Virus-induced decrease of insulin receptors in cultured human cells.

Viral infections may produce abnormalities in carbohydrate metabolism in normal subjects and profound changes in glucose homeostasis in insulin-dependent diabetics. Using an in vitro radio-receptor assay with 125I-labeled insulin and human-amnion (WISH) cells, the effect of viral infections on insulin receptors was examined. Both herpes simplex virus and vesicular stomatitis virus produced a 50% decrease in insulin binding. There was no evidence that this decrease was due to degradation of insulin. On quantitative analysis, this decrease in binding was found to be the result of a decrease in r

Reduction of cholesterol synthesis by methylxanthines in cultured glial cells.

Methylxanthines are shown in this study to decrease cholesterol synthesis in cultured C-6 glial cells. Aminophylline (10(-3) M) produced a rapid decline in cholesterol synthesis so that by 6 hr, synthesis in treated cells was less than 20% of that in untreated cells, and by 24 hr, less than 10%. Aminophylline induced parallel changes in 3-hydroxy-3-methylglutaryl coenzyme A (HMG-CoA) reductase activity, the rate-limiting enzyme in cholesterol synthesis. Theophylline and caffeine produced similar effects. Thus, methyxanthines caused a specific, enzyme-mediated reduction in cholesterol synthesis

Expression of the cystic fibrosis genotype in cultured somatic cells.

Observations on the characteristics of the cell with the cystic fibrosis (CF) genotype in culture are reviewed. Although numerous and diverse abnormalities have been described, none were specific for the CF gene. The relevance of each of these abnormalities to the clinical syndrome known as CF is discussed, emphasizing that the value thus far of such cell culture research has been to learn how the CF gene influences cellular function.

Survival-related DNA repair phenomena in cultured rat-kangaroo cells.

Cultured cells (line PtK-2) from the marsupial mammal rat-kangaroo, or potoroo (Potorous tridactylis), which photoreactivate (PR) both UV-induced dimers and lethality, excise few dimers, and are only slightly sensitized by post-UV exposure to caffeine, were subjected to caffeine and hydroxyurea (HU) treatments during the 30-min PR period. It was found that neither caffeine nor HU inhibited PR of lethality as measured by colony-forming ability. Further, the cells exhibited no photoprotective properties and 3 mM caffeine potentiated the same slight survival decrease in both photoreactivated and

Toxic effects of mitomycin-C on cultured ciliary process cells and trabecular meshwork cells.

Mitomycin-C has recently become an adjunct medication for inhibition of fibroblast proliferation in glaucoma filtering procedures. Prolonged postoperative ocular hypotony has been a frequent complication of trabeculectomy with mitomycin-C. In order to characterize the hypotony mechanism, we compared the toxic effects of mitomycin-C on cultured rabbit ciliary process cells and trabecular meshwork cells. The results indicate that mitomycin-C has a more marked effect on ciliary process cells on 3H-thymidine uptake than on trabecular meshwork cells at concentrations ranging from 10(-1) to 10(-5) m

Immunostimulated glial cells potentiate glucose deprivation-induced death of cultured rat cerebellar granule cells.

The present study investigates whether immunostimulated glial expression of inducible nitric oxide synthase influences the glucose deprivation-induced death of rat cerebellar granule cells (CGC). CGC/glia cocultures were immunostimulated by interferon-gamma (200 U/ml) and lipopolysaccharides (1 microg/ml) and 2 days later were challenged by glucose deprivation. Neurotoxicity was assessed by measuring the release of lactate dehydrogenase. Neither a 2-h glucose deprivation nor a 2-day immunostimulation altered the viability of CGC. A 2-day immunostimulation, however, markedly potentiated the glu

Adverse interactions of plastic nanoparticles with cultured primary microglial cells.

Due to ever-increasing production, plastics have become ubiquitous in the environment, with polystyrene (PS) being one of the most commonly produced types of plastics. Under the influence of mechanical pressure or UV radiation, plastic waste can decompose into smaller micro/nanoparticles (MPs/NPs), posing a threat to biological life, including humans. Recent findings indicate that MPs/NPs can penetrate biological barriers, including the blood-brain barrier (BBB) and affect cells of the central nervous system (CNS), including microglia, immune cells that play a key role in monitoring the neuron

Labeling Strategies for Monitoring Cytoskeleton Dynamics in Cultured Epithelial Cells.

