Structural insights into DHODH-mediated catalysis and drug resistance.

Dihydroorotate dehydrogenase (DHODH), which catalyzes the rate-limiting step in de novo pyrimidine biosynthesis, is a validated therapeutic target in cancer, autoimmune disorders and infectious diseases. DHODH is hypothesized to utilize a 'ping-pong' catalytic mechanism. However, available crystal structures of DHODH in complex with small molecule inhibitors are irreconcilable with this model, showing simultaneous occupancy of both substrate binding sites. To elucidate the structural basis for DHODH-mediated catalysis, we resolved the structures of two DHODH holoenzymes. These structures captu

Selective Hydroxylation of Collagen Model Peptides by Metal Catalysis.

Collagen is formed with a repeating structure of proline (Pro)-4-hydroxyproline-(4-Hyp)-glycine (Gly). In chemical synthesis, when synthesizing collagen sequences, it is necessary to protect the 4-Hyp side chain within them. Therefore, if (Pro-Pro-Gly)n could be synthesized followed by selective hydroxylation of proline residues using iron catalysts, improved synthetic efficiency could be expected. In this study, hydroxylation reactions were performed on Boc-Pro-OPac and Ac-Pro-OPac, where the N-termnus of proline is protected by Boc and Ac groups, respectively, and on Boc-Pro-Pro-OPac, a pept

Photoinduced Chemoselective Alkane Sulfinylation via Fe/Ni Dual Catalysis.

Sulfoxides occupy a central role in natural products and bioactive molecules, yet their direct construction from simple feedstocks remains challenging. Herein, we report a dual iron/nickel photocatalytic strategy for the selective C(sp3)-H sulfoxidation of liquid/gaseous alkanes using industrially abundant aryl sulfonyl chlorides. Broad substrate scope, good functional group tolerance, operational simplicity, high chemoselectivity, scale-up synthesis, and late-stage modification are demonstrated, providing a practical platform for the preparation of high-value sulfur-containing compounds.

Multistate Enzyme Design Enables Efficient and Stereoselective Multistep Catalysis.

Enzymes catalyze multistep reactions by stabilizing successive transition states within well organized, yet dynamic active sites. However, computational enzyme design typically targets a single transition state using rigid active-site models. Here, we introduce multistate enzyme design, which uses conformational ensembles to optimize active sites across an entire reaction coordinate. Applied to a de novo Morita-Baylis-Hillmanase, multistate enzyme design outperformed conventional single-state design, with the most active variant achieving >100-fold higher bi-substrate catalytic efficiency and

T-Shaped Stannyliumylidene Ion: Synthesis, Reactivity, and Redox Catalysis.

We report the synthesis and characterization of a T-shaped, 8-electron stannyliumylidene ion bearing a rigid acridane-based pincer ligand. This cationic Sn(II) complex exhibits pronounced ambiphilic reactivity, participating in electrophilic, nucleophilic, and σ-bond activation reactions. All derived compounds were characterized by nuclear magnetic resonance spectroscopy, single crystal x-ray diffraction analysis, and high-resolution mass spectrometry. Density functional theory calculations reveal the coexistence of a lone pair of electrons and a vacant 5p-orbital at the tin center, which rati

Working-state descriptors for theory-guided CO2 hydrogenation catalysis.

Thermal CO2 hydrogenation connects carbon-oxide recycling with low-carbon H2 use and established product families, including CO, methanol, methane and C2+ hydrocarbons. The design problem is that the selective catalyst is often not the ideal surface used at the start of a calculation. H2/CO2/CO/H2O feeds can redistribute phases, interfaces, adsorbates and promoter environments during measurement. First-principles descriptor maps and microkinetic models remain essential once candidate states are specified, but they cannot decide which states should enter the comparison. This Review asks how suc

Mechanochemical three-component radical sulfonamidation enabled by copper catalysis.

