Calcium and oxalate uptake by renal brush border membrane vesicles (BBMV) was examined in magnesium-deficient and pair-fed control rats. Uptake studies were carried out by rapid filtration technique and rate of influx of calcium and oxalate as a function of extravesicular concentration (0.1 nM--1.0 mM) examined. Calcium uptake by renal BBMV exhibited saturable kinetics while oxalate uptake followed a biphasic transport mechanism showing saturable kinetics at low oxalate concentrations and passive diffusion at higher concentrations. In magnesium deficiency the kinetics of calcium and oxalate up

A combined radiological, histological, and crystallographical study of bone, primarily the inferior femur, taken at autopsy from a 9 year old female with primary oxalosis, is reported. The essential feature is the deposition of calcium oxalate monohydrate (whewellite) crystals, inducing a foreign body reaction and bone remodelling. Synovial and cartilage surfaces are not involved. The changes occur primarily at sites of previous or present growth and seem to be related to the reaction of tissue to the deposition of blood-borne crystals. The radiological changes may be explained in part by the

BACKGROUND: Calcium oxalate (CaOx) stones are the most prevalent urological calculi with high incidence and recurrence. Effective preventive drugs remain lacking because of unclear pathogenesis. Although the imbalance of the dual renin-angiotensin system (RAS) axes is closely linked to oxidative stress and autophagy, its role in CaOx stone formation remains undefined.
METHODS: In vitro, human renal tubular epithelial cells were stimulated with CaOx crystals, with siRNA-mediated AT1R or MasR knockdown or pretreatment with Ang Ⅱ, Ang-(1-7), antioxidant Nacetylcysteine (NAC) or autophagy inhibit

INTRODUCTION: Desmodium styracifolium, known as Guangjinqiancao in Chinese, is a widely used traditional medicinal herb in Southern China for clearing heat and excreting dampness. It has been historically prescribed for the treatment of urological disorders, particularly urolithiasis (Shi Lin). While the clinical efficacy of D. styracifolium is well-documented, the precise mechanisms of its primary active fraction, total flavonoids of D. styracifolium (TFDS), regarding crystallization and inflammation remain to be fully elucidated. This study aims to investigate the therapeutic mechanism of TF

The increasing global prevalence of nephrolithiasis represents a significant public health concern. However, the role of environmental contaminants such as glyphosate remains unclear. Herein, epidemiological analysis, network toxicology, and experimental validation were integrated to investigate this association. Cross-sectional analysis of NHANES data demonstrated a significant dose-dependent relationship between urinary glyphosate levels and nephrolithiasis prevalence. Each unit increase in log-transformed glyphosate concentration was associated with a 25% increase in nephrolithiasis risk (O

Bile acids have been implicated in calcium oxalate (CaOx) nephrolithiasis. Here, we employ multi-omics approaches to identify isoallolithocholic acid (isoalloLCA) as the key bile acid elevated in the feces, serum, kidney, and urine of CaOx rats, confirmed by spatial metabolomics. 16S sequencing and metagenomic analyses indicate that elevated isoalloLCA levels in CaOx rats or individuals are likely of microbial origin. Mechanistically, isoalloLCA binds to PARP1 via specific molecular interactions validated by surface plasmon resonance and cellular thermal shift assays. Knockdown of PARP1 by ade

The incidence of kidney stones is increasing worldwide. However, the underlying mechanism of the process of kidney stone formation and the kidney damage caused are not well understood. Here, it is observed that Lgals3, a β-galactoside-binding protein, is significantly increased in tissues with calcium oxalate (CaOx) stones, and in both in vivo and in vitro models. Lgals3 expression is positively correlated with the deposition of CaOx crystals. Knockout of Lgals3 markedly reduces the deposition of CaOx crystal and renal fibrosis in vivo. Furthermore, Lgals3 deficiency decrease the glycolytic ra

BACKGROUND: Nephrolithiasis, a common urinary system disorder, exhibits high morbidity and recurrence rates, correlating with renal dysfunction and the increased risk of chronic kidney disease. Nonetheless, the precise role of disrupted cellular metabolism in renal injury induced by calcium oxalate (CaOx) crystal deposition is unclear. The purpose of this study is to investigate the involvement of the ubiquitin-like protein containing PHD and RING finger structural domain 1 (UHRF1) in CaOx-induced renal fibrosis and its impacts on cellular lipid metabolism.

BACKGROUND: Clinical therapeutic approaches to prevent and treat renal injury in patients with acute kidney injury (AKI) and chronic kidney disease (CKD) induced by calcium oxalate (CaOx) are limited. As a pivotal deacetylase, Sirtuin1 (Sirt1) exhibits notably anti-inflammatory effects, but its metabolic mechanism in regulating CaOx nephropathy remains unexplored.
METHODS: We analysed organic acid metabolism in kidney using the nontargeted metabolome and identified key targets by RNA-seq. Evaluate renal injury and oxidative stress using techniques such as Positron Emission Tomography-Computed

Nephrolithiasis is a prevalent urological disorder in which calcium oxalate (CaOx) stones are most common, and accumulating evidence suggests that oxidative stress and ferroptosis contribute to CaOx-induced tubular epithelial injury. In this study, glyoxylic acid–induced mouse nephrocalcinosis and CaOx-stimulated HK-2 cells were used to explore the role of gamma-glutamyltransferase 1 (GGT1), a key enzyme in glutathione metabolism. RNA sequencing and qRT-PCR analyses identified GGT1 as a significantly downregulated gene in both models, and bioinformatic prediction followed by molecular validati

