The carbohydrate utilization regulator Cbr1 coordinates nutrient-specific gene activation with selective carbon catabolite repression in a basidiomycete yeast.

Cells must sense and respond to nutrients to survive. To efficiently grow in mixed carbon environments, microbes repress genes necessary to utilize carbon sources that require substantial resources to catabolize when a simpler carbon source, such as glucose, is present. This process is known as carbon catabolite repression. Canonically, in fungi, nutrient sensing transcriptional networks are composed of carbon source-specific transcription factors that activate carbon source utilization genes and carbon catabolite repression regulators, which broadly repress all nonpreferred carbon source util

Cellulose-Binding Electron-Transfer Cytochrome Composing AA8 and CBM104 from the Wood-Rotting Fungus Armillaria tabescens.

Lytic polysaccharide monooxygenases (LPMOs) are activated by electrons supplied from external redox enzymes. Cellobiose dehydrogenase (CDH) and pyrroloquinoline quinone (PQQ)-dependent pyranose dehydrogenase (PDH) are electron donors to LPMOs. CDH and PDH each consist of two domains: a catalytic domain that oxidizes substrates to extract electrons, and an AA8 domain that receives and transfers these electrons to LPMOs. Although non-catalytic AA8 proteins comprising only an AA8 domain and a cellulose-binding module have been reported, their electron donors and physiological roles remain largely

Urinary GC-MS metabolomics reveals pregnancy-associated metabolic reprogramming and biomarkers in buffaloes.

BACKGROUND: Timely and accurate pregnancy detection in the dairy animals is essential for minimizing calving intervals to accelerate reproductive turnover and increase milk production from livestock farming. Urine, a non-invasive sample, is being investigated in many studies as it serves as a pool depicting the varied array of metabolites associated with intricate physiological changes. The study thus aimed to analyze GC-MS based urinary metabolites of pregnant (n = 13) and non-pregnant (n = 13) buffaloes using Bovine Metabolome Database. RESULTS: A total of 102 unique metabolites were annotat

Glucose Oxidoreductases for Glucose Monitoring: A Bird's Eye View of Glucose-Sensing Enzymes II.

Biosensors, utilizing glucose oxidoreductases as biological recognition elements (BREs), underpin current blood glucose monitoring (BGM) and continuous glucose monitoring (CGM) technologies. Glucose oxidase remains the gold standard due to its high selectivity and stability, yet its oxygen dependency limits it to first- and second-generation sensing. Fungal flavin adenine dinucletotide (FAD)-dependent glucose dehydrogenase (GDH) has emerged as the standard for mediator-based second-generation sensors due to its oxygen insensitivity, though it requires further engineering to match the stability

Laser-induced graphene arrays for high-throughput metabolic profiling reveal serum metabolic alterations in colon cancer.

Given the invasiveness of colonoscopy and the limited sensitivity and specificity of carcinoembryonic antigen for colon cancer detection, we developed a laser desorption/ionization mass spectrometry (LDI-MS) method based on a polyethersulfone film-based laser-induced graphene (PES-LIG) array. The PES-LIG is fabricated by one-step CO2 laser scribing on a PES film and can be directly used as an LDI substrate without any modification. Its layered structure facilitates the desorption/ionization of small-molecule metabolites, enabling uniform detection (RSD = 4.12%) with fmol-level sensitivity, hig

Anaerozeibacter quisquiliarum gen. nov., sp. nov., a novel mesophilic bacterium isolated from a laboratory-scale methanogenic landfill bioreactor digesting maize, and proposal of Anaerozeibacteraceae fam. nov., representing a new family within the order Eubacteriales.

Bacteria involved in the anaerobic degradation of lignocellulosic waste in landfill sites play crucial roles in carbon turnover and biogas generation. In this study, we isolated and characterized a novel anaerobic bacterium, strain meth-B3ᵀ, from a laboratory-scale methanogenic bioreactor fed with maize-based biomass. Cells were Gram-stain-negative, non-spore-forming, motile rods with optimal growth at 35 °C, pH 7.0 and 0.7% sodium chloride (NaCl). Strain meth-B3ᵀ utilized a broad spectrum of carbohydrates, amino acids and organic acids, including glucose, cellobiose, glycerol, sucrose, maltos

Reconstitution of cyanogenesis in barley (Hordeum vulgare L.) and its implications for resistance against the barley powdery mildew fungus.