Sulfonamides are privileged organic synthesis scaffolds, particularly in pharmaceutical and bioactive domains. Conventional synthetic strategies often rely on expensive metal cross-coupling protocols, the availability of pre-existing sulfur functionality, amine nucleophilicity, elevated reaction temperatures, and inherently unstable and expensive SO2 surrogates. Herein, we unveil a concise, mechanochemical driven, cuprous chloride (CuCl)-catalyzed three-component protocol that involves aryl radical precursors (aryldiazonium, diaryliodonium and thianthrenium salts), with K2S2O5 as a SO2 surroga

Deciphering Asymmetric Induction in Photoredox Catalysis by Chiral Counteranions.

We investigate the origin of stereocontrol in asymmetric counteranion-directed photoredox catalysis (ACPC) using a representative [2 + 2] cycloaddition mediated by a chiral imidodiphosphorimidate (IDPi) counteranion (Science 2023, 379, 494-499). Combining extensive conformational sampling, high-level DFT calculations, and multiscale modeling, we elucidate the mechanism and stereochemical landscape of this transformation. Both enantio- and diastereoselectivity are established in the first C-C bond-forming step: diastereoselectivity arises from intrinsic aryl-aryl interactions within the radical

Umpolung Activation of Bicyclobutanes via N-Heterocyclic Carbene Catalysis.

Bicyclo[1.1.0]butane (BCB) has predominantly been explored as an electrophile or radical acceptor owing to its inherent polarity. However, its reactivity as a nucleophile remains largely unexplored. Herein, we report an umpolung strategy that reverses the innate polarity of bicyclobutane aldehydes via N-heterocyclic carbene (NHC) catalysis. This transformation enables the construction of novel rigidified lactones and lactams via coupling with diverse electrophilic partners, including aldehydes, ketones, and imines. The synthetic versatility of this method highlights its potential for applicati

Enantioselective homocoupling of allenylic alcohols through divergent cooperative catalysis.

An enantioselective homocoupling of branched allenylic alcohols is developed under cooperative iridium and Lewis acid catalysis. In this reaction, racemic allenylic alcohol is transformed, under Lewis acidic Sc(OTf)3, into an α,β-unsaturated enol (cross dienol) through a Meyer-Schuster-type 1,3-hydroxy transposition. In an independent cycle, catalyzed by a combination of an Ir(i)/(phosphoramidite,olefin) complex and Sc(OTf)3, allenylic alcohol is proposed to produce an η2-Ir(i)-bound allenylic carbocation intermediate, which is intercepted by the in situ generated cross dienol. Overall, starti

Kharasch-Type Haloalkylation of Alkenes by Photoinduced Copper Catalysis.

The simultaneous construction of C-C and C-X bonds in a single step facilitates multistep synthesis through rapid carbon chain growth and subsequent transformations of halide functionalities. Traditional Kharasch addition requires simple polyhalogenated compounds or those with electron-withdrawing groups at the α-carbon. Herein, we present a Kharasch-type reaction utilizing a broad range of carboxylic acid-derived redox-active esters as the alkyl source, which enables the efficient introduction of highly functionalized alkyl groups. Our method produces α-halo carbonyls that enable versatile nu

Homolytic H2 dissociation for enhanced hydrogenation catalysis on oxides.

The limited surface coverage and activity of active hydrides on oxide surfaces pose challenges for efficient hydrogenation reactions. Herein, we quantitatively distinguish the long-puzzling homolytic dissociation of hydrogen from the heterolytic pathway on Ga2O3, that is useful for enhancing hydrogenation ability of oxides. By combining transient kinetic analysis with infrared and mass spectroscopies, we identify the catalytic role of coordinatively unsaturated Ga3+ in homolytic H2 dissociation, which is formed in-situ during the initial heterolytic dissociation. This site facilitates easy hyd

Synthesis of aminoalcohols from substituted alkenes via tungstenooxaziridine catalysis.

Herein we report the WO2Dipic(H2O) promoted oxyamination of alkenes using sulfonamides as the quantitative source of N. The reaction works for activated and unactivated alkenes in high yields, diastereoselectivities, and stereospecificity. A catalytic cycle involving the formation of tungstenooxaziridine complex 1 as the active catalyst and hydrolysis of tungstenooxazolidine intermediate A as the rate-determining-step has been proposed. Initial kinetic and competition experiments provide evidence for the proposed mechanism.