Calcium oxalate (CaOx) stones account for 90% of uroliths in cats and contribute to ureteral inflammation and fibrosis, although the underlying mechanism remains unclear. Apoptosis inhibitor of macrophage (AIM) is known to play a protective role against tubular injury in feline kidney disease. This study investigated whether AIM contributes to ureteral fibrosis by using AIM-felinized mice subjected to CaOx bead-induced ureteral injury. Male C57BL/6 mice (n = 54), including wild-type mice (mA), AIM-knockout (koA) mice, and AIM-felinized mice (fA), were assigned to either a unilateral ureteral o

Urinary stone disease is a widespread health problem in both adults and children, and its prevalence has been increasing worldwide. Various plants preparations have already been used since ancient times in order to treat urolithiasis. The aim of this study is to evaluate the antioxidant capacity and litholytic effect on kidney stones of Cydonia oblonga Miller. leaves. The infusion, methanol and acetone extracts were made from Cydonia oblonga Miller. leaf at different concentration. Estimation of mass fractions of total polyphenol, flavonoid, and flavonol contents, as well as the in vitro radic

Calcium oxalate (CaOx) stones are the most common type of kidney stones and are associated with high recurrence, short chain fatty acids (SCFAs), and inflammation. However, it remains uncertain whether SCFAs affect the formation of CaOx stones through immunomodulation. We first performed mass cytometry (CyTOF) and RNA sequencing on kidney immune cells with glyoxylate-induced CaOx crystals (to elucidate the landscape of the associated immune cell population) and explored the role of SCFAs in renal CaOx stone formation through immunomodulation. We identified 29 distinct immune cell subtypes in k

Hyperoxaluria-induced calcium oxalate (CaOx) deposition is the key factor in kidney stone formation, for which adipose-derived stromal cells (ADSCs) have been used as a therapeutic treatment. Studies revealed that miR-20b-3p is down-regulated in hypercalciuric stone-forming rat kidney. To investigate whether ADSC-derived miR-20b-3p-enriched exosomes protect against kidney stones, an ethylene glycol (EG)-induced hyperoxaluria rat model and an in vitro model of oxalate-induced NRK-52E cells were established to explore the protective mechanism of miR-20b-3p. The results showed that miR-20b-3p lev

Renal calculus is a global common urological disease that is closely related to crystal adhesion and renal tubular epithelial cell impairment. Gap junctions (GJs) and their components (connexins and Cxs) are involved in various pathophysiology processes, but their roles in renal calculi progression are not well defined. Our previous RNA microarray analysis suggests that GJs are one of the key predicted pathways involved in the renal calcium oxalate (CaOx) crystal rat model. In the current study, we found that the Cx43 and Cx32 expression and the GJ function decreased significantly after stimul

BACKGROUND: Overgrowth of calcium oxalate on Randall's plaque is a mechanism of stone formation among idiopathic calcium oxalate stone-formers (ICSFs). It is less clear how stones form when there is little or no plaque.
DESIGN, SETTING, PARTICIPANTS, & MEASUREMENTS: Participants were a consecutive cohort of ICSFs who underwent percutaneous nephroscopic papillary mapping in the kidney or kidneys containing symptomatic stones and a papillary tip biopsy from a representative calyx during a stone removal procedure between 2009 and 2013. The distribution of Randall's plaque coverage was analyzed a

This study examined the potential antilithic effects of a traditional Chinese medicine Urtica dentata Hand (UDH) in experimental rats and screened the optimal extract of UDH as a possible therapeutic agent for kidney stones. The rat model of urinary calcium oxalate stones was induced by intragastric (i.g.) administration of 2 mL of 1.25% ethylene glycol (EG) and 1% ammonium chloride (AC) for 28 days and was confirmed by Color Doppler ultrasound imaging. The rats in different experimental groups were then intragastrically given petroleum ether extract (PEE), N-butanol extract (NBE), aqueous ext

BACKGROUND: The tubular fluid of the mammalian kidney is often supersaturated with mineral salts, but crystallization rarely occurs under normal conditions. The unique ability of the kidney to avoid harmful crystal formation has long been attributed to the inhibitory activity of the urinary macromolecules, although few in vivo studies have been carried out to examine this hypothesis. Here we examined the role of Tamm-Horsfall protein (THP), the principal urinary protein, in urinary defense against renal calcium crystal formation, using a THP knockout model that we recently developed.

Calcium plays a crucial role in fruit development and quality formation, yet the regulatory mechanisms underlying calcium distribution and oxalic acid metabolism in litchi remain poorly understood. In this study, the effects of exogenous calcium on calcium accumulation, fruit quality, and oxalic acid metabolism in litchi were investigated through stem infusion experiments using calcium chloride (CaCl2) and ethylene glycol-bis(2-aminoethylether)-N,N,N',N'-tetraacetic acid (EGTA) treatments. The results demonstrated that exogenous CaCl2 application significantly enhanced calcium accumulation in

Oxalate severely impairs calcium and iron bioavailability by forming insoluble precipitates in the gastrointestinal tract, posing challenges for mineral nutrition in plant-based diets. This study investigated the thermodynamic and kinetic interactions between calcium, iron, and oxalate, and explored whether digested casein could enhance mineral bioavailability under oxalate challenge. Thermodynamic analysis revealed that iron(II) oxalate exhibited higher solubility product (Ksp) and weaker association (Ka = 4655) compared to calcium oxalate (Ka = 25126) at 25 ℃. Conductometric titration demons
