The development of immune-instructive biomaterials that actively guide host immune responses toward healing and homeostasis is a key step for advancing the next generation of biomedical implants and regenerative medicine. UV laser technology offers a non-contact, scalable, and versatile approach for directly patterning bacterial nanocellulose (BNC) topography at a cell-size scale. Distinct 40 μm-period topographies were generated on the BNC surface, including anisotropic patterns with distinct morphologies and an isotropic pattern of symmetrically arranged bumps. Using a genetic screening approach based on an inducible gene-activating system and cell sorting, Down syndrome critical region 3 (DSCR3) was isolated as a gene whose overexpression increased cell size. Fibroblasts derived from individuals with Down syndrome (DS) exhibit elevated DSCR3 expression at both the mRNA and protein levels, correlating with increased cell volume compared to fibroblasts from healthy donors. Despite a slower proliferation rate, DS fibroblasts demonstrate higher basal and maximal mitochondrial respiration, suggesting enhanced metabolic activity associated with increased

Cell surface-associated polysaccharides in Klebsiella are major virulence determinants and crucial targets for developing vaccines. Traditionally, the purification of these cell surface polysaccharides from Klebsiella pneumoniae involves a multi-step process comprising phenol extraction, nuclease digestion, ultracentrifugation, and repeated ethanol extractions. In this study, we evaluated size exclusion chromatography for effectively eliminating nucleic acid contamination while purifying high molecular weight cell surface-associated polysaccharides. Post-initial extraction, the nucleic acid co

We took a systems approach to the analysis of macrophage phenotype in regenerative and fibrotic volumetric muscle loss outcomes in mice together with analysis of systemic inflammation and of other leukocytes in the muscle, spleen, and bone marrow. Differences in expression of macrophage phenotype markers occurred as early as day 1, persisted to at least day 28, and were associated with increased numbers of leukocytes in the muscle and bone marrow, increased pro-inflammatory marker expression in splenic macrophages, and changes in the levels of pro-inflammatory cytokines in the blood. The most

Understanding of the interactions between macrophages and multifunctional nanoparticles is important for development of novel macrophage-based immunotherapies. Here, we investigated the effects of fluorescent thiol-organosilica particle size and surface properties on cell-particle interactions, including mitochondrial activity, using the mouse macrophage cell line J774A.1. Three different sizes of thiol-organosilica particles (150, 400, and 680 nm in diameter) containing fluorescein (OS/F150, OS/F400, and OS/F680) and particles surface functionalized with polyethylenimine (PEI) (OS/F150PEI, OS

Infection with the Gram-negative species Neisseria gonorrhoeae leads to inflammation that is responsible for the disease symptoms of gonococcal urethritis, cervicitis, and pelvic inflammatory disease. During growth these bacteria release significant amounts of peptidoglycan (PG) fragments which elicit inflammatory responses in the human host. To better understand the mechanisms involved in PG synthesis and breakdown in N. gonorrhoeae, we characterized the effects of mutation of mltG. MltG has been identified in other bacterial species as a terminase that stops PG strand growth by cleaving the

Various types of COVID-19 vaccines, including adenovirus, mRNA, and inactivated ones, have been developed and approved for clinical use worldwide. Inactivated vaccines are produced using a proven technology that is widely used for the production of vaccines for the prevention and control of infectious diseases, including influenza and poliomyelitis. The development of inactivated whole-virion vaccines commonly includes several stages: the production of cellular and viral biomass in cell culture; inactivation of the virus; filtration and ultrafiltration; chromatographic purification of the vira

Porous polymers are among the most promising porous materials for various application because they show the combined advantages of fluorescent porous materials and polymers. This study developed a cell imaging technique based on luminescent porous organosilicon polymers (LPOPs) that were synthesized via Friedel-Crafts reaction of octaphenylcyclotetrasiloxane with octavinylsilsesquioxanes. The porous organosilicon polymers possessed narrow pore-size distribution, high surface area, and monomodal nanopores centered at approximately 0.59 nm. The excellent properties to the porous polymers can be

AIMS AND OBJECTIVES: To determine spleen size using abdominal ultrasonography and haematological parameters in steady state children with sickle cell anaemia (SCA).
SUBJECTS AND METHODS: An analytical case control study of 150 children with SCA in steady state aged 6 months to 15 years was undertaken. Children with HbAA matched for sex and age were used as controls. The spleen sizes of children with SCA and the controls were determined using abdominal ultrasonography. Haematological parameters (RBC, Hb, MCV, MCH, MCHC,WBC, Platelet and Reticulocyte counts) were also assessed.
RESULTS: Fifty

