cortical regions and epileptogenic foci using subdural electrode stimulation before operation. These stimulation studies were useful in preserving postoperatively their language function. The following neuropsychological findings were obtained; (1) Stimulation of Broca area (Brodmann 44 and 45) did not produce anarthria (phonetic disintegration), but semantic and phonemic parahasias and word finding difficulties. (2) Stimulation of lower motor cortex elicited anarthria. (3) Buccofacial apraxia was not produced

Brain mapping in dysphonemic dyslexia: in resting and phonemic discrimination conditions.

Topographic mapping of EEG bands and alpha asymmetry were analyzed during resting conditions and an auditory phonemic discrimination task (APD) in carefully screened samples of 9.0- to 12-year-old control and dysphonemic-sequencing dyslexic children. The EEG was recorded over the entire scalp, with a linked ears reference. We found: (1) no differences between groups during resting conditions in any frequency band analyzed; (2) a left hemisphere alpha responsiveness in dyslexic children during the APD condition; and (3) a beta 2 decrease in the left posterior quadrant in dyslexics during the AP

Mapping eloquent cortex: A voxel-based lesion-symptom mapping study of core speech production capacities in brain tumour patients.

This study used voxel-based lesion-symptom mapping to examine the cortical and white matter regions associated with language production impairments in a sample of 63 preoperative tumour patients. We identified four cognitive functions considered crucial for spoken language production: semantic-to-lexical mapping (selecting the appropriate lexical label for the intended concept); phonological encoding (retrieving the word's phonological form); articulatory-motor planning (programming the articulatory motor movements); and goal-driven language selection (exerting top-down control over the words

Quantitative susceptibility mapping of deep brain nuclei in 22q11.2 deletion syndrome.

BACKGROUND: 22q11.2 Deletion Syndrome (22q11.2 DS) confers a high risk to dopamine-related disorders such as schizophrenia and Parkinson's disease. These disorders have recently been associated with abnormal iron concentrations in deep brain nuclei. In this study we hypothesized that abnormal iron concentrations may also appear in deep brain nuclei of individuals with 22q11.2 DS. METHODS: We analyzed iron concentrations in four dopamine-related nuclei (caudate, putamen, substantia nigra, and globus pallidus) of 32 individuals, including adolescents and adults, carriers of the 22q11.2 DS and 4

Mapping genetic convergence across brain structure, mental health, and cardiometabolic disease.

BACKGROUND: Individuals with psychiatric disorders frequently experience comorbid cardiometabolic conditions, complicating treatment and worsening health outcomes. Both psychiatric and cardiometabolic disorders have been individually associated with alterations in brain structure. Yet, it remains unclear whether these associations stem from a shared genetic basis that underlies their frequent co-occurrence. METHODS: We analyzed genome-wide association summary statistics from large international consortia of individuals of European ancestry, including psychiatric disorder GWAS with case-contro

Cerebral arterial mapping in the equine brain: a detailed anatomical study.

INTRODUCTION: Detailed knowledge of the cerebral arterial system in equine brain is required for advancing neuroanatomical understanding and supporting the interpretation of neurological disorders. The equine cerebrum, characterized by extensive cortical organization and functional specialization, requires a continuous arterial blood supply; however, detailed information on its arterial anatomy remains limited. This study aimed to describe the arterial supply to the equine cerebrum, including the origin, branching patterns, and distribution of the major cerebral arteries. METHODS: Ten adult h

[Tablet-based EloQ software for presenting tasks during brain function mapping].

UNLABELLED: Functional mapping of cortex and white matter tracts is important in surgery for epilepsy and supratentorial tumors. One of the limitations for speech mapping is difficult standardization, selection and presentation of stimuli during cortical stimulation. OBJECTIVE: To develop a user-friendly tablet application for creation of customized stimulus packages specific to speech cortex and white matter tracts for their presentation during mapping. RESULTS: The EloQ application contains predefined task packages for mapping of non-motor cortical functions of dominant hemisphere. It is s

Brain substrates of visual scene memory: a lesion-behavior mapping study.