Enantioselective Hydroaminoalkylation of Azaaryl Ketones through Asymmetric Photoredox Catalysis.

An enantioselective hydroaminoalkylation of azaaryl ketones under a transition-metal-free asymmetric photoredox catalysis platform is reported. A series of valuable azaarene-functionalized 1,2-amino alcohols featuring attractive quaternary carbon stereocenters have been synthesized in high yields with good to excellent enantioselectivities. The viability of readily accessible N-aryl glycines as reaction partners facilitates the conjugate modification of these products into important derivatives, thereby enhancing the synthetic utility of the current approach.

Reductive samarium (electro)catalysis enabled by SmIII-alkoxide protonolysis.

Samarium diiodide (SmI2) is a privileged, single-electron reductant deployed in diverse synthetic settings. However, generalizable methods for catalytic turnover remain elusive because of the well-known challenge associated with cleaving strong SmIII-O bonds. Prior efforts have focused on the use of highly reactive oxophiles to enable catalyst turnover. However, such approaches give rise to complex catalyst speciation and intrinsically limit the synthetic scope. Herein, we leveraged a mild and selective protonolysis strategy to achieve samarium-catalyzed, intermolecular reductive cross-couplin

Exploiting phase transitions in catalysis: reaction ofCO2andH2on dopedVO2-polymorphs.

VO2is well known for its reversible transition between two phases with tetragonal rutile and monoclinic structure. In a previous theoretical study (Stahl and Bredow 2022ChemPhysChem23e202200131) we showed that the adsorption energy of CO is different on surfaces of the two Mo-stabilized polymorphs. This can be exploited to promote catalytic reactions by removing CO from the catalyst surface. As proof-of-principle, we investigated the hydrogenation reaction ofCO2. For this purpose, the adsorption energies ofCO2and possible intermediates and productsH2O, HCOOH,H2COand CO were calculated. Signifi

Controlled Aerobic Oxidative Dimerization of Hydroxystilbenoids by Chromium Catalysis.

Aerobic oxidative dimerization of hydroxystilbenoids is described. A Cr-salen complex catalyzed the dimerization of hydroxystilbenoids in 1,1,1,3,3,3-hexafluoroisopropanol to form compounds comprising a natural product-like scaffold (quadrangularin) or its precursor depending on the aromatic substituents. The addition of a catalytic amount of scandium triflate [Sc(OTf)3] to the reaction system altered the reaction outcome to give a different natural product-like compound, a pallidol-type dimer.

Platelet Membrane-Enclosed Bioorthogonal Catalysis for Combating Dental Caries.

Platelets have shown promise as a means to combat bacterial infections, fostering the development of innovative therapeutic approaches. However, several challenges persist, including cargo loading issues, limited efficacy against biofilms, and concerns regarding the impact of payloads on the platelet carriers. Here, human platelet membrane vesicles (h-PMVs) encapsulating supramolecular metal catalysts (SMCs) as "nanofactories" to convert prodrugs into antimicrobial compounds within close proximity to bacteria are introduced. Having established the feasibility and effectiveness of the SMCs with

Copper Catalysis-Based Amperometric Microsensors for Carbon Dioxide Monitoring.

A fast response microsensor that can detect the distribution of CO2 at the microscale level is essential for the observation of biophysiological activity, carbon flux, and carbon burial. Inspired by the previous success of Cu catalysis, we attempted to use this metal Cu material to develop an amperometric microsensor that can meet the requirements. Specifically, the ambient gases diffuse through a silicone membrane into a trap casing filled with an acidic CrCl2 solution, where the otherwise interfering O2 interferent is removed by a redox with Cr2+. The gases then diffuse through a second sili

Enantioconvergent transformations of secondary alcohols through borrowing hydrogen catalysis.

Direct substitution of readily available alcohols is recognized as a key research area in green chemical synthesis. Starting from simple racemic secondary alcohols, the achievement of catalytic enantioconvergent transformations of the substrates will be highly desirable for efficient access to valuable enantiopure compounds. To accomplish such attractive yet challenging transformations, the strategy of the enantioconvergent borrowing hydrogen methodology has proven to be uniquely effective and versatile.
