Carbon monoxide expedites metabolic exhaustion to inhibit tumor growth.

One classical feature of cancer cells is their metabolic acquisition of a highly glycolytic phenotype. Carbon monoxide (CO), one of the products of the cytoprotective molecule heme oxygenase-1 (HO-1) in cancer cells, has been implicated in carcinogenesis and therapeutic resistance. However, the functional contributions of CO and HO-1 to these processes are poorly defined. In human prostate cancers, we found that HO-1 was nuclear localized in malignant cells, with low enzymatic activity in moderately differentiated tumors correlating with relatively worse clinical outcomes. Exposure to CO sensi

Bilateral brachial plexus injury following acute carbon monoxide poisoning.

BACKGROUND: Carbon monoxide (CO) intoxication is a leading cause of severe neuropsychological impairments. Peripheral nerve injury has rarely been reported. It consists usually in a demyelinating polyneuropathy or mononeuropathy affecting mainly the lower limbs. Isolated involvement of both upper extremities has been described in only 4 patients related to root damage. We report the first case of bilateral brachial plexus injury following CO poisoning and review all previous CO-induced neuropathy described in literature. CASE PRESENTATION: After being unconscious for three hours, a 42 years o

Carbon monoxide: endogenous mediator, potential diagnostic and therapeutic target.

The primary objectives of this article are to review the potential role of carbon monoxide (CO) as an endogenous mediator, diagnostic marker for pulmonary disorders, and therapeutic target in critical illness. The review will start by focusing on the importance of the heme oxygenase (HO)-CO axis as an endogenous system as it relates to the cardiovascular and pulmonary systems. It will elucidate the influence of HO gene expression on critical events like shock, sepsis, ischemia-reperfusion and others. Our focus will then shift and look at the potential diagnostic role of exhaled CO in major inf

[Acute poisoning with carbon monoxide (CO) and cyanide (CN)].

A critical analysis of acute CO and CN poisonings evidences that the lone toxicological similarity between CO and CN is their ability to avidly bind iron ions in the different hemoproteins. Otherwise, CO is a stable not-metabolized molecule while CN is a highly reactive molecule with extensive metabolism. CO impairs the ability of erythrocytes to transfer oxygen while CN has no effect. Both toxicants act in the mitochondria with CN resulting in the complete and sustained blockade of cellular respiration inducing severe lactic acidosis while CO does not. Acute CO poisoning mostly alters neurolo

Isocapnic hyperventilation increases carbon monoxide elimination and oxygen delivery.

Hyperventilation with mixtures of O2 and CO2 has long been known to enhance carbon monoxide (CO) elimination at low HbCO levels in animals and humans. The effect of this therapy on oxygen delivery (DO2) has not been studied. Isocapnic hyperventilation utilizing mechanical ventilation may decrease cardiac output and therefore decrease DO2 while increasing CO elimination. We studied the effects of isocapnic hyperventilation on five adult mechanically ventilated sheep exposed to multiple episodes of severe CO poisoning. Five ventilatory patterns were studied: baseline minute ventilation (RR. VT),

[Acute poisoning by carbon monoxide affecting the extrapyramidal system].

A case record of a 19-year old women acutely poisoned by carbon monoxide is presented. The state of the patient was very serious with loss of consciousness, cerebral oedema and acute circulatory failure, manifested by pulmonary oedema. The presence of carboxyhemoglobin in concentration of 45% was found in the blood. During the second week after the incident, extrapyramidal symptoms, manifested by hyperkinetic-hypokinetic syndrome with predominant involuntary movement, was observed. The diagnostic examinations of the central nervous system showed necrosis of globus pallidus, external capsule an

End tidal carbon monoxide concentration in childhood haemolytic disorders.

OBJECTIVES: Endogenous carbon monoxide (CO) is produced mainly by heme catabolism. As CO is excreted solely by the lung, a simple technique for measuring the end tidal carbon monoxide (ETCO) level was assessed as a method for screening for haemolytic disease in children. METHODS: Two end expiratory breath samples were collected from normal children and from children with haemolytic disease using a one way valve connector between a mouth piece and an anaesthetic bag. The samples were analysed by gas chromatography for CO and carbon for dioxide (CO2). The CO2 value was used to normalise the CO

Delayed death following carbon monoxide poisoning. A case report.

The body of a deceased woman was found in a car. The circumstances were typical for suicidal carbon monoxide (CO) poisoning by inhalation of exhaust gases, except that the vacuum cleaner hose had become detached from the exhaust. Autopsy revealed only decompositional changes. No CO or other drugs were detected. Hypoxic changes were present in the cortex and hippocampus, with an age of 2-3 days. It is consistent that this woman inhaled sufficient CO to cause coma, at which stage the hose became detached, and that she then exhaled CO over the next 2-3 days before finally dying. Delayed death fol

Acute carbon monoxide intoxication and hyperbaric oxygen in pregnancy.

