Carnosine supplementation reduces plasma soluble transferrin receptor in healthy overweight or obese individuals: a pilot randomised trial.

Abnormalities of iron homeostasis have been linked to insulin resistance, type 2 diabetes and cardiovascular disease. Carnosine, an over-the-counter food supplement with chelating properties, has been shown to decrease serum iron and improve glucose metabolism in diabetic rodents. We have previously demonstrated that carnosine supplementation prevented worsening of glucose metabolism in healthy overweight and obese middle-aged adults. Yet, the impact of carnosine on markers of iron metabolism in humans has not been investigated. We aimed to determine whether carnosine supplementation has an ef

Direct HPLC separation of carnosine enantiomers with two chiral stationary phases based on penicillamine and teicoplanin derivatives.

Carnosine is present in high concentrations in specific human tissues such as the skeletal muscle, and among its biological functions, the remarkable scavenging activity toward reactive carbonyl species is noteworthy. Although the two enantiomers show almost identical scavenging reactivity toward reactive carbonyl species, only d-carnosine is poorly adsorbed at the gastrointestinal level and is stable in human plasma. Direct methods for the enantioselective analysis of carnosine are still missing even though they could find more effective applications in the analysis of complex matrices. In th

Antioxidant, Anti-inflammatory, and Genomic Stability Enhancement Effects of Zinc l-carnosine: A Potential Cancer Chemopreventive Agent?

Cancer is one of the major causes of death worldwide, and the incidence and mortality rates of cancer are expected to rise tremendously in the near future. Despite a better understanding of cancer biology and advancement in cancer management, current strategies in cancer treatment remain costly and ineffective. Hence, instead of putting more efforts to search for new cancer cures, attention has now been shifted to the development of cancer chemopreventive agents as a preventive measure for cancer formation. It is well known that neoplastic transformation of cells is multifactorial, and the occ

Carnosine Attenuates the Development of both Type 2 Diabetes and Diabetic Nephropathy in BTBR ob/ob Mice.

We previously demonstrated that polymorphisms in the carnosinase-1 gene (CNDP1) determine the risk of nephropathy in type 2 diabetic patients. Carnosine, the substrate of the enzyme encoded by this gene, is considered renoprotective and could possibly be used to treat diabetic nephropathy (DN). In this study, we examined the effect of carnosine treatment in vivo in BTBR (Black and Tan, BRachyuric) ob/ob mice, a type 2 diabetes model which develops a phenotype that closely resembles advanced human DN. Treatment of BTBR ob/ob mice with 4 mM carnosine for 18 weeks reduced plasma glucose and HbA1c

Protective effects of L- and D-carnosine on alpha-crystallin amyloid fibril formation: implications for cataract disease.

Mildly denaturing conditions induce bovine alpha-crystallin, the major structural lens protein, to self-assemble into fibrillar structures in vitro. The natural dipeptide l-carnosine has been shown to have potential protective and therapeutic significance in many diseases. Carnosine derivatives have been proposed as potent agents for ophthalmic therapies of senile cataracts and diabetic ocular complications. Here we report the inhibitory effect induced by the peptide (l- and d-enantiomeric form) on alpha-crystallin fibrillation and the almost complete restoration of the chaperone activity lost

Carnosine-, calcitonin gene-related peptide- and tyrosine hydroxylase-immunoreactivity in the mouse olfactory bulb following peripheral denervation.

We report the effects of olfactory peripheral deafferentation by intranasal irrigation with ZnSO4 on carnosine and CGRP immunoreactivities in the mouse olfactory system. In the normal rodent olfactory epithelium carnosine immunoreactivity is associated with the olfactory receptor neurons. Conversely, CGRP immunoreactivity appears to be associated with the trigeminal innervation of the nasal cavity. Following lesion the magnitude of carnosine immunoreactivity in the olfactory epithelium is strongly reduced while CGRP immunoreactivity is unaffected. In the olfactory bulb, deafferentation causes

Conjugation of Dietary Carnosine to Phe-Leu Dipeptide Affords a Potent Inhibitor of Multifaceted Aβ-Induced Neurotoxicity.

Alzheimer's disease (AD) is a chronic neurological disorder manifested by cognitive decline, amyloid-β (Aβ) fibrillation, synaptic dysfunction, and neuronal loss. Furthermore, metal ion dysregulation is implicated in Aβ aggregation, oxidative stress, and neuronal toxicity, thereby exacerbating neurodegeneration. Thus, highly versatile chemical entities with a synergistic effect targeting Aβ aggregation, disintegrate Aβ fibrils, and impede Aβ-Cu2+ interactions hold considerable therapeutic potential against AD. With this aim, a novel tetrapeptide CarFL (β-Ala-His-Phe-Leu) was rationally designe

The neuroprotective effect of carnosine (β-alanyl-L-histidine) on retinal ganglion cell following ischemia-reperfusion injury.

