Quantification of zinc intoxication of Candida glabrata after phagocytosis by primary macrophages.

Zinc (Zn2+) is a trace element, playing pivotal roles during host-pathogen interactions. Macrophages can sequester Zn2+ and restrict bioavailability or increase phagolysosomal Zn2+ to kill pathogens. This method quantifies Zn2+-mediated clearance of the human fungal pathogen C. glabrata after phagocytosis by innate immune cells. Double staining with propidium iodide and a zinc-specific fluorescence dye allows for discrimination of live versus dead pathogens inside phagolysosomes. Moreover, elevated phagolysosomal Zn2+ decreases fungal viability as a function of intracellular Zn2+ concentration

Highly specific and rapid molecular detection of Candida glabrata in clinical samples.

The most common nosocomial fungal infections are caused by several species of Candida, of which Candida glabrata is the second most frequently isolated species from bloodstream infections. C. glabrata displays relatively high minimal inhibitory concentration values (MIC) to the antifungal fluconazole and is associated with high mortality rates. To decrease mortality rates, the appropriate treatment must be administered promptly. C. glabrata contains in its genome several non-identical copies of species-specific sequences. We designed three pairs of C. glabrata-specific primers for endpoint PCR

The involvement of the Candida glabrata trehalase enzymes in stress resistance and gut colonization.

Candida glabrata is an opportunistic human fungal pathogen and is frequently present in the human microbiome. It has a high relative resistance to environmental stresses and several antifungal drugs. An important component involved in microbial stress tolerance is trehalose. In this work, we characterized the three C. glabrata trehalase enzymes Ath1, Nth1 and Nth2. Single, double and triple deletion strains were constructed and characterized both in vitro and in vivo to determine the role of these enzymes in virulence. Ath1 was found to be located in the periplasm and was essential for growth

Genetic Basis of Azole and Echinocandin Resistance in Clinical Candida glabrata in Japan.

Infections caused by Candida glabrata have caused worldwide concern, especially when they are associated with increasing echinocandin and azole resistance. In this study, we analyzed the molecular mechanisms of azole and echinocandin resistance in C. glabrata isolates obtained from hospitalized patients in Japan from 1997 to 2019. All isolates were checked phenotypically for resistance and genotypically for mutations in PDR1, ERG11, hot spot 1 (HS1), HS2, and HS3 of FKS1, and HS1 and HS2 of FKS2, and all isolates were genotyped by multilocus sequence typing (MLST). Interestingly, 32.6% of the

Multi-route antifungal administration in the management of urinary Candida glabrata bezoar.

A 65-year-old lady was admitted with urosepsis and imaging suggesting right sided hydronephrosis secondary to a filling defect consistent with a fungal bezoar. An indwelling urinary catheter and a right percutaneous nephrostomy tube were inserted. Candida glabrata cultured from urine was resistant to fluconazole. Amphotericin B was instilled into the renal pelvis via the nephrostomy tube while intravenous liposomal amphotericin was administered daily along with oral flucytosine. This multi-modal antifungal administration was continued for 14 days. Clinical and biochemical improvement was achie

Genome-Wide Response to Drugs and Stress in the Pathogenic Yeast Candida glabrata.

Candida glabrata is the second most common cause of candidemia worldwide and its prevalence has continuously increased over the last decades. C. glabrata infections are especially worrisome in immunocompromised patients, resulting in serious systemic infections, associated to high mortality rates. Intrinsic resistance to azole antifungals, widely used drugs in the clinical setting, and the ability to efficiently colonize the human host and medical devices, withstanding stress imposed by the immune system, are thought to underlie the emergence of C. glabrata. There is a clear clinical need to u

The MNN2 Gene Knockout Modulates the Antifungal Resistance of Biofilms of Candida glabrata.

Candida glabrata biofilms are recognized to have high resistance to antifungals. In order to understand the effect of mannans in the resistance profile of C. glabrata mature biofilms, C. glabrata Δmnn2 was evaluated. Biofilm cell walls were analysed by confocal laser scanning microscopy (CLSM) and their susceptibility was assessed for fluconazole, amphotericin B, caspofungin, and micafungin. Crystal violet and Alcian Blue methods were performed to quantify the biomass and the mannans concentration in the biofilm cells and matrices, respectively. The concentration of β-1,3 glucans was also meas

Micafungin versus caspofungin in the treatment of Candida glabrata infection: a case report.

BACKGROUND: Micafungin and caspofungin, which are both echinocandins, elicit their antifungal effects by suppressing the synthesis of β-D-glucan, an essential component of fungal cell walls. If micafungin is not effective against a fungal infection, is it unreasonable to switch to caspofungin? CASE PRESENTATION: An 80-year-old Asian man presented to our hospital with brain and lung abscesses. Klebsiella pneumonia and Escherichia coli were identified by sputa culture and Streptococcus mitis was identified in the brain abscess culture obtained by drainage surgery. He was treated with antibiotic

Natural product based approaches to overcome Candida glabrata and emerging AMR threats.

The rise of C. glabrata as a serious, multidrug-resistant organism poses a significant and global challenge to the human health. The reasons C. glabrata has developed resistance to standard antifungal drugs, include the activation of efflux pumps, the production of biofilms, and changes in ergosterol biosynthesis. In light of the threat posed by C. glabrata, the potential of phytochemicals as therapeutic alternatives should be considered due to their diverse structures and ability to exhibit more than one type of antifungal activity. This review summarizes advances in the use of plant-based na

P4-ATPase Drs2 regulates virulence, stress adaptation, and autophagy in Candida glabrata.

Candida glabrata is an opportunistic pathogen with increasing clinical significance due to its innate antifungal resistance. This study reveals the critical role of the phospholipid flippase Drs2 (P4-ATPase) in maintaining fungal pathogenicity and stress adaptation. The DRS2 deletion mutant exhibited impaired growth, heightened drug susceptibility, and defective stress responses, including compromised oxidative stress tolerance and autophagy. Importantly, the mutant showed reduced survival in macrophages while eliciting stronger proinflammatory responses, along with attenuated virulence in a G

Candida glabrata associated primary pleuro-pulmonary infections: A report of two cases.

Candida glabrata, a respiratory tract commensal, is an opportunistic pathogen that rarely causes respiratory disease. Primary pleuropulmonary infection caused by Candida glabrata without systemic involvement is extremely uncommon. We report two cases of proven Candida glabrata-associated pleuropulmonary infections in poorly controlled diabetics. A 51-year-old lady with a lung abscess had Candida glabrata identified from a biopsy specimen. Another 42-year-old man had necrotizing pneumonia and empyema due to Candida glabrata identified from repeated pleural fluid cultures. Both patients showed r

Antibiofilm Activity of PDMS/TiO2 against Candida glabrata through Inhibited Hydrophobic Recovery.

Coatings with antibiofilm properties are desirable for biomedical applications. Titanium dioxide (TiO2) has been explored as an antimicrobial agent in materials development primarily due to it being an excellent photocatalyst. Candida glabrata (C. glabrata) is an emerging human fungal pathogen with known high resistance to oxidative stress. Here, we fabricated a polydimethylsiloxane/titanium dioxide (PDMS/TiO2) nanocomposite coating and tested its antibiofilm activities against C. glabrata. The resulting nanocomposite exhibited >50% reduction in C. glabrata biofilm formation with 2.5 wt % TiO2
