Perfluorooctanoic acid (PFOA) is a representative persistent organic pollutant widely present in the marine environment. However, its toxicological mechanisms during early developmental stages in marine benthic fish are not yet fully understood. In this study, the embryos and larvae of Paralichthys olivaceus were employed to investigate the toxic effects of PFOA during early development using a multi-omics approach. A 96-h acute exposure experiment was conducted, involving six exposure concentrations: 0 (control group), 5, 10, 20, 50, and 100 mg/L. The results revealed that the PFOA concentrat

Granular activated carbon (GAC) is widely used to treat contaminated per- and polyfluoroalkyl substances (PFAS) waste streams, resulting in the accumulation of large quantities of spent GAC that need to be landfilled or regenerated. A novel modified supercritical CO2 (scCO2) extraction for regeneration of spent GAC is developed. With the addition of organic solvents and acid modifiers, the procedure yielded >99% perfluorooctanoic acid (PFOA) desorption after a 60-min treatment in a continuous flow reactor. The mild extraction conditions at T ∼100 °C do not trigger the formation of volatile org

There is considerable interest in addressing soils contaminated with per- and polyfluoroalkyl substances (PFAS) because of the PFAS in the environment and associated health risks. The neutralization of PFAS in situ is challenging. Consequently, mobilizing the PFAS from the contaminated soils into an aqueous solution for subsequent handling has been pursued. Nonetheless, the efficiency of mobilization methods for removing PFAS can vary depending on site-specific factors, including the types and concentrations of PFAS compounds, soil characteristics. In the present study, the removal of perfluor

Due to their persistence and toxicity, perfluoroalkyl and polyfluoroalkyl substances (PFASs) constitute significant hazards to human health and the environment. Their effects include immune suppression, altered hormone levels, and osteoporosis. Recently, the most studied PFAS, perfluorooctanoic acid (PFOA), was shown to competitively binding to the Vitamin D receptor (VDR). VDR plays a crucial role in regulating genes involved in maintaining immune, endocrine, and calcium homeostasis, suggesting it may be a target for at least some of the health effects of PFAS. Hence, this study examined the

BACKGROUND: Recently, 1-octanol has been shown to have efficacy in treating patients with essential tremor (ET). The primary metabolite of 1-octanol is octanoic acid (OA), which is now thought to be the active substance that mediates tremor suppression. Our aim was to describe the maximum tolerated dose (MTD) of oral OA in patients with ET and assess the pharmacokinetics (PK) and pharmacodynamics (PD) profile of OA.
METHODS: The MTD was studied using an open-label, single-ascending 3 + 3 dose-escalation design. Predefined single doses ranged from 8 to 128 mg/kg, with grade 2 adverse events (A

Infection by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) can trigger excessive interleukin (IL)-6 signalling, leading to a myriad of biological effects including a cytokine storm that contributes to multiple organ failure in severe coronavirus disease 2019 (COVID-19). Using a mouse model, we demonstrated that nasal inoculation of nucleocapsid phosphoprotein (NPP) of SARS-CoV-2 increased IL-6 content in bronchoalveolar lavage fluid (BALF). Nasal administration of liquid coco-caprylate/caprate (LCC) onto Staphylococcus epidermidis (S. epidermidis)-colonized mice significantly at

BACKGROUND: Cutaneous leishmaniasis is one of the most important parasitic diseases in humans. In this disease, one of the responsible organisms is Leishmania major, which is transmitted by sandfly vector. There are specific differences in biochemical profiles and metabolite pathways in logarithmic and stationary phases of Leishmania parasites. In the present study, 1H NMR spectroscopy was used to examine the metabolites outliers in the logarithmic and stationary phases of promastigotes in L. major to enlighten more about the transmission mechanism in metacyclogenesis of L. major.