Barley (Hordeum vulgare L.) produces a leucine-derived cyanogenic beta-D-glucoside, epiheterodendrin that accumulates specifically in leaf epidermis. Barley leaves are not cyanogenic, i.e. they do not possess the ability to release hydrogen cyanide, because they lack a cyanide releasing beta-D-glucosidase. Cyanogenesis was reconstituted in barley leaf epidermal cells through single cell expression of a cDNA encoding dhurrinase-2, a cyanogenic beta-D-glucosidase from sorghum. This resulted in a 35-60% reduction in colonization rate by an obligate parasite Blumeria graminis f. sp. hordei, the ca

Koserella trabulsii, a new genus and species of Enterobacteriaceae formerly known as Enteric Group 45.

The name Koserella trabulsii is proposed for a group of Enterobacteriaceae formerly called Enteric Group 45. This group consists of 12 strains that were originally identified as atypical Hafnia alvei. K. trabulsii strains were negative for indole production, Voges-Proskauer, H2S production, urea hydrolysis, phenylalanine deaminase, and acid production from glycerol, lactose, sucrose, and D-sorbitol; they were positive for methyl red, citrate (Simmons), lysine and ornithine decarboxylases, arginine dihydrolase (negative in 1 to 2 days and positive in 3 to 7 days), and acid production from cello

Efficient high-temperature simultaneous saccharification and fermentation of lignocellulosic substrates enabled by engineering thermotolerance in an enzyme-secreting industrial Saccharomyces cerevisiae strain.

The high cost of commercial enzyme cocktails remains a major barrier for lignocellulosic (second-generation) bioethanol production. Simultaneous saccharification and fermentation (SSF) at elevated temperatures using enzyme-secreting yeast can reduce enzyme demand, but is constrained by the limited thermotolerance of industrial strains. In this study, thermotolerant isolates of an inhibitor-tolerant, xylose-utilizing, enzyme-secreting industrial Saccharomyces cerevisiae strain were generated using whole-genome transformation (WGT). Screening in mixed-sugar fermentations at 41 °C identified seve

Using a cellulose-complementary oligosaccharide as a tool to probe exposed cellulosic surfaces in cotton fibres and growing plant cell walls.

Cellulosic microfibrils in plant cell walls are largely ensheathed and probably tethered by hydrogen-bonded hemicelluloses. Ensheathing may vary developmentally as hemicelluloses are peeled to enable cell expansion. We characterised a simple method to quantify ensheathed versus naked cellulosic surfaces based on the ability to adsorb a radiolabelled 'cellulose-complementary oligosaccharide', [3H]cellopentaitol. Filter-paper (cellulose) adsorbed 40% and >80% of aqueous 5 nM [3H]cellopentaitol within ∼1 and ∼20 h respectively. When [3H]cellopentaitol was rapidly dried onto filter-paper, ∼50% of

Characterization of sugar receptor expression by neoglycoproteins in oral and oropharyngeal squamous cell carcinomas.

Recognition of the carbohydrate part of cellular glycoconjugates by sugar receptors like lectins may contribute to biosignaling and interactions between normal and transformed cells. Such recognitions may be essential for establishing phenotypic characteristics in neoplastic cells, including metastasis-associated properties. To evaluate various glycoconjugates in tumor diagnosis and clinical therapy, a panel of 18 biotinylated neoglycoproteins was prepared. This included conjugates of a histochemically inert carrier protein and crucial sugar moieties such as D-glucuronic acid, alpha- and beta-

Multiomics profiling of transcriptional and metabolic responses of Bacillus siamensis LSZ23 to corn straw.

