Myeloid cell IL-15 production in the brain supports bystander CD8+ T cell neuropathic immune responses following virus infection.

The central nervous system (CNS) includes a uniquely regulated immune response that supports homeostasis, response to injury, and response to pathogens. Recent work has shown that virus-associated immune responses in the CNS may contribute to neuronal injury and long-term outcomes such as neurocognitive decline. However, the fundamental mechanisms that regulate acute infiltration of immune cells from vascular compartments into the CNS are not well defined. Using an attenuated Venezuelan equine encephalitis virus TC83 (referred to as TC83) to inoculate via olfactory and intracranial injections,

Soluble factors released by peripheral blood-derived CAR-NK cells cause bystander myeloid cell activation.

INTRODUCTION: CAR-T cell therapy is associated with life-threatening inflammatory toxicities, partly due to the activation and secretion of inflammatory cytokines by bystander myeloid cells (BMCs). However, due to limited clinical data, it is unclear whether CAR-NK cells cause similar toxicities. METHODS: We characterized the soluble factors (SFs) released by activated human CAR-T and CAR-NK cells and assessed their role in BMC activation (BMCA). RESULTS: We found that SFs from both activated, peripheral blood-derived CAR-T (PB-CAR-T) and CAR-NK (PB-CAR-NK) cells induced BMCA; however, PB-CA

NADPH Oxidase as a Target for Modulation of Radiation Response; Implications to Carcinogenesis and Radiotherapy.

BACKGROUND: Radiotherapy is a treatment modality for cancer. For better therapeutic efficiency, it could be used in combination with surgery, chemotherapy or immunotherapy. In addition to its beneficial therapeutic effects, exposure to radiation leads to several toxic effects on normal tissues. Also, it may induce some changes in genomic expression of tumor cells, thereby increasing the resistance of tumor cells. These changes lead to the appearance of some acute reactions in irradiated organs, increased risk of carcinogenesis, and reduction in the therapeutic effect of radiotherapy. DISCUSSI

Analyses of gender disparities in receipt of bystander cardiopulmonary resuscitation after out-of-hospital cardiac arrests by patient's age group and bystander category.

AIM: To identify factors contributing to gender disparities in the receipt of bystander cardiopulmonary resuscitation (CPR), we examined patient age group and bystander category. METHODS: We analyzed witnessed out-of-hospital cardiac arrests of presumed cardiac etiology from a nationwide Japanese registry (2005-2023). Gender differences in receipt of bystander CPR were evaluated across five age groups (≤14, 15-49, 50-64, 65-74, ≥75 years) and five bystander categories. Logistic regression and effect modification analyses were performed to assess the relative contributions and relationships of

Repurposing payloads: next generation of antibody-drug conjugates.

Antibody-drug conjugates (ADCs) are reshaping solid tumor therapy, yet most programs converge on a narrow set of ultrapotent payloads. Trastuzumab deruxtecan demonstrates that cytotoxics abandoned due to systemic toxicity can achieve substantial benefit when redesigned for targeted delivery, controlled release, and tuned bystander effects. This perspective argues that future ADC development should prioritize pharmacologic precision, proliferation-restricted mechanisms, soft-drug pharmacokinetics, and sustained intracellular target engagement over maximal in vitro potency. The impact of vaccines on heterologous adaptive immunity.

BACKGROUND: Vaccines induce antigen-specific memory in adaptive immune cells that enables long-lived protection against the target pathogen. In addition to this, several vaccines have beneficial effects greater than protection against their target pathogen. These non-specific effects are proposed to be the result of vaccine-induced immunomodulation. In the case of bacille Calmette-Guérin (BCG) vaccine, this involves induction of innate immune memory, termed 'trained immunity', in monocytes and natural killer cells. OBJECTIVES: This review discusses current evidence for vaccine-induced immunom

Low-Dose Ionizing Radiation and Myalgic Encephalomyelitis/Chronic Fatigue Syndrome (ME/CFS): A Review of Recent Evidence and Future Research Directions Toward the Elucidation of a Metabolic, Immunologic, and Signaling Cascade.

Myalgic encephalomyelitis/chronic fatigue syndrome (ME/CFS) is an idiopathic, multisystem disorder marked by debilitating fatigue, post-exertional malaise, cognitive dysfunction and neuroinflammation. Its etiology remains unclear, yet emerging evidence implicates a complex interplay between immune dysregulation, metabolic impairment, mitochondrial bioenergetics, and environmental stressors such as viral infection or low-dose ionizing radiation (LDIR). To develop treatments for ME/CFS, it is essential to identify suitable targets for therapy. In this narrative review, we discuss recent findings

A novel strategy to enhance precise targeting of the RNA base editor mxABE.

Single nucleotide variations (SNVs) represent the most common form of pathogenic mutations in humans, while base editing technology offers an ideal solution for treating such pathogenic variants. RNA editing has become a hotspot in current gene therapy due to its reversible action, which avoids long-term risks by not permanently altering the genome, and the compact size of the editors. Among these, the mini-dCas13X.1-mediated RNA adenine base editing (mxABE) system demonstrates highly efficient RNA base editing; however, it still suffers from non-target nucleotide editing caused by the bystand

A national population-based study of patients, bystanders and contextual factors associated with resuscitation in witnessed cardiac arrest: insight from the french RéAC registry.

BACKGROUND: In out-of-hospital cardiac arrest (OHCA), bystander initiated cardiopulmonary resuscitation (CPR) increases the chance of return of spontaneous circulation and survival with a favourable neurological status. Socioeconomic disparities have been highlighted in OHCA field. In areas with the lowest average socioeconomic status, OHCA incidence increased, and bystander CPR decreased. Evaluations were performed on restricted geographical area, and European evaluation is lacking. We aimed to analyse, at a national level, the impact of area-level social deprivation on the initiation of CPR

Biology-informed reinterpretation of lattice radiotherapy using non-local bystander signalling and anatomy-aware vascular sink modelling.

