Management of cosmetic embarrassment caused by Malassezia spp. with fruticose lichen Cladia using phylogenetic approach.

During anti-Malassezia screening of plants by CLSI broth microdilution method, Cladia aggregata (Swartz) Nyl. (family Cladoniaceae), a fruticose lichen from Sikkim (northeast Himalayan region), has been found effective at minimum inhibitory concentrations (mg/mL) of 2.72, 0.63, and 1.28 against yeast-like fungi namely, M. furfur, M. globosa and M. sympodialis, respectively. These test pathogens are responsible for pityriasis versicolor (PV) and seborrheic dermatitis (SD) in humans. We tried to establish the reason for variable MICs against various Malassezia spp. using bioinformatical tools, t

Discovery, synthesis, and acaricidal activity evaluation of deep learning-assisted diphenyloxazoline thiosulfonate derivatives.

BACKGROUND: Phytophagous mites are widespread agricultural pests. Under persistent selection pressure from commercial acaricides, Tetranychus cinnabarinus has developed resistance to multiple acaricidal agents, reducing the control efficacy of conventional pesticides. It is therefore imperative to develop novel high-efficiency candidate acaricidal compounds. RESULTS: In this study, the diphenyloxazoline was selected the core scaffold. Lead compound 7a was obtained via deep learning-assisted molecular design. Based on this core scaffold, a series of thiosulfonate derivatives were further desig

A case of black dot ringworm with a review of Japanese cases.

Black dot ringworm (BDR), caused by Trichophyton violaceum var. glabrum (T. glabrum), was observed in a 28-year-old Japanese female who had been treated with prednisolone (22.5 mg/day) for systemic lupus erythematosus. It was successfully treated with oral terbinafine (125 mg/day) for 12 weeks. The causative fungus was identified by molecular analysis as well as morphological and biochemical examination. The chitin synthase 1 (CHS1) gene cleavage pattern of the clinical isolate with restricted enzyme HinfI was identical to that of T. violaceum. We reviewed previous reports of BDR to determine

Whole genome sequencing and annotation of a non-pathogenic Fusarium brachygibbosum TZ1MST22 strain isolated from Medicago sativa in Southern Tunisian oases.

Fusarium sambucinum species complex (FSAMSC) includes not only major mycotoxin-producing pathogens but also non-pathogenic endophytic species that can promote plant growth and stress tolerance. However, the molecular mechanisms underlying these contrasting lifestyles remain poorly understood, underscoring the need for deeper molecular studies. A non-pathogenic endophytic Fusarium strain, TZ1MST22, isolated from Medicago sativa plants in the Tozeur region of Southern Tunisia, was sequenced using the Illumina NovaSeq 6000 sequencing technology. Here, we report a 39.8 Mb draft genome of TZ1MST22,

Dual-Emission FRET-PCR Outperforms SYBR Green and EvaGreen for Accurate Discrimination of Primary Canine Dermatophytes: Microsporum canis, Nannizzia gypsea, and Trichophyton mentagrophytes.

Conventional diagnosis of dermatophytosis relies on fungal culture and microscopic examination, methods that are often time-consuming and lack sensitivity. This study aimed to develop and compare real-time PCR assays for the simultaneous detection and differentiation of three major dermatophytes in dogs: Microsporum canis, Nannizzia gypsea, and Trichophyton mentagrophytes. Three qPCR platforms targeting the chitin synthase 1 (CHS1) gene-SYBR Green, EvaGreen, and dual-emission fluorescence resonance energy transfer (FRET)-were evaluated. The FRET assay demonstrated the highest performance, achi

Proteomic Changes in Paracoccidioides brasiliensis Induced by Fludioxonil.

Paracoccidioides brasiliensis is a pathogenic fungus capable of thermodimorphic transition, causing systemic mycoses in humans. Fludioxonil, a phenylpyrrole fungicide, inhibits this transition and affects genes related to fungal cell wall composition. This study explored the proteomic response of P. brasiliensis yeast cells exposed to fludioxonil, identifying over 4100 proteins, approximately 50% of the total proteome, through DIA (data-independent acquisition) mass spectrometry. Treatment led to significant proteomic changes, with 28 proteins upregulated and 26 downregulated. Upregulated prot

CDP-DAG synthases regulate plant growth and broad-spectrum disease resistance.

Phosphatidic acid (PA) functions as a cell membrane component and signaling molecule in plants. PA metabolism has multiple routes, in one of which PA is converted into cytidine diphosphate diacylglycerol (CDP-DAG) by CDP-DAG synthases (CDSs). CDS genes are highly conserved in plants. Here, we found that knock-down of the CDS gene enhanced the resistance of Arabidopsis thaliana to multiple pathogens, with a growth penalty. When Arabidopsis leaves were treated with chitin or flg22, reactive oxygen species (ROS) production in cds mutants was significantly higher than that in the wild-type (WT). S

Molecular detection and species identification of dermatophytes by SYBR-Green real-time PCR in-house methodology using hair samples obtained from dogs and cats.

The classical dermatophytes diagnosis is based on mycological culture and microscopy observation both human and animal hair, skin, and nail samples. The aim of this work was to develop the new in-house real-time PCR with pan-dematophyte reaction for detection and identification of the main dermatophytes directly from hair samples, providing a simple and rapid diagnosis of dermatophytosis in dogs and cats. An in-house SYBR-Green real-time PCR was designed and used for detecting a DNA fragment encoding chitin synthase 1 (CHS1). A total of 287 samples were processed by culture, microscopic examin

Roles for Stress Response and Cell Wall Biosynthesis Pathways in Caspofungin Tolerance in Cryptococcus neoformans.