Modalities of oxygen therapy for pregnant women intoxicated with carbon monoxide (CO) are ill defined. Hyperbaric oxygen (HBO) is presumed to be hazardous to the pregnancy. On the other hand CO entails anoxic injuries in the mother and fetus. We have entered 44 pregnant women who sustained an acute carbon monoxide poisoning at home, into a prospective study in order to assess HBO tolerance. They were treated within 5.3 +/- 3.7 h (range: 1-12) of the intoxication with a combination of 2 h of HBO at a pressure of 2 atmospheres absolute (ATA) and 4 h of normobaric oxygen, irrespective of the clin

Diabetes insipidus after carbon monoxide poisoning and smoke inhalation.

Diabetes insipidus was encountered in a 58-year-old woman after injury in an closed space fire. Carboxyhemoglobin measured 26% after 30 minutes of assisted ventilation with an FIO2 of 1.0. The failure of antidiuretic hormone was a sign of a diffuse cerebral insult caused by carbon monoxide poisoning. The hormonal deficit was easily corrected by exogenous vasopressin. Mortality was related to the cerebral lesion.

Carbon monoxide enhances development of hypertension in Dahl rats.

The influence of carbon monoxide (CO) on the development of systemic hypertension was studied in Dahl rats selectively bred for susceptibility (DS) and resistance (DR) to NaCl-induced hypertension. This study was designed to examine the interactions among rat line (DS or DR), NaCl content of diet, and exposure to CO. The rats were exposed to 500 ppm CO or conditioned air, 21 hr/day, for 62 to 63 consecutive days. Carbon monoxide exposures affected blood pressure only in DS rats fed a high NaCl diet, where it enhanced the development of NaCl-induced hypertension. Whole-body weights were not aff

Carbon monoxide poisoning: mechanisms, presentation, and controversies in management.

Carbon monoxide (CO) remains the leading cause of death due to poisoning in the United States. CO produces toxicity by binding to hemoglobin, thereby reducing oxygen-carrying capacity, and by binding to myoglobin, which may impair cardiac output and result in cerebral ischemia. Severe CO poisoning results in coma or encephalopathy, but milder intoxication may occur with nonspecific symptoms suggestive of hysteria, hyperventilation, psychosis, or viral syndrome. Survivors of severe CO poisoning may have permanent neurologic or neuropsychiatric sequelae. Subtle memory deficits or personality cha

Carbon monoxide oxidation by Clostridium thermoaceticum and Clostridium formicoaceticum.

Cultures of Clostridium formicoaceticum and C. thermoaceticum growing on fructose and glucose, respectively, were shown to rapidly oxidize CO to CO(2). Rates up to 0.4 mumol min(-1) mg of wet cells(-1) were observed. Carbon monoxide oxidation by cell suspensions was found (i) to be dependent on pyruvate, (ii) to be inhibited by alkyl halides and arsenate, and (iii) to stimulate CO(2) reduction to acetate. Cell extracts catalyzed the oxidation of carbon monoxide with methyl viologen at specific rates up to 10 mumol min(-1) mg of protein(-1) (35 degrees C, pH 7.2). Nicotinamide adenine dinucleot

Prion diseases: copper deficiency states associated with impaired nitrogen monoxide or carbon monoxide transduction and translocation.

Literature concerning prion diseases and Cu metabolism was examined to determine merits of various suggestions concerning the relationship between these diseases and altered Cu metabolism. There are a number of recent suggestions that the normal non-pathogenic form of the prion protein (PrP(C)) contains Cu while the abnormal pathogenic form of this protein, PrP(SC), lacks Cu. Results of experiments showing oxidant sensitivity in the presence of ionically bonded Cu and millimolar concentrations of hydrogen peroxide were found to lack relevance. Demonstrating superoxide disproportionation and a

Entangled hydrophobic and hydrophilic networks in carbon monoxide dehydrogenase.

Carbon monoxide dehydrogenase (CODH) is a crucial enzyme involved in the global carbon cycle and fixation by catalysing the reversible interconversion of CO2 and CO. Although major structural features of the enzyme are known, key connections between the structure and function remain unclear. Here, we employ large-scale unconstrained all-atom molecular dynamics simulations to investigate the key catalytic features of CODH, including (i) the implications of CODH dimerization, (ii) substrate and water transport cavities, (iii) the impact of global and local hydration, and (iv) protonation-depende