PFASs are a group of pollutants ubiquitous in the environment, for which their effects on the microbial community composition and activity of anaerobic granular sludge are still poorly understood. With our study, we aimed to provide insight into the impact of four PFASs on the methane yield of anaerobic granular sludge (AGS) using acetate as the substrate. Anaerobic granular sludge was exposed to different concentrations of 1H,1H,2H,2H-Tridecafluorooctan-1-ol (6:2FTOH), 1H,1H,2H,2H-Perfluoro-1-decanol (8:2FTOH), Tridecafluorohexane-1-sulfonic acid (PFHxS) and Perfluorooctanoic acid (PFOA). The ecological safety of replacement per- and polyfluoroalkyl substances (PFASs), such as hexafluoropropylene oxide trimer acid (HFPO-TA), remains controversial and insufficiently assessed. In this study, we systematically compared the toxicological profiles of HFPO-TA and the legacy contaminant perfluorooctanoic acid (PFOA) at environmentally relevant concentrations, aiming to assess the potential risk of "regrettable substitution". Opsariichthys bidens were exposed to 5, 50, and 500 μg L-1 concentrations of HFPO-TA and PFOA for 60 days. Compared with PFOA, HFPO-TA induced more severe hepatic

Per- and polyfluoroalkyl substances (PFAS) are synthetic chemicals that persist in the environment and have potential health risks. Among them, 8:2 fluorotelomer alcohol (FTOH) undergoes microbial biotransformation in the environment into perfluorooctanoic acid (PFOA) and releases fluoride. While PFOA's toxic effects have been well-studied, the impact of PFOA (and its precursor 8:2 FTOH) on dental health is largely unknown. Moreover, it is not established whether, and to what extent, defluorination and fluoride bioaccumulation occur during the in vivo 8:2 FTOH metabolism. This study is the fir

Perfluorooctanoic acid (PFOA) is one of the most extensively investigated per- and polyfluoroalkyl substances (PFAS), a class of highly fluorinated organic compounds widely used in consumer products. While assessing the long-term health effects of chronic PFOA exposure in humans remains challenging, simple animal models like zebrafish serve as essential tools to identify potential toxicological risks. The aim of this study was to investigate the chronic effects of PFOA on bone metabolism in young adult zebrafish, as well as the potential transgenerational impacts on their offspring. Fish fed a

Perfluorooctanoic acid (PFOA), a legacy per- and polyfluoroalkyl substance (PFAS), has been associated with dyslipidemia and adverse cardiovascular phenotypes, yet the plaque-macrophage mechanisms that may connect exposure to lesion biology remain incompletely defined. Here, we integrated human plaque single-cell and spatial transcriptomics, bulk transcriptomic profiles of PFAS-exposed macrophages, cross-cohort regulon validation, human plaque histopathology, ApoE-/- mouse experiments, and targeted perturbation assays to examine how PFOA reshapes plaque-relevant macrophage state features and s

High exposure of per- and polyfluoroalkyl substances (PFAS) has been reported in main chemical production areas in China, while epidemiological study on exposure risk of PFAS is still limited. In this study, legacy and alternative PFAS were measured in serum samples from 161 adults living in Laizhou Bay, a famous chemical production area located in Shandong province, Northern China. Based on the concentrations of serum PFAS, the disrupting effects of PFAS on serum lipids and thyroid function were further explored. The results showed that the serum perfluorooctanoic acid (PFOA) (geometric mean

Perfluorinated compounds (PFCs), as an important class of new persistent organic pollutants, are widely distributed in the environment. Yet the effects of different types and concentrations of PFCs on soil microbial community in urban forest ecosystems are remain uncertain. Here, two typical PFCs, perfluorooctanoic acid (PFOA) and perfluorooctane sulfonic acid (PFOS), were selected to carry out a pot experiment in greenhouse with singly and joint treatment at different concentrations, to examine their effects on composition and diversity of soil microorganisms and availability of soil macronut

Intracerebral hemorrhage (ICH) is a severe stroke subtype caused by the rupture of blood vessels within the brain. Increased levels of S100B protein may contribute to neuroinflammation after ICH through activation of astrocytes and resident microglia, with the consequent production of proinflammatory cytokines and reactive oxygen species (ROS). Inhibition of astrocytic synthesis of S100B by arundic acid (AA) has shown beneficial effects in experimental central nervous system disorders. In present study, we administered AA in a collagenase-induced ICH rodent model in order to evaluate its effec

INTRODUCTION: Myasthenia gravis (MG) is an antibody-mediated disease that develops in the majority of patients mainly as a result of acetylcholine receptor (AChR) autoantibodies. This process is mediated by a series of immunoregulatory events. Therapeutic targets for MG include suppression of circulating antibodies or antibody production, suppression of complement activation, and immunomodulation of cytokines or T cells. Intravenous immunoglobulin (IVIg) has an effect on all of these mechanisms.
AREAS COVERED: This narrative review explores the broad immunomodulatory effects of IVIg in MG and