Brain structure and size require precise division of neural stem cells (NSCs), which self-renew and generate intermediate neural progenitors (INPs) and neurons. The factors that regulate NSCs remain poorly understood, and mechanistic explanations of how aberrant NSC division causes the reduced brain size seen in microcephaly are lacking. Here we show that Magoh, a component of the exon junction complex (EJC) that binds RNA, controls mouse cerebral cortical size by regulating NSC division. Magoh haploinsufficiency causes microcephaly because of INP depletion and neuronal apoptosis. Defective mi

We assessed midsagittal corpus callosum size in sickle cell disease (SCD) and its relationship to lesion volume, lesion location, and cognitive functioning. Twenty-eight children with SCD and 16 demographic controls completed magnetic resonance imaging (MRI) and neuropsychological testing. Corpus callosum (CC) size was smaller for children with silent infarcts (n = 8) or overt stroke (n = 8) than for those without visible infarcts (n = 12) or control participants. Lesion volume was a robust predictor of IQ and other cognitive scores; total CC size did not typically add explanatory power for th

TSC1 and TSC2 are responsible for the tumor suppressor gene syndrome tuberous sclerosis (TSC). Mammalian TSC genes have been shown to be involved in cell cycle regulation. Recently, in Drosophila, these data have been confirmed and TSC genes have further been demonstrated to affect cell size control. Here we provide supporting data for the fact that the latter function is conserved in mammals. Human TSC1 and TSC2 trigger mammalian cell size reduction and a dominant-negative TSC2 mutant induces increased size. These effects occur in all cell cycle phases, are dependent on the activity of the ph

Nissl profiles of cell somata in the lateral geniculate nuclei (LGN) of adult cats which had been reared from 3 to 4 weeks of age with monocular paralysis (MP) were compared to those from normally reared adult cats. In all subjects, measurements were made in 3 different regions of the binocular segments of the A and A1 laminae and in the monocular segment in both LGNs. In the cats reared with MP, reductions in average cell size of about 20%, relative to normal, were detected throughout the binocular segments of the A and A1 laminae in both LGNs whether their innervation was provided by the par

The effects of visual deprivation upon dorsal lateral geniculate (DLG) cell size were compared for seven kittens reared with monocular lid-suture (MD), seven with binocular lid-suture (BD), and six with one eye lid-sutured and the other eye enucleated soon after birth (MD-E). Six additional kittens were reared normally for comparison. For each kitten the cross-sectional areas of 300 cells were measured in one or both nuclei. Measurements were taken from the binocular segment of laminae A and A1 and the monocular segment of lamina A. In agreement with previous studies, cells in the binocular se

We compared exemplar strains from two hypervirulent clonal complexes, strain NMB-CDC from ST-8/11 cc and strain MC58 from ST-32/269 cc, in host cell attachment and invasion. Strain NMB-CDC attached to and invaded host cells at a significantly greater frequency than strain MC58. Type IV pili retained the primary role for initial attachment to host cells for both isolates regardless of pilin class and glycosylation pattern. In strain MC58, the serogroup B capsule was the major inhibitory determinant affecting both bacterial attachment to and invasion of host cells. Removal of terminal sialylatio

Bone represents a highly dynamic tissue whose development is strongly dependent on vasculogenic and angiogenic processes. Neovascularization also plays an important role in fracture healing and in tissue engineering applications aiming at restoring bone function. We have previously shown in a heterotopic subcutaneous implantation model of severe combined immunodeficiency (SCID) mice that implanted human umbilical vein endothelial cells (HUVECs) gave rise to the formation of a complex functional human neovasculature. In this study, we investigated the effect of HUVEC coimplantation on mesenchym

Petal size is crucial for both ornamental appeal and reproductive success in flowering plants, yet the molecular basis of its variation in many non-model species, including cotton, remains poorly understood. Here, we defined the period from -1 DPA to 0 DPA (10:00) as the rapid elongation phase for cotton (Gossypium hirsutum) petals. Disruption of R2R3-MYB gene GhYP in cotton prematurely arrested petal development (by 06:00 on 0 DPA) and reduced petal size through restraining epidermal cell expansion, whereas ectopic expression of its G. arboreum homolog GaYP in tobacco enlarged petals by promo

The plant cell wall is conventionally regarded as a physical barrier with a strict size exclusion limit. However, this static paradigm fails to explain the pervasive accumulation of oversized colloidal particles within plant tissues. Although the presence of oversized particles inside plant tissues has been increasingly reported, the underlying mechanism enabling these particles to overcome the physical barrier of the cell wall remains largely unresolved. Using oversized chromium (Cr) colloids and rice as a model system, this study integrated cross-scale characterizations, molecular dynamics s