Agricultural straw is an abundant yet underutilised lignocellulosic resource whose microbial valorisation requires cellulolytic strains that sustain enzyme production under substrate-imposed stress. Although Bacillus siamensis is valued for plant growth promotion and biocontrol, its lignocellulose-degrading capacity and regulation remain largely unexplored. B. siamensis LSZ23, isolated from humic soil in Inner Mongolia, China, produced a balanced cellulolytic system (CMCase 0.321, β-glucosidase 0.275, avicelase 0.240 and FPase 0.209 U/mL in 24 h LB culture) and reduced sterilised corn straw dr

Ultrasound-Assisted Ternary Deep Eutectic Solvent Extraction of Total Flavonoids from Artemisia argyi: GA-ANN Optimization, Greenness Assessment, and In Vitro Bioactivity Evaluation.

Total flavonoids (TF) from Artemisia argyi, a traditional edible-medicinal Asteraceae herb, were extracted via a food-grade ternary deep eutectic solvent (TDES, Glycerol/Levulinic Acid/Xylitol = 1:1:1 molar ratio, 30% w/w water) under ultrasound assistance. A hybrid response surface methodology-genetic algorithm-artificial neural network (RSM-GA-ANN) model optimized parameters to deliver a maximum TF yield of 107.7 mg/g-1.5-1.8-fold higher than conventional hydroalcoholic extraction. Greenness was quantified by AGREEprep (score = 0.70) and MoGAPI (score = 80/100), confirming the method's susta

Genomic diversity of Erwinia carotovora subsp. carotovora and its correlation with virulence.

We used genetic and biochemical methods to examine the genomic diversity of the enterobacterial plant pathogen Erwinia carotovora subsp. carotovora. The results obtained with each method showed that E. carotovora subsp. carotovora strains isolated from one ecological niche, potato plants, are surprisingly diverse compared to related pathogens. A comparison of 23 partial mdh sequences revealed a maximum pairwise difference of 10.49% and an average pairwise difference of 2.13%, values which are much greater than the maximum variation (1.81%) and average variation (0.75%) previously reported for

Antibiotic activity of an isocyanide metabolite of Trichoderma hamatum against rumen bacteria.

A metabolite of Trichoderma hamatum, 3-(3-isocyanocyclopent-2-enylidene)propionic acid, was tested for its effects on growth of and carbohydrate metabolism in 11 strains of functionally important rumen bacteria. To standardize the biological activity of this unstable metabolite, a rapid, aerobic disc diffusion assay was developed using Escherichia coli ATCC 11775. In an anaerobic broth dilution assay using a medium lacking rumen fluid and containing a soluble carbohydrate, the minimum inhibitory concentration of the metabolite which completely inhibited growth of the rumen bacteria for 18 h at

Exploiting Differential Signal Filtering (DSF) and Image Structure Filtering (ISF) Methods for Untargeted Mass Spectrometry Imaging of Bacterial Metabolites.

Matrix-assisted laser desorption/ionization (MALDI) mass spectrometry imaging (MSI) is a label-free technique, producing images where pixels contain mass spectra. The technique allows the visualization of the spatial distribution of (bio)molecules from metabolites to proteins, on surfaces such as tissues sections or bacteria culture media. One particularly exciting example of MALDI-MSI use rests on its potential to localize ionized compounds produced during microbial interactions and chemical communication, offering a molecular snapshot of metabolomes at a given time. Virtual Screening of Cathelicidin-Derived Anticancer Peptides and Validation of Their Production in the Probiotic Limosilactobacillus fermentum KUB-D18 Using Genome-Scale Metabolic Modeling and Experimental Approaches.

The development of anticancer peptides (ACPs) has emerged as a promising strategy in targeted cancer therapy due to their high specificity and therapeutic potential. Cathelicidin-derived antimicrobial peptides represent a particularly attractive class of ACPs, yet systematic evaluation of their anticancer activity remains limited. In this study, we conducted virtual screening of eight cathelicidin-derived peptides (AL-38, LL-37, RK-31, KS-30, KR-20, FK-16, FK-13, and KR-12) to assess their potential against colon cancer. Among these, LL-37 and FK-16 were identified as the most promising candid