Collective cell migration relies on coordinated cytoskeletal remodeling, yet the impact of live-cell actin filament probes on these dynamics remains poorly characterized. Here, we systematically compared the performance and cellular effects of fluorogenic jasplakinolide-based probes, SiR-actin and SiR-XActin, with the genetically encoded Lifeact reporter in epithelial monolayers. Using an injury-free wound healing assay combined with widefield images, particle image velocimetry, kymographs, and FUCCI-based cell cycle tracking system, we assessed how probe choice, efflux inhibition, and genetic

Ivermectin effects in cultured endothelial cells from porcine corpus luteum.

Ivermectin (IVM), a widely used antiparasitic, raises emerging concerns regarding reproductive toxicity. This study investigated the effects of clinically relevant IVM concentrations (20, 40, 60 ng/mL) on endothelial cells from porcine corpus luteum, a translational model for vascular function. Proliferation and metabolic activity were assessed via BrdU ELISA and ATP bioluminescence assay, VEGF-A secretion by ELISA, redox status through superoxide and nitric oxide, FRAP assay, and SOD activity by colorimetric methods. IVM significantly impairs endothelial homeostasis by inhibiting proliferatio

Clastogenicity of the food constituent higenamine in cultured V79 cells.

Higenamine is a catecholic benzyltetrahydroisoquinoline alkaloid occurring in plant-derived foods and food supplements, the latter potentially resulting in substantially higher exposure levels than conventional foods. Because of its catechol and alkylphenol structures, oxidative conversion to electrophilic quinone or quinone methide intermediates is plausible, raising concern about a potential genotoxic hazard. However, experimental data on the genotoxic potential of higenamine are lacking. The present study, therefore, investigated its potential to induce gene mutations and chromosomal damage

Ferrotoxicity and Its Amelioration by Calcitriol in Cultured Renal Cells.

Globally, acute kidney injury (AKI) is associated with significant mortality and an enormous economic burden. Whereas iron is essential for metabolically active renal cells, it has the potential to cause renal cytotoxicity by promoting Fenton chemistry-based oxidative stress involving lipid peroxidation. In addition, 1,25-dihydroxyvitamin D3 (calcitriol), the active form of vitamin D, is reported to have an antioxidative role. In this study, we intended to demonstrate the impact of vitamin D on iron-mediated oxidant stress and cytotoxicity of Vero cells exposed to iohexol, a low osmolar iodine

Cytotoxicity of Streptococcus agalactiae secretory protein on tilapia cultured cells.

Streptococcus agalactiae secrete virulence factors believed to be able of killing host tissues, especially under elevated water temperature. A direct effect of S. agalactiae secretory products on tilapia cells was tested on the tilapia kidney (TK-1) cell culture. The bacteria were cultured under four different temperature levels: 22, 29, 32 and 37°C; the cell-free portion was processed through SDS-PAGE; and distinct bands were identified by LC-MS/MS. At least, three virulence factors were identified, Bsp, PcsB and CAMP factor, with increasing levels as the cultured temperature rose. Expression

Effect of radiofrequency radiation in cultured mammalian cells: A review.

The use of mobile phone related technologies will continue to increase in the foreseeable future worldwide. This has drawn attention to the probable interaction of radiofrequency electromagnetic radiation with different biological targets. Studies have been conducted on various organisms to evaluate the alleged ill-effect on health. We have therefore attempted to review those work limited to in vitro cultured cells where irradiation conditions were well controlled. Different investigators have studied varied endpoints like DNA damage, cell cycle arrest, reactive oxygen species (ROS) formation,

The octadecaneuropeptide stimulates somatolactin release from cultured goldfish pituitary cells.

The present study aimed to investigate the distribution of the octadecaneuropeptide (ODN) in the goldfish brain and to look for a possible effect of ODN on somatolactin (SL) release from pituitary cells. A discrete population of ODN-immunoreactive neurones was localised in the lateral part of the nucleus lateralis tuberis. These neurones sent projections through the neurohypophyseal tract towards the neurohypophysis, and nerve fibres were seen in the close vicinity of SL-producing cells in the pars intermedia. Incubation of cultured goldfish pituitary cells with graded concentrations of ODN (1

Constitutive secretion of chemokines by cultured human trabecular meshwork cells.

Trabecular meshwork endothelial (TME) cells secrete a number of factors, such as enzymes and cytokines, which modulate the functions of the cells and the extracellular matrix of the conventional aqueous outflow pathway. TME cells usually secrete these factors in response to stimuli such as mechanical stretching, laser irradiation and pro-inflammatory cytokines. Here, we report that cultured human TME cells isolated from two non-glaucomatous individuals secrete significant quantities of the chemotactic cytokines IL8, CXCL6 and MCP1 in the absence of any stimulation. The secretion of these chemo