BACKGROUND: The brain network supporting visual-scene memory includes ventral (what) and dorsal (where, how) visual processing streams, and hubs within the medial temporal cortex, plus frontal, temporal, and parietal regions comprising the core-recollection memory system. However, the exact relationship between these regions and the capacity to memorize different types of visual-scene elements remains debated. Functional neuroimaging studies point to dissociable as well as common network components supporting perception and memory of different aspects of visual information. In the current neur

Gut-brain nexus: Mapping multimodal links to neurodegeneration at biobank scale.

Alzheimer's disease (AD) and Parkinson's disease (PD) are influenced by genetic and environmental factors. We conducted a biobank-scale study to (i) identify endocrine, nutritional, metabolic, and digestive disorders with potential causal or temporal associations with AD/PD risk before diagnosis; (ii) assess plasma biomarkers' specificity for AD/PD in the context of co-occurring gut related traits and disorders; and (iii) integrate multimodal datasets to enhance AD/PD prediction. Our findings show that several disorders were associated with increased AD/PD risk before diagnosis, with variation

Functional Brain Mapping of Body Size Estimation Using a 3D Avatar.

BACKGROUND: Body size estimation-the ability to judge the size and shape of one's own body-is a key perceptual component of body image. However, its neural basis, and the basis for inter-individual differences in accuracy, remain poorly understood, partly due to limitations in existing assessment tools. METHODS: We developed Somatomap 3D, an interactive fMRI-compatible task allowing participants to manipulate a rotatable 3D avatar by adjusting the size and shape of 26 individual body parts to match their perceived body. Twenty-eight healthy male and female adults completed the task during fMR

Anatomical mapping of whole-brain monosynaptic inputs to the orbitofrontal cortex.

The orbitofrontal cortex (ORB) exhibits a complex structure and diverse functional roles, including emotion regulation, decision-making, and reward processing. Structurally, it comprises three distinct regions: the medial part (ORBm), the ventrolateral part (ORBvl), and the lateral part (ORBl), each with unique functional attributes, such as ORBm's involvement in reward processing, ORBvl's regulation of depression-like behavior, and ORBl's response to aversive stimuli. Dysregulation of the ORB has been implicated in various psychiatric disorders. However, the neurocircuitry underlying the func

Mapping a brain parasite: occurrence and spatial distribution in fish encephalon.

Parasites, especially brain-encysting trematodes, can have an impact on host behaviour, facilitating the transmission to next host and completion of the life cycle, but insufficient research has been done on whether specific brain regions are targeted. Using Cardiocephaloides longicollis as a laboratory model, the precise distribution of metacercariae in experimentally-infected, wild and farmed fish was mapped. The brain regions targeted by this parasite were explored, also from a histologic perspective, and potential pathogenic effects were evaluated. Experimental infections allowed to reprod

Mapping Dysfunctional Circuits in the Frontal Cortex Using Deep Brain Stimulation.

Frontal circuits play a critical role in motor, cognitive, and affective processing - and their dysfunction may result in a variety of brain disorders. However, exactly which frontal domains mediate which (dys)function remains largely elusive. Here, we study 534 deep brain stimulation electrodes implanted to treat four different brain disorders. By analyzing which connections were modulated for optimal therapeutic response across these disorders, we segregate the frontal cortex into circuits that became dysfunctional in each of them. Dysfunctional circuits were topographically arranged from oc

Mapping dopaminergic projections in the human brain with resting-state fMRI.

The striatum receives dense dopaminergic projections, making it a key region of the dopaminergic system. Its dysfunction has been implicated in various conditions including Parkinson's disease (PD) and substance use disorder. However, the investigation of dopamine-specific functioning in humans is problematic as current MRI approaches are unable to differentiate between dopaminergic and other projections. Here, we demonstrate that 'connectopic mapping' - a novel approach for characterizing fine-grained, overlapping modes of functional connectivity - can be used to map dopaminergic projections