PURPOSE: To investigate whether carnosine can increase retinal ganglion cell (RGC) survival in ischemic mouse retina. METHODS: Retinal ischemia was induced by constant elevation of intraocular pressure (100-110 mmHg) for 60 min in C57BL/6 J mice pretreated with carnosine (1000 mg/kg) or saline. Hypoxia inducing factor-1 alpha (HIF-1α), glial fibrillary acidic protein (GFAP), and dynamin-related protein-1 (Drp-1) expressions were assessed at 6, 12, and 24 h after retinal ischemia. Bax and Bcl-2 expressions were also analyzed at 12 h after retinal ischemia. RGC survival was assessed by retrogra

Effect of histidine and carnosine on haemoglobin recovery in anaemia induced-kidney damage and iron-loading mouse models.

Histidine and carnosine can form complexes with divalent metal ions such as Fe2+, potentially providing stability to intracellular labile iron. Anaemia is a common comorbidity in the late stages of kidney disease, and patients are treated with erythropoiesis-stimulating agents (ESAs) and iron supplementation. However, iron supplementation is also associated with worse long-term outcomes. The purpose of this study is to investigate how histidine and carnosine supplementation can reduce symptoms of anaemia of chronic kidney disease (CKD) and the effects associated with iron-overloaded conditions

Carnosine as a protective metabolic mediator in inflammatory lung injury by inhibiting macrophage infiltration and M1-like polarization.

BACKGROUND: Inflammatory lung injury is a common pathological feature of pneumonia caused by various infectious and non-infectious agents. However, metabolic regulators that can mitigate inflammation and immune cell infiltration in diverse lung injury models remain poorly understood. METHODS: Using targeted metabolomic profiling of lung tissues collected on day 5 from two distinct murine models of lung inflammation-lipopolysaccharide (LPS)-induced and papain-induced-we identified carnosine as a commonly downregulated metabolite in both models. To evaluate its therapeutic potential, we adminis

Pulmonary inflammation, oxidative stress, and fibrosis in a mouse model of cholestasis: the potential protective properties of the dipeptide carnosine.

Cholestasis is a clinical complication that primarily influences the liver. However, it is well known that many other organs could be affected by cholestasis. Lung tissue is a major organ influenced during cholestasis. Cholestasis-induced lung injury could induce severe complications such as respiratory distress, serious pulmonary infections, and tissue fibrosis. Unfortunately, there is no specific pharmacological intervention against this complication. Several studies revealed that oxidative stress and inflammatory response play a role in cholestasis-induced lung injury. Carnosine (CARN) is a

Characterizing oxidative stress induced by Aβ oligomers and the protective role of carnosine in primary mixed glia cultures.

Alzheimer's disease (AD) is a neurodegenerative disorder characterized by cognitive decline and memory loss. A critical aspect of AD pathology is represented by oxidative stress, which significantly contributes to neuronal damage and death. Microglia and astrocytes, the primary glial cells in the brain, are crucial for managing oxidative stress and supporting neuronal function. Carnosine is an endogenous dipeptide possessing a multimodal mechanism of action that includes antioxidant, anti-inflammatory, and anti-aggregant activities. The present study investigated the effects of Aβ1-42 oligomer

Effects of carnosine, taurine, and betaine pretreatments on diethylnitrosamine-induced oxidative stress and tissue injury in rat liver.

Several chemicals such as N-diethylnitrosamine (DEN) promote hepatocellular cancer in rodents and induce hepatocyte injury. DEN affects the initiation stage of carcinogenesis together with enhanced cell proliferation accompanied by hepatocellular necrosis. DEN-induced hepatocellular necrosis is reported to be related to enhanced generation of reactive oxygen species. Carnosine (CAR), taurine (TAU), and betaine (BET) are known to have powerful antioxidant properties. We aimed to investigate the effects of CAR, TAU, and BET pretreatments on DEN-induced oxidative stress and liver injury in male r

Carnosine protects against cerebral ischemia/reperfusion injury through promoting microglial M2 polarization via SIRT1/NF-κB signaling pathway.

Ischemic stroke, a leading cause of disability and mortality worldwide, often results in secondary brain injury due to oxidative stress and neuroinflammation following thrombolysis with tissue plasminogen activator (t-PA). Carnosine (CAR), a naturally occurring dipeptide, exhibits anti-inflammatory and neuroprotective properties. But its mechanisms in cerebral ischemia-reperfusion injury (CIRI) remain unclear. In this study, we aim to investigate the role of microglial polarization regulated by SIRT1/NF-κB signaling pathway on CAR therapeutic effect in CIRI. Here, we demonstrate that CAR atten
