Chronic inflammatory discoid lupus erythematosus (DLE) and Jessner's lymphocytic infiltration of the skin (LIS) are both characterized by dermal infiltrates of activated T lymphocytes. However, an inflammatory involvement of the epidermis is only found in DLE. We therefore compared the phenotypic properties of the keratinocytes using immunohistochemical stainings of biopsies from typical DLE and LIS. Keratinocytes failed to express HLA-DR in LIS and surprisingly also in DLE. The adhesion molecule ICAM-1 was only expressed in DLE, with focal staining of the basal keratinocytes in close associat

The immune response to an allergen is not only dependent on the inflammatory stimulus, but also on the genetic disposition of the individual. Important effector cells in the immune response are myelomonocytic cells in their various differentiation stages. We recently described the expression of MRP8 and MRP14, two calcium-binding proteins of the S-100 family, by these cells during inflammatory activation. Here, we investigated whether their expression in murine contact dermatitis is dependent on the stimulus by which dermatitis is elicited, and if it is related to the genetic constitution of d

Protein aggregation in the form of amyloid fibrils has long been associated with the onset and development of various amyloidoses, including Alzheimer's, Parkinson's or prion diseases. Recent studies of their fibril formation process have revealed that amyloidogenic protein cross-interactions may impact aggregation pathways and kinetic parameters, as well as the structure of the resulting aggregates. Despite a growing number of reports exploring this type of interaction, they only cover just a small number of possible amyloidogenic protein pairings. One such pair is between two neurodegenerati

Age-related macular degeneration (AMD) is a complex disease in which inflammation is implicated as a key factor but the precise molecular mechanisms are poorly understood. AMD lesions contain an excess of the pro-inflammatory S100A9 protein, but its retinal significance was yet unexplored. S100A9 was shown to be intrinsically amyloidogenic in vitro and in vivo. Here, we hypothesized that the retinal effects of S100A9 are related to its supramolecular conformation. ARPE-19 cultures were treated with native dimeric and fibrillar S100A9 preparations, and cell viability was determined. Wild-type r

BACKGROUND: MRP8/14, a calcium-binding protein of the S100 family, is predominantly expressed in myeloid cells and exhibits proinflammatory and prothrombotic properties. Platelet-neutrophil interactions can trigger MRP8/14 release, but their role in atherosclerosis (AS) remains unclear.
AIMS: To investigate the effect of MRP8/14 on AS progression and the underlying mechanisms involved, focusing on neutrophil activation and the toll-like receptor 4 (TLR4)-ERK1/2-p90RSK and NRF2-ARE pathways.
STUDY DESIGN: Ex vivo and animal study.
METHODS: Neutrophils isolated from mouse bone marrow were sti

BACKGROUND AND AIM: Calprotectin (CLP) is a heterodimeric complex formed by two S100 proteins (S100A8/A9), which plays a pivotal role in innate immunity. Due to its intrinsic cytotoxic and proinflammatory properties, CLP controls cell differentiation, proliferation and NETosis and has been associated with a wide range of rheumatic diseases. Our review summarizes the widespread interest in circulating CLP (cCLP) as a biomarker of neutrophil-related inflammation, in autoimmune rheumatic disease (ARD) and non-ARD.
METHODS: A thorough literature review was performed using PubMed and EMBASE databa

NEW FINDINGS: What is the central question of this study? Are carotid bodies (CBs) modulated by the damage-associated molecular patterns (DAMPs) and humoral factors of aseptic tissue injury? What are the main findings and their importance? DAMPs (HMGB1, S100 A8/A9) and blood plasma from rats subjected to tibia surgery, a model of aseptic injury, stimulate the release of neurotransmitters (ATP, dopamine) and TNF-α from ex vivo rat CBs. All-thiol HMGB1 mediates upregulation of immune-related biological pathways. These data suggest regulation of CB function by endogenous mediators of innate immun

Calprotectin (S100A8/A9), a heterodimeric complex of calcium-binding proteins S100A8 and S100A9, is encoded by genes mapping to the chromosomal locus 1q21.3 of the epidermal differentiation complex. Whereas extracellular calprotectin shows proinflammatory and antimicrobial properties by signaling through RAGE and TLR4, intracytoplasmic S100A8/A9 appears to be important for cellular development, maintenance, and survival. S100A8/A9 is constitutively expressed in myeloid cells and the stratified mucosal epithelia lining the oropharyngeal and genitourinary mucosae. While upregulated in adenocarci

To shed light on the early immune response processes in severed peripheral nerves, we performed genome-wide transcriptional profiling and bioinformatics analyses of the proximal (P, regenerating) and distal (D, degenerating) nerve stumps on day 1 in the sciatic nerve axotomy model in rats. Multiple cell death-related pathways were activated in the degenerating D stump, whereas activation of the cytoskeletal motility and gluconeogenesis/glycolysis pathways was most prominent in the P stump of the axotomized nerve. Our bioinformatics analyses also identified the specific immunomodulatory genes o