Objective.Lattice radiotherapy is usually evaluated using physical dose metrics such as target coverage, peak-to-valley dose ratio (PVDR), and organ-at-risk (OAR) dose, but these metrics do not explicitly represent non-local biological signalling. This study developed a hypothesis-generating computational framework to test whether synthetic head-and-neck lattice dose patterns retain the same interpretation after non-local signalling and anatomy-aware vascular uptake are included.Approach.A reaction-diffusion model propagated ROS-like and cytokine-like signals from TOPAS-derived dose distributi

Non-targeted effects of ionizing radiation: implications for risk assessment and the radiation dose response profile.

Radiation risks at low doses remain a hotly debated topic. Recent experimental advances in our understanding of effects occurring in the progeny of irradiated cells, and/or the non-irradiated neighbors of irradiated cells (i.e., non-targeted effects associated with exposure to ionizing radiation), have influenced this debate. The goal of this document is to summarize the current status of this debate and speculate on the potential impact of non-targeted effects on radiation risk assessment and the radiation dose response profile.

Oligodendroglia in Neuromyelitis Optica Spectrum Disorder.

Neuromyelitis optica spectrum disorder (NMOSD) is an inflammatory autoimmune disease of the central nervous system, in which aquaporin-4 immunoglobulin G (AQP4-IgG) targets the water channel aquaporin-4 (AQP4) localized at astrocytic endfeet, thus triggering inflammatory lesions and tissue damage. The pathological characteristics of NMOSD are early loss of oligodendrocytes, extensive demyelination, and axonal injury. The pathogenesis of oligodendrocyte damage in NMOSD includes complement-dependent bystander effect, antibody-dependent cell-mediated cytotoxicity bystander effect, glutamate toxic

Chronic virus infection compromises memory bystander T cell function in an IL-6/STAT1-dependent manner.

Chronic viral infections are widespread among humans, with ∼8-12 chronic viral infections per individual, and there is epidemiological proof that these impair heterologous immunity. We studied the impact of chronic LCMV infection on the phenotype and function of memory bystander CD8+ T cells. Active chronic LCMV infection had a profound effect on total numbers, phenotype, and function of memory bystander T cells in mice. The phenotypic changes included up-regulation of markers commonly associated with effector and exhausted cells and were induced by IL-6 in a STAT1-dependent manner in the cont

Bystander placement of automated external defibrillators and out-of-hospital cardiac arrest outcomes: a propensity score-matched cohort study between 2021 and 2022.

Bystander automated external defibrillator (AED) placement is expected to benefit all out-of-hospital cardiac arrest (OHCA) cases, even in the absence of prehospital defibrillation. This study investigated the factors influencing bystander-AED placement and evaluated its impact on neurologically favourable outcomes in bystander-witnessed, out-of-home OHCA cases receiving bystander cardiopulmonary resuscitation (CPR). This retrospective cohort study analysed nationwide EMS-transported emergency and OHCA databases (2021-2022), including 22,443 bystander-witnessed, out-of-home OHCAs with bystande

Bystanders' willingness to assist using automated external defibrillators during cardiac arrest.

The "bystander effect," in which the presence of others inhibits rescue actions, has not been specifically examined in the context of cardiac arrest; understanding this effect in relation to rescue with automated external defibrillators (AEDs) is important. This study aims to identify the presence of others as a factor inhibiting rescue actions using an AED, from a social psychology perspective. We collected data through a web-based questionnaire involving registered residents in all 47 prefectures of Japan. The participants were presented with hypothetical scenarios of witnessing cardiac arre

Intercellular communication of cellular stress monitored by gamma-H2AX induction.

When cells are exposed to ionizing radiation (IR), unexposed cells that share media with damaged cells exhibit similar effects to irradiated cells including increased levels of DNA double-strand breaks (DSBs). Hypothesizing that this effect, known as the radiation-induced bystander effect, may be a specific instance of communication between damaged and undamaged cells regardless of damage source, we demonstrated that exposure of target cells to non-IR induces bystander damage in non-targeted cells as measured by gamma-H2AX and 53BP1 focal formation. Initially, bystander damage was found primar

Radiation-induced Non-targeted Effect and Carcinogenesis; Implications in Clinical Radiotherapy.

Bystander or non-targeted effect is known to be an interesting phenomenon in radiobiology. The genetic consequences of bystander effect on non-irradiated cells have shown that this phenomenon can be considered as one of the most important factors involved in secondary cancer after exposure to ionizing radiation. Every year, millions of people around the world undergo radiotherapy in order to cure different types of cancers. The most crucial aim of radiotherapy is to improve treatment efficiency by reducing early and late effects of exposure to clinical doses of radiation. Secondary cancer indu

Chlamydia trachomatis-infected cells and uninfected-bystander cells exhibit diametrically opposed responses to interferon gamma.

The intracellular bacterial pathogen, Chlamydia trachomatis, is a tryptophan auxotroph. Therefore, induction of the host tryptophan catabolizing enzyme, indoleamine-2,3-dioxgenase-1 (IDO1), by interferon gamma (IFNγ) is one of the primary protective responses against chlamydial infection. However, despite the presence of a robust IFNγ response, active and replicating C. trachomatis can be detected in cervical secretions of women. We hypothesized that a primary C. trachomatis infection may evade the IFNγ response, and that the protective effect of this cytokine results from its activation of tr
