Effect of calcium and phosphate in an artificial chemical model of root caries formation.

PURPOSE: To investigate the mineral density of artificial root caries lesions formed under varying concentrations of calcium phosphate, with or without the presence of fluoride. METHODS: Sixty-four human tooth root blocks were exposed to various lactic acid solutions for 96 h. These included a low concentration (0.16 mM) of calcium chloride and potassium dihydrogen phosphate (Group L), a low concentration (0.16 mM) of calcium chloride and potassium dihydrogen phosphate with 0.5 ppm fluoride (with sodium fluoride) (Group LF), a high concentration (2.2 mM) of calcium chloride and potassium dihy

The cariostatic mechanisms of fluoride.

UNLABELLED: This article discusses the possible cariostatic mechanisms of the action of fluoride. In the past, fluoride inhibition of caries was ascribed to reduced solubility of enamel due to incorporation of fluoride (F-) into the enamel minerals. The present evidence from clinical and laboratory studies suggests that the caries-preventive mode of action of fluoride is mainly topical. There is convincing evidence that fluoride has a major effect on demineralisation and remineralisation of dental hard tissue. The source of this fluoride could either be fluorapatite (formed due to the incorpor

Fluoride and bone: a second look. No use in osteoporosis.

(1) Data on fluoride efficacy in secondary prevention of osteoporotic fractures were contradictory when we last examined the file, in 1990. We now re-examine the clinical file on fluoride with more follow-up and new clinical trial data. (2) In 354 postmenopausal women with a history of vertebral fracture (secondary prevention), a comparative trial (the FAVOS trial) showed that moderate-dose fluoride for 2 years did not reduce the incidence of new vertebral fractures relative to calcium plus vitamin D. Furthermore, fluoride caused more cases of lower-limb pain. (3) In 110 postmenopausal women w

Defluoridation of water at high pH with use of brushite, calcium hydroxide, and bone char.

The aim of this study was to improve the efficiency of the bone-char method of water defluoridation by pre-treating the water with brushite and calcium hydroxide. Various amounts of brushite, calcium hydroxide, and bone char were suspended batchwise in 100 mL of distilled water containing 0.53 mmol/L fluoride for 24 h under gentle agitation. At suitable intervals, pH and the concentrations of fluoride, calcium, and phosphate in the water were determined and, when possible, the degrees of saturation with respect to brushite, hydroxyapatite, and fluorapatite calculated. Bone char used alone took

The ultrastructure of dentine from rat incisors following exposure to sodium fluoride and potassium pyroantimonate staining.

Weanling rats were given a single intraperitoneal injection of sodium fluoride and control animals normal saline for four consecutive days. The fluoride produced a consistent response in the mineralizing dentine of the incisors in which a hypermineralized band was succeeded by a hypomineralized band. Potassium pyroantimonate staining for calcium ions showed that following injection of fluoride, in contrast to the controls, there were large amounts of calcium pyroantimonate in the pre-dentine and throughout the odontoblasts. This suggests that fluoride temporarily affects the membrane enzyme sy

A computer simulation of the reactions between hydroxyapatite and fluoride in aqueous acidic solutions.

A precipitation reaction between hydroxyapatite (HAp) and fluoride solution was analyzed by computer simulation. The system comprised excess HAp and 0.05-0.2 M acetic buffer solutions containing 0.5-2.0 mM potassium fluoride. The amounts of HAp dissolved and calcium salts precipitated and activities of all the species present in the solution were calculated during the process. In the systems, calcium fluoride was initially precipitated when applying the fluoride solution to HAp, followed by precipitation of fluorapatite (FAp) and re-dissolution of the previously precipitated calcium fluoride a

Citrate-Stabilized Amorphous Calcium Phosphate Nanoparticles as an Effective Adsorbent for Defluorination.

Amorphous calcium phosphate (ACP), one of the most important calcium-phosphorus compounds, is widely used in dentistry, orthopedics, and medicine, but is rarely reported for fluoride removal from water. In view of this, sodium citrate-stabilized amorphous calcium phosphate (Cit-ACP) and Cit-ACP calcinated at different temperatures were successfully prepared for fluoride removal. The results showed that the adsorption data of the Cit-ACP sample could be well described by the Langmuir model, and the adsorption kinetic followed the pseudo-second-order model. The maximum adsorption capacity was 27

Influence of cold plasma activation on fluoride uptake and demineralization of enamel.

OBJECTIVES: This in vitro study investigated the influence of cold plasma activation time prior to 1.25% fluoride gel application on fluoride uptake and demineralization of enamel. MATERIALS AND METHODS: Human enamel specimens (n = 200) were either treated with a dielectric-barrier plasma discharge at a constant distance (d = 1 mm), pulse repetition frequency (f1 = 1 kHz), power (Pmean=36 mW) and voltage (Upp=28 kV) for 0.5, 1, 2, or 30 s or received no plasma activation (each n = 40). Subsequently, half of the specimens from each group were coated with fluoride gel (1.25% NaF/AmF, t = 5 min)

Elemental changes in tooth hard tissues during demineralization and fluoride-induced remineralization: an SEM-EDX study.

