Modulatory Effects of Buspirone on Psychostimulant-Induced Behavioral Alterations under Chronic Restraint Stress in Rats.

Psychostimulants such as methylphenidate (MPH) and modafinil (MOD) are effective in attention-deficit/hyperactivity disorder and narcolepsy but may induce behavioral activation, anxiety-like responses, and cognitive alterations, particularly under stress. Buspirone (BUS), a nonsedative anxiolytic acting as a partial 5-HT1A receptor agonist, may influence stimulant-related behavioral effects. This study evaluated whether BUS modifies behavioral outcomes induced by chronic MPH and MOD administration in rats exposed to repeated restraint stress. Adult male Sprague-Dawley rats received oral MPH (1

Buspirone administration: Influence on growth, spawning, immune response, and stress in female goldfish (Carassius auratus).

The current study evaluated the impact of buspirone supplementation on the growth, physiology, stress response, spawning, and immunity in female goldfish (Carassius auratus). For this purpose, buspirone was dissolved in absolute methanol and sprayed onto the feed to create four experimental groups: B0 (control), B25 (25 mg kg-1), B50 (50 mg kg-1), and B100 (100 mg kg-1). Fish were fed their respective diets for 56 days and subjected to stress using the air exposure method at the end of the experiment. Growth performance analysis revealed that fish in the B100 group exhibited significantly high

Buspirone hydrochloride: a potential regulator of PD-L1 for enhanced antitumor activity in melanoma-bearing mice.

PD-1/PD-L1 blockade therapy shows good efficacy in melanoma treatment. Yet, most patients still exhibit poor responses. Exploring more effective drugs remains worthwhile. In contrast to developing new drugs which entails a lengthy process, high costs and uncertainties, repurposing old drugs is regarded as a promising and safe method, attracting greater attention. In this study, we evaluated the anti-melanoma effect of buspirone hydrochloride, a novel anti-anxiety drug. The results demonstrated that buspirone hydrochloride effectively restrained cell proliferation and migration and decreased th

First Use of a New Device for Administration of Buspirone and Acetaminophen to Suppress Shivering During Therapeutic Hypothermia.

Therapeutic hypothermia or targeted temperature management has been used after cardiac arrest to improve neurological outcomes and mortality. However, a side effect of temperature modulation is a centrally mediated shivering response. The Columbia Anti-Shivering Protocol sets up a systematic method of intravenous (IV) and oral medication escalation to suppress this response and preserve the benefits of this therapy. We present the case of a 59-year-old male who began shivering after therapeutic hypothermia for cardiac arrest, leading to a persistent rise in core temperature despite adequate se

Development of high performance liquid chromatography method for buspirone in rabbit serum: Application to pharmacokinetic study.

A simple and sensitive high performance liquid chromatographic (HPLC) method for quantification of buspirone (BUSP) in rabbit serum was developed and validated. BUSP and internal standard (IS), diltiazem hydrochloride were extracted into dichloromethane and separated using an isocratic mobile phase, on a Kromasil C(8) column. The eluent was monitored by UV detector at 235 nm and at a flow rate of 1.0 mL min(-1). The linearity range of proposed method was 1-3000 ng mL(-1). The intra-day and inter-day coefficient of variation and percent error values of the assay method were less than 15% and me

Role of sex and sex steroids in mediating pituitary-adrenal responses to acute buspirone treatment in sheep.

Systematic characterisation of sex differences in the serotonergic modulation of the hypothalamic-pituitary-adrenal (HPA) axis may assist with our understanding of why stress-related disorders are disproportionately represented in women. In this study, we examined the acute effects of buspirone, a serotonergic 1A receptor subtype agonist, on the endocrine endpoints of adrenocorticotrophin (ACTH) and cortisol secretion in gonadectomised male and female sheep. Each sheep was treated with an acute i.v. injection containing vehicle or buspirone (0.03, 0.1 and 0.3 mg/kg) in the presence and absence

Buspirone Enhances Cell Survival and Preserves Structural Integrity during Oxidative Injury to the Retinal Pigment Epithelium.

Chronic oxidative stress impairs the normal functioning of the retinal pigment epithelium (RPE), leading to atrophy of this cell layer in cases of advance age-related macular degeneration (AMD). The purpose of our study was to determine if buspirone, a partial serotonin 1A (5-HT1A) receptor agonist, protected against oxidative stress-induced changes in the RPE. We exposed differentiated human ARPE-19 cells to paraquat to induce oxidative damage in culture, and utilized a mouse model with sodium iodate (NaIO3)-induced oxidative injury to evaluate the effect of buspirone. To investigate buspiron

Effect of Buspirone on Upper Gastrointestinal Disorders of Gut-Brain Interaction: A Systematic Review and Meta-analysis.