Aging is the most important risk factor for Parkinson's disease (PD). S100A9, a calcium-binding protein, is closely related to a variety of aging-related diseases, but its role in the pathogenesis of PD is still unclear. This study aims to investigate the role of S100A9 in aging-related mechanisms in PD. C57BL/6J mice were intraperitoneally injected with 1-methyl-4-phenyl-1,2,3, 6-tetrahydropyridine (MPTP; 15 mg/kg four times daily), followed by Paquinimod (a S100A9 inhibitor; 7 mg/kg, once a day for 7 days after model establishment, totaling 8 doses). We found that MPTP induced significant mo

Periodontitis drives irreversible destruction of periodontal tissue and is prone to exacerbating inflammatory disorders. Systemic immunomodulatory management continues to be an attractive approach in periodontal care, particularly within the context of 'predictive, preventive, and personalized' periodontics. The present study incorporated genetic proxies identified through genome-wide association studies for circulating immune cells and periodontitis into a comprehensive Mendelian randomization (MR) framework. Univariable MR, multivariable MR, subgroup analysis, reverse MR, and Bayesian model

Renal interstitial fibrosis is a key pathological feature associated with the progression of chronic kidney disease (CKD) progression. Macrophages play a central role in renal inflammation and fibrosis, and the S100 calcium binding protein A8/A9 (S100A8/A9) protein that they express participates in the immune response as an important regulatory inflammatory factor. Therefore, this study aimed to investigate the role of S100A8/A9 in M2-driven macrophage-to-myofibroblast transition (MMT) during progressive fibrosis in patients with CKD. We analyzed renal biopsy tissues from patients with CKD, co

Renal fibrosis, a pivotal pathological process in chronic kidney disease (CKD), is closely associated with inflammatory responses and gut microbiota dysbiosis. Curcumin (CUR), a natural polyphenol derived from turmeric, shows anti-inflammatory and microbiota-modulating properties. However, its potential to attenuate renal fibrosis via gut microbiota-derived metabolites remains unclear. In this study, we investigated the mechanisms underlying the renoprotective effects of CUR, using a murine model of unilateral ureteral obstruction and LPS-stimulated bone marrow-derived macrophages (BMDMs). CUR

BACKGROUND & AIMS: Biliary atresia (BA) is a progressive neonatal cholangiopathy characterized by unresolved inflammation and neutrophil extracellular traps (NET) formation. Although neutrophils contribute to bile duct injury, the upstream mechanisms driving their activation remain poorly defined. This study investigated whether platelet (PLT) hyperactivation promotes NET-mediated inflammation in BA and evaluated the therapeutic potential of targeting S100A8/A9.
METHODS: The role of PLT hyperactivation in neutrophil-mediated inflammation in BA was evaluated through retrospective clinical revi

BACKGROUND: Veno-arterial extracorporeal membrane oxygenation (VA-ECMO) is a life-saving intervention for refractory cardiogenic shock but is frequently complicated by thromboinflammation, leading to high morbidity and mortality. Neutrophil extracellular traps (NETs) and S100A8/A9 are involved in the thromboinflammatory process, yet their value as prognostic biomarkers for adverse outcomes in VA-ECMO patients remains to be further elucidated.
OBJECTIVE: This study aimed to investigate the dynamic changes of NETs and S100A8/A9 in VA-ECMO patients and evaluate their predictive value for adverse

Granular parakeratosis (GP) is an uncommon keratinization disorder that predominantly affects intertriginous areas and may clinically mimic atopic dermatitis or psoriasis. Histologically, GP is characterized by compact hyperkeratosis, parakeratosis, and retention of keratohyalin granules within the stratum corneum, although its histopathological spectrum is heterogeneous. Previous studies have consisted mainly of case reports, while mechanistic investigations remain limited, and whether GP represents a purely cornification-related abnormality is unclear. Photobiomodulation (PBM) modulates inflammation and promotes tissue repair. S100A8/A9 is a neutrophil-derived calcium-binding heterodimer that acts as a pro‑inflammatory damage‑associated molecular pattern, serving as a biomarker of inflammation. This study aimed to evaluate the effects of PBM at 660 nm and 808 nm wavelengths on S100A8/A9 levels in pulp blood (PB) and gingival crevicular fluid (GCF) in patients with symptomatic irreversible pulpitis (SIP). Patients diagnosed with SIP were block-randomised into three groups: Group 1 received 660 nm PBM (n = 14), Group 2 received 808 nm PBM (n =