BACKGROUND: Tooth enamel and dentin are the most highly mineralized tissues in the human body, and their structural stability depends on the balanced content of calcium (Ca), phosphorus (P), and fluoride (F). Demineralization leads to the loss of these ions, weakening the tooth structure and increasing susceptibility to caries. OBJECTIVE: This study aimed to evaluate elemental changes in Ca, P, and F in intact, demineralized, and remineralized teeth across the enamel, dentin-enamel junction and peripulpal dentin. MATERIALS AND METHODS: 45 extracted human teeth were longitudinally sectioned t

Investigations into the mechanism by which sodium fluoride stimulates prostaglandin production in guinea-pig uterus.

Sodium fluoride (10 mM) caused a slow increase in the outputs of PGF-2 alpha, 6-keto-PGF-1 alpha and, to a lesser extent, PGE-2 from the Day-7 and Day-15 guinea-pig uterus superfused in vitro. This stimulatory action of sodium fluoride was not prevented by using calcium-free Krebs' solution. There was also a faster stimulation of 6-keto-PGF-1 alpha output from the Day-7 guinea-pig uterus produced by sodium fluoride, and this quicker response was abolished by using calcium-free Krebs' solution. TMB-8 (an intracellular calcium antagonist) inhibited the stimulatory action of sodium fluoride on th

Effect of a water rinse on 'labile' fluoride and other ions in plaque and saliva before and after conventional and experimental fluoride rinses.

Labile reservoirs are important in maintaining ion concentrations in oral fluids, especially after a fluoride dentifrice application, where a persistent increase in fluid fluoride can mitigate or reverse caries progression. In this study, the effect of experimental and conventional fluoride rinses on the in vitro and in vivo water-induced release of fluoride, calcium, phosphate, acetate and hydrogen ions from oral reservoirs was examined. At the start of each experiment, 13 subjects rinsed either with a conventional 228-ppm fluoride NaF rinse, a 228-ppm fluoride controlled-release rinse (CR ri

Assessment of groundwater quality in a region of endemic fluorosis in the northeast of Brazil.

The aim of this study was to estimate the risk for caries and fluorosis in a desertification area, applying the calcium/fluoride concentration ratio of underground water and the quality of water in a selected geographical region. This study was performed in the municipality of São João do Rio do Peixe, located in the tropical semiarid lands of Brazil. A total of 111 groundwater samples were collected. Fluoride concentration varied from 0.11 to 9.33 mg/L. Thirty percent of all samples analyzed showed values above 1.5 mg/L, while 64 % were above the ideal limit of 0.7 mg/L. Mean calcium concentr

Scanning electron microscope and electron microprobe study of reactions of stannous fluoride and stannous chloride with dental enamel.

Stannous fluoride but not stannous chloride has shown a caries inhibiting effect in animal studies. The effect of aqueous solutions of stannous fluoride and stannous chloride on dental enamel surfaces was investigated by scanning electron microscope and analytical electron microprobe. Some large globules and a continuous layer of small globules were observed after stannous fluoride treatment. The large globules were soluble in both water and alkali, whereas the small ones were only soluble in alkali. Both fluoride and tin were found to be present on the enamel surfaces by the electron micropro

Novel topical fluoride delivery system I: remineralization of ground bovine teeth.

Laboratory studies were carried out on a newly conceived fluoride-containing remineralizing system with bovine teeth. The prototype fluoride delivery device involved micronized calcium fluoride maintained at the tooth surface with a cellulose film. Together with salivary calcium and phosphate (or simulated saliva), this system was able to generate and maintain the appropriate thermodynamic activity driving force for significant fluorapatite deposition in a reasonably short time (approximately 48 hr).

Fluoride in the environment and its metabolism in humans.

The presence of environmental fluoride and its impact on human health is well documented. When consumed in adequate quantity, fluoride prevents dental caries, assists in the formation of dental enamels, and prevents deficiencies in bone mineralization. At excessive exposure levels, ingestion of fluoride causes dental fluorosis skeletal fluorosis, and manifestations such as gastrointestinal, neurological, and urinary problems. The distribution of fluoride in the environment is uneven and largely is believed to derive from geogenic causes. The natural sources of fluoride are fluorite, fluorapati

Relationship between intracellular Ca²⁺ and ROS during fluoride-induced injury in SH-SY5Y cells.

The mechanisms underlying the neurotoxicology of endemic fluorosis still remain obscure. To explore lactate dehydrogenase (LDH) leakage, intracellular Ca²⁺ concentration ([Ca²⁺]i ) and reactive oxygen species (ROS) production induced by fluoride, human neuroblastoma (SH-SY5Y) cells were incubated with sodium fluoride (NaF, 20, 40, 80 mg/L) for 24 h, with 40 mg/L NaF for 3, 6, 12, 18, 24 h, and N-acetyl-L-cysteine (NAC), ethyleneglycol-bis-(β-aminoethyl ether)-N,N,N',N'-tetraacetic acid (EGTA), 1,2-bis(O-aminophenoxy)ethane-N,N,N',N'-tetraacetic acid tetra(acetoxymethyl) ester (BAPTA-AM) alone

Treatment of osteoporosis in men with fluoride alone or in combination with bisphosphonates.

Alendronate has been approved as a first antiresorptive drug for treatment of osteoporosis in men. Except for fluoride, in some countries so far there has been no approved anabolic substance for the treatment of male osteoporosis. From small studies in men and male patients included in studies on postmenopausal osteoporosis there is sufficient evidence that fluoride has the same osteoblast-stimulating potency in men and women. In our own study on 64 men with idiopathic osteoporosis without prevalent fractures, a low-dose intermittent fluoride regimen (15 mg fluoride ions 3 months on, 1 month o