Limited antifungal diversity and availability are growing problems for the treatment of fungal infections in the face of increasing drug resistance. The echinocandins, one of the newest classes of antifungal drugs, inhibit production of a crucial cell wall component. However, these compounds do not effectively inhibit the growth of the opportunistic fungal pathogen Cryptococcus neoformans, despite potent inhibition of the target enzyme in vitro Therefore, we performed a forward genetic screen to identify cellular processes that mediate the relative tolerance of this organism to the echinocandi

Genomic and functional analyses unveil the response to hyphal wall stress in Candida albicans cells lacking β(1,3)-glucan remodeling.

BACKGROUND: The cell wall is essential for the yeast to hypha (Y-H) transition that enables Candida albicans to invade human tissues and evade the immune system. The main constituent, β(1,3)-glucan, is remodeled by glucanosyltransferases of the GH72 family. Phr1p is responsible of glucan remodeling at neutral-alkaline pH and is essential for morphogenesis and virulence. Due to the pH-regulated expression of PHR1, the phr1Δ phenotype is manifested at pH > 6 and its severity increases with the rise in pH. We exploited the pH-conditional nature of a PHR1 null mutant to analyze the impact of gluca

Insect chitosan derived from Hermetia illucens larvae suppresses adipogenic signaling and promotes the restoration of gut microbiome balance.

Chitosan, the deacetylated form of chitin, is considered a valuable source of compounds in the feed and food industries. However, the impact of Hermetia illucens larvae chitosan (HCS) with specific physicochemical characteristics on obesity mediated by lipid accumulation and microbiome dysbiosis has not been fully elucidated. We purified HCS with a low molecular weight (84 kDa), low crystallinity, and a high deacetylation rate, characterizing it through several analytical techniques, including gel permeation chromatography, FT-IR, 1H NMR, FE-SEM, and XRD analysis. HCS effectively inhibited the

Natural products targeting the synthesis of β(1,3)-D-glucan and chitin of the fungal cell wall. Existing drugs and recent findings.

BACKGROUND: During the last three decades systemic fungal infections associated to immunosuppressive therapies have become a serious healthcare problem. Clinical development of new antifungals is an urgent requirement. Since fungal but not mammalian cells are encased in a carbohydrate-containing cell wall, which is required for the growth and viability of fungi, the inhibition of cell wall synthesizing machinery, such as β(1,3)-D-glucan synthases (GS) and chitin synthases (CS) that catalyze the synthesis of β(1-3)-D-glucan and chitin, respectively, represent an ideal mode of action of antifung

Functional metatranscriptomics of the rhizosphere: towards an understanding of metabolic processes of the microbial carbon pump.

The soil microbiome drives soil organic carbon (SOC) transformation, shaped and impacted by plant growth stage and soil management such as tillage. The soil microbial carbon pump conceptually links the degradation of plant- and microbe-derived compounds and the neosynthesis of microbial biomass, as a driver of SOC buildup. Using metatranscriptomic sequencing, we characterized actively expressed microbial functions associated with the microbial carbon pump in the Brassica napus rhizosphere across growth stages and under simulated erosion, using a tailored KEGG Orthology (KO) approach complement

Effects of 5'-3' Exonuclease Xrn1 on Cell Size, Proliferation and Division, and mRNA Levels of Periodic Genes in Cryptococcus neoformans.

Cell size affects almost all biosynthetic processes by controlling the size of organelles and disrupting the nutrient uptake process. Yeast cells must reach a critical size to be able to enter a new cell cycle stage. Abnormal changes in cell size are often observed under pathological conditions such as cancer disease. Thus, cell size must be strictly controlled during cell cycle progression. Here, we reported that the highly conserved 5'-3' exonuclease Xrn1 could regulate the gene expression involved in the cell cycle pathway of Cryptococcus neoformans. Chromosomal deletion of XRN1 caused an i

Multiple-strain Trichophyton mentagrophytes infection in a silver fox (Vulpes vulpes) from a breeding farm.

Dermatophyte infections are extremely frequent worldwide, and their epidemiological features and distribution make them one of the most frequent infections all over the world. We identified and analysed multiform T. mentagrophytes strains isolated from a silver fox (Vulpes vulpes) kept on a breeding farm. Identification of dermatophyte strains was carried out traditionally by correlating both the clinical manifestations of the infection with a micro- and macroscopic examination. To confirm the species affiliation fully, molecular differentiation methods were used. DNA was isolated from the der

Vulnerability to low salinity but compensatory responses to low pH in an intertidal limpet.

Rising anthropogenic carbon dioxide emissions have driven ongoing ocean warming and associated climate changes. In the Yellow Sea, this warming is associated with enhanced monsoonal rainfall, which increases freshwater inputs and lowers coastal salinity. Increased freshwater input can weaken seawater buffering capacity, thereby leading to lower pH conditions in coastal environments. Here, we examined the effects of low pH and low salinity on the intertidal limpet Patelloida pygmaea. Adult limpets were exposed for 31 days to four experimental artificial seawater conditions combining two pH leve

Enhanced eugenol biosynthesis through GmEGS overexpression confers broad-spectrum pathogen protection in soybean.

Soybean is a major source of plant protein and oil, yet its production is severely constrained by diverse pathogen infections. Here, we identify eugenol as a pathogen-induced defense metabolite in soybean with broad-spectrum antimicrobial activity against fungal, oomycete, and bacterial pathogens. Pathogen-associated molecular patterns (PAMPs), including Flg22, Chitin, and Elicitin, strongly induced soybean eugenol synthase (GmEGS) genes and promoted eugenol accumulation. Phylogenetic and expression analyses identified five GmEGS genes, among which GmEGS1a, GmEGS1c, and GmEGS2a were strongly i