BACKGROUND/AIMS: Buspirone shows promise in treating disorders of gut-brain interaction (DGBIs), particularly functional dyspepsia. However, findings have been mixed. METHODS: We systematically searched for prospective studies testing buspirone for any upper gastrointestinal DGBI in 4 databases (Cochrane, PubMed, Scopus, and PsycInfo). The primary outcome was any validated measure of gastrointestinal symptoms. Anxiety, depression and adverse events were secondary outcomes. For randomized controlled trials (RCTs), we performed random-effects meta-analysis of the standardized mean difference (S

Modulation of Cytochrome P450 Activity by 18β-Glycyrrhetic Acid and its Consequence on Buspirone Pharmacokinetics in Rats.

The aim of this study was to elucidate the inhibition mechanism of 18β-glycyrrhetic acid (GLY) on cytochrome P450 (CYP) activity and in vivo pharmacokinetic consequences of single GLY dose in rats. An in vitro CYP inhibition study in rat liver microsomes (RLM) was conducted using probe substrates for CYPs. Then, an in vivo pharmacokinetics of intravenous and oral buspirone (BUS), a probe substrate for CYP3A, was studied with the concurrent administration of oral GLY in rats. In the in vitro CYP inhibition study, CYP3A was involved in the metabolism of GLY. Moreover, GLY inhibited CYP3A activit

The anxiolytic buspirone shifts coping strategy in novel environmental context of mice with different anxious phenotype.

Patients suffering from anxiety disorders show increased fear when encounter a novel environment. Rodents, placed in new environmental context may respond either with increased novelty seeking (active), or enhanced anxiety (passive coping style), which may depend on the trait anxiety of the animal. Here, the connection between the initial level of anxiety and the behavioral responses in a novel environment was investigated. Two inbred mouse strains having either high- or low-anxiety related behavior (AX and nAX) were exposed to elevated plus maze (EPM), a standard test for assessing anxiety le

Effects of maternal ethanol consumption and buspirone treatment on 5-HT1A and 5-HT2A receptors in offspring.

In utero ethanol exposure results in a decreased concentration of serotonin (5-HT) in brain regions containing the cell bodies of 5-HT neurons and their cortical projections. The concentration of 5-HT reuptake sites is also reduced in several brain areas. The present study extended prior work by evaluating the effects of chronic maternal ethanol consumption and maternal buspirone treatment on 5-HT1A and 5-HT2A receptors in multiple brain areas of offspring. Receptors were quantitated early in postnatal development and at an age when the 5-HT networks are normally well-established. Because feta

Effects of buspirone on the resistance, development and passive transfer of immunity to Listeria monocytogenes in mice submitted to stress.

Mice exposed to a chronic auditory stressor and treated chronically with buspirone (1 mg/kg) showed a reduction in stress-induced suppression of the resistance and development of immunity to Listeria monocytogenes. Attempts to passively transfer immunity with spleen cells were also performed. Stressed, immunized mice had a reduced capacity to transfer immunity passively to nonimmunized mice and buspirone was found to partially suppress this inhibitory effect of stress.

Comparative assessment of efficacy and withdrawal symptoms after 6 and 12 weeks' treatment with diazepam or buspirone.

Fifty-one out-patients presenting with generalised anxiety disorder were included in a double-blind trial, and treated with either buspirone (a new non-benzodiazepine antianxiety drug) or diazepam over 6 or 12 weeks, after which they were abruptly withdrawn and continued on placebo to 14 weeks. Ratings of anxiety and other symptoms were administered fortnightly and additional withdrawal symptoms noted. Forty patients completed the study; 8 of the 11 drop-outs were taking buspirone. Both drugs reduced anxiety, diazepam more rapidly, but with greater withdrawal symptoms, particularly after 6 wee

A bedside-savior for insomnia and anxiety disorders: melatonin/buspirone hydrochloride compound oral fast-dissolving film.

Insomnia and anxiety disorders are highly prevalent conditions that significantly impair daily functioning. Notably, a bidirectional relationship exists between these two disorders: heightened cortical excitability secondary to anxiety disrupts sleep onset mechanisms, while prolonged sleep insufficiency impairs prefrontal cortical regulation of emotional processing, thereby exacerbating anxiety symptoms. This creates a vicious cycle that worsens both conditions. To address this clinical challenge, we developed a combination therapy using two pharmacological agents with complementary mechanisms

Buspirone for management of dyspnea in cancer patients receiving chemotherapy: a randomized placebo-controlled URCC CCOP study.

PURPOSE: Cancer-related dyspnea is a common, distressing, and difficult-to-manage symptom in cancer patients, resulting in diminished quality of life and poor prognosis. Buspirone, a non-benzodiazepine anxiolytic which does not suppress respiration and has proven efficacy in the treatment of generalized anxiety disorder, has been suggested to relieve the sensation of dyspnea in patients with COPD. The main objective of our study was to evaluate whether buspirone alleviates dyspnea in cancer patients. METHODS: We report on a randomized, placebo-controlled trial of 432 patients (mean age 64, fe
